Protein therapy using heme-oxygenase-1 fused to a polyarginine transduction domain attenuates cerebral vasospasm after experimental subarachnoid hemorrhage.

A sequence of 11 consecutive arginine residues (11R) is one of the best protein transduction domains for introducing proteins into cell membranes. Heme-oxygenase-1 (HO-1) is involved in heme catabolism and reduces the contractile effect of hemoglobin after subarachnoid hemorrhage (SAH). Therefore, we constructed 11R-fused HO-1 protein to achieve successful transduction of the protein into the cerebral arteries and examined the therapeutic effect of the 11R-HO-1 protein for cerebral vasospasm (CV) after SAH. We injected the 11R-HO-1 protein into the cisterna magna of male rats and, several hour

Epidermolysis bullosa: a genetic disease of altered cell adhesion and wound healing, and the possible clinical utility of topically applied thymosin beta4.

Inherited epidermolysis bullosa (EB), having an overall incidence of only about 19 in every 1 million live births, encompasses many phenotypically and genotypically distinct diseases characterized by the presence of recurrent blisters, mechanical fragility of the skin and other epithelial structures (most notably the cornea and gastrointestinal tract), and scar formation. Each disease is the result of mutations within any of 10 specific structural proteins (keratins, laminins, collagens, integrins) within the basilar keratinocyte or the skin basement membrane zone. Two of the more severe subty

Interactive role of protein kinase C-delta with rho-kinase in the development of cerebral vasospasm in a canine two-hemorrhage model.

BACKGROUND: We previously reported that protein kinase C (PKC)-delta was initially translocated from the cytosol to the membrane fraction (on day 4), followed by PKC-alpha, with the progression of cerebral vasospasm after subarachnoid hemorrhage (SAH) on day 7. Rho/Rho-kinase pathways have also been proposed to be involved in the vasospasm. Thus we investigated the interactive role of Rho-kinase and PKC in the development of cerebral vasospasm after SAH. METHODS: The cerebral vasospasm was produced using a 'two-hemorrhage' canine model. The animals were treated with Y-27632, a Rho-kinase inhi

Effects of masker frequency and duration in forward masking: further evidence for the influence of peripheral nonlinearity.

Forward masking has often been thought of in terms of neural adaptation, with nonlinearities in the growth and decay of forward masking being accounted for by the nonlinearities inherent in adaptation. In contrast, this study presents further evidence for the hypothesis that forward masking can be described as a linear process, once peripheral, mechanical nonlinearities are taken into account. The first experiment compares the growth of masking for on- and off-frequency maskers. Signal thresholds were measured as a function of masker level for three masker-signal intervals of 0, 10, and 30 ms.

Additivity of masking in normally hearing and hearing-impaired subjects.

The effects of combining two equally effective maskers were studied in normally hearing and elderly hearing-impaired subjects. The additivity of nonsimultaneous masking was investigated by measuring thresholds for a brief 4-kHz signal in the presence of a broadband-noise forward masker, a backward masker, and a combination of both. For the normally hearing subjects, combining two equally effective nonsimultaneous maskers resulted in up to a 15-dB greater increase in threshold than the 3 dB predicted by an energy-summation model ("excess masking"). However, the hearing-impaired subjects showed

[Sequential changes of vascular intimal ultrastructure in experimental cerebral vasospasm induced by oxyhemoglobin. Scanning electron microscopic study].

Organic changes of vessel wall may play an important role in the pathophysiology of vasospasm after subarachnoid hemorrhage. Sequential changes of intimal ultrastructure in experimental vasospasm induced oxyhemoglobin were investigated by scanning electron microscope. Thirty-four cats were used for this study. The basilar artery was exposed by transcervico-transclival approach. Oxyhemoglobin solution was applied under the arachnoid membrane around the artery in 16 cats at hourly intervals. Of 16 cats, 3 were sacrificed at 1 hour after the first application of the solution, 3 at 3 hours, 3 at 6

Comparing and Optimizing Tissue Clearing Protocols for Cochlear Immunostaining and 3D Reconstruction by Light-Sheet Microscopy.

PURPOSE: Current standard methods for immunohistochemical analysis of the inner ear rely on cryosectioning of the intact cochlea or dissection of the organ of Corti. Cryosections provide only limited structural information and do not allow quantification of hair cell numbers per basilar membrane length, whereas dissections require substantial technical expertise and often result in damage to the basal cochlear turns. Tissue clearing has largely overcome these limitations and has been widely applied to organs such as the brain, heart, liver, and kidney. However, despite several reports describi

Sex-specific differences in basilar artery vasospasm after subarachnoid hemorrhage: evidence from a rabbit model.

BACKGROUND: Subarachnoid hemorrhage (SAH) remains a devastating cerebrovascular disorder in which cerebral vasospasm is a key determinant of secondary injury and outcome. Sex differences are well-documented clinically, women exhibit a higher incidence of aneurysmal SAH and often worse outcomes, but whether biological sex influences vasospasm severity in controlled preclinical settings remains unclear. METHODS: In a randomized, controlled rabbit model, we investigated sex-specific differences in basilar artery (BA) vasospasm after SAH. Twenty-four New Zealand White rabbits (12 male, 12 female)

Vestibular sensory functional status of cochlear implanted ears versus non-implanted ears in bilateral profound deaf adults.

Patients with hearing loss who underwent cochlear implantation can present symptomatic or asymptomatic vestibular damages earlier or later after the surgery. The vestibular permanent lesions could be acute, produced by surgical trauma or could be progressive due to local morphological changes made by the presence of the portelectrode in the inner ear (fibrosis related, ossification, basilar membrane distortion, endolymphatic hydrops). Besides histopathological findings in inner ear of cochlear implanted patients, the vestibular permanent damages could be found by assessment of clinical vestibu

High frequency transient-evoked otoacoustic emission measurements using chirp and click stimuli.

