Exposure to low concentrations of ethanol alters global gene expression patterns in Vibrio parahaemolyticus.

Vibrio parahaemolyticus is a foodborne pathogen commonly associated with seafood worldwide. Ethanol is widely used as a disinfectant and preservative in the food industry. Low concentrations of ethanol can inhibit the growth of V. parahaemolyticus and alter its protein expression profile and stress resistance. However, it remains unknown how ethanol exposure affects the behavior and gene expression of V. parahaemolyticus. In this study, RNA sequencing revealed differential expression of 1020 genes in response to 1.5 % (v/v) ethanol, including biofilm-associated genes, c-di-GMP metabolism-relat

Listeria monocytogenes virulence factors, including listeriolysin O, are secreted in biologically active extracellular vesicles.

Outer membrane vesicles produced by Gram-negative bacteria have been studied for half a century but the possibility that Gram-positive bacteria secrete extracellular vesicles (EVs) was not pursued until recently due to the assumption that the thick peptidoglycan cell wall would prevent their release to the environment. However, following their discovery in fungi, which also have cell walls, EVs have now been described for a variety of Gram-positive bacteria. EVs purified from Gram-positive bacteria are implicated in virulence, toxin release, and transference to host cells, eliciting immune res

Sensitive colorimetric detection of Listeria monocytogenes based on isothermal gene amplification and unmodified gold nanoparticles.

Listeria monocytogenes (L. monocytogenes), one of most problematic food-borne bacteria, is mainly transmitted through the food chain and may cause listeriosis. Therefore, the development of rapid and sensitive L. monocytogenes detection technique has become an urgent task. In this study, we proposed a method using hyperbranching rolling circle amplification (HRCA) combined with gold nanoparticle (GNP) based colorimetric strategy to offer an isothermal, highly sensitive and specific assay for the detection of L. monocytogenes. First, a linear padlock probe targeting a specific sequence in the h

HlyF Produced by Extraintestinal Pathogenic Escherichia coli Is a Virulence Factor That Regulates Outer Membrane Vesicle Biogenesis.

Escherichia coli can cause extraintestinal infections in humans and animals. The hlyF gene is epidemiologically associated with virulent strains of avian pathogenic E. coli and human neonatal meningitis-associated E. coli. We demonstrated that culture supernatants of E. coli expressing HlyF induced autophagy in eukaryotic cells. This phenotype coincided with an enhanced production of outer membrane vesicles (OMVs) by bacteria expressing HlyF. The HlyF protein displays a predicted catalytic domain of the short-chain dehydrogenase/reductase superfamily. This conserved domain was involved the abi

Differential pathogenic contributions of tdh/trh genes in Vibrio parahaemolyticus revealed by rabbit and mice model comparative study.

Vibrio parahaemolyticus is a major cause of foodborne diarrhea, with virulence mainly attributed to thermostable direct hemolysin (TDH) and TDH-related hemolysin (TRH). To identify an animal model that closely mimics human pathology, this study established New Zealand infant rabbit and mice infection models to systematically compare pathological and immunological responses. Key parameters evaluated included clinical signs, intestinal colonization, histopathological damage, and multi-organ function indicators. Results showed that the infant rabbit model exhibited classical infection symptoms, i

Dryocrassin ABBA Inhibits the Function of LLO and SortaseA to Alleviate the Virulence of Listeria monocytogenes.

Listeria monocytogenes is a deadly foodborne pathogen, its virulence factors Listeriolysin O (LLO) and Sortase A (SrtA) play a critical role in its infection and have been identified as key targets for developing its inhibitor. This study reveals that the natural compound Dryocrassin ABBA (ABBA) alleviates the toxicity of L. monocytogenes by effectively inhibiting the functions of LLO and SrtA. ABBA bound to the binding pocket of LLO and inhibited the formation of its oligomers, which in turn reduced the ability of LLO to lyse mammalian erythrocytes. When co-cultured with L. monocytogenes, ABB

Selective blockage of Serratia marcescens ShlA by nickel inhibits the pore-forming toxin-mediated phenotypes in eukaryotic cells.

