Oxidative damage and protection by antioxidants in the frontal cortex of Alzheimer's disease is related to the apolipoprotein E genotype.

A great number of epidemiological studies have demonstrated that the frequency of the epsilon4 allele of the apolipoprotein E gene (APOE) is markedly higher in sporadic and in familial late onset Alzheimer disease (AD). In the frontal cortex of AD patients, oxidative damage is elevated. We address the hypothesis that the APOE genotype and reactive oxygen-mediated damage are linked in the frontal cortex of AD patients. We have related the APOE genotype to the levels of lipid oxidation (LPO) and to the antioxidant status, in frontal cortex tissues from age-matched control and AD cases with diffe

Antibodies against p40tax gene product of human T-lymphotropic virus type-I (HTLV-I) under various conditions of HTLV-I infection.

We investigated antibodies against pX gene product, p40tax, by ELISA using recombinant p40tax protein in HTLV-I seropositive carriers as well as patients with adult T cell leukemia (ATL) and HTLV-I-associated myelopathy (HAM). Seventy (49.0%) out of 143 HTLV-I healthy carriers were found to be positive for antibody against p40tax antigen and the follow-up samples at two-year intervals revealed constant reactivity by ELISA in each carrier. The onset of antibody production was delayed 4 to 12 weeks as compared with anti-HTLV-I in primary infection cases. The anti-p40tax-positive rate (90%) in HA

Reduced Foxp3 protein expression is associated with inflammatory disease during human t lymphotropic virus type 1 Infection.

The Foxp3 protein is a specific marker of CD4(+)CD25(+) regulatory T (T(reg)) cells, and its expression is critical to their development and function. Several studies have demonstrated the dysregulation of Foxp3 expression during human inflammatory diseases. Infection with human T lymphotropic virus type 1 (HTLV-1) is associated with the development of a number of inflammatory conditions, including myelopathy, although the majority of individuals who are infected with HTLV-1 remain asymptomatic. To examine the role played by T(reg) cells in the development of inflammatory disease during HTLV-1

Analysis revealed the molecular mechanism of oxidative stress-autophagy-induced liver injury caused by high alkalinity: integrated whole hepatic transcriptome and metabolome.

INTRODUCTION: High-alkalinity water is a serious health hazard for fish and can cause oxidative stress and metabolic dysregulation in fish livers. However, the molecular mechanism of liver damage caused by high alkalinity in fish is unclear. METHODS: In this study, 180 carp were randomly divided into a control (C) group and a high-alkalinity (A25) group and were cultured for 56 days. High-alkalinity-induced liver injury was analysed using histopathological, whole-transcriptome, and metabolomic analyses. RESULTS: Many autophagic bodies and abundant mitochondrial membrane damage were observed

Regulation of autophagy factors by oxidative stress and cardiac enzymes imbalance during arsenic or/and copper induced cardiotoxicity in Gallus gallus.

Basal autophagy has an indispensable role in the functioning and maintenance of cardiac geometry under physiological conditions. Recently, increasing evidence has demonstrated that arsenic (As)/copper (Cu) play important roles in the autophagy of the heart. The current study was to evaluate whether oxidative damage by As or/and Cu was correlated with autophagy through the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (AKT)/mammalian target of rapamycin (mTOR) pathway in the heart of birds. Arsenic trioxide (30mg/kg) or/and cupric sulfate (300mg/kg) were administered in a basal diet to

Network pharmacology, single-cell transcriptomics, machine learning and experimental validation identifying CHEK2 and HPGD as key therapeutic targets of berberine in diabetic nephropathy.

Diabetic nephropathy (DN) is a severe microvascular complication of diabetes with limited therapeutic options. Berberine exhibits renoprotective potential, yet its precise molecular targets and mechanisms of action in DN remain unclear. Berberine targets were retrieved from TCMSP, SEA, and SwissTargetPrediction databases. Bulk transcriptomic microarray datasets (GSE96804, GSE30122) and single-cell RNA sequencing (scRNA-seq) dataset (GSE131882) were obtained from the Gene Expression Omnibus (GEO). Differentially expressed genes (DEGs) and weighted gene co-expression network analysis (WGCNA) wer

Erigeron breviscapus alleviates senescence via AMPK-SIRT1 signaling by modulating FOXO3a-mediated antioxidant defense and p53-dependent apoptosis.

