One Substance, Multiple Assessments: How the Various European Environmental Risk Frameworks Affect the Outcome of Chemical Risk Assessments.

The European Union (EU) regulates chemicals to protect the environment, but different regulations apply depending on the intended use of the substances in question. Advances in chemical monitoring have revealed inconsistencies in environmental risk assessments, prompting the European Commission to advocate for a "one substance, one assessment" (OS-OA) approach. As part of the EU Partnership for the Assessment of the Risk from Chemicals (PARC), this study examines the implementation and outcome of environmental risk assessments carried out within different regulatory frameworks, focusing on the

Environmentally relevant body residues of current-use and legacy pesticide mixtures induce behavioral changes in Juvenile Chinook Salmon.

Pesticides are widely applied in agricultural, industrial, and residential settings, often reaching nearby aquatic ecosystems through runoff and spray drift, particularly during storm events. The Sacramento-San Joaquin Delta (Delta) in California is significantly impacted, with multiple pesticides being frequently detected in surface waters and sediments. These pesticides are associated with various adverse effects on aquatic species, from sublethal impairments to lethal outcomes. Juvenile Chinook Salmon (Oncorhynchus tshawytscha), which migrate downstream through these contaminated waterways,

Presence of localized elevated metabolic enzymes and kdr mutations in the voltage-gated sodium channel gene indicate early evidence of resistance of Phlebotomus argentipes to alpha-cypermethrin in Bihar, India.

BACKGROUND: Visceral leishmaniasis, caused by Leishmania donovani and transmitted by Phlebotomus argentipes, remains a major public health challenge in the Indian subcontinent. Sand fly populations are controlled by using different insecticides, particularly dichlorodiphenyltrichloroethane (DDT) and pyrethroids such as alpha-cypermethrin. Prolonged and irregular use of these insecticides raises concern about the development of resistance in sand fly populations. METHODS: This study aimed to compare metabolic detoxification and knockdown resistance (kdr) mutations in Vgsc and ace-1 between nin

Delta-Class Glutathione S-Transferase Modulates the Susceptibility of Myzus persicae to λ-Cyhalothrin.

Glutathione S-transferases (GSTs) constitute a critical enzyme superfamily responsible for xenobiotic detoxification in insects. The green peach aphid, Myzus persicae, is a globally significant agricultural pest that has exhibited declining field susceptibility to λ-cyhalothrin (LCT), a widely applied pyrethroid insecticide. While elevated GST activity has been correlated with LCT tolerance in various aphid populations, the precise molecular mechanisms driving this process have not been fully elucidated. In this study, we functionally characterized a delta-class GST gene (designated MpGSTd1) f

Analysis of alpha-cypermethrin pesticide residues along the value chain of tomato from Laikipia County, Kenya.

BACKGROUND: In Kenya, the extensive use of agrochemicals in fruits and vegetable farming raises concerns about pesticide residues in agricultural products including tomato fruits. There are still a number of highly hazardous pesticides including mancozeb, glyphosate acid, chlorpyrifos, acephate and cypermethrin amongst many others that are still used in Kenya despite being banned in Europe due to harmful effects on humans and other animals. This study aimed to evaluate the residual levels of cypermethrin pesticide in locally produced tomato in Laikipia County, Kenya. Also, the study identified

Synergistic antinematodal effects of fluopyram combined with chlorfenapyr and its potential mechanism against Bursaphelenchus xylophilus.

To enhance the chemical control of pine wilt disease (PWD) and mitigate resistance arising from the prolonged use of a single-agent pesticides, the nematicides fluopyram (FLU) and chlorfenapyr (CHL) were combined (FLU_CHL) to determine the optimal ratio and assess their effects on pine wood nematodes (PWNs) and the potential synergistic mechanisms. Results revealed that the optimal synergistic mass ratio of FLU to CHL was 1:3, with nematode survival rate, egg hatchability, and population size significantly lower than with FLU or CHL alone. At this ratio, FLU_CHL also induced morphological abno

Toxicity of Four Common Environmental Chemicals Across Caenorhabditis elegans Life Stages Supporting the One Health Concept.

Pesticides and pharmaceuticals are among the most common chemical groups in waterbodies and soils, and their universal distribution raises concerns about potential adverse effects on nontarget organisms and humans. Reproductive output disruption is of particular concern, as it transposes effects from the individual to the next generations at the population level, requiring precise identification of the affected processes. Caenorhabditis elegans , with its well-known lifecycle, is an excellent model organism for investigating such effects within the One Health framework. The present study evalu

Synergistic risk in the gut and liver: Insights into the toxic mechanisms and molecular interactions of combined exposure to triazophos and fenvalerate in zebrafish.

The simultaneous or sequential application of pesticides such as triazophos (TRI) and fenvalerate (FEN) in agriculture results in their residues co-existing in the environments. However, the impact of co-exposure to TRI and FEN on the gut-liver axis, along with the underlying mechanisms, remains unclear. Our results showed that exposure to FEN (96 h-LC50 value of 0.096 mg a.i. L-1) was more toxic to adult zebrafish compared to TRI (96 h-LC50 value of 6.75 mg a.i. L-1). Furthermore, the study aimed to reveal the toxic potencies of individual and combined exposure to TRI and FEN on the liver-gut

Acaricide resistance status of deltamethrin and coumaphos in Hyalomma anatolicum ticks collected from different districts of Haryana.

