DPP4-inhibition reduces pro-inflammatory cytokine production by alpha-beta and gamma-delta T cells in vitro and in the biliary atresia mouse model.

Etiology and pathogenesis of biliary atresia (BA) remain elusive. Evidence points to a T cell mediated autoimmune response contributing to the disease by driving the ongoing hepatic inflammation. CD26, also known as dipeptidylpeptidase 4 (DPP4), is an immunoregulatory protein also expressed on T-lymphocytes. We aimed to investigate the potential of pharmacological DPP4 inhibition on cytokine production of T cells and potential therapeutic benefits in experimental BA. We analyzed the expression and regulation of CD26/DPP4 on αβ and γδ T cells in mice suffering from rotavirus-induced BA. Enzymat

Extracorporeal membrane oxygenation in the context of the 2009 H1N1 influenza A pandemic.

BACKGROUND: Extracorporeal membrane oxygenation (ECMO) incorporates surgical techniques as adjuncts in the management of refractory respiratory dysfunction. For many years, its primary application was for support of neonatal infants in cardiorespiratory failure. As the 2009 H1N1 influenza A pandemic developed, more reports came in of severe respiratory dysfunction and even death that seemed to be occurring preferentially in younger adults. Centers with the capability began to use ECMO to salvage these patients. RESULTS: The H1N1 virus is a subtype of influenza A. The hemagglutinin receptor bi

Acceptability of Kangaroo Mother Care (KMC) in neonatal transport: a qualitative study on mothers' and healthcare professionals' perspectives on KMC transport in Ghana.

BACKGROUND: In limited-resource settings, the lack of ambulances and transport incubators increases the risk of adverse outcomes in transported preterm and low-birth-weight neonates. The Kangaroo Mother Care (KMC) position during transport using available transport modes may be a cost-effective alternative. Mothers' and healthcare professionals' perspectives on the intervention are key to its successful implementation. METHODS: This qualitative study using semi-structured interviews was conducted at Cape Coast Teaching Hospital, Ghana. The study involved mothers and healthcare professionals w

Time to positivity (TTP) of neonatal blood cultures: a trend analysis over a decade from Ireland.

Background and aims: Blood culture (BC) remains gold standard for the evaluation and diagnosis of neonatal sepsis. Time when BC becomes positive and the type of microorganism isolated are crucial in deciding the antimicrobial management. Likely pathogenicity of organisms growing in BC could potentially be predicted based on the "time to positivity" (TTP). We aimed to estimate the predictive value of isolating a likely pathogenic organism depending on TTP; evaluate the aetiological trend and neonatal mortality rate due to culture-proven neonatal sepsis for over a decade and verify whether the a

N-Acetylcysteine Reduces Tissue Injury Induced by Oxygen-Glucose Deprivation in an Organotypic Culture of Mouse Cerebral Cortex Slices.

BACKGROUND/OBJECTIVES: Hypoxic-ischemic encephalopathy is the leading cause of infant mortality and disability. Hypothermic therapy is effective in hypoxic-ischemic encephalopathy, albeit in a limited number of cases. Hypothermia requires advanced technologies and significant financial resources, which are difficult to sustain in low-income countries, with devastating consequences. Valid alternatives to hypothermia therapy are therefore needed. METHODS: In vitro organotypic cultures of mouse cerebral cortex slices were used to demonstrate the direct protective effect of N-acetylcysteine (NAC)

Selecting Infants With Cryptorchidism and High Risk of Infertility for Optional Adjuvant Hormonal Therapy and Cryopreservation of Germ Cells: Experience From a Pilot Study.

