Biological and Habitual Factors That Influence Sleep and Circadian Rhythm: A Narrative Review.

Sleep loss and circadian misalignment can adversely affect health. As such, a comprehensive understanding of what factors influence sleep loss and circadian misalignment benefits medical and scientific progress. While the interaction between light exposure and circadian entrainment and their relationship to circadian alignment is long-established, other factors that influence central or peripheral clocks (bodily oscillators) also contribute to circadian alignment and the potential for sleep loss. In this review, we will highlight the core factors that influence vulnerability or resilience to s

Chronobiological variables predict non-response to serotonin and noradrenaline reuptake inhibitors in fibromyalgia: a cross-sectional study.

Available data shows associations between chronotype, circadian rhythms, sleep quality and fibromyalgia (FM) presentation. However, no studies have explored links between the chronobiological variables and effectiveness of pharmacotherapy. We aimed to assess the chronotypes, circadian rhythms, sleep-wake cycle and sleep quality in FM and their links to treatment response to serotonin and noradrenalin reuptake inhibitors (SNRI). 60 FM patients: 30 responsive to SNRI (FM T[+]), 30 non-responsive to SNRI (FM T[-]) and 30 healthy controls participated. Subjects were assessed by physician and with

Sleep disturbances and female infertility: a systematic review.

BACKGROUND: Sleep disturbances are more prevalent among women with infertility. Current research increasingly highlights the significant relationship between sleep disturbances and female infertility, suggesting that sleep may be a key factor in reproductive health. In this review, we aim to delve into the complex interplay between sleep disturbances and female infertility, as well as to assess the underlying mechanisms involved, and seek to illuminate the causes of sleep-related fertility issues. The understanding of these contents may help clinicians enhance clinical strategies for managing

The Relationship Between Sleep Traits and Tinnitus in UK Biobank: A Population-Based Cohort Study.

OBJECTIVES: Understanding the association between sleep traits and tinnitus could help prevent and provide appropriate interventions against tinnitus. Therefore, this study aimed to assess the relationship between different sleep patterns and tinnitus. DESIGN: A cross-sectional analysis using baseline data (2006-2010, n = 168,064) by logistic regressions was conducted to evaluate the association between sleep traits (including the overall health sleep score and five sleep behaviors) and the occurrence (yes/no), frequency (constant/transient), and severity (upsetting/not upsetting) of tinnitus

Are sleep disturbances causally linked to the presence and severity of psychotic-like, dissociative and hypomanic experiences in non-clinical populations? A systematic review.

The present review aimed to 1) identify what sleep disturbances co-occur alongside psychotic-like, dissociative and hypomanic experiences; 2) assess the strength of potential associations between the severity of sleep disturbances and of the experiences studied; and 3) appraise evidence for a causal link. MedLine and PsycInfo were searched and 44 studies were deemed eligible. Results showed that insomnia was associated with all individual psychotic-like, dissociative and hypomanic experiences reviewed (effect size range: small-to-large). Parasomnias were associated with all psychotic-like expe

Haplotype function score improves biological interpretation and cross-ancestry polygenic prediction of human complex traits.

We propose a new framework for human genetic association studies: at each locus, a deep learning model (in this study, Sei) is used to calculate the functional genomic activity score for two haplotypes per individual. This score, defined as the Haplotype Function Score (HFS), replaces the original genotype in association studies. Applying the HFS framework to 14 complex traits in the UK Biobank, we identified 3619 independent HFS-trait associations with a significance of p < 5 × 10-8. Fine-mapping revealed 2699 causal associations, corresponding to a median increase of 63 causal findings per t

Chrono-modulated drug delivery systems for oncology: Mechanisms, translational evidence, and pharmaceutical innovations.

Conventional chemotherapy is limited by severe systemic toxicity, suboptimal pharmacokinetics, and nonspecific drug distribution. Chrono-modulated drug delivery systems (CDDS) address these challenges by aligning drug release with circadian rhythms, thereby enhancing efficacy and minimizing adverse effects. This strategy capitalizes on temporal fluctuations in drug metabolism, DNA repair mechanisms, cell proliferation, and immune function to refine pharmacokinetic and pharmacodynamic profiles. This review synthesizes recent preclinical and emerging clinical studies exploring chronomodulated ch

Quantifying the contribution of Neanderthal introgression to the heritability of complex traits.

Eurasians have ~2% Neanderthal ancestry, but we lack a comprehensive understanding of the genome-wide influence of Neanderthal introgression on modern human diseases and traits. Here, we quantify the contribution of introgressed alleles to the heritability of more than 400 diverse traits. We show that genomic regions in which detectable Neanderthal ancestry remains are depleted of heritability for all traits considered, except those related to skin and hair. Introgressed variants themselves are also depleted for contributions to the heritability of most traits. However, introgressed variants s

Diurnal Patterns of Accelerometer-Measured Physical Activity and Sleep in Relation to Cancer Risk: Findings From a Nationally Representative Study.

Physical activity (PA) and sleep are modifiable lifestyle factors, yet their circadian patterns in relation to cancer risk remain unclear. This study investigated their independent and joint associations with cancer risk. We analyzed data from 9038 adults aged ≥ 20 years in the 2011-2014 National Health and Nutrition Examination Survey. Circadian patterns of PA and sleep were derived from accelerometer data using k-means clustering. Weighted logistic regression assessed their independent and joint associations with cancer risk. Diurnal patterns were identified as early-morning (n = 2981), midd

The Effect of Night Shift Work on the Gut Microbiome Diversity: The EXPONIT Study.

