Pineal-gonadal relationship in the teleost Channa punctatus (Bloch): evidence for possible involvement of hypothalamic serotonergic system.

In Channa punctatus, pinealectomy results in an accelerated growth (stimulatory effect) of the ovary in preparatory phase (March), but had no significant effect in prespawning (May-June) or postspawning (September) phases. Administration of melatonin (25 micrograms/fish or 0.4 mg/kg BW at 2-day intervals for 30 days) inhibits the stimulatory effect of a long photoperiod and high temperature (16L:8D; 22 +/- 1 degree C) on the ovary in the early preparatory phase (February). In the late preparatory phase (April), the administration of both melatonin (0.5 mg/kg BW at 2-day intervals) and parachlo

Intestinal melatonin levels and gut microbiota homeostasis are independent of the pineal gland in pigs.

INTRODUCTION: Melatonin (MEL) is a crucial neuroendocrine hormone primarily produced by the pineal gland. Pinealectomy (PINX) has been performed on an endogenous MEL deficiency model to investigate the functions of pineal MEL and its relationship with various diseases. However, the effect of PINX on the gastrointestinal tract (GIT) MEL levels and gut microbiome in pigs has not been previously reported. METHODS: By using a newly established pig PINX model, we detected the levels of MEL in the GIT by liquid chromatography-tandem mass spectrometry. In addition, we examined the effects of PINX on

Melatonin and osmoregulation in fish: A focus on Atlantic salmon Salmo salar smoltification.

Part of the life cycle of several fish species includes important salinity changes, as is the case for the sea bass (Dicentrarchus labrax) or the Atlantic salmon (Salmo salar). Salmo salar juveniles migrate downstream from their spawning sites to reach seawater, where they grow and become sexually mature. The process of preparation enabling juveniles to migrate downstream and physiologically adapt to seawater is called smoltification. Daily and seasonal variations of photoperiod and temperature play a role in defining the timing of smoltification, which may take weeks to months, depending on t

A novel role for the pineal gland: Regulating seasonal shifts in the gut microbiota of Siberian hamsters.

The gut microbiota plays a significant role in a variety of host behavioral and physiological processes. The mechanisms by which the gut microbiota and the host communicate are not fully resolved but include both humoral and direct neural signals. The composition of the microbiota is affected by internal (host) factors and external (environmental) factors. One such signal is photoperiod, which is represented endogenously by nocturnal pineal melatonin (MEL) secretion. Removal of the MEL signal via pinealectomy abolishes many seasonal responses to photoperiod. In Siberian hamsters (Phodopus sung

Melatonin augments expression of the procollagen α1 (I) and α1 (III) genes in the infarcted heart scar of pinealectomized rats.

The pineal gland is involved in the regulation of collagen accumulation in peripheral wounds and scars of the infarcted heart. This study is aimed to provide an explanation of whether the pineal gland and melatonin (MLT) is involved in the regulation of α1 (I) and α1 (III) procollagen gene expression. A secondary aim is the investigation of whether the mechanism of changes could be explained by the direct influence of MLT on myofibroblasts isolated from the scar. Myocardial infarction was induced by left coronary artery ligation in all rats. Animals were divided into groups: control, vehicle-t

Measurement of melatonin in body fluids: standards, protocols and procedures.

BACKGROUND AND PURPOSE: The circadian rhythm of melatonin in saliva or plasma, or of the melatonin metabolite 6-sulfatoxymelatonin (a6MTs) in urine, is a defining feature of suprachiasmatic nucleus (SCN) function, the body's endogenous oscillatory pacemaker. The primary objective of this review is to ascertain the clinical benefits and limitations of current methodologies employed for detection and quantification of melatonin in biological fluids and tissues. DATA IDENTIFICATION: A search of the English-language literature (Medline) and a systematic review of published articles were carried o

Melatonin stimulates the release of growth hormone and prolactin by a possible induction of the expression of frog growth hormone-releasing peptide and its related peptide-2 in the amphibian hypothalamus.

