Supravital uptake of methylene blue by dendritic cells within stratified squamous epithelia: a light and electron microscope study.

Electron microscopic data on methylene blue staining of dendritic cells in the epithelia of the soft palate and skin of the mouse after supravital dye injection are presented. The ultra-structural details were compared with corresponding light microscopic findings. Methylene blue stained tissue was fixed by immersion in a paraformaldehyde-glutaraldehyde solution containing phosphomolybdic acid. The ensuing dye precipitate was stabilized by ammonium heptamolybdate. The light microscopic investigation revealed that selective staining of dendritic cells depended on the presence of ambient oxygen.

Singlet oxygen as an ultimately reactive species in Salmonella typhimurium DNA damage induced by methylene blue/visible light.

The specific recognition of various DNA modifications by repair enzymes is exploited for the analysis of DNA damage induced by visible light in the presence of methylene blue in Salmonella typhimurium. The relative frequencies of various endonuclease-sensitive sites and strand breaks are determined in the plasmid pAQ1 of the treated bacteria and are compared with those observed after exposure of isolated DNA to various conditions. This comparison of damage profiles indicates that the cellular DNA damage by illumination in the presence of methylene blue is caused predominantly by the direct act

The role of blue light-filtering and premium intraocular lenses on postoperative falls: A nationwide target trial emulation in Taiwan.

BACKGROUND: Falls are a leading cause of injury and premature death in older adults, and cataract surgery reduces fall risk. Concerns existed that blue-light filtering (BF) intraocular lenses (IOLs) might impair light transmittance and increase fall risk. We aimed to compare fall incidence and injury diagnoses among patients who received bilateral cataract surgery with premium BF, premium non-BF, and standard non-BF IOLs. MATERIAL AND METHODS: We emulated a target trial by enrolling 26,730 well-matched patients per IOL cohort who underwent bilateral cataract surgeries between 2011 and 2017 fr

Pupillary response to blue light as a biomarker of seasonal pattern in Major Depressive Episode: A clinical study using pupillometry.

Depressive disorders are characterized by disturbances in light signal processing. More specifically, an alteration of the melanopsin response is suggested. The post-illumination pupillary response (PIPR) to blue light (post-blue PIPR) is increasingly used as a marker of the activity of intrinsically photosensitive melanopsin ganglion cells (ipRGCs). We hypothesized that individuals with Major Depressive Episode (MDE) who exhibited a higher vulnerability to season patterns showed a decreased ability to transmit light signals to the brain. We explored the correlation between the post-blue PIPR

Nesfatin-1 protects the reproductive health of male Sprague Dawley rats exposed to blue and white LED lights.

There is little information on the effects of exposure to light emitting diode (LED) illumination on the welfare of laboratory animals. Nesfatin-1, a satiety-regulation peptide present in various tissues, is found in the central nervous system and participates in the stress response. The present study investigated whether exposure to blue and white LED lights for 14 weeks affected growth and reproductive, biochemical and histopathological parameters in male Sprague Dawley (SD) rats as well as whether subcutaneous (SC) injection of nesfatin-1 (0.5 mg/kg bodyweight) in the last two weeks of the

Damaging effects of UVA, blue light, and infrared radiation: in vitro assessment on a reconstructed full-thickness human skin.

INTRODUCTION: Exposure to solar radiation can cause a range of skin damage, including sunburn, erythema, skin carcinogenesis, the release of reactive oxygen species (ROS), inflammation, DNA damage, and photoaging. Other wavelengths beyond UVB, such as UVA, blue light, and infrared radiation, can also contribute to the harmful effects of solar radiation. Reconstructed full-thickness human skin has the potential to serve as effective predictive in vitro tools for evaluating the effects of solar radiation on the skin. The aim of this work was to evaluate the damaging effects of UVA, blue light, a

The Role of Oxidative Stress and Autophagy in Blue-Light-Induced Damage to the Retinal Pigment Epithelium in Zebrafish In Vitro and In Vivo.

Age-related macular degeneration (AMD) is the progressive degeneration of the retinal pigment epithelium (RPE), retina, and choriocapillaris among elderly individuals and is the leading cause of blindness worldwide. Thus, a better understanding of the underlying mechanisms in retinal tissue activated by blue light exposure is important for developing novel treatment and intervention strategies. In this study, blue-light-emitting diodes with a wavelength of 440 nm were applied to RPE cells at a dose of 3.7 ± 0.75 mW/cm2 for 24 h. ARPE-19 cells were used to investigate the underlying mechanism i

Impact of blue light therapy on plasma adrenocorticotropic hormone (ACTH) and hypertrichosis in horses with pituitary pars intermedia dysfunction.

Blue light therapy can be used in horses to alter the natural photoperiod and inhibit winter hair coat growth. Seasonal increases in ACTH occur in the fall season but are exaggerated in horses with pituitary pars intermedia dysfunction (PPID). Additionally, PPID horses frequently present with hypertrichosis. Thus, blue light therapy was proposed as a potential management tool for hypertrichosis and for investigating the impact of photoperiod manipulation on ACTH. Eighteen PPID horses, aged 18 to 31 yr, from a university-owned research herd were selected and assigned to either the control group

Antibacterial Photodynamic Treatment of Porphyromonas gingivalis with Toluidine Blue O and a NonLaser Red Light Source Enhanced by Dihydroartemisinin.

