Molecular characterization and infectious clone construction of beet capulavirus 1 infecting tomato in Germany.

Capulavirus betae (beet capulavirus 1, BCV1) is a recently described species of the genus Capulavirus (family Geminiviridae) that was first identified in sugar beet in France. Capulaviruses are new group of geminiviruses because they are transmitted by aphids. Here, we report the identification and molecular characterization of an isolate of BCV1 infecting cultivated tomato in Germany and describe the construction of an infectious clone. In August 2025, tomato plants displaying severe virus-like symptoms, including leaf deformation, leaf curling, stunting and chlorosis were collected from two

Multicolored Asian lady beetle hypersensitivity: a case series and allergist survey.

BACKGROUND: Multicolored Asian lady beetles (Harmonia axyridis) have been used as a biological control agent against crop-destroying aphids in the United States. Outside their natural habitat, H. axyridis seeks refuge in homes during fall and winter, leading to patient complaints and symptoms of rhinitis, wheezing, and urticaria on exposure to the beetles. OBJECTIVE: To gain a better understanding of the character and spectrum of allergic disease provoked by exposure to home-infesting lady beetles. METHODS: Eight patients with allergic symptoms suspected of being caused by H. axyridis and co

Field evaluation of synthetic and neem-derived alternative insecticides in developing action thresholds against cauliflower pests.

Synthetic chemical pesticides can enhance crop yields but also have undesired effects. Alternative 'botanical insecticides' may also have non-target effects on pollinators and biocontrol services. Employing action thresholds (ATs) can reduce pesticide (whether synthetic or botanical) use compared to fixed-interval applications. Here the azadirachtin-based botanical formulation NeemAzal and a neem seed extract (NSE) were evaluated in field spraying trials alongside commonly-used synthetics (Voliam Flexi [chlorentraniliprole plus thiamethoxam] and imidacloprid) in developing ATs for the regular

The Coiled-Coil and Nucleotide Binding Domains of BROWN PLANTHOPPER RESISTANCE14 Function in Signaling and Resistance against Planthopper in Rice.

BROWN PLANTHOPPER RESISTANCE14 (BPH14), the first planthopper resistance gene isolated via map-based cloning in rice (Oryza sativa), encodes a coiled-coil, nucleotide binding site, leucine-rich repeat (CC-NB-LRR) protein. Several planthopper and aphid resistance genes encoding proteins with similar structures have recently been identified. Here, we analyzed the functions of the domains of BPH14 to identify molecular mechanisms underpinning BPH14-mediated planthopper resistance. The CC or NB domains alone or in combination (CC-NB [CN]) conferred a similar level of brown planthopper resistance t

Biosynthesis and defensive function of Nδ-acetylornithine, a jasmonate-induced Arabidopsis metabolite.

Since research on plant interactions with herbivores and pathogens is often constrained by the analysis of already known compounds, there is a need to identify new defense-related plant metabolites. The uncommon nonprotein amino acid N(δ)-acetylornithine was discovered in a targeted search for Arabidopsis thaliana metabolites that are strongly induced by the phytohormone methyl jasmonate (MeJA). Stable isotope labeling experiments show that, after MeJA elicitation, Arg, Pro, and Glu are converted to Orn, which is acetylated by NATA1 to produce N(δ)-acetylornithine. MeJA-induced N(δ)-acetylorni

Sugars in the gut of the sandfly Phlebotomus orientalis from Dinder National Park, Eastern Sudan.

The sandfly Phlebotomus orientalis Parrot (Diptera: Psychodidae) is the vector of visceral leishmaniasis in eastern and Upper Nile regions of Sudan, where vector infection rates of over 7% have been reported. Sugars are known to be important for development of the parasite and for increasing the survival and oviposition rates of several species of sandflies. In the present study we have analysed the sugars present in the guts of individuals and groups of male and female P. orientalis and compared these with sugars from several potential local plant sources: Acacia seyal, Balanites aegyptiaca a

Design, Synthesis, and Insecticidal Activity of Novel m-Diamide Compounds Containing Sulfur.

