PER2 rs2304672, CLOCK rs1801260, and PER3 rs57875989 polymorphisms are not associated with juvenile myoclonic epilepsy.

Sleep disturbance is common in several epilepsy types, such as juvenile myoclonic epilepsy (JME). Genetic background could increase susceptibility to seizure and sleep abnormalities. From this perspective, a susceptibility gene for sleep disturbance or chronotype could contribute to the genetic susceptibility threshold for epilepsy and vice versa. Accordingly, we investigated whether functional clock gene polymorphisms (PER2 111C>G, CLOCK 3111T>C, and PER3 VNTR) might influence the risk for JME. All these polymorphisms have recently been reported to be associated with sleep disturbance, diurna

Identifying IN and OUT of bedtimes in patients with advanced cancer using self-reported methods, accelerometry and automated algorithms: Implications for measures of sleep and circadian rhythmicity.

The study evaluated several approaches for identifying IN and OUT of bedtimes, comparing their reliability, accuracy, and impact on calculating sleep and circadian parameters. In a 12-month observational study, 72 adults with advanced cancer wore wrist and thigh accelerometers for 72 hours and completed sleep diaries and questionnaires on chronotype, sleep quality, and daytime sleepiness. IN and OUT times were determined using patient diaries, wrist accelerometry event markers, and automated accelerometry algorithms. Automated algorithms identified IN and OUT times more consistently than patie

Integrating human-smartphone interactions and actigraphy in digital phenotyping of obesity and depression: Social-physical coherence algorithm development and validation study.

Circadian misalignment is associated with obesity and depression, yet few tools directly quantify the alignment between physical and social activity. We developed social-physical coherence, a digital-phenotyping measure defined as the magnitude-squared coherence between wrist-actigraphy physical activity and smartphone-derived social activity at the circadian (~24-hour) frequency, indexing how closely the two rhythms are aligned. In 135 adults (mean age 43.8 ± 12.3 years; 65.2% female; 42.2% with a history of major depression; 18.5% with obesity), physical and social activity were recorded for

TCM-Driven Circadian Modulation in Cancer: Engineering Spatiotemporal Drug Delivery for Precision Oncology.

Circadian disruption constitutes a hallmark of cancer, driving tumorigenesis, progression, and therapy resistance via dysregulation of core clock genes. Emerging evidence indicates that traditional Chinese medicine (TCM), primarily through Chinese herbal medicine (CHM), exerts promising chronotherapeutic effects by modulating tumor-intrinsic circadian rhythms. Bioactive CHM constituents restore the oscillatory expression of clock genes (eg, BMAL1, PER2), thereby resynchronizing cell cycle control, DNA repair, metabolism, and immune surveillance to suppress malignancy. This review provides a co

Circadian Rhythm and Psychiatric Features in Wolfram Syndrome: Toward Chrono Diagnosis and Chronotherapy.

Background/Objectives: Wolfram syndrome is a rare neurodegenerative disorder primarily known for its multisystemic manifestations. Although classically associated with diabetes insipidus, diabetes mellitus, optic atrophy, and deafness, emerging evidence suggests a consistent pattern of executive dysfunction in many affected individuals. Methods: Based on findings from a scoping review and results obtained through the Dysexecutive Questionnaire in a Spanish patient cohort, we propose that WFS1 gene mutations-via chronic endoplasmic reticulum stress-disrupt serotonergic and cholinergic neurotran

The moderating role of protective factors in shift work disorder and health outcomes: A cross-sectional study.

This study investigated whether sleep-specific (e.g. chronotype) and traditional (e.g. resilience) protective factors were associated with reduced shift work disorder (SWD) risk and explored their role as moderators in the relationship between SWD risk and health. Shift workers (n = 126) participated in a cross-sectional study evaluating SWD risk (i.e. low vs. high; SWD-screening Questionnaire), mental health (Patient Health Questionnaire-9; Generalized Anxiety Disorder Questionnaire-7), physical health (Subjective Health Complaints Inventory), sleep (Pittsburgh Sleep Quality Index; Insomnia S

Poorer sleep health is associated with altered brain activation during cognitive control processing in healthy adults.

This study investigated how proactive and reactive cognitive control processing in the brain was associated with habitual sleep health. BOLD fMRI data were acquired from 81 healthy adults with normal sleep (41 females, age 20.96-39.58 years) during a test of cognitive control (Not-X-CPT). Sleep health was assessed in the week before MRI scanning, using both objective (actigraphy) and self-report measures. Multiple measures indicating poorer sleep health-including later/more variable sleep timing, later chronotype preference, more insomnia symptoms, and lower sleep efficiency-were associated wi

Sleep/wake cycle alterations as a cause of neurodegenerative diseases: A Mendelian randomization study.

Sleep and/or wake cycle alterations are common in neurodegenerative diseases (ND). Our aim was to determine whether there is a causal relationship between sleep and/or wake cycle patterns and ND (Parkinson's disease (PD) age at onset (AAO), Alzheimer's disease (AD), and amyotrophic lateral sclerosis (ALS)) using two-sample Mendelian Randomization (MR). We selected 12 sleep traits with available Genome-Wide Association Study (GWAS) to evaluate their causal relationship with the ND risk through Inverse-Variance Weighted regression as main analysis. We used as outcome the latest ND GWAS with avai

Serum 25-hydroxyvitamin D concentrations, sleep characteristics and the risk of incident dementia.

