Longitudinal Changes in Retinotopic Rod Function in Intermediate Age-Related Macular Degeneration.

PURPOSE: Although impairment of rod function in the early stages of age-related macular degeneration (AMD) has been well recognized, data on longitudinal changes in rod function at multiple retinal locations remain limited. This study investigated the longitudinal changes in retinotopic rod function in eyes with intermediate AMD (iAMD). METHODS: Complete ophthalmic examination, multimodal imaging, and scotopic perimetry were performed at baseline and at 12-month follow-up. Perimetric scotopic retinal sensitivities for the 505-nm stimulus were repeatedly measured for 20 minutes after exposing

A near-infrared fluorescent probe based on photostable Si-rhodamine for imaging hypochlorous acid during lysosome-involved inflammatory response.

Hypochloric acid (HClO) is mainly distributed in acidic lysosomes of phagocytes and closely associated with numerous physiological and pathological processes, especially inflammatory response. Fluorescent probe has become an important tool for imaging HClO in lysosomes, but suffered from interference from autofluorescence in vivo, phototoxicity to biosamples and photobleaching phenomenon due to their short-wavelength excitation and emission. Unfortunately, up to now, no near-infrared (NIR) lysosome-targetable fluorescent probe has been reported for imaging HClO. In this paper, a near-infrared

Highly sensitive and accurate detection of C-reactive protein by CdSe/ZnS quantum dot-based fluorescence-linked immunosorbent assay.

BACKGROUND: The conventional and widely used enzyme-linked immunosorbent assays (ELISA), due to specificity and high-sensitivity, were suitable in vitro diagnosis. But enzymes are vulnerable to the external conditions, and the complex operation steps limit its application. Semiconductor quantum dots have been successfully used in biological and medical research due to the high photoluminescence and high resistance to photobleaching. In this study, we have developed a novel quantum dot-labeled immunosorbent assay for rapid disease detection of C-reactive protein (CRP). RESULTS: The assay for t

Collagen cross-linking using rose bengal and green light to increase corneal stiffness.

PURPOSE: Photochemical cross-linking of corneal collagen is an evolving treatment for keratoconus and other ectatic disorders. We evaluated collagen cross-linking by rose bengal plus green light (RGX) in rabbit eyes and investigated factors important for clinical application. METHODS: Rose bengal (RB, 0.1%) was applied to deepithelialized corneas of enucleated rabbit eyes for 2 minutes. The diffusion distance of RB into the stroma was measured by fluorescence microscopy on frozen sections. RB-stained corneas were exposed to green (532-nm) light for 3.3 to 9.9 minutes (50-150 J/cm(2)). Changes

Mutations in human αA-crystallin/sHSP affect subunit exchange interaction with αB-crystallin.

BACKGROUND: Mutation in αA-crystallin contributes to the development of congenital cataract in humans. Heterooligomerization of αA-crystallin and αB-crystallin is essential for maintaining transparency in the eye lens. The effect of congenital cataract causing mutants of αA-crystallin on subunit exchange and interaction with αB-crystallin is unknown. In the present study, interaction of the mutants of αA-crystallin with αB-crystallin was studied both in vitro and in situ by the fluorescence resonance energy transfer (FRET) technique. METHODOLOGY/PRINCIPAL FINDINGS: In vitro FRET technique was

Both lamin A and lamin C mutations cause lamina instability as well as loss of internal nuclear lamin organization.

We have applied the fluorescence loss of intensity after photobleaching (FLIP) technique to study the molecular dynamics and organization of nuclear lamin proteins in cell lines stably transfected with green fluorescent protein (GFP)-tagged A-type lamin cDNA. Normal lamin A and C proteins show abundant decoration of the inner layer of the nuclear membrane, the nuclear lamina, and a generally diffuse localization in the nuclear interior. Bleaching studies revealed that, while the GFP-tagged lamins in the lamina were virtually immobile, the intranuclear fraction of these molecules was partially

In vivo pharmacokinetics of 8-aminolevulinic acid-induced protoporphyrin IX during pre- and post-photodynamic therapy in 7,12-dimethylbenz(a)nthracene-treated skin carcinogenesis in Swiss mice: a comparison by three-compartment model.