To find efficient insecticides with novel structures, 16 novel m-diamide compounds containing sulfur were designed and synthesized based on the principle of biologically active factor splicing with cyproflanilide as the lead compound. Preliminary biological assay data showed that some of the target compounds exhibited insecticidal activity above 90% against Mythimna separata at 0.01 mg/L and Aphis craccivora at 100 mg/L, which were better than cyproflanilide. Meanwhile, most compounds still maintain 100% mortality at 1 mg/L for Plutella xylostella. In particular, compounds 5c, 5e, 5f, and 5g r

Development and validation of KASP markers for the greenbug resistance gene Gb7 and the Hessian fly resistance gene H32 in wheat.

Greenbug and Hessian fly are important pests that decrease wheat production worldwide. We developed and validated breeder-friendly KASP markers for marker-assisted breeding to increase selection efficiency. Greenbug (Schizaphis graminum Rondani) and Hessian fly [Mayetiola destructor (Say)] are two major destructive insect pests of wheat (Triticum aestivum L.) throughout wheat production regions in the USA and worldwide. Greenbug and Hessian fly infestation can significantly reduce grain yield and quality. Breeding for resistance to these two pests using marker-assisted selection (MAS) is the m

A bug's life: change and transformation in early modern China.

Chanting wasps and shape-shifting worms were all in a day's work for sixteenth-century Chinese naturalists such as Li Shizhen (1518-1593). In an effort to understand the metamorphoses of both nature and the human body, he and other early modern Chinese scholars looked towards tiny creatures like roundworms, lice and demon bugs. For them, such animals could reveal the most intimate secrets of the universe.

The insect effector CSPs activate an atypical plant TNL immune receptor.

CSP-mediated activation of RCSP involves noncanonical structural and catalytic mechanisms-including BB-loop deletion, α5α6-loop insertion, and atypical catalytic residues within the RCSP-TIR domain. Chemosensory proteins (CSPs), a conserved effector family in phloem-feeding insects such as aphids and the brown planthopper (Nilaparvata lugens), induce dwarfism in Nicotiana benthamiana through recognition by the Toll-interleukin-1 receptor (TIR) domain-containing nucleotide-binding leucine-rich repeat receptor (TNL) RCSP. However, CSP-mediated RCSP activation and enzymatic mechanism remain unkno

The Glucosinolates: A Sulphur Glucoside Family of Mustard Anti-Tumour and Antimicrobial Phytochemicals of Potential Therapeutic Application.

This study reviewed aspects of the biology of two members of the glucosinolate family, namely sinigrin and glucoraphanin and their anti-tumour and antimicrobial properties. Sinigrin and glucoraphanin are converted by the β-sulphoglucosidase myrosinase or the gut microbiota into their bioactive forms, allyl isothiocyanate (AITC) and sulphoraphanin (SFN) which constitute part of a sophisticated defence system plants developed over several hundred million years of evolution to protect them from parasitic attack from aphids, ticks, bacteria or nematodes. Analyses of the Cell Wall Modification of Wheat Cultivars Infested with Diuraphis noxia.

Wheat infested with Diuraphis noxia (Russian wheat aphid: RWA) respond by inducing a range of defense processes at the morphological, physiological, molecular, and biochemical levels. The physiological response includes the cell wall modification, which is generally weakened by the RWA infestation in the susceptible cultivar, while in the resistant cultivar, the cell wall modification leads to a more robust and strengthened wall. Direct measurements of the cell wall have been a challenge, but in the last decade, extraction methods for cell wall components, i.e., cellulose, hemicellulose and li

Small impact of the host plant physiological state on the genome formula of a nanovirus.

