Precision Visualization of Tumor Microenvironment via a Lysosome-Anchored Molecular Rotor with Viscosity-Triggered Fluorescence Ignition.

Intracellular viscosity is a key biomarker of the tumor microenvironment. However, most existing probes lacked tumor-targeting capabilities, limiting the accuracy of tumor detection and imaging analysis. Herein, we present a new fluorescent probe (NM-Lys), engineered for the highly selective and sensitive sensing of intracellular viscosity. Operating through a restricted intramolecular motion mechanism, NM-Lys exhibited a remarkable fluorescence enhancement (up to 21-fold) at 645 nm in high-viscosity environments. NM-Lys demonstrated outstanding selectivity for viscosity against potential inte

A Near-Infrared Fluorescent Probe for Imaging Nitroxyl Dynamics in Ferroptosis-Associated Drug-Induced Liver Injury.

As a global health challenge, drug-induced liver injury (DILI) is an acute inflammatory disease closely linked to ferroptosis. Ferroptosis, characterized by iron-dependent lipid peroxidation, has been implicated in diverse pathological conditions, however, its precise molecular mechanism in liver injury remains to be fully elucidated. As a key active nitrogen species, the concentration fluctuation of nitroxyl (HNO) is closely correlated with ferroptosis. Nevertheless, in situ and real-time monitoring of endogenous HNO remains highly challenging owing to its transient lifespan and prominent rea

Lipid droplet-targeted ratiometric fluorescent probe for visualizing labile Fe2+ in alcohol-induced liver injury and silymarin-mediated protection.

The dysregulation of labile ferrous ions (Fe2+) is closely associated with oxidative stress, lipid peroxidation, and ferroptosis, all of which contribute to liver injury. The lipid droplet (LD) is not only a neutral lipid storage organelle but also a dynamic subcellular platform involved in lipid metabolism and stress responses. However, direct visualization of LD-associated labile Fe2+ during alcohol-induced liver injury remains challenging due to the lack of suitable LD-targeted Fe2+ imaging tools. Herein, we developed LD-Fe, a novel LD-targeted ratiometric fluorescent probe for Fe2+ imaging

Bright rRNA-Associated Merocyanine Probes Highlight Tumor Extensions in Ovarian Cancer.

We report a structure-activity relationship of merocyanine quinolinium dyes optimized as fluorogenic rRNA-associated probes. A series of analogs (11a-h) bearing electron-withdrawing groups (EWGs) or electron-donating groups (EDGs) on the phenolic "push" ring (ortho vs meta; mono vs disubstitution) was synthesized to evaluate electronic and steric effects on photophysical performance. Emission extended to 658 nm, fluorescence turn-on reached 570-fold, and brightness to 8 mM-1 cm-1. ortho-EWGs and meta-EDGs enhanced fluorescence, whereas steric hindrance (e.g., meta-dichloro) reduced planarity a

A nitroreductase-triggered NIR fluorescent probe for selective visualization in orthotopic breast cancer.

Nitroreductase (NTR) plays a crucial role in the hypoxic metabolism of breast tumors and serves as an important indicator of tumor aggressiveness and therapeutic response. However, visualization of NTR activity remains challenging due to the limited photophysical properties of existing probes, particularly their short emission wavelengths and small Stokes shifts. Here, we report a near-infrared (NIR) activatable fluorescent probe, DHM-NO2, which we constructed by coupling the large-Stokes-shift fluorophore DHM-OH with a nitrobenzyl recognition unit. DHM-NO2 exhibited negligible background fluo

An oral-to-urinalysis fluorescent prodrug platform for IBD theranostics.

We developed an oral-to-urinalysis theranostic approach for inflammatory bowel disease (IBD) that aims to improve procedural practicality and safety by enabling diagnosis and treatment monitoring without invasive sampling. The strategy exploits disease-associated changes in intestinal chemistry that alter absorption, using the hydrophobicity shift between a fluorescent prodrug (MB-ASA) and its activated product methylene blue (MB). MB-ASA was synthesized by conjugating MB to 5-aminosalicylic acid (5-ASA), a first-line IBD therapy, via a ROS-responsive urea linkage. Owing to its high hydrophobi

Sulfur-substituted hemicyanine-based NIR fluorescent probe for hypochlorous acid enables imaging of liver injury and plasma diagnosis of sepsis.

