Transcriptional profiling of cattle infected with Trypanosoma congolense highlights gene expression signatures underlying trypanotolerance and trypanosusceptibility.

BACKGROUND: African animal trypanosomiasis (AAT) caused by tsetse fly-transmitted protozoa of the genus Trypanosoma is a major constraint on livestock and agricultural production in Africa and is among the top ten global cattle diseases impacting on the poor. Here we show that a functional genomics approach can be used to identify temporal changes in host peripheral blood mononuclear cell (PBMC) gene expression due to disease progression. We also show that major gene expression differences exist between cattle from trypanotolerant and trypanosusceptible breeds. Using bovine long oligonucleotid

Responses of bovine chimaeras combining trypanosomosis resistant and susceptible genotypes to experimental infection with Trypanosoma congolense.

West African N'Dama cattle have developed a genetic capacity to survive, reproduce and remain productive under trypanosomosis risk. The cellular and molecular bases of this so-called trypanotolerance are not known, but the trait is manifested by the N'Dama's greater capacity to control parasitaemia and anaemia development during an infection. In order to examine the role of the haematopoietic system in trypanotolerance, we have exploited the tendency for the placentas of bovine twin embryos to fuse. Placental fusion in cattle results in bone marrow chimaerism in twins. By comparison with the N

Trypanocide usage in the cattle belt of southwestern Uganda.

BACKGROUND: Systematic infrastructure and regulatory weaknesses over many decades, in communities struggling with animal African trypanosomiasis (AAT) would be expected to create an environment that would promote drug misuse and risk development of drug resistance. Here, we explore rural community practices of livestock keepers, livestock extension officers and drug shop attendants to determine whether appropriate practice was being followed in administration of trypanocides and other drugs. METHODS: A questionnaire-based survey was undertaken in southwestern Uganda in 2022 involving 451 farm

Assessment of impacts of tsetse and trypanosomosis control measures on cattle herd composition and performance in southern region, Ethiopia.

This study was conducted to assess the impact of tsetse and trypanosomosis control measures on cattle herd size and composition, herd dynamics, and milk yield in Wolaita and Gamogofa Zones, southern Ethiopia. The study showed that the average number of cattle herd size in tsetse challenged areas was significantly higher than those in tsetse-controlled areas. The number of non-pregnant dry cows, bulls and oxen were significantly higher in tsetse challenged areas than the other two study areas. The rate of cattle addition to and disposal from the herd were significantly higher in tsetse challeng

Comparing FAMACHA eye color chart and Hemoglobin Color Scale tests for detecting anemia and improving treatment of bovine trypanosomosis in West Africa.

African animal trypanosomosis (AAT) is considered the most important cattle disease in sub-Saharan Africa but its diagnosis in the field is difficult, resulting in inappropriate treatments, excessive delay in treatments and under-treatment. A field study in West Africa investigated the usefulness of anemia in the diagnosis of trypanosomosis. A total of 20,772 cattle blood samples were taken from 121 villages in 3 countries. The average packed cell volume (PCV) of trypanosomosis positive cattle was 23%, versus 28% for negative cattle. In a sub-set of animals, other causes of anemia were investi

Lack of specific alleles for the bovine chemokine (C-X-C) receptor type 4 (CXCR4) gene in West African cattle questions its role as a candidate for trypanotolerance.

A panel of 81 Asian, African and European cattle (Bos taurus and B. indicus) was analysed for the whole sequence of the CXCR4 gene (3844bp), a strong candidate for cattle trypanotolerance. Thirty-one polymorphic sites identified gave 31 different haplotypes. Neutrality tests rejected the hypothesis of either positive or purifying selection. Bayesian phylogenetic tree showed differentiation of haplotypes into two clades gathering genetic variability predating domestication. Related with clades definition, linkage disequilibrium analyses suggested the existence of one only linkage block on the C

Bovine trypanosomosis and its vectors in two districts of Bench Maji zone, South Western Ethiopia.

A cross-sectional study was carried out from November 2008 to February 2009 in Guraferda and Sheko districts of Bench Maji Zone, South Western Ethiopia. The objective of the study was to determine the prevalence of bovine trypanosomosis and the density of its vectors. An overall prevalence of trypanosome infection in the study area was 4.4%. Trypanosoma congolense (36.36%) was the dominant trypanosome species followed by Trypanosoma vivax (18.18%) and Trypanosoma brucei (9.09%). Mean packed cell volume value of parasitemic animals (21.8%) was significantly (P<0.05) lower than that of aparasite

Modulation of the phenotype and function of bovine afferent lymph cells during infection with Trypanosoma congolense.

