Seroprevalence of some vector-borne infections of dogs in Hungary.

The first comprehensive study on the prevalence of canine vector-borne pathogens (Anaplasma phagocytophilum, Borrelia burgdorferi, Ehrlichia canis, and Dirofilaria immitis) was carried in Hungary because, except for babesiosis and dirofilariosis caused by Dirofilaria repens, there were no data on their regional distribution and prevalence. In 2011 and 2012, 1305 blood samples were collected from randomly selected, apparently healthy pet dogs in 167 localities of 19 counties of Hungary. All sera samples from dogs were screened for simultaneous qualitative detection of circulating antibodies to

Detection and differentiation of filarial parasites by universal primers and polymerase chain reaction-restriction fragment length polymorphism analysis.

Filarial nematode parasites are a serious cause of morbidity in humans and animals. Identification of filarial infection using traditional morphologic criteria can be difficult and lead to misdiagnosis. We report on a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)-based method to detect and differentiate a broad range of filarial species in a single PCR. The first internal transcribed spacer 1 (ITS1) along with the flanking 18S and 5.8S ribosomal DNA (rDNA) were isolated and cloned from Wuchereria bancrofti, Brugia malayi, and Brugia pahangi. Sequence analysis id

A case of autochthonous human Dirofilaria infection, Germany, March 2014.

Allium grayi Regel is a seasonally limited small perennial herb of the subfamily Allioideae, and is endemic to Matsu, Taiwan. The species possesses nutritious, ornamental value, and biological pharmacological activity. This study evaluated for the first time to induce polyploidy using the dissected bulb of A. grayi as explants in vitro, with the expectation of increasing the bulb size. Sterile bulbs were divided into four equal parts and pre-cultured under different durations before being soaked in a one g/L colchicine solution for 12 or 24 hours. Survival, regeneration, variation, and tetrapl

Molecular Analysis of Canine Filaria and Its Wolbachia Endosymbionts in Domestic Dogs Collected from Two Animal University Hospitals in Bangkok Metropolitan Region, Thailand.

Canine filariasis is caused by several nematode species, such as Dirofilaria immitis, Dirofilaria repens, Brugia pahangi, Brugia malayi, and Acanthocheilonema reconditum. Zoonotic filariasis is one of the world's neglected tropical diseases. Since 2000, the World Health Organization (WHO) has promoted a global filarial eradication program to eliminate filariasis by 2020. Apart from vector control strategies, the infection control of reservoir hosts is necessary for more effective filariasis control. In addition, many studies have reported that Wolbachia is necessary for the development, reprod

Evaluation of recombinant Babesia gibsoni thrombospondin-related adhesive protein (BgTRAP) for the sero-diagnosis of canine babesiosis.

Canine babesiosis, caused by Babesia gibsoni is one of the most significant tick-borne illnesses across the world. Light microscopy as well as polymerase chain reaction may fail in the diagnosis of disease when the level of parasitaemia is very low during subclinical and chronic cases. The serological techniques using a recombinant protein will be useful for the accurate and sensitive surveillance of the disease, especially in chronic cases. The present study describes the evaluation of recombinant N-terminal B. gibsoni Thrombospondin-related adhesive protein (BgTRAP) based indirect ELISA for

Molecular detection of vector-borne pathogens from mosquitoes collected in two zoological gardens in Germany.

In Germany, knowledge of disease agents transmitted by arthropods in zoological gardens is scarce. In the framework of ecological studies, mosquitoes were therefore collected in German zoological gardens and examined for mosquito-borne pathogen DNA and RNA. In total, 3840 mosquitoes were screened for filarial nematodes and three groups of viruses (orthobunyaviruses, flaviviruses, alphaviruses) while 405 mosquitoes were tested for avian malaria parasites. In addition to the filarial nematode species Dirofilaria repens (n = 1) and Setaria tundra (n = 8), Sindbis virus (n = 1) and the haemosporid

A high resolution melting real time PCR for mapping of filaria infection in domestic cats living in brugian filariosis-endemic areas.

We present here a real time PCR with high resolution melting (HRM) analysis for determining the prevalence and distribution of filarial species in domestic cats residing in brugian filariosis endemic areas of Narathiwat province, Thailand. Filarial species can be clearly distinguished in a single well using a single pair of primers. Blood samples were taken from a total of 2039 domestic cats living in endemic areas. Microfilariae were detected in 5.7% of the sample, while the overall prevalence of filaria infection by HRM analysis was 6.6%. The filariae species found in the infected cats were

Laboratory Comparison of Rapid Antigen Diagnostic Tests for Lymphatic Filariasis: STANDARD Q Filariasis Antigen Test (QFAT) Versus Bioline Filariasis Test Strip (FTS).

Accurate rapid diagnostic tests (RDTs) are needed to diagnose lymphatic filariasis (LF) in global elimination programmes. We evaluated the performance of the new STANDARD Q Filariasis Antigen Test (QFAT) against the Bioline Filariasis Test Strip (FTS) for detecting W. bancrofti antigen (Ag) in laboratory conditions, using serum (n = 195) and plasma (n = 189) from LF-endemic areas (Samoa, American Samoa and Myanmar) and Australian negative controls (n = 46). The prior Ag status of endemic samples (54.9% Ag-positive) was determined by rapid test (ICT or FTS) or Og4C3 ELISA. The proportion of sam

Vector-borne pathogens of zoonotic concern in hunting dogs of southern Italy.