Hypochlorous acid (HClO), as a key reactive oxygen species in the human immune defense, can be found in tissue damage and bloodstream infections, which can serve as a biomarker for diagnosis of blood sepsis and drug-induced liver injury (DILI). Here, based on the advantages of NIR fluorescent probe, an HClO-activatable sulfur-substituted hemicyanine-based NIR fluorescent probe for imaging of drug-induced liver injury and plasma diagnosis of sepsis was developed in this paper. HClO-CyOH-S exhibits turn-on NIR fluorescence response towards HClO with an excellent selectivity and good sensitivity.

Precise diagnosis of hepatocellular carcinoma and acute kidney injury through near-infrared imaging utilizing a fluorescence probe for monitoring α-l-fucosidase.

Herein, hepatocellular carcinoma (HCC) and acute kidney injury (AKI) were accomplished using α-l-fucosidase (AFU)-responsive fluorescent probe, HD-AFU, which incorporates the canonical AFU recognition moiety and a structurally optimized hemicyanine-thioxanthene signaling unit. In comparison with recently reported probes, HD-AFU demonstrated robust and reliable performance in monitoring AFU activity across HCC and AKI models. This probe enabled mitochondrial-targeted intracellular imaging to track AFU dynamics during progression and therapeutic intervention. In the orthotopic transplanted tumor

Water-Soluble Viscosity-Activated Molecular Rotor for In Situ Imaging of Liver Damage in the NIR-II Window.

Microviscosity in the hepatic microenvironment changes during steatosis, matrix remodeling, and drug-induced injury but remains challenging to monitor in vivo. Here, we report JMR-1002-PEG, a water-soluble near-infrared II (NIR-II, 1000-1700 nm) fluorescent molecular rotor for viscosity-responsive liver imaging. The design of JMR-1002-PEG hinges on the strategic optimization of the donor-π-acceptor structure, where achieving NIR-II emission required careful modulation of the conjugated system while maintaining sufficient flexibility for viscosity-sensitive behavior. In mice, the probe reports

Negatively Charged Peptides-Assisted Self-Assembled Mitochondria-Targeting Nanoprobe for Detecting Enterokinase and Image-Guided Navigation in Human Breast Cancer.

Surgical resection remains the primary treatment in human breast cancer. However, the accurate and rapid delineation of tumor margins intraoperatively is still a significant challenge. Herein, we designed a near-infrared fluorescent probe DCIP-ENTK by incorporating dicyanoisophorone dye with a negatively charged peptide, which can be specifically cleaved by enterokinase (ENTK, a tumor biomarker for breast cancer). In vitro experiments confirmed that the probe could quickly (<10 min), specifically, and sensitively (LOD = 0.031 U/mL) respond to ENTK. Notably, this probe was engineered as a pepti

Dual-mode fluorescence and colorimetric detection of bilirubin using a curcumin-methylene blue metal-organic framework composite.

Neonatal jaundice requires rapid and reliable bilirubin monitoring to prevent severe complications. Here, a dual-mode (fluorescence-colorimetric) sensing platform is developed by encapsulation of curcumin within an amine-functionalized zirconium metal-organic framework (UiO-66-NH₂) in the presence of methylene blue. The MOF scaffold provides aqueous stability and adsorption sites for bilirubin, while curcumin serves as the fluorescence reporter and methylene blue enables complementary optical modulation. Bilirubin induces concentration-dependent fluorescence quenching. The sensor exhibits repr

Dual-Locked Near-Infrared Hemicyanine Probe toward ATP and HClO for Improved Inflammatory Bowel Disease Diagnosis.

Inflammatory bowel disease (IBD) is pathologically characterized by oxidative stress and disrupted energy metabolism, which is closely related to the overproduction of ATP and hypochlorite (ClO-). However, fluorescent probes capable of simultaneous detection of ATP and ClO- remain scarce. Herein, we report an ATP/ClO--activatable hemicyanine-based fluorescent probe, AClO-CHcy, for accurate diagnosis of IBD. This dual-locked probe features two independent recognition units: a cyclization switch for ATP sensing and a thiocarbamate group responsible for ClO- response, displaying intense near-infr

"IEDDA"-activated fluorescent sensing for protein cysteine redox modifications profiling.