OBJECTIVES: Serum 25-hydroxyvitamin D concentrations have been associated with the risk of dementia, but the results are inconsistent. Previous studies have reported that vitamin D metabolism is related to sleep characteristics. We investigated the association between serum 25-hydroxyvitamin D concentrations and the risk of dementia, as well as whether sleep characteristics and sleep patterns modified this association. STUDY DESIGN: In this prospective population-based cohort study, serum 25-hydroxyvitamin D concentrations were measured. Sleep characteristics, including sleep duration, chrono

The role of the transdiagnostic sleep and circadian intervention in adolescent bedtime and sleep quality: a pilot study.

PURPOSE: Circadian misalignment is prevalent among adolescents and contributes to sleep deficiency and mental health struggles. There is a need for interventions targeting sleep and circadian difficulties in adolescents. This pilot study examined the effectiveness of a Transdiagnostic Intervention for Sleep and Circadian Dysfunction (TSC) in a community-based adolescent sample with circadian misalignment. We hypothesized that TSC would improve sleep timing and duration, lessen circadian misalignment, improve subjective sleep quality, shorten sleep onset latency, and improve mental health outco

Circadian Rhythm Disruption and Osteoporosis in Postmenopausal Women: An Observational Study from a Tertiary Care Center in India.

BACKGROUND: Osteoporosis, a common bone disease among postmenopausal women, where bone is weak by diminished bone mineral density (BMD), increasing the fracture risk. Our body's natural rhythm, called the "circadian rhythm," which is controlled by the brain and body, helps in bone formation and also in breakdown, disruption of this rhythm may affect bone health. This study explores how problems with circadian rhythm might be linked with osteoporosis in postmenopausal women. OBJECTIVE: To assess the prevalence of osteoporosis among postmenopausal women and to see if it is related to changes in

Genetic architecture of a Circadian Imbalance Index: genome-wide association, phenome-wide association, and Mendelian randomisation analyses.

BACKGROUND: Circadian disruption affects multiple aspects of human health, but the genetic architecture of individual susceptibility remains unclear. We examined the genetics of the Circadian Imbalance Index (CII), an additive 0-5 score combining evening chronotype, short/long sleep, high neuroticism, atypical caffeinated coffee intake, and low vitamin D. METHODS: We ran a genome-wide association study (GWAS) of CII in 312,935 European-ancestry UK Biobank participants, compared signals with component-specific and leave-one-component-out (LOCO) GWASs, and tested sex and shift work interactions

Hemispheric efficiency during the day: the influence of circadian preference and time-of-day on verbal and visuospatial tasks.

INTRODUCTION: Cognitive performance fluctuates daily due to the interaction between circadian and homeostatic processes, which may differently activate the two hemispheres. This study investigated the effects of time-of-day, circadian typology, and the synchrony effect on semantic categorization and visuospatial processing tasks with potential hemispheric lateralization. METHODS: A sample of 113 healthy young women (20.60 ± 2.60 years) performed two divided visual field tasks (semantic categorization and vertical landmark) in morning (08:00) and early afternoon (13:00) sessions. RESULTS: In

Examining the relationship between sleep disturbance and ICD-11 personality disorder in a community sample.

There is a substantial literature on the associations between personality disorder (PD) and sleep dysfunction, but the majority of studies have focused on borderline PD. Studies that have taken a broader perspective have indicated that associations between PD and sleep problems are diffuse. The current study examined associations between the International Classification of Diseases, 11th revision (ICD-11) PD (severity and traits) with a diverse set of sleep problems as well as the role of adverse childhood experiences (ACEs) and executive functioning in explaining the overlap between PD and sl

Sweetened caffeine drinking revealed behavioral rhythm independent of the central circadian clock in male mice.

Caffeine consumption is associated with the evening chronotype, and caffeine administration in mice results in prolonged period of the circadian rhythm in locomotor activity. However, as caffeine is bitter, sweetened caffeine is preferred by humans and mice; yet, its impact on the circadian clock has not been explored. In this study, mice were provided with freely available sweetened caffeine to investigate its effects on behavioral rhythms and peripheral clocks. Mice that freely consumed sweetened caffeine shifted from nocturnal to diurnal activity rhythms. In addition to the light-dark entra

Disentangling the Genetic Landscape of Peripartum Depression: A Multi-Polygenic Machine Learning Approach on an Italian Sample.

BACKGROUND: The genetic determinants of peripartum depression (PPD) are not fully understood. Using a multi-polygenic score approach, we characterized the relationship between genome-wide information and the history of PPD in patients with mood disorders, with the hypothesis that multiple polygenic risk scores (PRSs) could potentially influence the development of PPD. METHODS: We calculated 341 PRSs for 178 parous mood disorder inpatients affected by major depressive disorder (MDD) or bipolar disorder (BD) with (n = 62) and without (n = 116) a history of PPD. We used partial least squares reg

Selective amplification of ipRGC signals accounts for interictal photophobia in migraine.

Second only to headache, photophobia is the most debilitating symptom reported by people with migraine. While the melanopsin-containing intrinsically photosensitive retinal ganglion cells (ipRGCs) are thought to play a role, how cone and melanopsin signals are integrated in this pathway to produce visual discomfort is poorly understood. We studied 60 people: 20 without headache and 20 each with interictal photophobia from migraine with or without visual aura. Participants viewed pulses of spectral change that selectively targeted melanopsin, the cones, or both and rated the degree of visual di
