Near-infrared spectral generation and regression modeling with a hybrid CVAE-1D-CNN framework: application to soil organic matter estimation.

BACKGROUND: Near-infrared (NIR) spectroscopy combined with chemometric modeling is a widely used rapid and non-destructive analytical technique, showing promise in estimating soil organic matter (SOM). However, acquiring a sufficient number of experimental spectra and corresponding SOM values is time-consuming, expensive, and often impractical, which can compromise the accuracy of regression models. Therefore, there is an urgent need for a strategy that can overcome data scarcity while maintaining robust estimation. RESULTS: This study development a hybrid framework that integrates a conditio

DVMMHGNN: A Dual View Multi-Modal Heterogeneous Graph Neural Network with Contrastive Learning for Microbe-Informed Drug Repurposing.

Drug repurposing (DR) offers an efficient and cost-effective strategy for pharmaceutical development by identifying new therapeutic applications for existing drugs. The effectiveness of this approach relies on accurately uncovering potential drug-disease associations; however, capturing the complex biological interactions underlying these associations remains a major challenge. Current computational approaches frequently overlook the critical regulatory role of the microbiota in modulating drug action pathways. Moreover, many methods fail to preserve semantic consistency during multimodal biol

AI-driven pipeline discovers ombuin as a novel M1 macrophage polarization inhibitor for sepsis treatment.

Sepsis is a life-threatening condition driven by dysregulated immune responses to infection with excessive M1 macrophage polarization-driven cytokine storm which plays a key role in the early progression of sepsis. Targeting macrophage polarization represents a promising therapeutic strategy to improve sepsis outcomes. Conventional drug discovery is hampered by high costs, long timelines and low success rates, posing significant challenges to the identification of novel M1 polarization inhibitors. In this study we constructed a novel transformer-variational autoencoder (TVAE) that integrated c

Eco-FinOps: a causal-agentic framework for energy-efficient and explainable cloud cost optimization.

Two converging crises-uncontrolled operational expenditure and significant environmental hazards-have been aggravated by the exponential scaling of cloud infrastructure. This has led to a greater focus on the efficiency of Financial Operations (FinOps). Current monitoring methods exhibit a major flaw: passive dashboards require constant human monitoring and lead to alert fatigue, whereas deep learning models require substantial Graphics Processing Unit (GPU) resources and operate as opaque "black boxes." This lack of interpretability contradicts the sustainability goals these tools promise to

CDK5-p25 Peptide Inhibitor Samples an Intrinsically Disordered Ensemble in Solution.

The 24-residue peptide p5 (KEAFWDRCLSVINLMSSKMLQINA) selectively inhibits the pathological CDK5-p25 complex implicated in Alzheimer's disease and other neurodegenerative disorders, yet the structural basis for this selectivity remains poorly understood. In this study, replica exchange with solute tempering (REST2) molecular dynamics simulations with explicit solvent were performed over 350 ns using the CHARMM36m force field to extensively characterize the conformational landscape of p5 in aqueous solution at physiological temperature. Gaussian Mixture Variational Autoencoder (GMVAE) clustering

SMAS: Structural MRI-based AD Score using Bayesian supervised VAE.

This study introduces the Structural MRI-based Alzheimer's Disease Score (SMAS), a novel index intended to quantify Alzheimer's Disease (AD)-related morphometric patterns using a deep learning Bayesian-supervised Variational Autoencoder (Bayesian-SVAE). The SMAS index was constructed using baseline structural MRI data from the DELCODE study and evaluated longitudinally in two independent cohorts: DELCODE (n=415) and ADNI (n=190). Our findings indicate that SMAS has strong associations with cognitive performance (DELCODE: r=-0.83; ADNI: r=-0.62), age (DELCODE: r=0.50; ADNI: r=0.28), hippocampal

Proteomic Learning of Gamma-Aminobutyric Acid (GABA) Receptor-Mediated Anesthesia.

Anesthetics are crucial in surgical procedures and therapeutic interventions, but they come with side effects and varying levels of effectiveness, calling for novel anesthetic agents that offer more precise and controllable effects. Targeting Gamma-aminobutyric acid (GABA) receptors, the primary inhibitory receptors in the central nervous system, could enhance their inhibitory action, potentially reducing side effects while improving the potency of anesthetics. In this study, we introduce a proteomic learning of GABA receptor-mediated anesthesia based on 24 GABA receptor subtypes by considerin

Efficient multi-view fusion and flexible adaptation to view missing in cardiovascular system signals.