BACKGROUND: Protein cysteine sulfenylation is a pivotal reversible modification in redox signaling and disease pathology. However, comprehensive monitoring of these events remains challenging, as most current "always-on" fluorescent probes suffer from high background signals and lack the capability for dynamic, real-time tracking in living systems. RESULTS: Here, we developed a background-free "turn-on" sensing strategy based on the inverse electron-demand Diels-Alder (IEDDA) reaction. We designed selective probes, BCN-SH and BCN-SOH, to tag protein thiols and sulfenic acids, respectively, by

Antioxidant 3-hydroxyflavone analogues: Integrated experimental and theoretical insights into lead fluoro-sensing and toxicity mitigation in living systems.

BACKGROUND: Lead (Pb2+) toxicity represents a major public health challenge, underscoring the need for advanced detection and remediation strategies. RESULTS: We introduce two synthetic 3-hydroxyflavone (3-HF) analogues: 3',4'-methylenedioxy-3-hydroxyflavone (1) and 4'-N,N-dimethyl-3-hydroxyflavone (2) as multifunctional "sense-and-treat" agents for Pb2+. These probes detect Pb2+ via a characteristic excited-state intramolecular proton transfer (ESIPT) fluorescence response, demonstrating excellent sensitivity with detection limits of 1.8 μM for (1) and 4.9 μM for (2), and high selectivity ov

Schiff base chemosensors for neutral analytes: optical transduction mechanisms, analytical performance, and application prospects.

The selective optical detection of neutral organic and biologically relevant analytes remains a central challenge in analytical chemistry, as the absence of formal charge and structural diversity among targets render conventional ion-responsive sensing frameworks largely inadequate. Schiff base-derived chemosensors, defined by their synthetically accessible azomethine (-C[double bond, length as m-dash]N-) core, have emerged as particularly well-suited molecular scaffolds for addressing this gap, owing to their modular structural tunability, straightforward synthesis from commercially available

Multi-biological activity evaluation of Sn(П), Zn(П) and Fe(П) complexes based on thiocarbohydrazide schiff bases: synthesis, spectroscopic investigations and fluorescence studies.

Two new Schiff base ligands (L1 and L2) were synthesized by condensing thiocarbohydrazide (TCH) with o-anisaldehyde or p-anisaldehyde in ethanol. Their mono- and bi-nuclear complexes with Sn(II), Zn(II), and Fe(II) were prepared for potential fluorescence and biological applications. Characterization was performed using FT-IR, NMR, UV-Vis spectroscopy, mass spectrometry, molar conductance, TGA, X-ray diffraction and SEM. XRD results indicated good crystallinity with crystallite sizes of 20-50 nm. Fluorescent intensity of free TCH ligands increased upon complexation with Sn, Zn, and Fe, suggest

Facilely fabricating fluorescent iron-doped polymer dots nanozyme with oxidase-like activity for dual-mode analysis of glutathione S-transferase and its inhibitor.

Glutathione S-transferase (GST) is a key enzyme involved in detoxification, antioxidation, and signal transduction, and its abnormal activity is closely associated with tumor drug resistance, liver damage, and inflammatory diseases. In this study, a bifunctional nanozyme based on novel iron-doped polymer dots (Fe-PDs) was constructed, which exhibits both yellow-green fluorescence and excellent oxidase-like activity. The Fe-PDs can oxidize N,N-diethyl-p-phenylenediamine (DPD) to generate a red oxidized product oxDPD (with an absorption peak at 551 nm), while simultaneously quenching their own f

A highly sensitive genetically encoded red cAMP sensor for multiplex imaging in vivo.

Circularly permuted green fluorescent protein (cpGFP)-based high-performance cAMP sensors have enabled real-time monitoring of cAMP dynamics with high spatiotemporal resolution in living animals. However, their utility is hampered by significant spectral overlap with other green/yellow fluorescent indicators and blue/cyan light-activated optogenetic actuators, limiting their compatibility in multiplexed imaging applications. While existing red cAMP sensors offer great spectral separation, they often suffer from a limited dynamic range ( < 1.5-fold in HEK293T cells), low cellular brightness, ag