Delta-aminolevulinic acid-photodynamic therapy (ALA-PDT) has emerged as a useful technique in the treatment of superficial basal cell carcinoma, actinic keratosis, squamous cell carcinoma and tumors of other organs. Earlier reports mention that there is reappearance of protoporphyrin IX (PpIX) after photoirradiation of tumors. This property of reappearance of PpIX is being utilized to treat nodular tumors by fractionated light dose delivery. However, there is still no unanimously accepted reason for this reappearance phenomenon and the rate of resynthesis after PDT. On account of this, studies

A platform for complementation and characterization of familial haemophagocytic lymphohistiocytosis 3 mutations.

Mutations in UNC13D cause the severe immune disorder familial haemophagocytic lymphohistiocytosis type 3 (FHL3). The gene product munc13-4 is expressed in hematopoietic cells and is essential for degranulation. Little information is available on genotype-phenotype relationships of UNC13D mutations. Some mutants may have residual functionality which qualifies them as promising targets for attempts to enhance function pharmacologically. A problem for such analysis is the scarcity of patient material. We established assays in the RBL-2H3 cell line to assess functionality of lentivirally transduce

Photodynamic anti-biofilm PHEMA/Rose Bengal hydrogels enhanced by iodide: evidence for hydrogen peroxide and triiodide formation.

Biofilm-associated infections remain a major challenge for biomedical devices, motivating the development of light-activated antimicrobial materials. Here we report the antibiofilm activity (against Staphylococcus aureus and Pseudomonas aeruginosa) of a photodynamic hydrogel consisting of Rose Bengal (RB) physically entrapped within a crosslinked poly(2-hydroxyethyl methacrylate) (PHEMA) matrix. Under visible-light irradiation, RB-PHEMA alone produced only modest reductions (ca. 1 log), whereas addition of iodide markedly enhanced antibiofilm performance, yielding a 4.2-log reduction for S. au

Molecular mechanism of cholesterol-dependent membrane fusion in SARS-CoV-2 entry.

Enveloped virus invasion relies on spike glycoprotein-mediated membrane fusion. Cholesterol that serves crucial roles in modulating protein conformations and membrane properties, plays an essential role in the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) cell entry. However, the precise regulatory mechanism of cholesterol in SARS-CoV-2 fusion remains unknown. Here, using an in vitro vesicle-vesicle content mixing assay, we demonstrated that the addition of cholesterol enhanced SARS-CoV-2 spike-mediated vesicle-vesicle fusion, with this enhancement being dependent on the C-termi

Chopped optical biosensing enables temporal background subtraction for high-sensitivity fluorescence immunoassays.

Fluorescence-based immunoassays are widely used for sensitive biomarker detection, but their performance at low analyte concentrations is often limited by background signals originating from unbound fluorophores, Raman scattering, and sample autofluorescence. Here, we introduce chopped optical biosensing (COB), an analytical framework that improves fluorescence immunoassay sensitivity by temporally separating background and target fluorescence. In COB, fluorophore-labeled targets captured on magnetic beads are concentrated into a small detection region using a permanent magnet. An optical chop

Interaction analysis of tight junction proteins by Förster resonance energy transfer is recommended in live cells rather than after paraformaldehyde fixation.

The tight junction regulates the paracellular passage of solutes and water. It is primarily constituted by the family of claudins which assemble to huge multimers and basically seal the paracellular cleft. Certain claudins, however, can function as subunits of selective paracellular channels and thereby mediate paracellular ion and water flux across epithelia. In the thick ascending limb of Henle's loop (TAL), ion handling depends on the spatial separation of paracellular sodium and magnesium transport. Magnesium is conducted via a claudin-16/19 complex, whereas sodium permeation is enabled by

Engineered Calmodulin Assembles into Core-Shell Particles Serving as Robust Fluorescent Calcium Sensors.

Calcium is a strictly regulated physiological ion whose levels in blood and saliva act as biomarkers for a wide range of ailments, including renal, endocrine, and bone diseases. Current diagnostic approaches mainly rely on centralised laboratory assays that are accurate but slow and depend on infrastructural support. Thus, robust biosensors capable of rapid, selective calcium detection in complex biofluids are needed to enable real-time diagnostics with minimal invasive sampling. In this study, we engineered the soluble fluorescent sensor NCaMP7, a fusion of calmodulin and split neongreen fluo

The Balbiani body is formed by microtubule-controlled molecular condensation of Buc in early oogenesis.

