Serological epitope profile of anti-Ro52-positive patients with systemic autoimmune rheumatic diseases.

BACKGROUND: Ro52 is an interferon-inducible protein of the tripartite motif family. Antibodies against Ro52 have been described in patients with different autoimmune diseases, such as systemic lupus erythematosus and Sjögren's syndrome, that are often associated with anti-Ro60 antibodies. The Ro52 autoantigen is extraordinarily immunogenic, and its autoantibodies are directed against both linear and conformational epitopes. The aim of this study was to evaluate the prevalence of antibodies to the five Ro52 domains, as well as to Ro52 176- to 196-amino acid (aa) and 200-239-aa peptides, in diff

Extensive circadian and light regulation of the transcriptome in the malaria mosquito Anopheles gambiae.

BACKGROUND: Mosquitoes exhibit 24 hr rhythms in flight activity, feeding, reproduction and development. To better understand the molecular basis for these rhythms in the nocturnal malaria vector Anopheles gambiae, we have utilized microarray analysis on time-of-day specific collections of mosquitoes over 48 hr to explore the coregulation of gene expression rhythms by the circadian clock and light, and compare these with the 24 hr rhythmic gene expression in the diurnal Aedes aegypti dengue vector mosquito. RESULTS: In time courses from An. gambiae head and body collected under light:dark cycl

Reduction of cross-reactive carbohydrate determinants in plant foodstuff: elucidation of clinical relevance and implications for allergy diagnosis.

BACKGROUND: A longstanding debate in allergy is whether or not specific immunoglobulin-E antibodies (sIgE), recognizing cross-reactive carbohydrate determinants (CCD), are able to elicit clinical symptoms. In pollen and food allergy, ≥20% of patients display in-vitro CCD reactivity based on presence of α1,3-fucose and/or β1,2-xylose residues on N-glycans of plant (xylose/fucose) and insect (fucose) glycoproteins. Because the allergenicity of tomato glycoallergen Lyc e 2 was ascribed to N-glycan chains alone, this study aimed at evaluating clinical relevance of CCD-reduced foodstuff in patients

Vesicular stomatitis.

Vesicular stomatitis is a disease of livestock caused by some members of the Vesiculovirus genus (Family Rhabdoviridae), two of which are called 'vesicular stomatitis virus'. Clinical disease presents as severe vesiculation and/or ulceration of the tongue, oral tissues, feet, and teats, and results in substantial loss of productivity. Except for its appearance in horses, it is clinically indistinguishable from foot-and-mouth disease. Unlike foot-and-mouth disease, it is very infectious for man and can cause a temporarily debilitating disease. Vesicular stomatitis occurs seasonally every year i

Nano-enabled RNA interference at the plant-virus interface: antiviral mechanisms, nanocarrier delivery and translational challenges.

Viral diseases remain among the most difficult constraints in horticultural crops because they reduce yield, impair market quality, persist in vegetatively propagated material and are intensified by mixed infections, insect vectors and climate-driven shifts in disease epidemiology. Conventional measures, including resistant cultivars, sanitation, vector management, pesticide use and virus-free planting material, remain essential but often lack the precision and flexibility required for rapidly evolving plant-virus pathosystems. RNA interference (RNAi) provides a sequence-specific antiviral str

Invertebrate Automated Phenotyping Platform (INVAPP): An Automated High-Throughput System with Applications in Understanding and Combating Human Diseases.

The nematode Caenorhabditis elegans is a eukaryotic genetic model organism introduced for studies of animal development and behavior (Brenner S, Genetics 77:71-94, 1974). It is also proving useful to expedite our understanding of human diseases and to explore potential therapies (Ahringer J, Curr Opin Genet Dev 7:410-415, 1997; Culetto E, Sattelle DB, Hum Mol Genet 9:869-877, 2000). Monitoring phenotypic changes and the impact of drug candidates is particularly convenient in the case of C. elegans models of neuromuscular or neurological disorders, where changes in motility and growth are often

Draft genome sequences and comparative analyses of Spiroplasma citri strains from various hosts.

