Voltage-dependent potassium channels in activated rat microglia.

1. Voltage-dependent currents of untreated (proliferating) and lipopolysaccharide (LPS)-treated rat microglial cells in culture were recorded using the whole-cell patch-clamp technique. 2. Membrane potentials showed prominent peaks at -35 mV and -70 mV. Membrane potentials of LPS-treated cells alternated between the two values. This may be due to a negative slope region of the I-V relation resulting in two zero current potentials. 3. From a holding potential of -70 mV, hyperpolarizing steps evoked an inwardly rectifying current both in proliferating and in LPS-treated cells, while depolarizing

Nitric oxide, prostanoid and non-NO, non-prostanoid involvement in acetylcholine relaxation of isolated human small arteries.

The main purpose of the study was to clarify to which extent nitric oxide (NO) contributes to acetylcholine (ACh) induced relaxation of human subcutaneous small arteries. Arterial segments were mounted in myographs for recording of isometric tension, NO concentration and smooth muscle membrane potential. In noradrenaline-contracted arteries, ACh induced endothelium-dependent relaxations. The NO synthase inhibitor, N(G)-nitro-L-arginine (L-NOARG) had a small significant effect on the concentration-response curves for ACh, and in the presence of L-NOARG, indomethacin only caused a small addition

Sapecin B alters kinetic properties of rapidly inactivating K(+) channels in rat pituitary GH(3) cells.

BACKGROUND: Rhizocephalan interaction with their decapod hosts is a superb example of host manipulation. These parasites are able to alter the host's physiology and behavior. Host-parasite interaction is performed, presumably, via special modified rootlets invading the ventral ganglions. METHODS: In this study, we focus on the morphology and ultrastructure of these special rootlets in Polyascus polygeneus (Lützen & Takahashi, 1997), family Polyascidae, invading the neuropil of the host's nervous tissue. The ventral ganglionic mass of the infected crabs were fixed, and the observed sites of th

Potentiation of regulatory volume decrease by P2U purinoceptors in HSG-PA cells.

HSG-PA human salivary gland duct cells exhibit progressively increased regulatory volume decrease (RVD) in response to decreased medium osmolarity. The P2U purinoceptor agonist UTP causes a potentiation of RVD, the extent of which is most pronounced in 220 mosM medium and is least apparent in 180 mosM medium. We examined the underlying mechanisms for this effect. Exposure of HSG-PA cells to UTP promotes Ca2+ mobilization, hyperpolarization, and net K+ efflux, suggesting the participation of Ca(2+)-activated K+ channels in RVD. To delineate the anion counterpart of K+ movement during RVD, cell

The effect of racemic ketamine on the large conductance Ca(+2)-activated potassium (BK) channels in GH3 cells.

Recently, inhalation anesthetics have been reported to block BK channels in adrenal chromaffin cells. To determine if BK block was characteristic only of inhalation anesthetics or was also a property of other general anesthetics we examined the effects of ketamine, an intravenous general anesthetic which is structurally different than inhalation anesthetics. Cell-attached and excised patch single channel and standard whole cell recording techniques were used to examine the effect of racemic ketamine on the BK channel activity in GH3 cells. When solutions containing 150 mM KCl are used in both

Nitric oxide-mediated inhibitory response of rat proximal colon: independence from changes in membrane potential.

1. We studied the relation of nitric oxide-mediated relaxation of smooth muscle to changes in membrane potential of cells in the proximal colon of rats. 2. The resting membrane potential and electrical field stimulation (EFS)-induced junction potentials were recorded from the circular and longitudinal muscle cells. 3. Localized distension with a small balloon caused relaxation of the circular muscle on the anal side of the distended region (descending relaxation). Relaxation of the longitudinal muscle was also induced by EFS. 4. Inhibitory junction potentials (i.j.ps) were recorded from all ci

Modulation of K+ and Ca2+ channels by histamine H1-receptor stimulation in rabbit coronary artery cells.