Serratia marcescens is an opportunistic pathogen with increasing incidence in clinical settings. This is mainly attributed to the timely expression of a wide diversity of virulence factors and intrinsic and acquired resistance to antibiotics, including β-lactams, aminoglycosides, quinolones, and polypeptides. For these reasons, S. marcescens has been recently categorised by the World Health Organization as one priority to strengthen efforts directed to develop new antibacterial agents. Therefore, it becomes critical to understand the underlying mechanisms that allow Serratia to succeed within

Staphylococcus aureus α-toxin triggers the synthesis of B-cell lymphoma 3 by human platelets.

The frequency and severity of bacteremic infections has increased over the last decade and bacterial endovascular infections (i.e., sepsis or endocarditis) are associated with high morbidity and mortality. Bacteria or secreted bacterial products modulate platelet function and, as a result, affect platelet accumulation at sites of vascular infection and inflammation. However, whether bacterial products regulate synthetic events in platelets is not known. In the present study, we determined if prolonged contact with staphylococcal α-toxin signals platelets to synthesize B-cell lymphoma (Bcl-3),

Probing the pH- and Voltage-Driven Intramolecular Folding Dynamics of the c-MycC52 Promoter i-Motif at the Single-Molecule Level.

The human c-Myc proto-oncogene is a transcription factor that regulates protein expression and is controlled by GC-rich regulatory sequences capable of adopting non-canonical DNA secondary structures. Here, we use the α-hemolysin (α-HL) protein nanopore to probe pH- and voltage-dependent effects on the stability of the i-motif formed by the 52-nucleotide cytosine-rich c-Myc promoter (c-MycC52). Under acidic conditions, electrophoretically driven c-MycC52 i-motifs are transiently trapped within the α-HL vestibule, generating pH-dependent electrical current signatures that are volumetrically and

Comparative genomics of Sneathia species: insights into pathogenicity and adaptation to the human niche.

BACKGROUND: Sneathia are urogenital anaerobes linked to bacterial vaginosis, preterm birth, chorioamnionitis, urethritis, and HPV-mediated cervical carcinogenesis. Two species have been identified: Sneathia vaginalis and Sneathia sanguinegens. Yet, due to their fastidious nature, our knowledge of their pathogenicity is limited. METHODS: Comparative genomic analysis of 12 Sneathia genomes, including two newly sequenced via whole-genome sequencing, was used to evaluate the pathogenic potential of S. vaginalis and S. sanguinegens. RESULTS: Sneathia species have small genomes (1.25-1.35 Mbp) wit

Whole-genome characteristics, antimicrobial resistance and virulence of pathogenic Aeromonas veronii from diseased Macrobrachium rosenbergii.

Aeromonas veronii is an opportunistic pathogen capable of infecting various aquatic organisms. In this study, a pathogenic strain, designated MRG1, was isolated from diseased Macrobrachium rosenbergii and identified as A. veronii through morphological, biochemical, and molecular analyses, including whole-genome average nucleotide identity (ANI). Injection challenge trials indicated a 7-day median lethal dose (LD50) of A. veronii strain MRG1 for M. rosenbergii was 1.33 × 106 CFU/mL. Histological examination indicated that MRG1 infection caused pathological alterations, including deformation of

Brazilin reduces methicillin-resistant Staphylococcus aureus virulence and pathogenicity by decreasing the secretion of the α-hemolysin.

BACKGROUND: Methicillin-resistant Staphylococcus aureus (MRSA) is a super-resistant bacterium with strong pathogenicity, causing broad range of infections in various tissues. α-Hemolysin (Hla) is the main virulence factor of S. aureus. Brazilin (BN), is a homoisoflavonoid derivative, obtained from the wood of Caesalpinia echinata Lam (Brazil-wood), Caesalpinia sappan L (Leguminosae), and Caesalpinia violacea Standl, has been proven to exert excellent antibacterial and anti-virulence effects against S. aureus. However, the underlying mechanisms remain still unclear. OBJECTIVE: This study aims

Distinct Expression Levels of ALS, LIP, and SAP Genes in Candida tropicalis with Diverse Virulent Activities.