The progression of deep learning and the widespread adoption of sensors have facilitated automatic multi-view fusion (MVF) about the cardiovascular system (CVS) signals. However, prevalent MVF model architecture often amalgamates CVS signals from the same temporal step but different views into a unified representation, disregarding the asynchronous nature of cardiovascular events and the inherent heterogeneity across views, leading to catastrophic view confusion. Efficient training strategies specifically tailored for MVF models to attain comprehensive representations need simultaneous conside

Deep learning classification of reading disability with regional brain volume features.

Developmental reading disability is a prevalent and often enduring problem with varied mechanisms that contribute to its phenotypic heterogeneity. This mechanistic and phenotypic variation, as well as relatively modest sample sizes, may have limited the development of accurate neuroimaging-based classifiers for reading disability, including because of the large feature space of neuroimaging datasets. An unsupervised learning model was used to reduce deformation-based data to a lower-dimensional manifold and then supervised learning models were used to classify these latent representations in a

Unsupervised abnormality detection in neonatal MRI brain scans using deep learning.

Analysis of 3D medical imaging data has been a large topic of focus in the area of Machine Learning/Artificial Intelligence, though little work has been done in algorithmic (particularly unsupervised) analysis of neonatal brain MRI's. A myriad of conditions can manifest at an early age, including neonatal encephalopathy (NE), which can result in lifelong physical consequences. As such, there is a dire need for better biomarkers of NE and other conditions. The objective of the study is to improve identification of anomalies and prognostication of neonatal MRI brain scans. We introduce a framewo

Virtual disease landscape using mechanics-informed machine learning: Application to esophageal disorders.

Esophageal disorders are related to the mechanical properties and function of the esophageal wall. Therefore, to understand the underlying fundamental mechanisms behind various esophageal disorders, it is crucial to map mechanical behavior of the esophageal wall in terms of mechanics-based parameters corresponding to altered bolus transit and increased intrabolus pressure. We present a hybrid framework that combines fluid mechanics and machine learning to identify the underlying physics of various esophageal disorders (motility disorders, eosinophilic esophagitis, reflux disease, scleroderma e

Comparative Analysis of Current Deep Learning Networks for Breast Lesion Segmentation in Ultrasound Images.

Automatic lesion segmentation in breast ultrasound (BUS) images aids in the diagnosis of breast cancer, the most common type of cancer in women. Accurate lesion segmentation in ultrasound images is a challenging task due to speckle noise, artifacts, shadows, and lesion variability in size and shape. Recently, convolutional neural networks have demonstrated impressive results in medical image segmentation tasks. However, the lack of public benchmarks and a standardized evaluation method hampers the networks' performance comparison. This work presents a benchmark of seven state-of-the-art method

Life-threatening ventricular arrhythmia prediction in patients with dilated cardiomyopathy using explainable electrocardiogram-based deep neural networks.

AIMS: While electrocardiogram (ECG) characteristics have been associated with life-threatening ventricular arrhythmias (LTVA) in dilated cardiomyopathy (DCM), they typically rely on human-derived parameters. Deep neural networks (DNNs) can discover complex ECG patterns, but the interpretation is hampered by their 'black-box' characteristics. We aimed to detect DCM patients at risk of LTVA using an inherently explainable DNN. METHODS AND RESULTS: In this two-phase study, we first developed a variational autoencoder DNN on more than 1 million 12-lead median beat ECGs, compressing the ECG into 2

GM-Pep: A High Efficiency Strategy to De Novo Design Functional Peptide Sequences.

Although peptides are regarded as ideal therapeutic agents, only a small proportion of the marketed drugs are peptides. In the past decade, pharmacists have paid great attention to the development of peptide therapeutics. Except a few approved chemically/rationally designed peptides, most attempts failed due to unsatisfactory efficacy or safety. Luckily, computation methods, such as artificial intelligence, have been utilized to accelerate the discovery of therapeutic peptides by predicting the activity, toxicity, and absorption, distribution, metabolism, and excretion of polypeptides. Diagnosis of mesothelioma with deep learning.

Malignant mesothelioma (MM) is a rare but aggressive cancer. The definitive diagnosis of MM is critical for effective treatment and has important medicolegal significance. However, the definitive diagnosis of MM is challenging due to its composite epithelial/mesenchymal pattern. The aim of the current study was to develop a deep learning method to automatically diagnose MM. A retrospective analysis of 324 participants with or without MM was performed. Significant features were selected using a genetic algorithm (GA) or a ReliefF algorithm performed in MATLAB software. Subsequently, the current