We recently identified a novel hypothalamic neuropeptide stimulating GH release in bullfrogs and termed it frog GH-releasing peptide (fGRP). The fGRP precursor encodes fGRP and its related peptides (fGRP-RP-1, -RP-2, and -RP-3), and fGRP-RP-2 also stimulates GH and prolactin (PRL) release. Cell bodies and terminals containing these neuropeptides are localized in the suprachiasmatic nucleus (SCN) and median eminence, respectively. To understand the physiological role of fGRP and fGRP-RP-2, we investigated the mechanisms that regulate the expression of these neuropeptides. This study shows that

Control of secretory lipid droplets in the harderian gland by testosterone and the photoperiod: comparison of two species of hamsters.

Harderian glands of mammals secrete lipid. They are markedly sexually dimorphic in Syrian (golden) hamsters (Mesocricetus auratus): female glands consist almost entirely of one cell type (type I) with small lipid droplets, whereas glands of males have both type I and type II cells, with large lipid droplets. Siberian (Djungarian) hamsters (Phodopus sungorus) have sexually monomorphic Harderian glands, with both type I and type II cells. We used a morphometric technique to quantify the proportions of small (type 1) and large (type 2) lipid droplets in these two species, in relation to the prese

Regional and diurnal studies of melatonin and melatonin binding sites in the duck gastro-intestinal tract.

Since melatonin and putative melatonin receptors can be detected in a variety of peripheral tissues, direct endocrine and paracrine actions of melatonin on the physiological functions of different organ systems in response to internal and external stimuli probably exist. As an extension of our earlier work on the pharmacological characterization of 2-[125I]iodomelatonin binding sites in the duck jejunum, the regional and diurnal variations of melatonin and putative melatonin receptors of different segments of the duck gastro-intestinal tract were studied in an attempt to understand the role of

Photoperiodic synchronization of a circannual reproductive rhythm in sheep: identification of season-specific time cues.

Seasonal reproduction in the ewe is generated by an endogenous circannual rhythm of reproductive neuroendocrine activity. Exposure to as few as 70 days of photoperiodic information a year is sufficient to synchronize the rhythm. The present study was conducted to identify which portions of the photoperiodic cycle are utilized for synchronization. For this purpose, we used pinealectomized ewes that could not respond reproductively to changes in day length. Selected photoperiodic information was provided via infusion of melatonin, a hormone that provides the neuroendocrine code for day length in

Pineal and habenula calcification in schizophrenia.

Animal data indicate that melatonin secretion is stimulated by the paraventricular nucleus (PVN) of the hypothalamus and that lesions of the PVN mimic the endocrine effects of pinealectomy. Since the PVN lies adjacent to the third ventricle, I propose that periventricular damage, which is found in schizophrenia and may account for the third ventricular dilatation seen on computed tomographic (CT), may disrupt PVN-pineal interactions and ultimately enhance the process of pineal calcification (PC). To investigate this hypothesis, I conducted CT study on the relationship of PC size to third ventr

Transplantation of the pineal gland in the mammalian third cerebral ventricle.

Fine structural correlates and functional parameters were measured in pinealectomized rats following grafting of the pineal gland into the third cerebral ventricle. Pinealectomy caused a significant decrease in serum melatonin concentration of animals compared to that in normal controls. No significant difference was observed in the serum melatonin concentration between pinealectomized rats and those receiving sham transplantation with fragments of occipital cortex. By 6 weeks nearly 50% of pinealectomized rats receiving pineal transplants demonstrated a significant increase in the serum melat

Corpus luteum progesterone secretion in rats undergoing pineal-mediated reproductive regression.