Candia tropicalis is an increasingly important human pathogen, causing nosocomial fungemia among patients with neutropenia or malignancy. However, limited research has been published concerning its pathogenicity. Based on the phenotypes of C. tropicalis in our previous study, we selected nine representative strains with different activities of virulence factors (adhesion, biofilm formation, secreted aspartic proteinases, and hemolysins), and one reference strain, ATCC750. The present study aimed to investigate the filamentation ability, the expression of virulence genes (ALST1-3, LIP1, LIP4, a

Intracellular and cell-to-cell spread of Listeria monocytogenes involves interaction with F-actin in the enterocytelike cell line Caco-2.

Listeria monocytogenes penetrates and multiplies within professional phagocytes and other cells such as the Caco-2 human enterocytelike cell line. Listeriolysin O, a membrane-damaging cytotoxin accounts for intracellular multiplication through lysis of the membrane-bound phagocytic vacuole. This work demonstrates that once released within the cytosol, L. monocytogenes acquires the capacity to spread intracellularly and infect adjacent cells by interacting with host cell microfilaments. Such evidence was obtained by using drugs which disrupt the cell cytoskeleton. Nocodazole, which blocks polym

[Infections of the respiratory tract with Pseudomonas aeruginosa in cystic fibrosis].

The main cause of death in cystic fibrosis (CF) patients is progressive pulmonary insufficiency frequently associated with chronic infections of the respiratory tract by Pseudomonas aeruginosa. Bacteria of this species synthesize numerous extracellular products contributing to its pathogenicity. An alginate-like exopolysaccharide is characteristic for mucoid mutants predominating among P. aeruginosa isolates from CF patients. It interferes with immune defense mechanisms of the host and probably protects the bacteria against certain antibiotics. Furthermore, it is involved in the formation of b

Uropathogenic Escherichia coli superinfection enhances the severity of mouse bladder infection.

Urinary tract infections (UTIs) afflict over 9 million women in America every year, often necessitating long-term prophylactic antibiotics. One risk factor for UTI is frequent sexual intercourse, which dramatically increases the risk of UTI. The mechanism behind this increased risk is unknown; however, bacteriuria increases immediately after sexual intercourse episodes, suggesting that physical manipulation introduces periurethral flora into the urinary tract. In this paper, we investigated whether superinfection (repeat introduction of bacteria) resulted in increased risk of severe UTI, manif

Large screening of CA-MRSA among Staphylococcus aureus colonizing healthy young children living in two areas (urban and rural) of Portugal.

BACKGROUND: The incidence of pediatric infections due to community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA), including children with no identifiable risk factors, has increased worldwide in the last decade. This suggests that healthy children may constitute a reservoir of MRSA in the community. In this study, nested within a larger one on nasopharyngeal ecology, we aimed to: (i) evaluate the prevalence of MRSA colonizing young children in Portugal; and (ii) compare results with those obtained in a study conducted a decade ago, when this prevalence was <0.5%. METHODS: I

[New knowledge of the molecular biology of enterohemorrhagic Escherichia coli (EHEC) O157].

Since 1982, enterohaemorrhagic Escherichia coli (EHEC) have been identified as a cause of diarrhoea and haemorrhagic colitis. The most serious complication of the infection is the haemolytic-uraemic syndrome (HUS) that develops in 5 to 10% of children with diarrhoea. Shiga toxins (Stx) are the most important presently known virulence factors of EHEC. After reaching the bloodstream, the toxins cause damage of endothelial cells but also of tubular cells in the kidneys which may result in renal failure. In EHEC O157 isolates from patients, we were able to identify seven different combinations of

Dynamics of urinary tract infections: a comprehensive study on antimicrobial susceptibility, virulence profiling and molecular epidemiology of uropathogenic Escherichia coli from Pakistan.

BACKGROUND: Urinary tract infections (UTIs), primarily caused by Escherichia coli, pose a global health challenge owing to rising antimicrobial resistance (AMR), particularly carbapenem-resistant (Carb-RT) and extended-spectrum beta-lactamase (ESBL)-producing uropathogenic E. coli (UPEC). This study analyzed AMR trends, virulence determinants, and molecular epidemiology of UPEC isolates from community and hospital settings in Pakistan. METHODS AND RESULTS: A total of 2700 urine samples were screened in this study. UPEC isolates underwent antimicrobial susceptibility testing, PCR screening for