Spiroplasma citri is a plant pathogenic bacterium that infects a wide range of hosts. In Iran, S. citri is responsible for causing several important diseases, including citrus stubborn, sesame yellows, and safflower necrotic yellows. The beet leafhopper Circulifer haematoceps is a common vector for S. citri transmission in the Mediterranean and Middle Eastern regions. For the first time, we sequenced the whole genome of three Iranian strains D10, D12, and D4 isolated from plants (citrus and sesame) and the insect hosts, originating from Fars Province, for a better understanding of their geneti

Spatial Dynamics and Sterilization Range of Incompatible Aedes albopictus Males: Advancing Toward an Optimized IIT Approach.

The Incompatible Insect Technique (IIT) is a species-specific, eco-friendly mosquito control method that relies on releasing Wolbachia-infected males, which induce cytoplasmic incompatibility (CI), rendering eggs inviable when mating with wild females. Aiming at optimizing IIT protocols in terms of cost-effectiveness, data on incompatible male dispersal and survival and the distance- and time-related impact of induced sterility are fundamental. This study plans to fill this gap and reports findings from a two-year field trial (2022-2023) at the ENEA-Casaccia Research Center, based on single-sp

Clodronate Liposomes Untangle the Role of Hemocytes in Apis mellifera Response to Temperature Variation and Microbial Infection.

INTRODUCTION: The cellular immune response, mediated by hemocytes, is a fundamental component of honey bee (Apis mellifera) health. However, the specific contribution of hemocyte subtypes to resilience against combined stressors remains poorly characterized. This study investigated the effect of temperature and bacterial challenge on hemocyte abundance. We employed clodronate liposomes (CLD) for the first time in honey bees as a novel tool for the selective depletion of hemocytes to investigate this gap. METHODS: Five-day-old (nurses) and fifteen-day-old (foragers) honey bees were treated wit

Sulfur fumigation of botanical drugs: impact on chemical composition and pharmacological properties, and advances in detection technologies.

Traditional Chinese medicine (TCM), a fundamental aspect of traditional medicine, is highly regarded for its natural efficacy and diverse applications. However, sulfur fumigation, a prevalent processing technique used to prevent insect and mold infestations, poses significant risks, including the presence of Sulfur Dioxide (SO2) residues, alterations in chemical compositions, diminished therapeutic effects, and heightened toxicity. As the demand for TCM continues to rise, ensuring its quality and safety becomes increasingly critical. This review explores the significance of analytical and regu

Physiological and metabolic alterations induced by commercial neonicotinoid formulations in Daphnia magna.

Neonicotinoid insecticides are widely used agents in agriculture to control a broad range of insect pests. Although use of neonicotinoid pesticides has resulted in the widespread contamination of surface waters, sublethal toxicity data of these products in relation to non-target aquatic biota are still poor. Therefore, the objective of this study was to assess the effects of two neonicotinoid pesticides with widespread use on the basic physiological functions: the thoracic limb activity and heart rate of Daphnia magna, and to screen for their potential to affect the cytochrome P450 monooxygena

Estimating the effect of the wMel release programme on the incidence of dengue and chikungunya in Rio de Janeiro, Brazil: a spatiotemporal modelling study.

BACKGROUND: Introgression of genetic material from species of the insect bacteria Wolbachia into populations of Aedes aegypti mosquitoes has been shown in randomised and non-randomised trials to reduce the incidence of dengue; however, evidence for the real-world effectiveness of large-scale deployments of Wolbachia-infected mosquitoes for arboviral disease control in endemic settings is still scarce. A large Wolbachia (wMel strain) release programme was implemented in 2017 in Rio de Janeiro, Brazil. We aimed to assess the effect of this programme on the incidence of dengue and chikungunya in

Spatio-temporal distribution of Spiroplasma infections in the tsetse fly (Glossina fuscipes fuscipes) in northern Uganda.

