Effect of itch, scratching and mental stress on autonomic nervous system function in atopic dermatitis.

Atopic dermatitis is a stress-responsive disorder that involves the autonomic nervous system. The current study used heart rate variability to examine the effect of itch, scratching and mental stress in atopic patients with moderate to severe disease. Twenty-one patients with active disease and 24 healthy volunteers participated in the study. Heart rate variability measurements were taken at 5 min intervals at rest and after each of 3 acute stress tests, which included histamine-induced itch at the forearm, scratching around the itch site, and the Trier Social Stress Test. Atopic patients disp

Deferoxamine-B-Functionalized Silicon Nanowire Field-Effect Transistor Interfaces for Ultralow-Concentration Fe3+ Detection in Semiconductor Process Chemicals.

Maintaining ultralow metal-ion purity in process chemicals is crucial in semiconductor fabrication, where trace Fe3+ contamination can lead to dielectric breakdown, yield loss, and unintended doping. Inductively coupled plasma mass spectrometry (ICP-MS) provides excellent qualitative and quantitative sensitivity; however, it requires specialized infrastructure, is time-consuming and costly, and is not well suited for rapid on-site screening. Herein, we present a deferoxamine-B (DFO-B)-functionalized silicon nanowire field-effect transistor (SiNW-FET) engineered for the ultrasensitive, label-fr

Diet-induced pre-diabetes slows cardiac conductance and promotes arrhythmogenesis.

BACKGROUND: Type 2 diabetes is associated with abnormal electrical conduction and sudden cardiac death, but the pathogenic mechanism remains unknown. This study describes electrophysiological alterations in a diet-induced pre-diabetic rat model and examines the underlying mechanism. METHODS: Sprague-Dawley rats were fed either high-fat diet and fructose water or normal chow and water for 6 weeks. The electrophysiological properties of the whole heart was analyzed by in vivo surface ECG recordings, as wells as ex vivo in Langendorff perfused hearts during baseline, ischemia and reperfussion. A sensitive DNA capacitive biosensor using interdigitated electrodes.

This paper presents a label-free affinity-based capacitive biosensor using interdigitated electrodes. Using an optimized process of DNA probe preparation to minimize the effect of contaminants in commercial thiolated DNA probe, the electrode surface was functionalized with the 24-nucleotide DNA probes based on the West Nile virus sequence (Kunjin strain). The biosensor has the ability to detect complementary DNA fragments with a detection limit down to 20 DNA target molecules (1.5aM range), making it suitable for a practical point-of-care (POC) platform for low target count clinical applicatio

Proton-gated channels in PC12 cells.

Acid-sensing ion channels (ASICs) are expressed in various sensory and central neurons. The functional role of these channels remains elusive. Complex subunit combinations and lack of specific blockers for native receptors are likely to contribute to the difficulty of resolving the function of ASICs. Finding a neuronal cell line, which expresses a single population of ASICs, should prove to be useful in delineating the function of individual ASICs. Using patch-clamp, Ca(2+)-imaging, and RT-PCR techniques, we have explored the existence of ASICs in PC12 cells, a clonal neuronal cell line. Fast

Intelligent soft robotic gripper for non-destructive grasping and attribute recognition via multi-modal waveguide tactile sensors.

The intelligent soft robotic gripper integrated with tactile sensors significantly enhances the robot's execution capabilities in complex tasks, resolving critical shortcomings of traditional mechanical grippers-namely, fragile item breakage from rigid impacts, irregular object slippage, and inefficiency due to recognition errors. While electrical sensors (e.g., piezoresistive, capacitive) struggle with structural complexity, signal crosstalk, and environmental interference, optical waveguide tactile sensing offers superior sensitivity, rapid dynamics, and electromagnetic immunity. However, ex

A Multicenter Single-Arm Objective Performance Criteria Trial to Determine the Efficacy and Safety of High-Frequency Irreversible Electroporation as Primary Treatment for Localized Prostate Cancer: A Study Protocol.

