Astaxanthin ameliorates benzalkonium chloride-induced dry eye disease through suppressing inflammation and oxidative stress via Keap1-Nrf2/HO-1 signaling pathways.

BACKGROUND: Dry eye disease (DED) predominantly results from elevated tear film osmolarity, which can not only cause ocular inconvenience but may lead to visual impairments, severely compromising patient well-being and exerting substantial economic burdens as well. Astaxanthin (AST), a member of the xanthophylls and recognized for its robust abilities to combat inflammation and oxidation, is a common dietary supplement. Nonetheless, the precise molecular pathways through which AST influences DED are still poorly understood. METHODS: Therapeutic targets for AST were identified using data from

Preparation and characterization of ovomucin self-assembly nanoparticles under glycerol compression for astaxanthin delivery: Sustained release and antioxidant activity.

Astaxanthin (AST) is a natural hydrophobic nutrient with various biological activities, but its low solubility limits its application. In this study, self-assembly nanoparticles were prepared by ovomucin (OVM) and Ca2+ with the enhancement of glycerol to deliver AST. Glycerol compressed the particle size of nanoparticles from 175.7 ± 1.8 to 142.9 ± 0.6 nm, and the nanoparticles had a strong negative charge (-28.9 ± 0.6 mV). Ultraviolet-visible spectroscopy and X-ray diffraction (XRD) confirmed the successful encapsulation of AST in an amorphous form with a high encapsulation efficiency (82.9%

Protective effects of astaxanthin solid lipid nanoparticle as a promising candidate against acute kidney injury in rats.

Acute kidney injury (AKI) is a sudden onset of renal injury that occurs within a few hours or days. Ischemia-reperfusion (IR) is a major cause of AKI. There are multiple dysregulated mechanisms behind the pathogenesis of AKI and IR which urges the need for finding multi-targeting therapies. Natural products are multi-targeting agents with promising sources of anti-inflammation, antioxidant, and antiapoptosis. Among them, astaxanthin (AST) is a keto-carotenoid with a high antioxidant potential. Using solid lipid nanoparticles (SLNs) as a novel formulation of AST helps to increase its efficacy a

Protective effect of astaxanthin against SnS2 nanoflowers induced testes toxicity by suppressing RIPK1-RIPK3-MLKL signaling in mice.

The reproductive toxicity of SnS2 nanoflowers (SnS2 NFs) has been studied in our previous experiment, but the underlying mechanism is still not clear. Astaxanthin (ASX) is a red carotenoid pigment with antioxidant, anticancer and anti-inflammatory properties, showing neuroprotective properties via its antioxidant capacity. To examine the ASX effect on sub-chronic testis injury induced by SnS2 NFs, we randomly and equally divided 40 Kunming male mice into four groups (control, ASX control, NF and NF + ASX groups). Then, ASX dissolved in olive oil was administered intragastrically for 30 consecu

Free radical formation in novel carotenoid metal ion complexes of astaxanthin.

The carotenoid astaxanthin forms novel metal ion complexes with Ca(2+), Zn(2+), and Fe(2+). MS and NMR measurements indicate that the two oxygen atoms on the terminal cyclohexene ring of astaxanthin chelate the metal to form 1:1 complexes with Ca(2+) and Zn(2+) at low salt concentrations <0.2 mM. The stability constants of these complexes increased by a factor of 85 upon changing the solvent from acetonitrile to ethanol for Ca(2+) and by a factor of 7 for Zn(2+) as a consequence of acetonitrile being a part of the complex. Optical studies showed that at high concentrations (>0.2 mM) of salt, 2

Astaxanthin Supplementation Lowers Respiratory Exchange Ratio During Submaximal Cycling in Females: A Randomized, Double-Blind, Placebo-Controlled Crossover Trial.

Astaxanthin (AX) is a lipid-soluble carotenoid that has produced mixed results in increasing fat oxidation during submaximal exercise in male cohorts. Females exhibit a higher reliance on fat oxidation, potentially making them a demographic more likely to benefit from AX supplementation. Following an initial VO2max test to determine maximum power (Wmax), twenty recreationally active females (age: 22 ± 2 y) participated in a two-arm double-blind, counterbalanced, crossover design study with a 2-week supplementation period of either AX (12 mg·day-1) or placebo (PLA) separated by a 2-week washout

The dual action of astaxanthin: activating the Nrf2 pathway and suppressing NF-κB to combat AFB1-induced liver injury in broilers.

Aflatoxin B1 (AFB1), a mycotoxin that primarily targets the liver, poses a serious threat to the poultry industry. Astaxanthin (AST), a potent natural antioxidant, has been shown in our previous research to alleviate the toxic effects of AFB1 on chick embryo liver cells (CELs). This study advances our prior observation by providing the first in vivo evidence to investigate tissue damage caused by AFB1 in broilers and the protective effect and mechanism of AST, and compared its efficacy with that of biphenyl dimethyl dicarboxylate (DDB), a clinical hepatoprotective drug. The broilers were divid

Protective Effects of Astaxanthin on PFOS-Induced Liver Injury via Fatty Acid and Cholesterol Metabolic Pathways.

Perfluorooctanesulfonate (PFOS) is a persistent bioaccumulative pollutant that can induce significant liver injury and functional impairment. Astaxanthin (ASX) is a naturally occurring carotenoid with potent bioactive properties and hepatoprotective effects. However, whether ASX can mitigate PFOS-induced liver injury and its underlying molecular mechanisms remain unclear. The results indicate that ASX supplementation significantly alleviatess PFOS-induced liver injury. Transcriptomic analysis suggests that lipid metabolic disorder may be a primary driver of PFOS-induced liver damage, and ASX m

Redox-sensitive miRNAs and Humanin could mediate effects of exercise and astaxanthin on oxidative stress and inflammation in type 2 diabetes.

Type 2 diabetes mellitus (T2DM) is driven by oxidative stress (OS) and inflammation (IF), accelerating disease progression. This study examined whether combined aerobic and resistance training (CT) and astaxanthin (AST) supplementation synergistically improve oxidant and inflammatory status as well as metabolic indices in T2DM, focusing on the mediatory role of Humanin (HN) and microRNAs (miRNA-122, miRNA-126-3p, and miRNA-146a). Ninety women with T2DM were randomly assigned to six groups (n = 15 each): control (C), placebo (P), AST supplementation (S), combined training (CT), CT + placebo (CT
