Microbial Anti-Inflammatory Molecule (MAM) secreted by Faecalibacterium prausnitzii ameliorates colitis through autophagy and gut microbiota modulation.

OBJECTIVE: Faecalibacterium prausnitzii is a major commensal bacterium that plays a crucial role in intestinal homeostasis. Its secretes an microbial anti-inflammatory molecule (MAM) that functions as a potential therapeutic agent in inflammatory bowel disease (IBD). However, the molecular mechanisms through which MAM exerts its beneficial effects remain incompletely understood. Here, we investigate whether MAM modulates autophagy and to evaluate its therapeutic potential in a murine model of colitis. METHODS: A genetically engineered Lactococcus lactis strain expressing MAM was administered t

Evaluation of antimicrobial efficacy against spoilage microorganisms and impact on beverage color and turbidity of commercial preservatives derived from edible mushrooms.

The antimicrobial efficacy and the impact on sensory quality of preservatives from different natural sources can vary dramatically. This makes it challenging for manufacturers to incorporate natural preservatives in shelf-stable beverage formulation. This study compares new-to-market natural glycolipids (NG) derived from Dacryopinax spathularia and white button mushroom extract (WBME) containing chitosan from Agaricus bisporus. The antimicrobial efficacy was evaluated in a microbial spoilage challenge study wherein three-species suspensions of yeasts (Saccharomyces cerevisiae YG25, Zygosacchar

Development of innate immune memory by non-immune cells during Staphylococcus aureus infection depends on reactive oxygen species.

INTRODUCTION: The mechanisms underlying innate immune memory (trained immunity) comprise epigenetic reprogramming of transcriptional pathways associated with alterations of intracellular metabolism. While the mechanisms of innate immune memory carried out by immune cells are well characterized, such processes in non-immune cells, are poorly understood. The opportunistic pathogen, Staphylococcus aureus, is responsible for a multitude of human diseases, including pneumonia, endocarditis and osteomyelitis, as well as animal infections, including chronic cattle mastitis that are extremely difficul

GadR4 mediates the acid resistance and pathogenicity of Listeria monocytogenes 10403S by negatively regulating the gadT2/gadD2 cluster.

Listeria monocytogenes is an important foodborne pathogen that can survive under acidic conditions. The glutamate decarboxylase (GAD) system is one of the acid resistance systems of L. monocytogenes. It usually comprises two glutamate transporters (GadT1/T2) and three glutamate decarboxylases (GadD1/D2/D3). Among them, gadT2/gadD2 contributes most significantly to the acid resistance of L. monocytogenes. However, the regulation mechanisms of gadT2/gadD2 still remain unclear. The results of this study indicated that gadT2/gadD2 deletion significantly decreases the survival rate of L. monocytoge

A Single Cysteine Residue in the Translocation Pathway of the Mitosomal ADP/ATP Carrier from Cryptosporidium parvum Confers a Broad Nucleotide Specificity.

Cryptosporidiumparvum is a clinically important eukaryotic parasite that causes the disease cryptosporidiosis, which manifests with gastroenteritis-like symptoms. The protist has mitosomes, which are organelles of mitochondrial origin that have only been partially characterized. The genome encodes a highly reduced set of transport proteins of the SLC25 mitochondrial carrier family of unknown function. Here, we have studied the transport properties of one member of the C. parvum carrier family, demonstrating that it resembles the mitochondrial ADP/ATP carrier of eukaryotes. However, this carrie

Role of surface protein SasG in biofilm formation by Staphylococcus aureus.

The SasG surface protein of Staphylococcus aureus has been shown to promote the formation of biofilm. SasG comprises an N-terminal A domain and repeated B domains. Here we demonstrate that SasG is involved in the accumulation phase of biofilm, a process that requires a physiological concentration of Zn(2+). The B domains, but not the A domain, are required. Purified recombinant B domain protein can form dimers in vitro in a Zn(2+)-dependent fashion. Furthermore, the protein can bind to cells that have B domains anchored to their surface and block biofilm formation. The full-length SasG protein

Intestinal microbiome: a new target for chalaziosis treatment in children?

Intestinal dysbiosis (changes in the gut commensal microbiome) is related to several ophthalmic diseases. The aim of this study was to verify whether oral specific probiotics can alter the clinical course of chalaziosis and its recurrence. A prospective comparative pilot study involving 26 children suffering from chalaziosis was conducted. Children were randomly divided into two groups. The first group received medical treatment (lid hygiene, warm compression and dexamethasone/tobramycin ointment for at least 20 days), and the second group received medical treatment plus a daily supplementatio

SdrF, a Staphylococcus epidermidis surface protein, contributes to the initiation of ventricular assist device driveline-related infections.

Staphylococcus epidermidis remains the predominant pathogen in prosthetic-device infections. Ventricular assist devices, a recently developed form of therapy for end-stage congestive heart failure, have had considerable success. However, infections, most often caused by Staphylococcus epidermidis, have limited their long-term use. The transcutaneous driveline entry site acts as a potential portal of entry for bacteria, allowing development of either localized or systemic infections. A novel in vitro binding assay using explanted drivelines obtained from patients undergoing transplantation and

Antibiotic Resistance-Susceptibility Profiles of Enterococcus faecalis and Streptococcus spp. From the Human Vagina, and Genome Analysis of the Genetic Basis of Intrinsic and Acquired Resistances.

