Tick-borne diseases cause significant losses to livestock production in tropical and subtropical regions. In Tanzania, detailed studies on tick-borne pathogens in cattle using sensitive molecular detection methods are scarce. In this study, we investigated the occurrence of Theileria spp., Babesia spp., Anaplasma spp. and Ehrlichia spp. in 245 blood samples collected from cattle on Pemba Island, Tanzania. We used polymerase chain reaction (PCR) and gene sequencing to detect and identify pathogens. PCR screening revealed overall infection rates of 62.4% for Theileria spp., 17.6% for Babesia big

Canine piroplasmoses, caused by Babesia spp., Theileria spp. and Rangelia vitalii, are emerging vector-borne diseases with a worldwide distribution, transmitted by ticks. The aim of this study was to determine the prevalence and perform molecular characterization of piroplasmids in domestic dogs from Asunción city, Paraguay. Blood samples were taken from 384 domestic dogs from Asunción city, Paraguay. DNA was purified from dog blood samples and submitted to nested PCR assays for piroplasmids (18S rRNA) and sequenced for identification and phylogenetic analysis. Overall piroplasmid prevalence i

Tick-borne hemoparasites (TBHs) are a group of pathogens of concern in animal management because they are associated with a diversity of hosts, including both wild and domestic species. However, little is known about how frequently TBHs are shared across the wildlife-livestock interface in natural settings. Here, we compared the TBHs of wild Grant's gazelle (Nanger granti) and domestic sheep (Ovis aries) in a region of Kenya where these species extensively overlap. Blood samples collected from each species were screened for piroplasm and rickettsial TBHs by PCR-based amplification of 18S/16S r

This study was designed to investigate an outbreak of high mortality that occurred in naïve Assaf sheep introduced into a Latxa sheep flock in the Basque Country, a region where piroplasmosis is endemic. To identify the causes of this outbreak, a panel of different methods, including traditional pathological, biopathological and parasitological analyses combined with recently developed molecular methods, was used. These novel molecular methods included a multiplex real-time PCR assay to screen for the presence of the most important tick-borne pathogens (piroplasms and anaplasmas), followed by

Equine piroplasmosis due to Theileria equi and Babesia caballi is endemic in Trinidad. A case of equine piroplasmosis due to T. equi was diagnosed in a thoroughbred foal at 10h post-partum. A high parasitaemia (63%) of piroplasms was observed in a Wright-Giemsa(®) stained thin blood smear from the foal. In addition, the 18S rRNA gene for Babesia/Theileria was amplified from DNA extracted from the blood of the foal and the mare. Amplified products were subjected to a reverse line blot hybridization assay (RLB), which confirmed the presence of T. equi DNA in the foal. The mare was negative by RL

A quantitative real-time polymerase chain reaction (qPCR) assay using a TaqMan minor groove binder (MGB) probe was developed for the detection of Babesia caballi infection in equids from South Africa. Nine previously published sequences of the V4 hypervariable region of the B. caballi 18S rRNA gene were used to design primers and probes to target unique, conserved regions. The B. caballi TaqMan MGB qPCR assay was shown to be efficient and specific. The detection limit, defined as the concentration at which 95% of positive samples can be detected, was determined to be 0.000114% parasitized eryt

Between October and November 2006, a total of 278 bovine blood samples were examined, and 104 (37.4%) were positive for piroplasms by microscopy. A reverse line blot hybridisation with polymerase chain reaction detected Theileria annulata, T. buffeli, Babesia bovis and B. bigemina in cattle accounting for 48.6% of positive samples. The most frequently found species was T. buffeli, which was present in 39.2% of the samples. T. annulata was found in 48 samples (17.3%). Babesia infections were less frequently detected: B. bovis was found in 6.8% of the samples and B. bigemina in 4.3%. Mixed infec

Very limited information is available on epizootiology of haematozoan infections in French domestic animals. In an attempt to address this issue, prevalence of piroplasmida was studied in carnivores and ruminants, whereas prevalence of Hepatozoon spp. was only investigated in carnivores. In total, 383 animals were included in the survey (namely 116 cats, 108 dogs, 91 sheep and 68 cows). Parasite diagnosis was carried out using molecular methods such as PCR and sequencing of the 18S rRNA gene. In addition, ruminant samples were analyzed with the reverse line blotting technique (RLB). Results of

