Antifungal susceptibility, enzymatic activity, PCR-fingerprinting and ITS sequencing of environmental Cryptococcus laurentii isolates from Uberaba, Minas Gerais, Brazil.

Cryptococcus laurentii has been classically considered a saprophytic species, although several cases of human infection have been already reported. This study aimed to evaluate the phospholipase, proteinase and hemolysins activity, the antifungal susceptibility profile, the genetic variability by M13 and (GACA)(4) fingerprinting and the internal transcribe spacer (ITS) sequencing of 38 C. laurentii isolates recovered from captive bird droppings and surrounding hospital areas. All of them exhibited phospholipase activity, while the hemolytic activity was evidenced in 34 (89.4%) isolates. None o

Bacillus cereus enterotoxins change epithelial barrier function and ion transport in a Caco-2/HT29-MTX co-culture model, provoking inflammation, apoptosis and membrane damage.

Diarrheal disease caused by Bacillus cereus is driven by pore-forming enterotoxins, with hemolysin BL (Hbl) and non-hemolytic enterotoxin (Nhe) as major virulence factors. Toxin structure and mode of action are well described, while especially human intestinal epithelial responses remain incompletely characterized. With the co-culture system of Caco-2 and mucus-producing HT29-MTX cells (9:1), this study describes a suitable model for investigating enterotoxin effects. Cell line-, dose- and enterotoxin-dependent changes in cell viability were detected. Undifferentiated Caco-2 generally showed h

Broad-Spectrum Antimicrobial and Antibiofilm Activities of Halogenated Anilines Against Uropathogenic Escherichia coli and ESKAPE Pathogens.

Uropathogenic Escherichia coli (UPEC) is one of the leading causes of nosocomial infections and urinary tract infections (UTIs), capable of inducing a spectrum of conditions ranging from acute bladder cystitis to chronic pyelonephritis. The virulence arsenal of UPEC includes factors, such as curli fimbriae, hemolysin, motility elements and metallophores. Moreover, UPEC can form biofilm-like communities and quiescent intracellular reservoirs within host tissues, contributing to recurrent and persistent infections. This study investigates the antibiofilm and antimicrobial activities of two halog

SARS-CoV-2 Post-Infection and Sepsis by Saccharomyces cerevisiae: A Fatal Case Report-Focus on Fungal Susceptibility and Potential Virulence Attributes.

The pandemic caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been responsible for approximately 6.8 million deaths worldwide, threatening more than 753 million individuals. People with severe coronavirus disease-2019 (COVID-19) infection often exhibit an immunosuppression condition, resulting in greater chances of developing co-infections with bacteria and fungi, including opportunistic yeasts belonging to the Saccharomyces and Candida genera. In the present work, we have reported the case of a 75-year-old woman admitted at a Brazilian university hospital with an

Biological effects of two genetically defined leukotoxin mutants of Mannheimia haemolytica.

Mannheimia(Pasteurella)haemolytica serotype 1 is the primary causative agent responsible for bovine pneumonic mannheimiosis, also known as shipping fever in cattle. The bacterium produces a variety of virulence factors, foremost of which is the exotoxic leukotoxin. The leukotoxin is a calcium-dependent cytolysin that is a member of the RTX (repeats in toxin) family and exhibits a narrow cell-type and species specificity and has biological effects only on ruminant leukocytes and platelets. The genetic organization of the leukotoxin is comprised of four genes: lktC, lktA, lktB and lktD. The lktA

Environmental transition navigates phenotype switching, affecting the virulence and multidrug-resistant profile of pathogenic Morganella morganii.

The bacteria's ability to respond to environmental changes is critical for their survival. This allows them to form intricate communities, withstand stress, and initiate virulence responses in hosts during infection, a phenomenon known as phenotypic switching. In this study, we investigated the role of shaking conditions on phenotype switch in multidrug-resistant and pathogenic Morganella morganii both under in vitro and in vivo conditions. The results demonstrate that M. morganii grown in non-shaking conditions, possibly causing low fluid shear, developed floccules or cellular aggregates, and

Resistance to Cry14A family Bacillus thuringiensis crystal proteins in Caenornabditis elegans operates via the nhr-31 transcription factor and vacuolar-type ATPase pathway.

Bacillus thuringiensis (Bt) has been successfully used commercially for more than 60 years for biocontrol of insect pests. Since 1996, transgenic plants expressing Bt crystal (Cry) proteins have been used commercially to provide protection against insects that predate on corn and cotton. More recently, Bt Cry proteins that target nematodes have been discovered. One of these, Cry14Ab, has been expressed in transgenic soybean plants and found to provide significant protection against the soybean cyst nematode, Heterodera glycines. However, to date there has been no description of high-level resi

Use of listeriolysin O and internalin A in a seroepidemiological study of listeriosis in Swiss dairy cows.

