First detection of voltage-gated sodium channel mutations in Phlebotomus argentipes collected from Bangladesh.

BACKGROUND & OBJECTIVES: Phlebotomus argentipes is the main vector of visceral leishmaniasis in Bangladesh and is controlled using deltamethrin, a synthetic pyrethroid, through indoor residual spraying (IRS). A mutation at L1014 (leucine at codon 1014) of the voltage-gated sodium channel (VGSC), known as a knockdown resistance (kdr) gene, is thought to be an important pyrethroid resistance mechanism. This study detected mutations at codon 1014, and at codons 1011, 1016, and 1020, which are kdr sites in other insects. The kdr relationship with deltamethrin resistance in P. argentipes from an IR

Involvement of small heat shock proteins (sHsps) in developmental stages of fall armyworm, Spodoptera frugiperda and its expression pattern under abiotic stress condition.

Fall armyworm (FAW), Spodoptera frugiperda a recent invasive pest in India is reported to cause significant damage by feeding voraciously on maize and other economic crops from tropical to temperate provinces. It is becoming an arduous challenge to control the pest as it can survive in a wide range of temperature conditions and is already said to develop resistance towards certain insecticides. The small Heat shock proteins (hereafter, sHsps) are known to play an important role in adaptation of insects under such stress conditions. Our present study involved characterization of the three sHsps

The deleterious toxic effects of bifenthrin on male fertility.

Bifenthrin (BF), a broad-spectrum synthetic pyrethroid insecticide, has been generally used to eradicate harmful insects. However, according to the U.S. EPA, BF has been classified as a "Class C" carcinogenic ingredient. Furthermore, a previous study reported that BF was considered as endocrine-disrupting chemicals and causes reproductive toxicity in mammals. Despite the various effects of BF, there is a scarcity of studies about its adverse effects on male fertility. Therefore, this study was conducted to determine the effects of BF on sperm functions at various concentrations (0.1, 1, 10, an

A first, naturally occurring substitution at the second pyrethroid receptor of voltage-gated sodium channel of Aedes aegypti.

BACKGROUND: Aedes aegypti is a remarkably effective mosquito vector of epidemiologically important arboviral diseases including dengue fever, yellow fever and Zika. The present spread of resistance against pyrethroids, the primary insecticides used for mosquito control, in global populations of this species is of great concern. The voltage-gated sodium channel (VGSC) in the nervous system is the known target site of pyrethroids in insects. Past studies have revealed several amino-acid substitutions in this channel that confer pyrethroid resistance, which are known as knockdown resistance (kdr)

Delayed mortality effects of cold fronts during the winter season on Aedes aegypti in a temperate region.

The expansion of the invasive mosquito Aedes aegypti L. (Diptera: Culicidae) towards temperate regions in the Americas is causing concern because of its public health implications. As for other insects, the distribution limits of Ae. aegypti have been suggested to be related to minimum temperatures and to be controlled mainly by cold tolerance. The aim of this study was to assess the daily mortality of immature stages of Ae. aegypti under natural winter conditions in Buenos Aires, Argentina, in relation to preceding thermal conditions. The experiment was performed outdoors, and one cohort of l

Proteins, Transcripts, and Genetic Architecture of Seminal Fluid and Sperm in the Mosquito Aedes aegypti.

The yellow fever mosquito, Aedes aegypti,, transmits several viruses causative of serious diseases, including dengue, Zika, and chikungunya. Some proposed efforts to control this vector involve manipulating reproduction to suppress wild populations or to replace them with disease-resistant mosquitoes. The design of such strategies requires an intimate knowledge of reproductive processes, yet our basic understanding of reproductive genetics in this vector remains largely incomplete. To accelerate future investigations, we have comprehensively catalogued sperm and seminal fluid proteins (SFPs) t

De novo characterization of microRNAs in oriental fruit moth Grapholita molesta and selection of reference genes for normalization of microRNA expression.

