Fetal alcohol exposure blocks full masculinization of the dorsolateral nucleus in rat spinal cord.

Male rats prenatally exposed to a combination of stress and ethanol show severely impaired ejaculatory patterns. This study examined two sexually dimorphic nuclei in the lumbar spinal cord implicated in the control of male copulatory reflexes in rats whose mothers were exposed to alcohol, to stress, or to both treatments during pregnancy. Alcohol exposure led to a marked decrease (22%) in the number of motor neurons in the dorsolateral nucleus (DLN) of the adult male offspring, but no significant change in cell count was detectable in the sexually dimorphic nucleus of the bulbocavernosus (SNB)

Nuclear membrane disruption in neurodegenerative diseases: Emerging perspectives.

The nucleus, as the largest organelle within the cell, serves as the central hub for storing, replicating, and transcribing genetic information, thereby orchestrating vital cellular processes. In eukaryotic cells, the nuclear membrane is composed of several structural components: the outer and inner nuclear membranes, the nuclear pore complexes, and the underlying nuclear lamina, which together preserve the stability of the intracellular environment. Neurodegenerative disorders, such as Alzheimer's disease and related dementias, are characterized by the gradual degeneration and loss of neurona

[Immunocytochemical study of CuZn superoxide dismutase in the myocardium of normal subjects and patients of rheumatic heart disease].

Using the methods of immunocytochemistry and immune electron microscopy, we studied the subcellular localization of GuZn superoxide dismutase (CuZn SOD) in the myocardium of 8 normal subjects and the changes of CuZn SOD of the myocardium in 51 patients of rheumatic heart disease, and at same time, we also determined the CuZn SOD contents in the myocardium with radioimmunoassay. This study demonstrated that most normal myocardial cells contained CuZn SOD. The ventricular myocardial cells stained somewhat stronger than the atrial myocardial cells. CuZn SOD was mainly distributed in the cytoplasm

Different cortical and subcortical astroglial responsiveness in rats with acute liver failure.

Hepatic encephalopathy (HE) is a neuropsychiatric complication of liver failure. Previous studies described astroglia alterations in HE, but regional changes have not been well investigated. This study addresses regional astroglial response by exploring glial fibrillary acidic protein (GFAP) immunoreactivity in cortical structures including somatosensory (S1Tr and S1BF), piriform (Pir), and perirhinal (PRh) cortices, and subcortical regions including corpus callosum (CC), ventromedial thalamus (VMT), mammillothalamic tract (MTT), and dorsomedial hypothalamic nucleus (DHN) in rats with acute li

Macromolecular Crowding in Cytoplasm and Cellular Organelles.

A high concentration of diverse macromolecules causes a densely packed environment, influencing both function and structural change of the cell organelles, cytoplasm, and molecules within. The concept of molecular crowding was for the first time introduced in the 70 s but was only accepted at the end of the 90 s. Crowding demonstrates how the high concentration of macromolecules, such as proteins, nucleic acids, and polysaccharides, influences the behavior and interactions of these molecules. This systematic review explored recent literature that impacted the understanding of molecular crowdin

The spatial transcriptome of the late-stage embryonic and postnatal mouse brain reveals spatiotemporal molecular markers.

The neocortical development process includes cell proliferation, differentiation, migration, and maturation, supported by precise genetic regulation. To understand these processes at the cellular and molecular levels, it is necessary to characterize the fundamental anatomical structures by gene expression. However, markers established in the adult brain sometimes behave differently in the fetal brain, actively changing during development. The spatial transcriptome is a powerful analytical method that enables sequence analysis while retaining spatial information. However, a deeper understanding

Localization of the main noradrenergic neuron groups in the pons and medulla of the rabbit and the importance of cathodal lesions for prolonged survival.

Methods for stereotaxically localizing the major noradrenergic (NA) cell groups (i.e. A1, A2, A5 and A6 + A7) in the rabbit are described. Using a modified Kopf head holder we used surface landmarks including the obex for making lesions of the A1 and A2 cells in the medulla. Localization of the pontine cell groups was done by mapping intracerebral structures including the facial nerve for A5 and the motor nucleus of the trigeminal nerve for A6 + A7. In the initial experiments we made A1 lesions by passing anodal currents through stainless steel electrodes, which was associated with pulmonary o

Effects of iron-, manganese-, or magnesium-deficiency on the growth and morphology of Euglena gracilis.

Iron-, manganese-, or magnesium-deficiency has been induced in Euglena gracilis. Each arrests cell proliferation, decreases the intracellular content of the deficient metal, and increases that of several other metals. Light and electron microscopy of stationary phase cells reveal that Fe-deficient (-Fe) cells are similar in size and shape to control organisms. Magnesium-deficient (-Mg) cells, however, are larger, and approximately 14% are multilobed, containing 2 to 12 lobes of equal size emanating from a central region. Individual (-Mg) cells and each lobe of multilobed cells contain a single

Comparison of Signals from Cerebellar Purkinje Cells and Deep Nuclei during Temporal Prediction in Primates.

The cerebellum is crucial for predictive motor control and learning, but growing evidence suggests that it also generates forward models for nonmotor functions. Previous studies have reported periodic neuronal activity in the cerebellar dentate nucleus when animals predicted the timing of regularly presented visual stimuli. Since the dentate nucleus is an output structure, this activity likely reflects computations in the cerebellar cortex. Considering that Purkinje cells (PCs), the sole output of the cerebellar cortex, are more numerous than cerebellar nuclear neurons and that nuclear neurons

Plant plastids: from evolutionary origins to functional specialization and organelle interactions.