Dogs are commonly exposed to vector-borne pathogens (VBPs), yet few data are available on hunting dogs, which are often at high risk of infection due to their involvement in field activities. To investigate the occurrence of VBPs and evaluate the relative performance of different diagnostic tools, blood and serum samples were collected from hunting dogs (n = 1,433) in rural areas of southern Italy. All samples were tested by Knott's technique for filarioids, serologically (SNAP® 4Dx® Plus) for Anaplasma spp., Borrelia burgdorferi sensu lato, Dirofilaria immitis and Ehrlichia spp. and molecular

Vector-borne pathogens in dogs from the Republic of Kosovo.

BACKGROUND: Canine vector-borne pathogens (CVBP) are transmitted by arthropod vectors such as ticks, fleas, mosquitoes, and phlebotomine sand flies and are of global veterinary and medical importance. Dogs are important reservoir hosts, which may develop potentially life-threatening clinical signs. The Balkan area harbors diverse vector fauna and associated CVBPs, and data, particularly from the Republic of Kosovo, are scarce. Considering the high number of stray and privately owned dogs primarily kept outside, living in close contact with dogs might promote spillover of zoonotic pathogens to

Molecular detection of Leishmania infantum, filariae and Wolbachia spp. in dogs from southern Portugal.

BACKGROUND: Leishmaniosis caused by the protozoan Leishmania infantum and dirofilariosis caused by the nematodes Dirofilaria immitis or Dirofilaria repens are vector-borne zoonoses widely present in the Mediterranean basin. In addition, some studies reported that the endosymbiont Wolbachia spp. play a role in the biology and pathogenesis of filarial parasites. The aim of this work was to evaluate the frequency of mono- and co-infections by L. infantum, filariae and Wolbachia spp. and their association with clinical signs in dogs from the south of Portugal. Leishmanial, filarial and Wolbachia s

Buffy coat smear or Knott's test: which to choose for canine microfilaria screening in field studies?

BACKGROUND: In recent years, an increasing number of cases of canine dirofilariasis have been reported worldwide. However, the rate of infection in dogs is largely unknown in many remote areas, and the importance of field studies for determination of the prevalence of canine dirofilariasis in such areas is well recognized. The detection of microfilariae by the modified Knott's test (MKT) is a recommended screening method for canine dirofilariasis. OBJECTIVES: The purposes of this study were to compare the diagnostic sensitivity of the MKT with the buffy coat smear method (BCS), and to evaluat

Diagnosis of human filariases (except onchocerciasis).

The traditional method of diagnosing filarial infections is to examine blood or skin samples for microfilariae and for many this is still the standard procedure. However, since the present global campaign to eliminate lymphatic filariasis new diagnostic tools have emerged like PCR, antigen detection using finger-prick blood taken during the day and ultrasound to visualize adult worms. The last two can be applied in endemic countries with limited resources and enable the detection of early infections. As well as their value in control schemes, the latter is particularly important for the indivi

[Mosquitoes as vectors for exotic pathogens in Germany].

As a result of intensified globalization of international trade and of substantial travel activities, mosquito-borne exotic pathogens are becoming an increasing threat for Europe. In Germany some 50 different mosquito species are known, several of which have vector competence for pathogens. During the last few years a number of zoonotic arboviruses that are pathogenic for humans have been isolated from mosquitoes in Germany including Usutu, Sindbis and Batai viruses. In addition, filarial worms, such as Dirofilaria repens have been repeatedly detected in mosquitoes from the federal state of Br

Study of dirofilariasis in a selected area in the Western Province.

INTRODUCTION: Human dirofilariasis is a zoonotic infection caused by the filarial worm, Dirofilaria (Nochtiella) repens, whose primary host is the dog. This infection is on the increase over the past decade in Sri Lanka and the prevalence of canine dirofilariasis in the country is also believed to be high. We present here a study on public awareness of dirofilariasis and the prevalence of this infection in dogs in Negombo, an urban area that has a high domestic canine population. OBJECTIVE: To assess the awareness of dirofilariasis infection among residents and study the prevalence of this in

Microfilaremic infection in canine filariosis in Colombia: a challenge in morphological and molecular diagnostics.

Canine filariosis is caused by filiform nematodes and affects several species of animals as well as humans. The disease produces a wide range of symptoms that can often be confused with other diseases, which increases the complexity of its diagnosis. The search for methodologies to facilitate its diagnosis is a challenge, and specific and differential identification of the parasite species causing the disease holds key to a successful diagnosis. In Colombia, there is a problem of underdiagnosis of filariosis in microfilaremic dogs infected by Dirofilaria immitis and Acanthocheilonema reconditu

Serological detection of circulating Angiostrongylus vasorum antigen and specific antibodies in dogs from central and northern Italy.

The most frequently employed method for the diagnosis of Angiostrongylus vasorum in dogs is the detection of first stage larvae (L1) in faeces. The sensitivity of coproscopy, however, is limited in case of low parasite load, intermittent larval excretion, and during pre-patency. An epidemiological survey on dogs was conducted applying serological methods in two Italian regions where angiostrongylosis is endemic in foxes. 265 dog serum samples from Tuscany (central Italy - site A) and 447 from Liguria (north-western Italy - site B) were tested with a sandwich-ELISA for detection of circulating

Natural infection of filarioid nematodes in mosquitoes: An approach to neglected disease xenosurveillance and prevention in intercontinental human transit areas in Darién.

BACKGROUND: Filarioid nematodes are significant vector-borne parasites affecting both humans and animals. Despite their importance, their distribution, ecological dynamics, and health implications remain poorly characterized in the Neotropics. This knowledge gap is particularly critical in high-risk areas like the Darién, a vital migratory corridor connecting the diverse ecosystems of South and Central America, where unregulated migration intersects with complex ecological and social dynamics, creating optimal conditions for the emergence and spread of filarial infections.