INTRODUCTION: Orchiopexy for congenital cryptorchid testes is recommended between ½ and 1 year of age to preserve testicular germ cell maturation. Early operation is not enough to preserve fertility in 22 and 36% of cases. Aim of this study was to set up a protocol for optional adjuvant hormonal therapy after orchiopexy and thereafter cryopreservation of testicular biopsies from infants with bilateral cryptorchidism and high infertility risk. MATERIALS AND METHODS: We included 17 boys with bilateral cryptorchidism, normal FSH, and impaired germ cell number per tubular transverse section (G/T)

Enhanced metabolism of 1-beta-D-arabinofuranosylcytosine in Down syndrome cells: a contributing factor to the superior event free survival of Down syndrome children with acute myeloid leukemia.

Down syndrome (DS) children with acute myeloid leukemia (AML) have significantly higher event-free survival (EFS) rates compared with non-DS children when treated with protocols containing 1-beta-D-arabinofuranosylcytosine (ara-C). Sensitivity and metabolism of ara-C was examined in myeloblasts from DS and non-DS patients with AML, DS infants with the transient myeloproliferative disorder, and Epstein-Barr Virus (EBV) transformed lymphoblastoid cell lines with and without trisomy 21. DS myeloblasts were approximately 10-fold more sensitive to ara-C (measured by the 3-[4,5-dimethyl-thiazol-2-yl

In vitro maturation is associated with increased early embryo arrest without impairing morphokinetic development of useable embryos progressing to blastocysts.

STUDY QUESTIONS: Does polycystic ovarian syndrome (PCOS) or in vitro maturation (IVM) treatment affect embryo development events and morphokinetic parameters after time-lapse incubation? SUMMARY ANSWER: There was an increase in some abnormal phenotypic events in PCOS-IVM embryos as well as an increase in early arrest of PCOS-IVM and PCOS-ICSI embryos; however, IVM treatment or PCOS status did not alter morphokinetic development of embryos suitable for transfer of vitrification. WHAT IS KNOWN ALREADY: IVM has been less successful than standard IVF in terms of clinical pregnancy, implantation

Optimization of the viability PCR for accurate detection of Staphylococcus aureus in food samples.

Staphylococcus (S.) aureus is a prominent foodborne pathogen that can cause food poisoning due to its staphylococcal toxins. Controlling the viable levels of S. aureus is crucial for ensuring food safety. The detection of S. aureus during routine quality control is still primarily conducted using traditional culture-based methods, which are time-consuming and unable to detect viable but non-culturable cells. Viability PCR (vPCR) - a combination of traditional (or quantitative) PCR with photo-reactive DNA-intercalating dye(s) - has been introduced as an alternative to detect viable cells by exc

Group B streptococci vaginal colonization and drug susceptibility pattern among pregnant women attending in selected public antenatal care centers in Addis Ababa, Ethiopia.

BACKGROUND: Group B Streptococcus (GBS) is the leading cause of septicemia, meningitis, and pneumonia in neonates. Maternal colonization with GBS is the principal risk factor for early-onset disease in infants. Group B Streptococcus is now an important cause of maternal and neonatal morbidity and mortality in many parts of the world. In Ethiopia, few studies have been done on GBS colonization among pregnant women. The aim of this study was to determine the prevalence of GBS colonization, antimicrobial susceptibility patterns and assess risk factors among pregnant women. METHODS: A prospective

Effects of anti-human neutrophil antibodies in vitro. Quantitative studies.

Opsonic, antiphagocytic, cytotoxic, and metabolic effects of homologous and heterologous antibodies against human neutrophils were analyzed by means of quantitative assays to facilitate detection of antibody activity, and to probe membrane function of these cells. Normal human neutrophils were purified by gradient centrifugation, sensitized with heat-inactivated antineutrophil antisera, and incubated with rabbit alveolar macrophages in balanced salt solution containing nitroblue tetrazolium. The macrophages engulfed sensitized neutrophils and reduced nitroblue tetrazolium to formazan in phagoc

Development loop-mediated isothermal amplification assay for detection of enteropathogenic Escherichia coli (EPEC).