1. The modulation of whole-cell K+ and Ca2+ currents by stimulation of histamine H1-receptors in freshly isolated single smooth muscle cells from the rabbit coronary artery was characterized using the patch-clamp technique at 35 degrees C. Single-channel K+ currents were also analysed using the cell-attached patch configuration. 2. The histamine H1-receptor agonist, 2-(2-aminoethyl)pyridine (AEP) (0.1 mM), increased the amplitude of voltage-activated inward Ba2+ currents, recorded using the perforated-patch recording technique, which could be completely blocked by the dihydropyridine antagonis

Somatostatin stimulates Ca(2+)-activated K+ channels through protein dephosphorylation.

The neuropeptide somatostatin inhibits secretion from electrically excitable cells in the pituitary, pancreas, gut and brain. In mammalian pituitary tumour cells somatostatin inhibits secretion through two distinct pertussis toxin-sensitive mechanisms. One involves inhibition of adenylyl cyclase, the other an unidentified cyclic AMP-independent mechanism that reduces Ca2+ influx by increasing membrane conductance to potassium. Here we demonstrate that the predominant electrophysiological effect of somatostatin on metabolically intact pituitary tumour cells is a large, sustained increase in the

Endothelium-independent vasodilation induced by kolaviron, a biflavonoid complex from Garcinia kola seeds, in rat superior mesenteric arteries.

Previous studies have established the hepatoprotective, gastroprotective, hypolipidemic and hypoglycemic effects of kolaviron (KV), a biflavonoid complex from Garcinia kola seeds. In this study, we investigated the mechanisms involved in the vasorelaxant effects of KV in isolated superior mesenteric arteries from normotensive rats. KV (1, 10, 30, 100, 300, 500 and 1,000 microg/ml) concentration-dependently inhibited the contractions induced by phenylephrine (PHE) (10 microM) and KCl (80 mM) in both endothelium-intact (E(max) = 58.3 +/- 1.7% and 51.4 +/- 1.3%, respectively) and -denuded rings (

A marine snail neurotoxin shares with scorpion toxins a convergent mechanism of blockade on the pore of voltage-gated K channels.

kappa-Conotoxin-PVIIA (kappa-PVIIA) belongs to a family of peptides derived from a hunting marine snail that targets to a wide variety of ion channels and receptors. kappa-PVIIA is a small, structurally constrained, 27-residue peptide that inhibits voltage-gated K channels. Three disulfide bonds shape a characteristic four-loop folding. The spatial localization of positively charged residues in kappa-PVIIA exhibits strong structural mimicry to that of charybdotoxin, a scorpion toxin that occludes the pore of K channels. We studied the mechanism by which this peptide inhibits Shaker K channels

Nitric oxide-independent activation of soluble guanylyl cyclase contributes to endotoxin shock in rats.

We investigated whether a complete inhibition of nitric oxide (NO) formation caused by bacterial endotoxin (lipopolysaccharide, LPS) in vivo prevents the hypotension and restores the vascular hyporeactivity to normal in vivo and ex vivo. The combination of dexamethasone (Dex; 3 mg/kg at 30 min before LPS) plus aminoguanidine (AG; 15 mg/kg at 2 h after LPS) inhibited the overproduction of nitrate (an indicator of NO) in the plasma and aortic smooth muscle and also prevented the development of the delayed hypotension in rats treated with LPS for 6 h. However, the vascular hyporeactivity to norep

Tityustoxin K alpha blocks voltage-gated noninactivating K+ channels and unblocks inactivating K+ channels blocked by alpha-dendrotoxin in synaptosomes.

Two nonhomologous polypeptide toxins, tityustoxin K alpha (TsTX-K alpha) and tityustoxin K beta (TsTX-K beta), purified from the venom of the Brazilian scorpion Tityus serrulatus, selectively block voltage-gated noninactivating K+ channels in synaptosomes (IC50 values of 8 nM and 30 nM, respectively). In contrast, alpha-dendrotoxin (alpha-DTX) and charybdotoxin (ChTX) block voltage-gated inactivating K+ channels in synaptosomes (IC50 values of 90 nM and 40 nM, respectively). We studied interactions among these toxins in 125I-alpha-DTX binding and 86Rb efflux experiments. Both TsTX-K alpha and

Adenosine-activated potassium current in smooth muscle cells isolated from the pig coronary artery.