Recombinant listeriolysin O and internalin A were used as antigens in an enzyme-linked immunosorbent assay (ELISA) for the specific detection of anti-Listeria monocytogenes antibodies in cattle. The results showed sensitivities and specificities of 82 and 92%, respectively, for the listeriolysin O ELISA, and 100 and 90%, respectively, for the internalin A ELISA, respectively. The test may be useful for the confirmation of listeria-related abortions and mastitis but does not seem to be indicated for use in the diagnosis of listeria-related encephalitis in cattle. [Investigation of Antibiotic Resistance and Virulence Factors of Enterococcus faecium and Enterococcus faecalis Strains Isolated from Clinical Samples].

Enterococci, which are commonly found in the environment, cause serious infections despite the absence of well-defined virulence factors and toxins. Knowing the virulence properties of enterococci is important to understand the complex pathogenic structures. In this study, we aimed to investigate the virulence factors (asa1, hyl, cylA, efa, ebp, ace, esp, gelE, sprE, fsrA, fsrB, fsrC genes, gelatinase activity, hemolysin, hydrogen peroxide and biofilm production) and antibiotic resistance of Enterococcus faecium and Enterococcus faecalis strains isolated from clinical specimens. Machine Learning Analysis of Type VII Secretion System Expression and Its Relationships With Virulence Traits and Antibiotic Resistance in Staphylococcus aureus.

Background Staphylococcus aureus (S. aureus) remains a high-priority pathogen due to its extensive virulence arsenal, immune evasion strategies, and rising multidrug resistance, particularly in methicillin-resistant strains (MRSA). The type VII secretion system (T7SS), encoded by the ess locus, contributes to persistence, immune modulation, and interbacterial competition. Although recognized as a key virulence determinant, its regulatory interplay with other virulence traits and resistance mechanisms remains unclear. Understanding these relationships is essential to identifying novel antivirul

Novel anti-virulence compounds disrupt exotoxin expression in MRSA.

Hemolysins are lytic exotoxins expressed in most strains of S. aureus, but hemolytic activity varies between strains. We have previously reported several novel anti-virulence compounds that disrupt the S. aureus transcriptome, including hemolysin gene expression. This report delves further into our two lead compounds, loratadine and a structurally related brominated carbazole, and their effects on hemolysin production in methicillin-resistant S. aureus (MRSA). To gain understanding into how these compounds affect hemolysis, we analyzed these exotoxins at the DNA, RNA, and protein level after i

Multivalent Nanobodies for Potent and Broad Neutralization of Staphylococcus aureus Toxins.

UNLABELLED: Staphylococcus aureus is a leading cause of lethal bacteremia and pneumonia, which are driven by potent virulence factors such as T-cell superantigens and alpha hemolysin. S. aureus has among the highest rates of antibiotic resistance, yet no vaccines or alternative therapies are available despite decades of research. Here, we developed a repertoire of potent, high affinity nanobodies (Nbs) targeting key toxins in S. aureus infection, including superantigens (SAgs) SEB, SEC, TSST-1, and Hla. Comprehensive cryo-EM and AlphaFold3 analyses of these Nbs, which were elicited with clinic

Identification of New Virulence Factors and Vaccine Candidates for Yersinia pestis.

Earlier, we reported the identification of new virulence factors/mechanisms of Yersinia pestis using an in vivo signature-tagged mutagenesis (STM) screening approach. From this screen, the role of rbsA, which encodes an ATP-binding protein of ribose transport system, and vasK, an essential component of the type VI secretion system (T6SS), were evaluated in mouse models of plague and confirmed to be important during Y. pestis infection. However, many of the identified genes from the screen remained uncharacterized. In this study, in-frame deletion mutants of ypo0815, ypo2884, ypo3614-3168 (cyoA

Listeria monocytogenes virulence factors that stimulate endothelial cells.

Listeria monocytogenes infection of endothelial cells upregulates surface expression of adhesion molecules and stimulates neutrophil adhesion to infected cell monolayers. The experiments presented here tested the roles of specific bacterial virulence factors as triggers for this inflammatory phenotype and function. Human umbilical vein endothelial cell (HUVEC) monolayers were infected with wild-type L. monocytogenes or L. monocytogenes mutants; then surface expression of E-selectin and neutrophil adhesion were measured. The results showed that delta hly and prfA mutants were the most crippled,

Genetic and phenotypic evaluation of Candida albicans strains isolated from subgingival biofilm of diabetic patients with chronic periodontitis.

Candida spp. are commensal microorganisms that are part of the microflora of different sites within the oral cavity. In healthy subjects, who have an unaltered immunological status, these yeasts do not cause disease. However, in immunosuppressed individuals whose condition may have been caused by diabetes mellitus, Candida spp. can express different virulence factors and may consequently become pathogenic. Studies have detected the presence of Candida spp. in periodontal sites of patients with chronic periodontitis, especially those that are immunologically compromised. However, the role of th