Alterations in the phenotype and function of cells isolated from bovine afferent lymph were studied following tsetse-transmitted Trypanosoma congolense infection. Little alteration was observed in the output of the CD2+ T cells in the lymph, and within this population the CD4:CD8 ratio remained relatively constant. By contrast, a marked decrease was observed in the output of gamma delta T cells over the first 7 days following infection. The number of B cells increased between 2 and 6 days post-infection, and thereafter returned to pre-infection values. Little change was observed within the aff

Sensitivity and specificity of antigen-capture ELISAs for diagnosis of Trypanosoma congolense and Trypanosoma vivax infections in cattle.

Sensitivity and specificity of the FAO/IAEA antigen-ELISA kits for diagnosis of bovine trypanosomosis were investigated using sera from experimental cattle infected by tsetse challenge with cloned populations of Trypanosoma congolense (three populations) or T. vivax (one population). The kits are based on monoclonal antibodies that recognise internal antigens of tsetse-transmitted trypanosomes. Ten cattle were infected with each trypanosome population for at least 60 days, and in combination with uninfected cohorts (n = 16) were used in a double-blind study design. Sensitivity and specificity

Trypanotolerance and the value of conserving livestock genetic resources.

Studies have been made in two main areas of genetic research on African trypanotolerant (N'Dama) cattle. The first is of the significance for performance and the heritability of trypanotolerance traits and the second is the search for markers of the breed type and the traits of interest. Results demonstrate significant effects of the ability of an animal to control parasitaemia and anaemia on its performance. Initial estimates suggested that parasitaemia measures had a very low heritability, but ability to maintain packed cell volume levels when detected as parasitaemic and to generate an immu

Sensitive and specific detection of Trypanosoma vivax using the polymerase chain reaction.

The nucleic acid probes that are currently in use detect and distinguish Trypanosoma vivax parasites according to their geographic origin. To eliminate the need for using multiple DNA probes, a study was conducted to evaluate the suitability of a tandemly reiterated sequence which encodes a T. vivax diagnostic antigen as a single probe for detection of this parasite. The antigen is recognized by monoclonal antibody Tv27 currently employed in antigen detection ELISA (Ag-ELISA). A genomic clone which contained a tetramer of the 832-bp cDNA sequence was isolated and shown to be more sensitive tha

Genetic diversity and population structure of Trypanosoma brucei: clonality versus sexuality.

Genomic fingerprinting by arbitrarily primed PCR was used to analyze the genetic variability among 59 Trypanosoma brucei stocks representing the three T. brucei subspecies isolated from various hosts and different countries in Africa. 14 oligonucleotide primers revealed 355 polymorphic binary characters which were used for phenetic and phylogenetic analysis and to perform recombination tests exploring the linkage disequilibrium in the sample. There was good concordance between arbitrarily primed PCR polymorphisms and isoenzyme data previously collected for many of the same strains [1]. However

Pastoralists' Vulnerability to Trypanosomiasis in Maasai Steppe.

Trypanosomiasis is a neglected tropical disease of both livestock and humans. Although pastoral communities of the Maasai Steppe have been able to adapt to trypanosomiasis in the past, their traditional strategies are now constrained by changes in climate and land regimes that affect their ability to move with their herds and continually shape the communities' vulnerability to trypanosomiasis. Despite these constraints, information on communities' vulnerability and adaptive capacity to trypanosomiasis is limited. A cross-sectional study was therefore conducted in Simanjiro and Monduli district

Characterization of Calflagin, a Flagellar Calcium-Binding Protein from Trypanosoma congolense.

BACKGROUND: Identification of species-specific trypanosome molecules is important for laboratory- and field-based research into epidemiology and disease diagnosis. Although Trypanosoma congolense is the most important trypanosome pathogen of cattle in Africa, no species-specific molecules found in infective bloodstream forms (BSF) of the parasites have been identified, thus limiting development of diagnostic tests. METHODS: Immuno-mass spectrometric methods were used to identify a protein that is recognized by a T. congolense-specific monoclonal antibody (mAb) Tc6/42.6.4. The identified molec