1. The perforated patch technique with nystatin or amphotericin was used to record whole cell currents activated by adenosine in smooth muscle cells isolated enzymatically from pig coronary arteries. 2. Adenosine (5-40 microM) activated an outward current at a holding potential of 0 mV in 5 mM [K+]o and an inward current at -60 mV in 143 mM [K+]o. The dependence of the reversal potential for the adenosine-activated current on [K+]o suggests that it flows through K+ channels, while its current-voltage relation is consistent with the channels showing little voltage dependence. 3. The adenosine-a

Responsiveness of a human parotid epithelial cell line (HSY) to autonomic stimulation: muscarinic control of K+ transport.

Salivary electrolyte secretion is under the control of the autonomic nervous system. In this paper we report that HSY, an epithelial cell line derived from the acinar-intercalated duct region of the human parotid gland, responds to muscarinic-cholinergic (generation of Ca2+ signal) and beta-adrenergic (generation of cAMP signal), but not to alpha-adrenergic (lack of Ca2+ signal), receptor stimulation. The muscarinic response was studied in detail. Carbachol (10(-4) M, muscarinic agonist) or A23187 (5 microM, calcium ionophore) stimulation of HSY cells increases both 86Rb (K+) influx and efflux

Scorpion toxins specific for potassium (K+) channels: a historical overview of peptide bioengineering.

Scorpion toxins have been central to the investigation and understanding of the physiological role of potassium (K⁺) channels and their expansive function in membrane biophysics. As highly specific probes, toxins have revealed a great deal about channel structure and the correlation between mutations, altered regulation and a number of human pathologies. Radio- and fluorescently-labeled toxin isoforms have contributed to localization studies of channel subtypes in expressing cells, and have been further used in competitive displacement assays for the identification of additional novel ligands

Single channel study of a Ca(2+)-activated K+ current associated with ras-induced cell transformation.

1. Ras-transformed fibroblasts have a whole-cell Ca(2+)-activated K+ current which is either absent or unavailable for activation in their non-transformed counterparts. To better understand the physiological significance of this K+ current the single channel basis for the current was characterized in ras-transformed cells. 2. More than 90% of inside-out patches from ras-transformed balb 3T3 cells had a channel type which was Ca(2+)-activated (threshold < 0.2 microM internal Ca2+), K(+)-selective (permeability ratio PNa:PK < 0.02), and inwardly rectifying in symmetric 150 mM KCl solutions (cond

Small-conductance calcium-activated potassium (SK) channels in the amygdala mediate pain-inhibiting effects of clinically available riluzole in a rat model of arthritis pain.

BACKGROUND: Arthritis pain is an important healthcare issue with significant emotional and affective consequences. Here we focus on potentially beneficial effects of activating small-conductance calcium-activated potassium (SK) channels in the amygdala, a brain center of emotions that plays an important role in central pain modulation and processing. SK channels have been reported to regulate neuronal activity in the central amygdala (CeA, output nucleus). We tested the effects of riluzole, a clinically available drug for the treatment of amyotrophic lateral sclerosis, for the following reason

Structural and functional features of noxiustoxin: a K+ channel blocker.

Binding of [125I]-Noxiustoxin to rat brain synaptosome membranes and displacement with synthetic peptides corresponding to the amino acid sequence of Noxiustoxin, show that the N-terminal segment of this toxin is implicated in the recognition of brain K(+)-channels. Cleavage of NTX with cyanogen bromide, endopeptidase V8 from Staphylococcus aureus and endopeptidase Lys-C support these findings. On the contrary, a synthetic C-terminal tetradecapeptide of charybodotoxin (ChTX), shows that in this K(+)-channel blocker, the C-terminal region, rather than the N-terminal is capable of displacing 125