INTRODUCTION: The classical pathway for the therapy of low- to intermediate-risk localized prostate cancer is radical prostatectomy or radiation therapy, which has shown a high incidence of complications, including erectile dysfunction, urinary incontinence, and bowel injury. An alternative pathway is to perform an ablation by some energy to the localized lesion, known as focal therapy. High-frequency irreversible electroporation (H-FIRE) is nonthermal energy that can be used in cancer ablation to deliver pulsed high-voltage but low-energy electric current to the cell membrane and to invoke ce

μ-Opioid inhibition of Ca2+ currents and secretion in isolated terminals of the neurohypophysis occurs via ryanodine-sensitive Ca2+ stores.

μ-Opioid agonists have no effect on calcium currents (I(Ca)) in neurohypophysial terminals when recorded using the classic whole-cell patch-clamp configuration. However, μ-opioid receptor (MOR)-mediated inhibition of I(Ca) is reliably demonstrated using the perforated-patch configuration. This suggests that the MOR-signaling pathway is sensitive to intraterminal dialysis and is therefore mediated by a readily diffusible second messenger. Using the perforated patch-clamp technique and ratio-calcium-imaging methods, we describe a diffusible second messenger pathway stimulated by the MOR that inh

Voltage-dependent and odorant-regulated currents in isolated olfactory receptor neurons of the channel catfish.

The electrical properties of olfactory receptor neurons, enzymatically dissociated from the channel catfish (Ictalurus punctatus), were studied using the whole-cell patch-clamp technique. Six voltage-dependent ionic currents were isolated. Transient inward currents (0.1-1.7 nA) were observed in response to depolarizing voltage steps from a holding potential of -80 mV in all neurons examined. They activated between -70 and -50 mV and were blocked by addition of 1 microM tetrodotoxin (TTX) to the bath or by replacing Na+ in the bath with N-methyl-D-glucamine and were classified as Na+ currents.

Inhibition of calcineurin phosphatase promotes exocytosis of renin from juxtaglomerular cells.

To examine the role of the calcium/calmodulin-dependent phosphatase calcineurin in regulation of renin release, we assayed exocytosis using whole-cell patch clamp of single juxtaglomerular cells in culture. The calcineurin inhibitor, cyclosporine A (CsA), significantly increased juxtaglomerular cell membrane capacitance, an index of cell surface area and an established measure of exocytosis in single-cell assays. This effect was mimicked by intracellular delivery of a calcineurin inhibitory peptide, the calcium chelator ethylene glycol tetraacetic acid (EGTA), or the calmodulin inhibitor W-13.

Beneficial effect of a moisturizing cream as adjunctive treatment to oral isotretinoin or topical tretinoin in the management of acne.

Despite their beneficial effects on the treatment of acne vulgaris, topical and oral retinoids may cause severe local irritation (retinoid dermatitis) due to their mechanism of action, thereby jeopardizing patient adherence, and thus compromising treatment efficacy. Alleviating dryness and improving skin comfort by using a moisturizer concomitantly to retinoids could enhance efficacy. In the present study, 30 subjects receiving either oral isotretinoin for at least 2 months or topical tretinoin for at least one month applied a moisturizing cream (Cetaphil Moisturizing Cream) twice daily for 15

Role of CuZn superoxide dismutase on carotid body function in heart failure rabbits.

AIMS: Peripheral chemoreflex sensitivity is potentiated in both clinical and experimental chronic heart failure (CHF). NADPH oxidase-derived superoxide mediates angiotensin II (Ang II)-enhanced carotid body (CB) chemoreceptor sensitivity in CHF rabbits, and tempol, the superoxide dismutase (SOD) mimetic, inhibits this Ang II- and CHF-enhanced superoxide anion effect. Here we investigated the role of cytoplasmic SOD [CuZn superoxide dismutase (CuZnSOD)] in the CB on chemoreceptor activity and function in CHF rabbits. METHODS AND RESULTS: CuZnSOD protein expression was decreased in CBs from CHF

Bioelectronic block of stellate ganglia mitigates pacing-induced heterogeneous release of catecholamine and neuropeptide Y in the infarcted pig heart.