Erigeron breviscapus Hand-Mazz. (EBHM) is a well-documented herbal medicine with recognized antioxidant, anti-apoptotic, and anti-inflammatory bioactivities. However, its anti-senescence effects and the underlying mechanisms remain poorly understood. Here, we investigated the anti-senescence potential of EBHM employing Caenorhabditis elegans and SAMP8 mice. In C. elegans, EBHM treatment prolonged the average lifespan of C. elegans to a maximum of 18.68%, while also significantly enhancing stress resistance and motor function. The lifespan-extending effects of EBHM were abolished in mutants of

Hypoxia increases susceptibility of grass carp to Aeromonas hydrophila infection by inducing oxidative stress and impairing antimicrobial defense.

Hypoxia is a major environmental stressor that threatens fish health in intensive aquaculture, particularly when it co-occurs with bacterial infection. To clarify how hypoxia influences host responses to Aeromonas hydrophila, we established a combined stress model in grass carp using hypoxia (2.0 mg L-1 dissolved oxygen) and bacterial challenge. Survival, hepatic bacterial load, histopathology, hematological indices, antioxidant and immune enzyme activities, and the expression of hypoxia- and immune-related genes were evaluated systematically. The median lethal dose (LD50) of A. hydrophila was

Itaconic Acid Alleviates Perfluorooctanoic Acid-Induced Oxidative Stress and Intestinal Damage by Regulating the Keap1/Nrf2/Ho-1 Pathway and Reshaping the Gut Microbiota.

Itaconic acid (IA) is recognized for its potential application in treating intestinal diseases owing to the anti-inflammatory and antioxidant properties. Perfluorooctanoic acid (PFOA) can accumulate in animals and result in oxidative and inflammatory damages to multi-tissue and organ, particularly in the intestinal tract. This study aimed to explore whether IA could mitigate intestinal damage induced by PFOA exposure in laying hens and elucidate its potential underlying mechanisms. The results showed that IA improved the antioxidant capacity of laying hens and alleviated the oxidative damage i

Tea tree oil inhibits hydrogen sulfide-induced oxidative damage in chicken lungs through CYP450s/ROS pathway.

A large amount of hydrogen sulfide (H2S) is produced in the process of chicken breeding, which can cause serious inflammation and oxidative damage to the respiratory system of chickens. Tea tree oil (TTO) has antioxidant and anti-inflammatory properties. No studies have been reported on the use of TTO in H2S-induced lung injury in chickens. Therefore, in this study, 240 one-day-old Roman pink laying hens were randomly and equally divided into 3 groups: control group (CON), H2S exposure group (AVG, containing H2S), and TTO treatment group (TTG, containing H2S and 0.02 mL/L TTO) to establish an

Paternal BPA and BPS exposure induce testicular dysfunction in pubertal male offspring: roles of OCTN2 carnitine transporter disruption and exacerbated oxidative stress/apoptosis.

Although numerous studies have reported the male reproductive toxicity of bisphenol A (BPA) and its substitute bisphenol S (BPS), the impacts of paternal exposure to these chemicals on the male reproductive system of offspring and the underlying molecular mechanism remain inadequately explored. In this study, we investigated male reproductive toxicity in pubertal offspring resulting from paternal exposure to environmentally relevant doses of BPA (0.45 μg/kg body weight [bw]/day) or BPS (0.15 μg/kg bw/day). Our results showed that paternal exposure to BPA or BPS reduced pubertal offspring testo

Effects of dietary supplementation with pumpkin powder on egg quality and antioxidant function in laying hens.

β-carotene, a carotenoid with strong antioxidant and immunity-enhancing properties, is a safe and widely available precursor of vitamin A in nature. Pumpkin powder, as a familiar agricultural product, is rich in vitamins, amino acids, and minerals, which also contain β-carotene. Currently, there remain few published reports on the effects of pumpkin powder on antioxidant function, reproductive performance, and egg quality in laying hens, representing a significant gap in existing research. 270 laying hens were divided into three groups; the CON group was fed a basal diet, the BCA group was fed

Clostridium butyricum Alleviates Enterotoxigenic Escherichia coli K88-Induced Oxidative Damage Through Regulating the p62-Keap1-Nrf2 Signaling Pathway and Remodeling the Cecal Microbial Community.

Clostridium butyricum (CB) can enhance antioxidant capacity and alleviate oxidative damage, but the molecular mechanism by which this occurs remains unclear. This study used enterotoxigenic Escherichia coli (ETEC) K88 as a pathogenic model, and the p62-Keap1-Nrf2 signaling pathway and intestinal microbiota as the starting point to explore the mechanism through which CB alleviates oxidative damage. After pretreatment with CB for 15 d, mice were challenged with ETEC K88 for 24 h. The results suggest that CB pretreatment can dramatically reduce crypt depth (CD) and significantly increase villus h
