New insights on β-glycan synthases using in vitro GT-array (i-GT-ray) platform.

Cellulose and hemicellulose are the major structural β-glycan polysaccharides in cell walls of land plants. They are characterized by a backbone of β-(1,3)- and/or β-(1,4)-linked sugars such as glucose, mannose, or xylose. The backbones of these polymers are produced by processive glycosyltransferases (GTs) called synthases having multiple transmembrane domains anchoring them to the membrane. Thus, they are among the most difficult membrane proteins to test in vitro and to purify. Recently, we developed an in vitro GT-array (i-GTray) platform and showed that non-processive type II membrane GTs

Eco-friendly, stable, and high-performance biochar prepared by a twice-modification scheme: Saccharification of raw materials & thermal air oxidation of biochar.

Organic pollutants, such as phenolic compounds, pose significant risks to both the environment and human health. While biochar is an effective adsorbent for removing these pollutants, its dissolved solid (DS) components can lead to the loss of functional groups, structural disintegration, unstable performance, and environmental issues. This study introduces a twice-modification scheme designed to produce a biochar (BC-M) that combines high stability with superior performance. The process begins with the preparation of a stable biochar from cellulase-treated lignocellulose. This precursor bioch

Using A Protoplast Transformation System to Enable Functional Studies in Mangifera indica L.

Mangoes (Mangifera indica L.) are an important kind of perennial fruit tree, but their biochemical testing method and transformation technology were insufficient and had not been rigorously explored. The protoplast technology is an excellent method for creating a rapid and effective tool for transient expression and transformation assays, particularly in plants that lack an Agrobacterium-mediated plant transformation system. This study optimized the conditions of the protoplast isolation and transformation system, which can provide a lot of help in the gene expression regulation study of mango

Evaluation of Reference Genes in Glenea cantor (Fabricius) by Using qRT-PCR.

Kapok is the main host of Glenea cantor (Fabricius), which causes serious damage and is difficult to control. In severe cases, it often causes the kapok trees to die continuously, which seriously affects the results of urban landscaping. To provide reference for the functional research on related genes in G. cantor, we screened the stable expression of candidate reference genes at different developmental stages (i.e., eggs, larvae, pupae, and adults), in various adult tissues (i.e., head, thorax, abdomen, feet, antennae, and wings), and sexes (i.e., male pupae, female pupae, male adults, and f

Fungal traits that drive ecosystem dynamics on land.

Fungi contribute extensively to a wide range of ecosystem processes, including decomposition of organic carbon, deposition of recalcitrant carbon, and transformations of nitrogen and phosphorus. In this review, we discuss the current knowledge about physiological and morphological traits of fungi that directly influence these processes, and we describe the functional genes that encode these traits. In addition, we synthesize information from 157 whole fungal genomes in order to determine relationships among selected functional genes within fungal taxa. Ecosystem-related traits varied most at r

Identification of a global repressor gene, rsmA, of Erwinia carotovora subsp. carotovora that controls extracellular enzymes, N-(3-oxohexanoyl)-L-homoserine lactone, and pathogenicity in soft-rotting Erwinia spp.

The production of extracellular enzymes such as pectate lyase (Pel), polygalacturonase (Peh), cellulase (Cel), and protease (Prt) is activated by the cell density (quorum)-sensing signal, N-(3-oxohexanoyl)-L-homoserine lactone (HSL); plant signals; and aep genes during postexponential growth of Erwinia carotovora subsp. carotovora 71. Studies with mutants of E. carotovora subsp. carotovora 71 derepressed in exoenzyme production led to the identification of a negative regulator gene, rsmA (rsm, repressor of secondary metabolites). Modulation of Maillard reaction pathways and aroma profiles in peanut oil using enzymatically tailored peanut meal hydrolysates.

This study establishes a mechanistic framework for directing Maillard-driven aroma formation in peanut oil through targeted enzymatic precursor engineering, exploring the potential of precursor engineering for flavor modulation in peanut oil. Defatted peanut meal was subjected to selective hydrolysis using distinct glycosidases (cellulase, hemicellulase, pectinase, α-amylase, and amyloglucosidase) in combination with proteases to systematically reconstruct the precursor microenvironment (reducing sugars and free amino acids). The resulting hydrolysates were thermally reacted with cold-pressed

Establishment of a CRISPR/Cas9-mediated system for targeted editing of the MFS gene in mint.

The key message of this study is that we established a CRISPR/Cas9-mediated genome-editing system for Mentha haplocalyx "738" by optimizing protoplast transient assay and screening effective regulatory elements. Targeted knockout of the MFS gene generated edited mint plants with reduced menthofuran content, offering a strategy for quality improvement of mint essential oil. The commercial value of mint (Mentha spp.) essential oil is often diminished by the presence of undesirable metabolites, notably menthofuran, which impairs flavor and raises safety concerns. This study aimed to develop a rob

Diverse thermophilic anaerobes from Indian hot springs show potential for bioenergy production from lignocellulosic biomass.

Thermophilic anaerobic bacteria degrade lignocellulosic biomass and produce biofuels under high-temperature, oxygen-limited conditions, yet their cultivable diversity and functional potential in Indian hot springs remain underexplored. Water and sediment samples were collected from ten geographically distinct Indian hot springs spanning a temperature range of 25-85 °C, and the sites were characterised using standard physicochemical measurements. Of the four substrates tested, cellulose, xylan, and neutral detergent fibre supported notable hydrogen-producing enrichments at 55-85 °C, while ligni