In nonseasonal breeders, such as the laboratory rat, the potentiation of the antigonadal action of light deprivation by olfactory bulbectomy is manifested, among other things, by a diminished secretion of testosterone in males and estradiol in females. Little is known about the actions of the pineal gland on progesterone secretion. Since secretion of the luteotrophic hormones (LH and PRL) decreases in blind-anosmic rats and because the rat corpus luteum exhibits different degrees of LH and/or PRL dependency, the purpose of these experiments was to study the corpus luteum progesterone secretion

Effect of external factors on gonadal activity and body mass of male golden hamsters (Mesocricetus auratus).

Effects of photoperiod, temperature and food and water availability on body and testicular mass of golden hamsters were investigated. Lack of water or food inhibited body growth. Short photoperiod (LD 8:16) enhanced body growth in males fed 'poor diet' but it had no effect in animals on 'high quality diet'. Cold exposure diminished body growth in animals with access to water but not in those deprived of water. It is suggested that lowered motor activity is mainly responsible for body mass gain of animals kept under the short photoperiod. All stressful factors (water deprivation, 'poor diet' or

The expression of inflammatory cytokines on the aorta endothelia are up-regulated in pinealectomized rats.

This study was designed to investigate the effect of melatonin on the expression of aortic inflammatory cytokines and its underlying mechanisms in rats. Melatonin deficiency rats (Px, N = 16) were created by pinealectomy and were fed with normal diet for 16 weeks after the surgery, and compared with sham-operated rats (Con, N = 14). Serum lipid profile, glucose metabolism parameters, serum oxidative stress and inflammatory biomarkers were evaluated. The expression of inflammatory cytokines in the aorta endothelia was analyzed. To evaluate the signal transduction pathways of melatonin on the ex

Modulation by pineal gland of ouabain high-affinity binding sites in rat cerebral cortex.

To investigate the participation of the pineal gland and its hormone melatonin on Na(+)-K(+)-ATPase (the sodium pump) in rat brain, we used Scatchard plots to analyze the changes in rat cerebral cortex of [3H]ouabain high-affinity binding in groups of intact, pinealectomized (PX), and sham-PX rats. Only one type of binding site, with a dissociation constant of approximately 3 nM and site number (Bmax) of approximately 250 fmol/mg protein, was apparent with our assay conditions. PX or sham-PX rats (subjected to surgery 15 days earlier) were killed at six different time intervals during the 24-h

Synchronization of the circannual reproductive rhythm of the ewe by discrete photoperiodic signals.

Although many species display endogenous circannual rhythms of biological activity that are synchronized by day length, the specific photoperiodic requirements for synchronizing such rhythms are not established for any species. We tested the hypothesis that the circannual reproductive rhythm of sheep can be synchronized by exposure to just one or two discrete blocks of photoperiodic information each year. Ewes were pinealectomized to prevent their ability to transduce photoperiodic information into altered reproductive neuroendocrine activity. During the 53/4 yr following pinealectomy, specifi

Anterograde nerve degeneration after superior cervical ganglionectomy coexists with a decrease in arginine vasopressin release in rats.

After bilateral superior cervical ganglionectomy (SCGx) in adult male rats, norepinephrine content of the pituitary neurointermediate lobe (NIL) decreased at 12-24 h after surgery to attain concentrations 40-60% of controls between 24 and 60 h after surgery. To assess arginine vasopressin (AVP) secretion during this time, plasma and NIL-AVP levels were measured by radioimmunoassay. In sham SCGx controls, plasma AVP increased about 2-fold within 6 h after surgery and decreased thereafter, to attain presurgical values by 60 h after surgery. In SCGx rats, a significant increase in plasma AVP conc

The Pineal Gland Is Required for Optimal Nighttime Immunity to Bacterial Infection in Larval Zebrafish.

The immune response to bacterial infection demonstrates circadian rhythmicity, with heightened antibacterial immunity often observed during the daytime. This circadian-gated immune response is believed to have evolved to synchronize peak antibacterial immunity with the increased threat of infection when active. Using a larval zebrafish infection model, we have previously shown that light has a positive effect on antibacterial immunity, with enhanced survival and neutrophil recruitment observed when infections are delivered during the light phase. However, the specific contribution of the light