Vertebrate oocyte polarity has been observed for two centuries and is essential for embryonic axis formation and germline specification, yet its underlying mechanisms remain unknown. In oocyte polarization, critical RNA-protein (RNP) granules delivered to the oocyte's vegetal pole are stored by the Balbiani body (Bb), a membraneless organelle conserved across species from insects to humans. However, the mechanisms of Bb formation are still unclear. Here, we elucidate mechanisms of Bb formation in zebrafish through developmental biomolecular condensation. Using super-resolution microscopy, live

IDR-driven LLPS of GAS2L3 scaffolds CHMP4B condensates to accelerate cytokinesis in hepatocellular carcinoma cells.

INTRODUCTION: Dysregulated cell cycle progression is a hallmark of cancer, promoting uncontrolled cell proliferation. Growth arrest-specific 2 like 3 (GAS2L3), which is highly expressed in hepatocellular carcinoma (HCC) cells, correlates with poor clinical outcomes. However, the role of GAS2L3 in HCC pathogenesis remains unclear. OBJECTIVES: This study aimed to investigate the molecular mechanisms by which GAS2L3 contributes to cytokinesis and hyperproliferation of HCC cells. METHODS: Loss-of-function assays of GAS2L3 were performed to assess midbody assembly, a process essential for cytokin

YTHDF2-mediated stabilization of SREBF1 promotes lipid metabolic reprogramming and ferroptosis-associated radioresistance in anaplastic thyroid carcinoma.

Anaplastic thyroid carcinoma (ATC) is the most aggressive endocrine malignancy and frequently resistant to radiotherapy. Lipid metabolic reprogramming has been implicated in ferroptosis evasion and therapeutic resistance in cancers; however, the specific mechanisms of lipid metabolic rewiring in regulating radioresistance in ATC remain unclear. This study employed an integrated multi-omics profiling, including RNA sequencing (RNA-seq), methylated RNA immunoprecipitation sequencing (MeRIP-seq), and RNA immunoprecipitation sequencing (RIP-seq), lipidomics profiling, and mRNA stability assays, to

The Allosteric Inhibitor Pentachloropseudilin Inhibits Myosin 1C ATPase Activity and Recapitulates Retinitis Pigmentosa Phenotypes in Mice.

Unconventional myosins are molecular motors that move along actin filaments in an ATPase-dependent manner, thereby influencing intracellular cargo transport. Dysfunction of myosins due to loss-of-function mutations causes human disease phenotypes such as deafness, retinitis pigmentosa, renal failure, and hypertrophic cardiomyopathy. However, some genetic models lacking unconventional myosins do not recapitulate the retinal phenotypes observed in humans, necessitating alternative approaches. We previously showed that the myosin motor protein MYO1C is required for the trafficking of rhodopsin in

Highly accurate image registration for 3D multiplexed cyclic imaging using dense labeling in expandable tissue gels.

Multiplexed cyclic imaging in expandable tissue gels has been extensively studied to visualize numerous biomolecules at a nanoscale resolution in situ. Previous studies have employed sparse labels, such as DAPI or lectin staining, as registration markers. However, these sparse labels do not adequately capture the full extent of deformation across the entire region of interest. To overcome this challenge, we propose the use of dense labels, specifically fluorophore N-hydroxysuccinimide (NHS)-ester staining, as registration markers to achieve highly accurate image registration. We first tested s

Beta-lactam antibiotics under light: optical and photochemical characterization.

Antibiotics (ATBs) remain a cornerstone in the treatment of bacterial infections, yet clinical practice increasingly incorporates photonic therapies such as photobiomodulation (PBM) and photodynamic therapy (PDT). These modalities, widely used in infectious and inflammatory conditions, are frequently administered concomitantly with ATBs. However, the potential photochemical interactions between β-lactam ATBs and light-based therapies remain poorly understood, raising questions about efficacy and safety in combined treatment settings. In this study, we characterized the optical and photochemica

The ER-Resident Ras Inhibitor 1 (Eri1) of Candida albicans Inhibits Hyphal Morphogenesis via the Ras-Independent cAMP-PKA Pathway.

Ras signaling and glycosylphosphatidylinositol (GPI) biosynthesis are mutually inhibitory in S. cerevisiae (Sc). The inhibition is mediated via an interaction of yeast Ras2 with the Eri1 subunit of its GPI-N-acetylglucosaminyl transferase (GPI-GnT), the enzyme catalyzing the very first GPI biosynthetic step. In contrast, Ras signaling and GPI biosynthesis in C. albicans (Ca) are mutually activated and together control the virulence traits of the human fungal pathogen. What might be the role of Eri1 in this pathogen? The present manuscript addresses this question while simultaneously characteri