To study the acaricide resistance status and possible mechanisms of action in conferring resistance to commonly used acaricides (deltamethrin and coumaphos), Hyalomma anatolicum ticks were collected from 6 dairy farms of Hisar and Charkhi Dadri districts of Haryana. By using standard larval packet test, H. anatolicum tick larvae of Charkhi Dadri isolates were found to be susceptible (100% mortality) to both the acaricides. Level-I resistance against coumaphos was recorded from four isolates, whereas, level-II was observed in only one isolate, collected from Hisar. One isolates (Kaimri) from Hi

A mutation associated with resistance to synthetic pyrethroids is widespread in US populations of the tropical lineage of Rhipicephalus sanguineus s.l.

The brown dog tick, Rhipicephalus sanguineus sensu lato (s.l.), is an important vector for Rickettsia rickettsii, causative agent of Rocky Mountain spotted fever. Current public health prevention and control efforts to protect people involve preventing tick infestations on domestic animals and in and around houses. Primary prevention tools rely on acaricides, often synthetic pyrethroids (SPs); resistance to this chemical class is widespread in ticks and other arthropods. Rhipicephalus sanguineus s.l. is a complex that likely contains multiple unique species and although the distribution of thi

Polyaniline-coated kapok fibers for convenient in-syringe solid-phase microextraction and determination of organochlorine and pyrethroid pesticide residues in aqueous samples.

Pesticides used in agriculture have low polarity and a tendency to accumulate in fatty tissues, posing potential risks to human health. Effective pre-treatment is crucial due to complex sample matrices and low concentrations of pesticide residues typically encountered in instrument analysis. In this study, polyaniline-coated kapok fiber (PANI-KF) was synthesized successfully using in-situ oxidative polymerization for use as sorbents in in-syringe SPME of pyrethroid pesticides (PYRs) and organochlorine pesticides (OCPs) from aqueous samples. Coating the natural KF with PANI maintained the hollo

Pseudomonas mosselii: a potential alternative for managing pyrethroid-resistant Aedes aegypti.

BACKGROUND: Aedes aegypti is a widespread mosquito in tropical and subtropical regions that causes significant mortality and morbidity in humans by transmitting diseases, such as dengue fever and Zika virus disease. Synthetic insecticides, such as pyrethroids, have been used to control Ae. aegypti, but these insecticides can also affect nontarget organisms and contaminate soil and water. This study aimed to investigate the mosquitocidal activity of Pseudomonas mosselii isolated from pond sludge against larvae of Ae. aegypti. RESULTS: Based on the initial results, similar time-course profiles

Biomarkers of oral subacute toxicity of deltamethrin in exposed male Albino rats.

Deltamethrin is one of the most effective pyrethroid compounds, widely employed in veterinary medicine, public health, and farming. Deltamethrin-triggered oxidative stress largely causes serious harm to an organism. Acute toxicity of this compound was extensively investigated, while less information is available on its oral sub-acute effects. This study assessed, in the male Albino rats, the effects of oral gavage of either 0.874 mg/kg (0.01 LD50) or 8.740 mg/kg (0.10 LD50) of deltamethrin for successive 14 days to investigate its effects on biomarkers and to detect the tissue injury in rats f

The Effect of Plasmodium falciparum (Welch) (Haemospororida: Plasmodiidae) Infection on the Susceptibility of Anopheles gambiae s.l. and Anopheles funestus (Diptera: Culicidae) to Pyrethroid Insecticides in the North-Western and South-Eastern, Tanzania.

The rapid development of insecticide resistance in malaria vectors threatens insecticide-based interventions. It is hypothesized that infection of insecticide-resistant vectors with Plasmodium parasites increases their vulnerability to insecticides, thus assuring the effectiveness of insecticide-based strategies for malaria control. Nonetheless, there is limited field data to support this. We investigated the effect of the Plasmodium falciparum infection on the susceptibility of Anopheles gambiae s.l. and Anopheles funestus to pyrethroids in south-eastern (Kilombero) and north-western (Muleba)

The water-exiting behavior and survival of predaceous diving beetles in responses to lambda-cyhalothrin, chlorantraniliprole, and thiamethoxam.

Predaceous diving beetles (dytiscids) are important top insect predators in various natural, agricultural, and artificial water bodies. How they respond to human disturbances such as insecticide contamination to their habitats has been understudied. In this study, we investigated the lethal effects of lambda-cyhalothrin, chlorantraniliprole, and thiamethoxam at nominal field concentrations in 3 cm-deep water in a hypothetical paddy field (Ci,3) on adult Dytiscus sinensis Feng (Coleoptera: Dytiscidae). Lambda-cyhalothrin exhibited significant lethal effects on D. sinensis adults with its Ci,3 e

First report on knockdown resistance mutations in wild populations of Aedes aegypti from Argentina determined by a novel multiplex high-resolution melting polymerase chain reaction method.

BACKGROUND: The mosquito Aedes aegypti is an urban vector of dengue and other arboviruses. During epidemics of these viruses, pyrethroid insecticides are used for the control of adult mosquitoes. The worldwide resistance of Ae. aegypti to these insecticides is a cause of failure of vector control campaigns. The primary target of pyrethroids is the voltage-gated sodium channel. Point mutations in the gene coding for this channel, called knockdown resistance (kdr) mutations, are associated with pyrethroid resistance. Two kdr mutations, V1016I and F1534C, have increased in frequency in natural po