Night shift work may alter the gut microbiome through mechanisms involving circadian misalignment, sleep disturbance, and changes in dietary behavior. However, existing studies on this topic have been limited in sample size and scope. We analyzed stool samples from 240 participants (mean age 42 years, 80% women), of whom 53% were night shift workers. Gut microbiota composition was assessed using 16S rRNA gene sequencing to derive measures of relative abundance, alpha diversity, and beta diversity. Associations between night shift work and microbial composition and alpha diversity were examined

Metabolic signatures of sleep health and stroke risk: A prospective cohort study with mediation analysis.

OBJECTIVES: Healthy sleep is associated with lower stroke risk, but the underlying metabolic mechanisms remain poorly understood. We investigated whether a nuclear magnetic resonance-derived metabolic signature mediates this association. METHODS: This prospective cohort study included 200,729 UK Biobank participants (mean age 56.48±8.10 years, 53.80% female) free of stroke at baseline. A healthy sleep score (0-6) was derived from 6 behaviors as follows: chronotype, sleep duration, insomnia, snoring, daytime dozing, and ease of getting up. Sleep-related metabolites among 249 plasma nuclear mag

Chronotherapeutic and Circadian Rhythm-Based Strategies in Diabetes Management: A Critical Opinion Based on Evidences.

Diabetes mellitus is one of the world's leading health problems, and standard approaches to therapy do not take into consideration the impact of circadian misalignment on metabolic health. Chronobiological processes are involved in regulating glucose homeostasis, insulin release, hepatic glucose production, lipids, and energy metabolism; hence, disruptions such as those caused by non-rhythmic sleep patterns, shifts at work, irregular meal times, and light exposure could cause insulin resistance and even type 2 diabetes. The current critical review discusses the light-melatonin interaction and

The Potential of Chronotherapy and Nanotherapy-Based Strategies for Glioblastoma Treatment.

Glioblastoma is the most common and aggressive brain tumour in adults, and despite ongoing efforts, effective treatment remains limited. Standard therapies often face challenges because of the tumour's specific biology, its aggressive nature, and the presence of certain physiological barriers in the brain that impede chemotherapeutics from reaching their target. Emerging research in circadian biology highlights the role of the internal circadian clock in tumour progression and treatment response. Evidence suggests that aligning therapy to patients' chronotypes could potentially improve treatme

Salivary Melatonin Changes in Oncological Patients: A Systematic Review.

Melatonin is known as a regulator of circadian sleep and waking rhythm. This hormone secreted by the pineal gland also has protective, oncostatic, and antioxidant properties. This systematic review was designed to answer the question "Is there a relationship between salivary melatonin changes and oncological diseases?". Following the inclusion and exclusion criteria, ten studies were included, according to PRISMA statement guidelines. In all included studies, the diagnostic material was unstimulated whole saliva, in which the melatonin changes were determined by different laboratory methods. M

The Influence of Alertness on the Spatial Deployment of Visual Attention is Mediated by the Excitability of the Posterior Parietal Cortices.

With a reduced level of alertness, healthy individuals typically show a rightward shift when deploying visual attention in space. The impact of alertness on the neural networks governing visuospatial attention is, however, poorly understood. By using a transcranial magnetic stimulation twin-coil approach, the present study aimed at investigating the effects of an alertness manipulation on the excitability of the left and the right posterior parietal cortices (PPCs), crucial nodes of the visuospatial attentional network. Participants' visuospatial attentional deployment was assessed with a free

Reallocating morning physical activity to later-day activity and its association with type 2 diabetes incidence: The Maastricht Study.

AIMS: The timing of physical activity may influence metabolic health through interactions with circadian rhythms, yet its role in type 2 diabetes mellitus (T2DM) development is unclear. We investigated associations between time-of-day-specific physical activity and incident T2DM, and whether theoretically reallocating activity from morning to later in the day was associated with changes in T2DM risk. MATERIALS AND METHODS: We included 4615 participants from The Maastricht Study cohort without diabetes (age 59.2 ± 8.6 years; 56.3% women). Device-based physical activity was measured over 7 days

Daily associations of subjective and objective sleep parameters with restorative sleep and morning sleepiness in Japanese working adults.

Restorative sleep and morning sleepiness are essential factors associated with daytime functioning among workers, yet their variability and determinants remain unclear. Previous studies have primarily focused on either subjective or objective sleep measures, with many cross-sectional investigations, although fewer longitudinal studies have considered the interaction with psychological and chronobiological factors. The aim of this study was to investigate both subjective and objective sleep parameters, along with psychological and chronobiological factors, and examine their influence on daily r

Differences in circadian rhythm changes between myopic and non-myopic college students over 2 years.

This study aimed to characterize and compare the differences in circadian rhythm changes during 2 years between college students with myopia and non-myopia based on a longitudinal cohort study. Wake-up time and bedtime were obtained through a self-administered questionnaire. Chronotype was assessed using the reduced Morningness-Eveningness Questionnaire (rMEQ). Circadian rhythm timing was determined by dim-light melatonin onset (DLMO), measured through hourly saliva collection from 21:00 to 01:00. A total of 450 college students (146 [32.4%] males) with a mean age of 18.65 ± 1.05 years were in