The genome formula refers to the specific and reproducible pattern of relative accumulation of genome segments in within-host populations of viruses with segmented genomes. It has been proposed that the genome formula may have an adaptive function by allowing rapid adaptation of viral genes' expression, through variation in their relative copy number, in response to a changing environment. Consistently, the faba bean necrotic stunt virus (FBNSV) genome formula has been demonstrated to be strongly dependent on the host genotype, since it changes significantly when the same viral isolate moves f

Novel Insecticidal Butenolide-Containing Methylxanthine Derivatives: Synthesis, Crystal Structure, Biological Activity Evaluation, DFT Calculation and Molecular Docking.

The design of novel agrochemicals starting from bioactive natural products is one of the most effective ways in the discovery and development of new pesticidal agents. In this paper, a series of novel butenolide-containing methylxanthine derivatives (Ia-Ir) were designed based on natural methylxanthine caffeine and stemofoline, and the derivatized insecticide flupyradifurone of the latter. The structures of the synthesized compounds were confirmed via 1H-NMR, 13C NMR, HRMS and X-ray single crystal diffraction analyses. The biological activities of the compounds were evaluated against a variety

A recombinant fusion protein containing a spider toxin specific for the insect voltage-gated sodium ion channel shows oral toxicity towards insects of different orders.

Recombinant fusion protein technology allows specific insecticidal protein and peptide toxins to display activity in orally-delivered biopesticides. The spider venom peptide δ-amaurobitoxin-PI1a, which targets insect voltage-gated sodium channels, was fused to the "carrier" snowdrop lectin (GNA) to confer oral toxicity. The toxin itself (PI1a) and an amaurobitoxin/GNA fusion protein (PI1a/GNA) were produced using the yeast Pichia pastoris as expression host. Although both proteins caused mortality when injected into cabbage moth (Mamestra brassicae) larvae, the PI1a/GNA fusion was approximatel

Insect-specific irreversible inhibitors of acetylcholinesterase in pests including the bed bug, the eastern yellowjacket, German and American cockroaches, and the confused flour beetle.

Insecticides directed against acetylcholinesterase (AChE) are facing increased resistance among target species as well as increasing concerns for human toxicity. The result has been a resurgence of disease vectors, insects destructive to agriculture, and residential pests. We previously reported a free cysteine (Cys) residue at the entrance to the AChE active site in some insects but not higher vertebrates. We also reported Cys-targeting methanethiosulfonate molecules (AMTSn), which, under conditions that spared human AChE, caused total irreversible inhibition of aphid AChE, 95% inhibition of

Discovery of FM-2665 as a Potent Arylpyrazole Insecticidal and Acaricidal Candidate: Rational Design via Pyridine Bioisosterism and Terminal Alkyl Optimization.

Sixteen novel arylpyrazole derivatives were designed using tigolaner as a scaffold reference and synthesized to examine the effects of pyridine bioisosterism and terminal alkyl modification. Among the synthesized compounds, FM-2665 (A14) showed the highest overall activity against Plutella xylostella, Tetranychus cinnabarinu, and Myzus persicae, with LC50 values of 0.127, 0.604, and 0.496 mg/L, respectively. On the basis of the LC50 values, FM-2665 was 6.68-fold more potent than tigolaner against P. xylostella. Molecular docking and DFT calculations suggested that additional predicted contacts

Cross-kingdom signaling manipulation by insect-associated microbial symbionts: Linking molecular mechanisms to pest management strategies.

Microbial symbionts associated with herbivorous insects can modulate plant hormone networks and reconfigure induced defenses during feeding. This review synthesizes current knowledge on how symbiont effectors converge on conserved jasmonic acid (JA)/salicylic acid (SA)/ethylene (ET) signaling hubs to suppress or reprogram plant immunity. These microbial partners secrete a diverse arsenal of bioactive molecules, including effector proteins (e.g., the histidine-rich Ca2+-binding protein ApHRC from Serratia symbiotica in pea aphids), enzymes (e.g., gut microbiome-derived proteases in Anticarsia g
