Analgesia after lateral thoracotomy in dogs. Epidural morphine vs. intercostal bupivacaine.

One of two analgesic treatments was randomly assigned to 40 dogs undergoing lateral thoracotomy. Group E (20 dogs) received an epidural injection of morphine (0.1 mg/kg) before surgery. Group I (20 dogs) received injections of bupivacaine around five intercostal nerves when the thorax was about to be closed. All dogs were given an opioid as part of their preoperative medication (meperidine or butorphanol), followed by thiopental for induction and halothane or methoxyflurane for maintenance of anesthesia. Scores were assigned for pre-operative demeanor and response to restraint and injection. A

Phosphodiesterase inhibitors potentiate opiate-antagonist discrimination by morphine-dependent rats.

This study was performed to examine the relevance of the quasi-withdrawal syndrome in nondependent rats to the syndrome precipitated by naltrexone in rats physically dependent upon morphine. Morphine-dependent rats trained to discriminate between SC injections of naltrexone (0.1 mg/kg) and saline were pretreated with 10 mg/kg of a phosphodiesterase inhibitor: 3-isobutyl-1-methylxanthine (IBMX), Ro 20-1724, or papaverine. The naltrexone stimulus-generalization curve and dose-response curve for loss of body weight were shifted to the left by IBMX and Ro 20-1724, which produce quasi-withdrawal, b

Hematological Parameters from the Feline Blood Donor to the Blood Unit: What Changes Are to Be Expected?

Feline blood donation requires sedation to allow for good blood collection, avoiding venous damage and unnecessary donor stress. In the present study, we describe the variation of hematological parameters in a population of 74 healthy blood-donor cats that underwent the same sedation protocol, including medetomidine, alfaxalone, and butorphanol. Changes in hematological parameters were evaluated in blood samples collected from conscious cats (group A), sedated cats (group B), and feline whole-blood units (FBUs) (group C). Significant differences were observed between unsedated and sedated cats

Same-day echocardiography in dogs sedated with intravenous dexmedetomidine followed by reversal with intramuscular atipamezole.

OBJECTIVE: To estimate the time until reliable echocardiography in healthy dogs by evaluating differences in measured echocardiographic variables in dogs before sedation, during sedation with dexmedetomidine, and after reversal with atipamezole. METHODS: 13 dogs were prospectively enrolled. Two-dimensional echocardiographic measurements on left atrial and ventricular dimensions and ventricular systolic function were obtained at each time point. Dogs underwent IV sedation with 10 µg/kg of dexmedetomidine and 0.2 mg/kg of butorphanol, and dexmedetomidine was reversed with atipamezole IM. Echoca

Blood biochemistry and hematological changes in rats after administration of a mixture of three anesthetic agents.

Currently, given the concerns regarding animal welfare, it is required that anesthesia or analgesia be used during surgery in experimental animals. Therefore, it is important to understand how anesthesia affects the health conditions of experimental animals. In this study, rat blood biochemistry and hematological changes were examined following administration of a mixture of three anesthetic agents-medetomidine, midazolam and butorphanol (MMB). One of three MMB dose combinations was subcutaneously administered to rats. After 1 hr, rats were treated with atipamezole, to reverse the anesthetic e

The effect of body position, sedation, and thoracic bandaging on functional residual capacity in healthy deep-chested dogs.

The objective of this study was to determine the effect of body position, chest wrap, and sedation on functional residual capacity (FRC) in 6 healthy dogs. Functional residual capacity was determined by helium dilution (re-breathing) whilst in different clinically relevant conditions. These conditions included the standing (sternal) and lateral positions in unsedated dogs and then again both standing and lateral following chest bandaging, and sedation with acepromazine, IV and butorphanol, IV. The mean FRC at each measurement point was determined, as was the change in FRC (delta FRC) from one

Interactions Between Opioids and Dextroamphetamine on Locomotor Activity: Influence of an Opioid's Relative Efficacy at the Mu Receptor.

Opioids and stimulants are often used in combination for both recreational and non-recreational purposes. High-efficacy mu opioid agonists generally increase the behavioral effects of stimulants, whereas opioid receptor antagonists generally attenuate the behavioral effects of stimulants; however, less is known regarding the interactions between stimulants and opioids possessing low to intermediate efficacy at the mu receptor. The purpose of this study was to examine the role of an opioid's relative efficacy at the mu receptor in altering the behavioral effects of dextro(d-)amphetamine. To thi

Paroxysmal Neuropathic Pruritus in Patients With Chiari Malformation Type I: A Rare Phenotype.

Chiari malformation type I (CM-I) is a group of deformities in the posterior fossa and hindbrain, including the pons, cerebellum, and medulla oblongata. Paroxysmal pruritus in CM-I has been reported only once before in the literature. This study was a cross-sectional study over 12 months at a tertiary care pediatric hospital involving children aged one to 18 years with CM-I presenting with paroxysmal itching. Three patients with CM-I presented with severe episodes of paroxysmal itching. Patient 3 was started on carbamazepine therapy for seizures, and incidentally, his itching subsided. The pru

Genes and pathways co-associated with the exposure to multiple drugs of abuse, including alcohol, amphetamine/methamphetamine, cocaine, marijuana, morphine, and/or nicotine: a review of proteomics analyses.

Drug addiction is a chronic neuronal disease. In recent years, proteomics technology has been widely used to assess the protein expression in the brain tissues of both animals and humans exposed to addictive drugs. Through this approach, a large number of proteins potentially involved in the etiology of drug addictions have been identified, which provide a valuable resource to study protein function, biochemical pathways, and networks related to the molecular mechanisms underlying drug dependence. In this article, we summarize the recent application of proteomics to profiling protein expressio

Evaluation of three midazolam-xylazine mixtures. Preliminary trials in dogs.

The depressant effects of midazolam and xylazine on the central nervous system (CNS) were evaluated in 12 dogs. Xylazine was administered to six dogs (1.1 mg/kg intravenously [IV]) followed in 5 minutes by midazolam (1.0 mg/kg intramuscularly [IM]). In a second group of six dogs, xylazine (2.2 mg/kg IM) was followed in 5 minutes by midazolam (1.0 mg/kg IV). Both drug regimens induced rapid and profound sedation or anesthesia. Duration of action varied with the doses and routes of administration. Dogs given the high dose of xylazine IM had an arousal time of 95.4 +/- 8.9 minutes and a walking t

Evaluation of different intramuscular injectable anesthetic combinations in rabbits: Impact on anesthetic depth, physiological parameters, and EEG recordings.

Rabbits have a high anesthesia-related mortality rate because of their small size, high metabolic rate and challenging airway management. This study aimed to investigate different combinations of intramuscularly administered anesthetics in New Zealand White rabbits, focusing on their effects on anesthetic depth, physiological parameters, and electroencephalogram (EEG) recordings. Defined doses ketamine (K), esketamine (SK), medetomidine (M), dexmedetomidine (D), midazolam (Mi), and butorphanol (B) were investigated and compared in five different combinations: KM (25/0.25 mg/kg), SKM (17/0.25 m