The sympathetic nervous system modulates cardiac contractile and electrophysiological function and contributes to adverse remodelling following myocardial infarction (MI). Axonal modulation therapy (AMT), directed at the sympathetic chain, blocks efferent sympathetic outflow to the heart and is a strategy to transiently and controllably mitigate chronic MI-associated sympatho-excitation. In porcine models, we evaluated scalable AMT, directed at the paravertebral chain, in blocking reflex-mediated pacing-induced sympatho-excitation post-MI. The level of sympatho-excitation was assessed by dynam

Bioelectronic modulation of the thymic "genetic mirror": 448 kHz radiofrequency stimulation as a novel strategy for immune tolerance induction in type 1 diabetes.

BACKGROUND: Type 1 Diabetes Mellitus (T1DM) arises from a fundamental breakdown of central immune tolerance within the thymus. Medullary thymic epithelial cells (mTECs) express the Autoimmune Regulator (AIRE) protein, which drives the promiscuous transcription of peripheral tissue-restricted antigens - including insulin - thereby creating a "genetic mirror" essential for the clonal deletion of autoreactive T-cells. Genetic susceptibility loci that reduce intrathymic insulin expression allow high-affinity insulin-reactive T-cells to escape negative selection and enter the periphery, where they

Design of multicore-shell structured Co3O4-NiO nanocomposites as high-performance cathodes for zinc-ion hybrid supercapacitors.

Mixed-metal oxides (MMOs) hold great promise as cathodes for zinc-ion hybrid supercapacitors (Zn-HSCs); however, the lack of comprehensive insight into how their compositional tuning affects the electrochemical performance hinders the systematic design of MMO-based high-performance materials. To examine the impact of MMOs' composition on the performance of Zn-HSCs, a series of cathode materials were selected, including carbon (acetylene black), Co3O4, NiO, and Co3O4-NiO nanocomposites with different Co2+ : Ni2+ molar ratios (Co-Ni(x : y), where x : y = 1 : 0.1, 1 : 0.2, 1 : 0.3, 1 : 0.5, and 1

A Novel Design of a Portable Birdcage via Meander Line Antenna (MLA) to Lower Beta Amyloid (Aβ) in Alzheimer's Disease.

Late Onset Alzheimer's Disease (LOAD) is the most common cause of dementia, characterized by the deposition of plaques primarily of neurotoxic amyloid-[Formula: see text] ([Formula: see text]) peptide and tau protein. Our objective is to develop a noninvasive therapy to decrease the toxic A[Formula: see text] levels, using repeated electromagnetic field stimulation (REMFS) in the brain of Alzheimer's disease patients. We previously examined the effects of REMFS on [Formula: see text] levels in primary human brain (PHB) cultures at different frequencies, powers, and specific absorption rates (S

Chronic enalapril treatment increases transient outward potassium current in cardiomyocytes isolated from right ventricle of spontaneously hypertensive rats.

It has been well established that chronic pressure overload resulting from hypertension leads to ventricular hypertrophy and electrophysiological remodeling. The transient outward potassium current (I to) reduction described in hypertensive animals delays ventricular repolarization, leading to complex ventricular arrhythmias and sudden death. Antihypertensive drugs, as angiotensin-converting enzyme inhibitors (ACEi), can restore I to and reduce the incidence of arrhythmic events. The purpose of this study was to evaluate the differential effects of long-term treatment with ACEi or direct-actin

The spatial pattern of cochlear amplification.

Sensorineural hearing loss, which stems primarily from the failure of mechanosensory hair cells, changes the traveling waves that transmit acoustic signals along the cochlea. However, the connection between cochlear mechanics and the amplificatory function of hair cells remains unclear. Using an optical technique that permits the targeted inactivation of prestin, a protein of outer hair cells that generates forces on the basilar membrane, we demonstrate that these forces interact locally with cochlear traveling waves to achieve enormous mechanical amplification. By perturbing amplification in