MicroRNAs (miRNAs) are a group of endogenous non-coding small RNAs that have critical regulatory functions in almost all known biological processes at the post-transcriptional level in a variety of organisms. The oriental fruit moth Grapholita molesta is one of the most serious pests in orchards worldwide and threatens the production of Rosacea fruits. In this study, a de novo small RNA library constructed from mixed stages of G. molesta was sequenced through Illumina sequencing platform and a total of 536 mature miRNAs consisting of 291 conserved and 245 novel miRNAs were identified. Most of

MosqTent: An individual portable protective double-chamber mosquito trap for anthropophilic mosquitoes.

Here, we describe the development of the MosqTent, an innovative double-chamber mosquito trap in which a human being attracts mosquitoes while is protected from being bitten within the inner chamber of the trap, while mosquitoes are lured to enter an outer chamber where they are trapped. The MosqTent previously collected an average of 3,000 anophelines/man-hour compared to 240 anophelines/man-hour for the human landing catch (HLC), thereby providing high numbers of human host-seeking mosquitoes while protecting the collector from mosquito bites. The MosqTent performed well by collecting a high

Do tsetse flies only feed on blood?

Tsetse flies (Diptera: Glossinidae) are the vectors of trypanosomes causing sleeping sickness in humans, and nagana (animal trypanosomosis) in domestic animals, in Subsaharan Africa. They have been described as being strictly hematophagous, and transmission of trypanosomes occurs when they feed on a human or an animal. There have been indications however in old papers that tsetse may have the ability to digest sugar. Here we show that hungry tsetse (Glossina palpalis gambiensis) in the lab do feed on water and on water with sugar when no blood is available, and we also show that wild tsetse ha

Transcriptome profile analysis of cell proliferation molecular processes during multicellular trichome formation induced by tomato Wov gene in tobacco.

BACKGROUND: Trichomes, developing from the epidermis of nearly all terrestrial plants, provide good structural resistance against insect herbivores and an excellent model for studying the molecular mechanisms underlying cell fate determination. Regulation of trichomes in Rosids has been well characterized. However, little is known about the cell proliferation molecular processes during multicellular trichome formation in Asterids. RESULTS: In this study, we identified two point mutations in a novel allele (Wov) at Wo locus. Ectopic expression of Wov in tobacco and potato induces much more tri

A synthesis of the effects of pesticides on microbial persistence in aquatic ecosystems.

Pesticides have a pervasive presence in aquatic ecosystems throughout the world. While pesticides are intended to control fungi, insects, and other pests, their mechanisms of action are often not specific enough to prevent unintended effects, such as on non-target microbial populations. Microorganisms, including algae and cyanobacteria, protozoa, aquatic fungi, and bacteria, form the basis of many food webs and are responsible for crucial aspects of biogeochemical cycling; therefore, the potential for pesticides to alter microbial community structures must be understood to preserve ecosystem s

Physiology of invertebrate oxytocin and vasopressin neuropeptides.

Neuropeptides and regulatory peptide hormones control many developmental, physiological and behavioural processes in animals, including humans. The nonapeptides oxytocin and arginine vasopressin are produced and released by the pituitary gland and have actions on many organs and tissues. Receptive cells possess particular receptors to which the peptides bind as ligands, leading to activation of G-protein-coupled receptors, hence cellular responses. In humans and other mammalian species, oxytocin and vasopressin mediate a range of peripheral and central physiological functions that are importan

Effect of starvation on lipophorin density in fifth instar larval Manduca sexta.

Lipophorin (Lp) is a major insect lipoprotein and is responsible for lipid transport between organs. In this study, the effect of starvation on Lp properties was analyzed in larval Manduca sexta during the fifth instar. Lp hemolymph concentrations in larvae at days 1 and 2 were around 2-3 mg/ml and at day 3 it increased to 8 mg/ml. When larvae were starved for 24 h, they did not grow, but their body mass and hemolymph volume did not decrease significantly. Differences in Lp densities were observed. In fed larvae, from days 1 to 4, two major Lp populations were found with densities of 1.124 ± 0

Olfactory cues from plants infected by powdery mildew guide foraging by a mycophagous ladybird beetle.