Detection and identification of Theileria and Babesia species in 920 apparently healthy small ruminants in eastern Turkey, as well as parasite genetic diversity, was investigated using a specifically designed reverse line blot (RLB) assay. The hypervariable V4 region of the 18S ribosomal RNA (rRNA) gene was amplified and hybridized to a membrane onto which catchall and species-specific oligonucleotide probes were covalently linked. Three Theileria and one Babesia genotype were identified. Comparison of the Theileria genotypes revealed 93.6-96.2% similarity among their 18S rRNA genes. Two Theil

Ninety-three (93) horses were investigated for serum antibodies to Theileria equi (T. equi) and Babesia caballi (B. caballi) using the immunofluorescent antibody test (IFAT). Seventy-seven (82.8%) horses were seropositive; 31 (33.3%) were positive to T. equi compared to 64 (68.8%) to B. caballi while 18 (19.4%) horses were seropositive to both parasites. No significant differences in antibody frequencies among females and males for either T. equi or B. caballi were noted. Differences in seropositivity to B. caballi among age groups were not significant. Antibodies to T. equi were more frequent

Tick-borne pathogens (TBPs) represent a major constraint to livestock health and productivity worldwide. Water buffaloes (Bubalus bubalis) are an important livestock species in Türkiye; however, molecular data on TBPs in this host remain limited. This study aimed to investigate the prevalence and molecular diversity of Babesia, Theileria, Anaplasma, and Ehrlichia species in water buffaloes from Iğdır Province, Eastern Türkiye. A total of 200 blood samples were analysed using genus-specific PCR assays targeting the 18S and 16S rRNA genes, followed by Reverse Line Blot (RLB) hybridisation and Sa

Caprine piroplasmosis is caused by various species belonging to Babesia and Theileria genera and it is a major pathological condition affecting goats globally, particularly in tropical and subtropical regions, including Türkiye. In endemic areas, these infections cause huge losses and are major threat to the sustainability of goat farming. Despite their impact, data on the molecular prevalence and genetic diversity of these pathogens in Türkiye remain limited. This study aimed to determine the molecular prevalence of caprine piroplasm agents in 400 goats sampled from nine provinces of Southeas

Fifteen dairy buffaloes of a farm in the state of Kerala, India developed fatal oriental theileriosis within 2 months of their procurement. Typical piroplasms of Theileria orientalis were observed in the erythrocytes of all affected animals by Giemsa-Leishman staining of blood smears. Case fatality rate was 87·5% (seven out of eight) in the clinically progressed cases. Therapeutic management with anti-theilerial drugs buparvaquone and oxytetracycline led to recovery of seven other animals in less advanced stages of the disease. The aim of this study was to determine the reasons for increased v

Miltefosine, a membrane-active synthetic ether-lipid analogue, has antiproliferative and antiparasitic effects. In this study, the inhibitory effects of miltefosine were evaluated against three Babesia species and Theileria equi in vitro and against Babesia microti in mice. The drug showed significant growth inhibition from an initial parasitemia of 1% for Babesia bovis, Babesia bigemina, Babesia caballi, and T. equi with IC50 values of 25, 10.2, 10.4, and 99 μM, respectively. Complete inhibition was observed at 200 μM of miltefosine on the third day of culture for the three Babesia species an

Heat-shock proteins (HSPs) refer to a group of proteins whose synthesis is enhanced upon sudden increase in temperature or exposure to a variety of other stressors. In this study, Theileria annulata (T. annulata) HSP90 was identified and characterized as a first step to understand the function of this molecule in T. annulata-infected cells. Our results indicated the existence in the genome of T. annulata of two HSP90 genes: one located in chromosome one (TaHSP90-Chr1) and the other in chromosome four (TaHSP90-Chr4). The amino acid alignment between the two isoforms has shown identity and simil

A herd-based study was carried out in Central Equatoria State, Southern Sudan, to study epidemiological aspects of tick-borne diseases. Six herds of cattle situated in three different locations were selected and investigated every 3 months during the year 2005. Blood smears for Giemsa staining and blood spots on filter paper for deoxyribonucleic acid extraction were collected from 600 apparently healthy indigenous cattle. A total of 69 (11.5%) samples showed the presence of piroplasms in Giemsa-stained blood smears, and polymerase chain reaction increased the detection limit to 297 (49.5%). Us

Nucleic acid-based methods offer a variety of tools for the detection of parasites. This field of veterinary and medical sciences is rapidly evolving, leading to greater sensitivity and higher throughput. One of the reasons justifying such a fast development is the fact that tests targeting several taxa can be created. The present article deals with the applications of molecular diagnostics of tick-borne diseases in Parasitology. Special attention is given to Babesia and Theileria species infecting livestock. The commonly used molecular methods in diagnostic of tick-transmitted hematic protozo