BACKGROUND: Enteropathogenic Escherichia coli (EPEC) is a major etiological agent of persistent diarrhea in pediatric and adult population globally and it is a leading cause of infant mortality in developing countries. Bacterial culture and biochemical assays are insufficient for accurately identifying EPEC strains. In contrast, PCR serves as a major molecular diagnostic method for EPEC. However, the application of PCR is limited due to high cost, time taking and technical requirements. METHOD AND RESULTS: In this study loop mediated isothermal amplification LAMP) was developed for rapid, spe

Brief exposure of neonatal testis cells to EGF or GDNF alters the regenerated tissue.

UNLABELLED: We have previously shown that implantation of testis cell aggregates under the back skin of immunodeficient mice results in de novo regeneration of testis tissue. We used this unique model to investigate the effects of epidermal growth factor (EGF) and glial cell-derived neurotrophic factor (GDNF) on testis cord development. Neonatal piglet testis cells were briefly (<1 h) exposed to either low (L: 0.02 μg/mL) or high (H: 2 μg/mL) doses of EGF, GDNF, or vehicle (control), before implantation in recipient mice. Randomly selected implants were removed from each mouse at 1, 2, 4, and

COVID-19 in Children, Pregnancy and Neonates: A Review of Epidemiologic and Clinical Features.

The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic has spread rapidly across the globe. In contrast to initial reports, recent studies suggest that children are just as likely as adults to become infected with the virus but have fewer symptoms and less severe disease. In this review, we summarize the epidemiologic and clinical features of children infected with SARS-CoV-2 reported in pediatric case series to date. We also summarize the perinatal outcomes of neonates born to women infected with SARS-CoV-2 in pregnancy. We found 11 case series including a total of 33

Transcription Profiles Associated with Inducible Adhesion in Candida parapsilosis.

Candida parapsilosis has emerged as a frequent cause of invasive candidiasis with increasing evidence of unique biological features relative to C. albicans As it adapts to conditions within a mammalian host, rapid changes in gene expression are necessary to facilitate colonization and persistence in this environment. Adhesion of the organism to biological surfaces is a key first step in this process and is the focus of this study. Building on previous observations showing the importance of a member of the ALS gene family in C. parapsilosis adhesion, three clinical isolates were cultured under

Adult immunohistochemical markers fail to detect intratubular germ cell neoplasia in prepubertal boys with cryptorchidism.

PURPOSE: Intratubular germ cell neoplasia is a precursor to testicular germ cell cancer. The condition is characterized by large germ cells with large nuclei with a hyperchromatic, coarse chromatin pattern, large prominent nucleoli and abundant pale cytoplasm. In prepubertal boys these cells are located centrally and peripherally mixed with normal cells in the seminiferous tubules. We evaluated the impact of adult intratubular germ cell neoplasia marking immunohistochemistry in screening for intratubular germ cell neoplasia in boys with cryptorchidism. MATERIALS AND METHODS: Histology section

Selective allergy to sheep's and goat's milk proteins.

BACKGROUND: Cow's milk proteins are amongst the most common causes of food allergy in infants, and caseins are probably the main allergens. The existence of a high degree of cross-reactivity between milk caseins from different animals has been reported. We describe a 2-year-old boy who experienced allergic reactions after eating and touching sheep's cheese, but who tolerated cow's milk and cow's milk dairy products. He had never ingested milk or milk derivatives from sheep or goat. METHODS: Skin prick tests were carried out using whey fractions of cow's milk, whole milk and casein from goat,

[The AIDS epidemic and its importance in obstetrics and gynecology].

There is hardly any other disease where differences of opinion are so dramatic as they are in AIDS. Safely established epidemiologic data are interpreted most controversially and heatedly discussed. Gynaecology and obstetrics are no exception; here, too, scientific discussion is marred by emotional overtones. The article attempts to survey update information and to sum up epidemiologic knowledge while trying to strike a balance. Since even the nomenclature is not always clear or uniform, the most important definitions and abbreviations are listed. Special attention is focussed on the problems