Powdery mildews (Erysiphales) are economically important plant pathogens that attack many agricultural crops. Conventional management strategies involving fungicide application face challenges, including the evolution of resistance and concerns over impacts on non-target organisms, that call for investigation of more sustainable alternatives. Mycophagous ladybird beetles (Coleoptera: Coccinellidae) feed on powdery mildew and have considerable potential as biological control agents; however, the foraging ecology and behavior of these beetles is not well understood. Here we document the olfactor

A genetic basis for the manipulation of sink-source relationships by the galling aphid Pemphigus batae.

We examined how the galling aphid Pemphigus batae manipulates resource translocation patterns of resistant and susceptible narrowleaf cottonwood Populus angustifolia. Using carbon-14 ((14)C)-labeling experiments in common garden trials, five patterns emerged. First, although aphid galls on resistant and susceptible genotypes did not differ in their capacity to intercept assimilates exported from the leaf they occupied, aphids sequestered 5.8-fold more assimilates from surrounding leaves on susceptible tree genotypes compared to resistant genotypes. Second, gall sinks on the same side of a shoo

Characterization of olfactory genes in the antennae of the Southern house mosquito, Culex quinquefasciatus.

Odorant reception in insects is mediated by different families of olfactory proteins. Here we focus on the characterization of odorant-binding proteins (OBPs), "plus-C" odorant-binding proteins ("plus-C" OBPs), chemosensory proteins (CSPs) and sensory neuron membrane proteins (SNMPs) families from the Southern house mosquito, Culex quinquefasciatus, a vector of pathogens implicated in multiple human diseases. Using bioinformatics and molecular approaches, we have identified a diversity of genes in the genome of Culex quinquefasciatus and examined their expression profiles by RT-PCR and real-ti

An analysis of plant-aphid interactions by different microarray hybridization strategies.

Aphids have long been considered 'stealthy' herbivores that subvert a plant's induced defenses and manipulate its source-sink signaling, but these hypotheses are largely untested at a transcriptional level. We analysed gene expression in native tobacco plants (Nicotiana attenuata) infested with Myzus nicotianae aphids, without resorting to the use of clip-cages, with a cDNA microarray containing 240 defense-related N. attenuata genes. Using a hybridization scheme ('ratio analysis' and 'state analysis') broadly applicable in two-factor analyses, we examined how the aphids influenced source--sin

Isolation and expression of a Pax-6 gene in the regenerating and intact Planarian Dugesia(G)tigrina.

The Pax-6 gene encodes a transcription factor containing both a paired and a homeodomain and is highly conserved among Metazoa. In both vertebrates and invertebrates, Pax-6 is required for eye morphogenesis, development of parts of the central nervous system, and, in some phyla, for the development of olfactory sense organs. Ectopic expression of Pax-6 from insects, mammals, cephalopods, and ascidians induces ectopic eyes in Drosophila, suggesting that Pax-6 may be a universal master control gene for eye morphogenesis. Platyhelminthes are an ancient phylum, originating from the base of spirali

Experimental infection of ectoparasitic arthropods with Rickettsia prowazekii (GvF-16 strain) and transmission to flying squirrels.

Epizootiologic studies conducted during the past few years showed the existence of widespread natural infection of the southern flying squirrel, Glaucomys volans, with epidemic typhus rickettsiae, Rickettsia prowazekii. The ecological findings strongly implicated transmission of the etiologic agent by an arthropod vector. Studies were conducted under controlled laboratory conditions to determine whether ectoparasites naturally associated with flying squirrels (squirrel fleas, lice, mites and ticks) were capable of acquiring, maintaining and transmitting the infection. Also studied were the cat