The spread of antibiotic resistance is a major public health concern worldwide. Commensal bacteria from the human genitourinary tract can act as reservoirs of resistance genes playing a role in their transfer to pathogens. In this study, the minimum inhibitory concentration of 16 antibiotics to 15 isolates from the human vagina, identified as Enterococcus faecalis, Streptococcus anginosus, and Streptococcus salivarius, was determined. Eight isolates were considered resistant to tetracycline, five to clindamycin and quinupristin-dalfopristin, and four to rifampicin. To investigate the presence

The Effect of Whey on Performance, Gut Health and Bone Morphology Parameters in Broiler Chicks.

Whey is a highly nutritious byproduct of the cheese industry that can be used effectively in the animal feed industry. However, the use of whey in poultry diets is limited by its high lactose and mineral contents. The objective of the present study was to evaluate the effect of different concentrations of whey in poultry diets on the performance, intestinal microbiota and physico-chemical parameters of the intestinal ecosystem, as well as on the bone morphology and its strength in broiler chicks. One hundred and twenty-eight, day-old, male broiler chicks were randomly allocated into four treat

Impact of GAstV-2 infection on intestinal microbiota and renal gene transcriptome in goslings.

Goose astrovirus-2 (GAstV-2) is the major etiological agent responsible for hyperuricemia and visceral gout in goslings, which causes substantial economic losses to the goose-raising industry. However, the complete pathogenic mechanisms underlying GAstV-2-induced hyperuricemia have not been fully elucidated. In this study, 1-day-old healthy Yangzhou goslings were orally inoculated with GAstV-2 to establish a viral infection model. The effects of GAstV-2 infection on growth performance, serum uric acid levels, cecal microbiota structure, and renal gene expression of goslings were systematically

Characterization of the Immune Response of MERS-CoV Vaccine Candidates Derived from Two Different Vectors in Mice.

Middle East respiratory syndrome (MERS) is an acute, high-mortality-rate, severe infectious disease caused by an emerging MERS coronavirus (MERS-CoV) that causes severe respiratory diseases. The continuous spread and great pandemic potential of MERS-CoV make it necessarily important to develop effective vaccines. We previously demonstrated that the application of Gram-positive enhancer matrix (GEM) particles as a bacterial vector displaying the MERS-CoV receptor-binding domain (RBD) is a very promising MERS vaccine candidate that is capable of producing potential neutralization antibodies. Antilisterial activity of raw sheep milk from two native Epirus breeds: Culture-dependent identification, bacteriocin gene detection and primary safety evaluation of the antagonistic LAB biota.

Raw milk from native small ruminant breeds in Epirus, Greece, is a valuable natural source of autochthonous lactic acid bacteria (LAB) strains with superior biotechnological properties. In this study, two bulk milks (RM1, RM2) from two local sheep yards, intended for traditional Kefalotyri cheese production, were preselected for bacteriocin-like antilisterial activity by in vitro tests. Their antagonistic LAB biota was quantified followed by polyphasic (16S rRNA gene sequencing; IGS for Enterococcus; a multiplex-PCR for Leuconostoc) identification of 42 LAB (RM1/18; RM2/24) isolates further ev

Regulation of PI-2b Pilus Expression in Hypervirulent Streptococcus agalactiae ST-17 BM110.

The widely spread Streptococcus agalactiae (also known as Group B Streptococcus, GBS) "hypervirulent" ST17 clone is strongly associated with neonatal meningitis. The PI-2b locus is mainly found in ST17 strains but is also present in a few non ST17 human isolates such as the ST-7 prototype strain A909. Here, we analysed the expression of the PI-2b pilus in the ST17 strain BM110 as compared to the non ST17 A909. Comparative genome analyses revealed the presence of a 43-base pair (bp) hairpin-like structure in the upstream region of PI-2b operon in all 26 ST17 genomes, which was absent in the 8 n

A rapid biosensor for viable B. anthracis spores.

A simple membrane-strip-based biosensor assay has been combined with a nucleic acid sequence-based amplification (NASBA) reaction for rapid (4 h) detection of a small number (ten) of viable B. anthracis spores. The biosensor is based on identification of a unique mRNA sequence from one of the anthrax toxin genes, the protective antigen ( pag), encoded on the toxin plasmid, pXO1, and thus provides high specificity toward B. anthracis. Previously, the anthrax toxins activator ( atxA) mRNA had been used in our laboratory for the development of a biosensor for the detection of a single B. anthraci

ABC and MFS efflux transporters mediate antibiotic resistance and bile salt tolerance in Enterococcus faecalis.

Bacteria that reside in the gastrointestinal tract must adapt to the antibacterial properties of bile salts (BSs) and other compounds. We investigated five putative transporters of the enteric-colonizing Enterococcus faecalis for their roles in adaptation to BS and antibiotic resistance. Transcript levels of ef2593 and ef0575 increased over fivefold, and ef0420 increased 12-fold when exposed to 0.02% and 0.1% BS concentrations. No change in the expression of ef2592 or ef1814 was seen. E. faecalis wild-type MMH594 overexpressing efflux pumps by plasmid constructs (EF2592, EF2593, EF0575, EF0420

A Pfs48/45-based vaccine to block Plasmodium falciparum transmission: phase 1, open-label, clinical trial.

BACKGROUND: The stalling global progress in malaria control highlights the need for novel tools for malaria elimination, including transmission-blocking vaccines. Transmission-blocking vaccines aim to induce human antibodies that block parasite development in the mosquito and mosquitoes becoming infectious. The Pfs48/45 protein is a leading Plasmodium falciparum transmission-blocking vaccine candidate. The R0.6C fusion protein, consisting of Pfs48/45 domain 3 (6C) and the N-terminal region of P. falciparum glutamate-rich protein (R0), has previously been produced in Lactococcus lactis and elic
