Ginkgolide B Protects Cardiomyocytes from Angiotensin II-Induced Hypertrophy via Regulation of Autophagy through SIRT1-FoxO1.

Ginkgolide B (GB) is an active ingredient extracted from Ginkgo biloba leaves. However, the effects of GB on cardiac hypertrophy remain unclear. The study is aimed at determining whether GB could alleviate cardiac hypertrophy and exploring its underlying molecular mechanism. Rat cardiomyocyte cell line H9c2 cells were pretreated with GB and incubated with angiotensin II (Ang II) to simulate an in vitro cardiac hypertrophy model. Cell viability, cell size, hypertrophy markers, and autophagy were determined in H9c2 cells after Ang II treatment. Proteins involved in autophagy and the SIRT1 pathwa

Ginkgolic acid promotes autophagy-dependent clearance of intracellular alpha-synuclein aggregates.

The accumulation of intracytoplasmic inclusion bodies (Lewy bodies) composed of aggregates of the alpha-synuclein (α-syn) protein is the principal pathological characteristic of Parkinson's disease (PD) and may lead to degeneration of dopaminergic neurons. To date there is no medication that can promote the efficient clearance of these pathological aggregates. In this study, the effect on α-syn aggregate clearance of ginkgolic acid (GA), a natural compound extracted from Ginkgo biloba leaves that inhibits SUMOylation amongst other pathways, was assessed in SH-SY5Y neuroblastoma cells and rat p

Chaetoglobosins from Chaetomium globosum, an endophytic fungus in Ginkgo biloba, and their phytotoxic and cytotoxic activities.

In preceding studies, cultivation of Chaetomium globosum, an endophytic fungus in Ginkgo biloba, produced five cytochalasan mycotoxins, chaetoglobosins A, G, V, Vb, and C (1-5), in three media. In the present work, five known chaetoglobosins, C, E, F, Fex, and 20-dihydrochaetoglobosin A (5-9), together with the four known compounds (11-14), were isolated from the MeOH extracts of the solid culture of the same endophyte. The structures of these metabolites were elucidated on the basis of spectroscopic analysis. Treatment of chaetoglobosin F (7) with (diethylamino)sulfur trifluoride (DAST) in di

Pentoxifylline uses in inner ear diseases.

BACKGROUND: Labyrinth or the inner ear consists of the cochlea (for hearing) and vestibular system (for balance), with disorders affecting hearing, balance, or both, and symptomatology including hearing loss, tinnitus, and vertigo. Regulatory-approved medications for inner ear diseases are rare worldwide relative to the frequency of those diseases. There are no FDA-approved medications for any inner ear disease. This is due to multiple reasons, including the lack of conclusive evidence for various drugs that have been investigated. We aim to contribute to the review endeavor by addressing pent

Primary Vascular Dysregulation in Normal-Tension Glaucoma: Pathophysiology and Management.

Normal-tension glaucoma (NTG) is a progressive optic neuropathy occurring in the absence of elevated intraocular pressure (IOP), implying that mechanisms beyond mechanical stress drive axonal loss. Primary vascular dysregulation (PVD), a constitutional impairment of vascular tone regulation independent of structural disease, is increasingly recognized as a central pathogenetic contributor to NTG. This review examines the evidence linking PVD to NTG, explores underlying mechanisms, and evaluates current and emerging management strategies targeting vascular dysregulation. A comprehensive narrati

Targeting the LRRK2 G2019S Mutation Found in Parkinson's Disease: Efficiency of Biosynthesized Solid Lipid Nanoparticles for Therapeutic siRNA-Mediated Gene Therapy.

BACKGROUND: Parkinson's disease (PD) is a progressive neurodegenerative disorder with which the leucine-Rich repeat kinase 2 glycine 2019 serine (LRRK2 G2019S) mutation is strongly associated. This mutation elevates kinase activity, disrupts mitochondrial function, increases reactive oxygen species (ROS) production, and impairs DNA repair mechanisms, all of which contribute to the pathogenesis of PD. Thus, addressing these pathological features through targeted delivery systems holds promise for more effective therapies. METHODS: This study aimed to investigate the use of Ginkgo biloba leaf e

Neuropharmacological evaluation of Ginkgo biloba phytosomes in rodents.

On oral administration, Ginkgo biloba phytosomes significantly reduced pentobarbitone-induced sleeping time, produced an alteration in the general behaviour pattern, increased spontaneous motility and inhibited the chlorpromazine-induced blockade of conditioned and unconditioned responses in rodents. They exhibited both antiamnestic and antidepressant activities in the scopolamine-induced amnesia test and behavioural despair test, respectively. However, the phytosomes failed to show anticonvulsant activity. The observations suggest that the G. biloba phytosomes possess moderate antiamnestic/no

Ginkgolide B as a Candidate Gerotherapeutic: Evidence, Evidentiary Limits, and Translational Requirements.

Aging is a systemic process in which redox imbalance, cellular senescence, progenitor-cell dysfunction and chronic inflammation converge on frailty and functional decline, and geroscience seeks compounds that modulate several at once. What is more often missing is an explicit standard for deciding when a compound qualifies as a candidate gerotherapeutic. Ginkgolide B (GB) is a structurally defined diterpene trilactone from Ginkgo biloba and a potent antagonist of the platelet-activating factor receptor. This Review scores GB against ten stated candidacy criteria and grades the supporting evide