In vitro experiments confirmed that antibacterial photodynamic treatment (aPDT) inactivates periodontal pathogens. However, more effective sterilization is needed in the complex oral environment. This study tested whether dihydroartemisinin (DHA) enhanced the photokilling effect of aPDT on Porphyromonas gingivalis (P. gingivalis) in planktonic and biofilm states. aPDT combining toluidine blue O (TBO) with 630 nm red light was performed on bacterial suspensions and biofilms in vitro with different final concentrations of DHA (10, 20 and 40 μg mL-1 ). Randomised controlled trial of the efficacy of a blue-enriched light intervention to improve alertness and performance in night shift workers.

OBJECTIVES: Night workers often experience high levels of sleepiness due to misalignment of the sleep-wake cycle from the circadian pacemaker, in addition to acute and chronic sleep loss. Exposure to light, in particular short wavelength light, can improve alertness and neurobehavioural performance. This randomised controlled trial examined the efficacy of blue-enriched polychromatic light to improve alertness and neurobehavioural performance in night workers. DESIGN: Participants were 71 night shift workers (42 males; 32.8±10.5 years) who worked at least 6 hours between 22:00 and 08:00 hours

Neonatal Blue Light Phototherapy and Melanocytic Nevus Count in Children: A Systematic Review and Meta-Analysis of Observational Studies.

BACKGROUND: Neonatal blue light phototherapy (NBLP) is an established method of managing neonatal hyperbilirubinemia. Approximately 5% of newborns are exposed to NBLP. Evidence of whether NBLP predisposes to the development of melanocytic nevi later in life has been conflicting. OBJECTIVES: The goal of the current study was to conduct a systematic review and meta-analysis to quantitatively assess the effect of NBLP on melanocytic nevus count. METHODS: We searched for observational studies in Medline, EMBASE, and the Cochrane Central Register from their inception to April 15, 2015. Meta-analy

Molecular cloning, mRNA expression, and immunocytochemical localization of a putative blue-light photoreceptor CRY4 in the chicken pineal gland.

In non-mammalian vertebrates, the pineal gland contains an endogenous circadian oscillator and serves as a photosensitive neuroendocrinal organ. To better understand the pineal phototransduction mechanism, we focused on the chicken putative blue-light photoreceptive molecule, Cryptochrome4 (cCRY4). Here we report the molecular cloning of pineal cCry4 cDNA, the in vivo expression of cCry4 mRNA, and the detection of cCRY4 protein. cCry4 is transcribed in a wide variety of chick tissues out of which the pineal gland and retina contain high levels of cCry4 mRNA. In the pineal gland, under 12 h lig

Blue Light Aminolevulinic Acid Photodynamic Therapy Did Not Induce Detectable CPD or 6-4PP Photolesions in Human Dermal Fibroblasts.

Photodynamic therapy (PDT) is an established dermatologic therapeutic modality used to induce apoptosis within targeted tissue. Despite increasing clinical use, the genotoxic safety of blue light (BL) PDT has not yet been established. We examined whether combination BL (417 ± 5 nm) and 5-aminolevulinic acid (5-ALA) PDT induces cyclobutane pyrimidine dimers (CPDs) and 6-4 photoproducts (6-4PPs) in human dermal fibroblasts. CRL-2617 and AG-13145 fibroblasts were treated with 0, 0.5, or 1 mM 5-ALA, then irradiated with BL at 10, 30, or 45 J/cm2. Unlike robust photolesion formation in the UVB-irra

Dynamin-Related Protein 1-Dependent Disruption of Mitochondrial Homeostasis Drives Blue Light-Induced Epithelial-Mesenchymal Transition in Retinal Aging.

Age-related macular degeneration (AMD) stands as a leading cause of blindness in the elderly, yet the fundamental aging processes that underpin its pathogenesis remain incompletely defined. The dysfunction of retinal pigment epithelial (RPE) cells is a central event in AMD, a process that shares key hallmarks with broader cellular aging, particularly the progressive decline in mitochondrial function. In this study, we investigated how a common environmental stressor-blue light-triggers a key pathological transformation, epithelial-mesenchymal transition (EMT), in RPE cells by specifically disr

Transparent, 3D-printed microneedle patches that co-deliver ALA and guide blue light for preclinical photodynamic therapy of acne.

Acne is a common chronic inflammatory skin disease associated with Cutibacterium acnes (C. acnes). Although photodynamic therapy (PDT) effectively improves acne, the transdermal delivery of photosensitizers and limited light penetration through the skin restrict its therapeutic efficacy. In this study, we developed a dual-functional flexible microneedle patch using 3D printing technology, capable of simultaneously delivering the photosensitizer 5-aminolevulinic acid (ALA) and blue light. The microneedle patch exhibits favorable mechanical properties (a fracture force of 2.47 N per patch and a