Plastids are highly diverse organelles that play critical roles in supporting many forms of life on Earth. Among them, chloroplasts house the machinery for photosynthesis, providing phototrophic capabilities to eukaryotes such as plants, algae, and photosynthetic protists. The functions of plastids are indispensable for the survival and development of life, and they are widely recognized as endosymbiotic organelles with a single origin. They exhibit morphological diversity, tissue specificity, and the ability to adapt to specific cellular functions. Despite this level of understanding, signifi

Histological and Molecular Characterization of the Inferior Olivary Nucleus and Climbing Fibers in the Goldfish, Carassius auratus.

The cerebellum receives inputs via the climbing fibers originating from the inferior olivary nucleus in the ventral medulla. In mammals, the climbing fibers entwine and terminate onto both major and peripheral branches of dendrites of the Purkinje cells. In this study, the inferior olivary nucleus and climbing fiber in the goldfish were investigated with several histological techniques. By neural tracer application to the hemisphere of the cerebellum, labeled inferior olivary neurons were found in the ventral edge of the contralateral medulla. Kainate stimulated Co + + uptake and gephyrin immu

Thyroid hormone nuclear receptor levels are influenced by the acetylation of chromatin-associated proteins.

The thyroid hormone receptor is a chromatin-associated protein which appears to mediate the actions of the thyroid hormones in mammalian cells. Unlike steroid hormone receptors, a cytoplasmic form of the receptor has not been identified, and the factors which govern the nuclear concentrations of the receptor are poorly understood. Using cultured GH1 cells, a rat pituitary cell line, we having previously demonstrated that thyroid hormones reduces the concentration of its receptor by a mechanism which involves the association of the ligand with the receptor binding site (Samuels, H.H., Stanley,

[Pain and emotional dysregulation: Cellular memory due to pain].

Genetic factors are involved in determinants for the risk of psychiatric disorders, and neurological and neurodegenerative diseases. Chronic pain stimuli and intense pain have effects at a cellular and/or gene expression level, and will eventually induce "cellular memory due to pain", which means that tissue damage, even if only transient, can elicit epigenetically abnormal transcription/translation and post-translational modification in related cells depending on the degree or kind of injury or associated conditions. Such cell memory/transformation due to pain can cause an abnormality in a fu

Nuclear phosphoinositides and their roles in cell biology and disease.

Since the late 1980s, a growing body of evidence has documented that phosphoinositides and their metabolizing enzymes, which regulate a large variety of cellular functions both in the cytoplasm and at the plasma membrane, are present also within the nucleus, where they are involved in processes such as cell proliferation, differentiation, and survival. Remarkably, nuclear phosphoinositide metabolism operates independently from that present elsewhere in the cell. Although nuclear phosphoinositides generate second messengers such as diacylglycerol and inositol 1,4,5 trisphosphate, it is becoming

Lenacapavir-induced Lattice Hyperstabilization is Central to HIV-1 Capsid Failure at the Nuclear Pore Complex and in the Cytoplasm.

Lenacapavir (LEN) is the first HIV-1 capsid inhibitor approved for clinical use. It inhibits multiple steps of the viral life cycle; however, the molecular details of the effect of LEN on capsid structure and the mechanistic steps of the inhibition are not understood. Recent studies show that intact cone-shaped capsids and capsids with LEN-induced breaks can dock at nuclear pore complexes (NPC), but only intact capsids enter the nucleus. In this work, we combined large-scale coarse-grained molecular dynamics simulations and live-cell imaging to investigate the stepwise mechanism of docking of

The dengue vector Aedes aegypti contains a functional high mobility group box 1 (HMGB1) protein with a unique regulatory C-terminus.

The mosquito Aedes aegypti can spread the dengue, chikungunya and yellow fever viruses. Thus, the search for key molecules involved in the mosquito survival represents today a promising vector control strategy. High Mobility Group Box (HMGB) proteins are essential nuclear factors that maintain the high-order structure of chromatin, keeping eukaryotic cells viable. Outside the nucleus, secreted HMGB proteins could alert the innate immune system to foreign antigens and trigger the initiation of host defenses. In this work, we cloned and functionally characterized the HMGB1 protein from Aedes aeg

An autoradiographic study of the efferent connections of the lateral hypothalamic area in the rat.

The efferent connections of the lateral hypothalamic area (LHA) have been analyzed in a series of 30 rat brains with injections of 3H-amino acids into different parts of the area and the surrounding regions. Our findings indicate that all parts of the LHA contribute ascending and descending fibers to the medial forebrain bundle, and also project medially to certain of the adjoining hypothalamic nuclei. All levels of the LHA appear to send some fibers to a continuous group of structures that extends from the medial septal-diagonal band complex rostrally, through the lateral preoptic and lateral

Alternative control: what's WASp doing in the nucleus?

Wiskott-Aldrich syndrome (WAS) is a rare X-linked recessive immunodeficiency disorder of childhood that is caused by mutations in the WAS gene. WAS encodes WASp, a protein that is known to function in the cytoplasm of hematopoietic cells and is required for the induced differentiation of CD4+ T helper type 1 (TH1) lymphocytes. Now, a paper in Science Translational Medicine describes another mechanism for impaired immunity in WAS by showing that WASp localizes in the nucleus and regulates histone modifications and chromatin structure, thereby modulating expression of the TH1 master gene TBX21 (

Determinants of nerve cell patterns during development: a review.

The aim of the work reviewed is to define some of the mechanisms which are implicated in the control of neural cell pattern formation in the developing central nervous system. This question was examined by studies of brain embryonic development in normal and reeler mutant mice, which are characterized by profuse architectonic anomalies. The adult reeler phenotype is characterized by extreme abnormalities of cell positioning in the telencephalic and cerebellar cortices as well as by distinct architectonic anomalies in non-cortical structures such as the inferior olive, the facial nerve nucleus