Transient-evoked otoacoustic emissions (TEOAEs) at high frequencies are a non-invasive physiological test of basilar membrane mechanics at the basal end, and have clinical potential to detect risk of hearing loss related to outer-hair-cell dysfunction. Using stimuli with constant incident pressure across frequency, TEOAEs were measured in experiment 1 at low frequencies (0.7-8 kHz) and high frequencies (7.1-14.7 kHz) in adults with normal hearing up to 8 kHz and varying hearing levels from 9 to 16 kHz. In combination with click stimuli, chirp stimuli were used with slow, medium and fast sweep

Dose-response hearing loss for white noise in the Sprague-Dawley rat.

The effect of noise exposure on the inner ear is well documented. However, the sensitivity to noise-induced damage varies with animal species. The purpose of this investigation was to generate a hearing loss dose-response curve for a 20-day white noise exposure in the Sprague-Dawley rat. Eight male rats were exposed to 110, 100, 95 or 85 dB sound pressure level (SPL) for 6 hr/day, 5 days/week for 4 weeks in a sound-attenuated chamber fitted with a bank of overhead speakers. Controls were placed in an identical chamber without speakers. Four weeks after the exposure period ended, brainstem audi

Atraumatic Scala Tympani Cochleostomy; Resolution of the Dilemma.

OBJECTIVES: While an accurate placement in cochleostomy is critical to ensure appropriate insertion of the cochlear implant (CI) electrode into the scala tympani (ST), the choice of preferred cochleostomy sites widely varied among experienced surgeons. We present a novel technique for precise yet readily applicable localization of the optimum site for performing ST cochleostomy. MATERIAL AND METHODS: Twenty fresh frozen temporal bones were dissected using the mastoidectomy-posterior tympanotomy approach. Based on the facial nerve and the margins of the round window membrane (RWM), the cochleo

The effects of aging on otoacoustic emissions.

Otoacoustic emissions were measured in 42 normal hearing subjects ranging from 20 to 80 years old. For each subject spontaneous, click-evoked, tone-burst-evoked, stimulus frequency and distortion product emissions were measured across a wide intensity range for frequencies between 1 and 3 kHz. Although there are significant differences between age groups, the results indicate no age effect independent of hearing sensitivity on any type or parameter of otoacoustic emissions (OAE). The effect of increasing age is confounded with the effect of decreasing sensitivity such that post hoc analyses ar

Sentinels or saboteurs, the Janus face of cochlear-resident macrophages in hearing loss: spatiotemporal distribution, pathophysiological functions, and translational potential.

BACKGROUND: Neuroinflammation is a critical mechanism underlying various forms of sensorineural hearing loss (SNHL). Contrary to the conventional view that cochlea is an immune-privileged organ shielded by the blood-labyrinth barrier, it is now evident that it hosts a diverse immune landscape dominated by cochlear-resident macrophages (CoRMs). These CoRMs share a common developmental origin with central nervous system microglia. Despite this phylogenetic homology, CoRMs have evolved distinct phenotypic and functional profiles to adapt to the unique peripheral sensory environment of cochlea. Re

Combinatorial Atoh1, Gfi1, Pou4f3, and Six1 gene transfer induces hair cell regeneration in the flat epithelium of mature guinea pigs.

Flat epithelium (FE) is a condition characterized by the loss of both hair cells (HCs) and supporting cells and the transformation of the organ of Corti into a simple flat or cuboidal epithelium, which can occur after severe cochlear insults. The transcription factors Gfi1, Atoh1, Pou4f3, and Six1 (GAPS) play key roles in HC differentiation and survival in normal ears. Previous work using a single transcription factor, Atoh1, to induce HC regeneration in mature ears in vivo usually produced very few cells and failed to produce HCs in severely damaged organs of Corti, especially those with FE.

Exponential fitting of spread of excitation response measurements in cochlear implants.

BACKGROUND: Hearing performance in cochlear implant (CI) users is variable. An objective measure which can allow a prediction of this performance is desirable. Spread of neural excitation (SoE) curves are an objective measure that can be obtained using the fitting software of cochlear implants and might be able to be used as a predictor. A novel method to interpret SoE curves is presented. NEW METHOD: Spread of excitation measurements for three recording sites were fitted using two exponential functions. An asymmetric width measure was developed, defined as the distance in mm to the point, wh

[Objective frequency-specific measurement of hearing threshold using narrow-band chirp stimuli with level-adaptive simultaneous masking].

BACKGROUND: In the past, various simulation and measurement paradigms have been introduced and evaluated in order to improve frequency-specific measurement of the hearing threshold using early auditory evoked potentials (EAEP). A promising approach for improvement of detection of stimulus response is the usage of frequency-modulated chirp signals, which optimize the temporal synchrony of neuronal responses along a region of the basilar membrane. AIM OF THE STUDY: This study validated the performance of three generated narrow-band chirp stimuli in combination with a level-adaptive simultaneous

A Tanshinone IIA loaded hybrid nanocomposite with enhanced therapeutic effect for otitis media.

Otitis media, commonly known as middle ear inflammation, is among one of the most common maladies and results in significant morbidity such as loss of hearing. In view of the bacteria invasion such as Staphylococcus aureus causes the majority forms of otitis media, drug treatment generally uses antibacterial by topical or systematic approach. However, the effectiveness of antibacterial is diminishing because of the rapid emergence of antibiotic-resistant bacterial strains. Here, we designed and fabricated a silver nanoparticle (AgNPs)-based multicomponent hybrid nanocomposite termed as TSIIA @