Tsetse flies (Glossina spp.) are vectors of parasitic trypanosomes, which cause human (HAT) and animal African trypanosomiasis (AAT) in sub-Saharan Africa. In Uganda, Glossina fuscipes fuscipes (Gff) is the main vector of HAT, where it transmits Gambiense disease in the northwest and Rhodesiense disease in central, southeast and western regions. Endosymbionts can influence transmission efficiency of parasites through their insect vectors via conferring a protective effect against the parasite. It is known that the bacterium Spiroplasma is capable of protecting its Drosophila host from infectio

Wolbachia infection in Argentinean populations of Anastrepha fraterculus sp1: preliminary evidence of sex ratio distortion by one of two strains.

BACKGROUND: Wolbachia, one of the most abundant taxa of intracellular Alphaproteobacteria, is widespread among arthropods and filarial nematodes. The presence of these maternally inherited bacteria is associated with modifications of host fitness, including a variety of reproductive abnormalities, such as cytoplasmic incompatibility, thelytokous parthenogenesis, host feminization and male-killing. Wolbachia has attracted much interest for its role in biological, ecological and evolutionary processes as well as for its potential use in novel and environmentally-friendly strategies for the contr

Bio-pesticide applications may impair the pollination services and survival of foragers of honey bee, Apis cerana Fabricius in oilseed brassica.

Honey bees and brassica plants are co-evolved and due to the peculiar floral characters, mustard (Brassica juncea) plants are strongly dependent on bees for survival. Mustard is one of the most important oilseeds across the world. Insect pests often cause huge economic losses in mustard and their management, especially during flowering stage is very crucial to achieve maximum yield, although this step often displays undesirable effects on the foraging bees. Effects of synthetic pesticides on bees are widely documented and extensively reported. Although the numbers of pesticides/bio-pesticides

Gene expression and plant hormone levels in two contrasting rice genotypes responding to brown planthopper infestation.

BACKGROUND: The brown planthopper (BPH; Nilaparvata lugens Stål) is a destructive piercing-sucking insect pest of rice. The plant hormones salicylic acid (SA) and jasmonic acid (JA) play important roles in plant-pest interactions. Many isolated rice genes that modulate BPH resistance are involved in the metabolism or signaling pathways of SA, JA and ethylene. 'Rathu Heenati' (RH) is a rice cultivar with a high-level, broad-spectrum resistance to all BPH biotypes. Here, RH was used as the research material, while a BPH-susceptible rice cultivar 'Taichung Native 1' (TN1) was the control. A cDNA

The Genomic Architecture of Novel Simulium damnosum Wolbachia Prophage Sequence Elements and Implications for Onchocerciasis Epidemiology.

Research interest in Wolbachia is growing as new discoveries and technical advancements reveal the public health importance of both naturally occurring and artificial infections. Improved understanding of the Wolbachia bacteriophages (WOs) WOcauB2 and WOcauB3 [belonging to a sub-group of four WOs encoding serine recombinases group 1 (sr1WOs)], has enhanced the prospect of novel tools for the genetic manipulation of Wolbachia. The basic biology of sr1WOs, including host range and mode of genomic integration is, however, still poorly understood. Very few sr1WOs have been described, with two such

Keloid and Hypertrophic Scars Are the Result of Chronic Inflammation in the Reticular Dermis.

Keloids and hypertrophic scars are caused by cutaneous injury and irritation, including trauma, insect bite, burn, surgery, vaccination, skin piercing, acne, folliculitis, chicken pox, and herpes zoster infection. Notably, superficial injuries that do not reach the reticular dermis never cause keloidal and hypertrophic scarring. This suggests that these pathological scars are due to injury to this skin layer and the subsequent aberrant wound healing therein. The latter is characterized by continuous and histologically localized inflammation. As a result, the reticular layer of keloids and hype
