Elevated CO2, warming and drought differentially impact reproductive and vegetative economic traits in two grassland species.

BACKGROUND AND AIMS: Since the Industrial Revolution, rising atmospheric CO2, warming and more frequent droughts have significantly impacted ecosystems. While the responses of leaf functional traits to these climate change factors have been widely studied, reproductive traits remain relatively understudied, despite their key role in the diversification and distribution of flowering plants. Here we investigated how elevated CO2, warming, drought and their interactions affect floral, leaf and seed traits in two model grassland species. We also examined how these factors influence trait coordinat

N-propargylglycine restores survival by preventing calcium oxalate stone formation, tubular injury, and kidney dysfunction in a lethal mouse model of primary hyperoxaluria type 2.

INTRODUCTION: New therapeutics are needed to address the rapid progression of calcium oxalate (CaOx) nephrolithiasis and life-threatening kidney failure afflicting infants and young adults with one of the three different genetic types of Primary Hyperoxaluria (PH) types 1, 2, and 3. Glyoxylate and hydroxypyruvate reductase knockout (Grhpr KO) mice recapitulate the pathophysiology of PH type 2 (PH2), developing accelerated hyperoxaluria and CaOx kidney stone formation. Previous studies have shown that this process can be mitigated by introducing an additional genetic knockout of the liver and k

Neferine, a major alkaloid from the traditional Chinese medicine Lian Zi Xin, alleviates renal fibrosis by targeting GSK3β and β-catenin signaling.

ETHNOPHARMACOLOGICAL RELEVANCE: Neferine (Nef) is the primary bioactive alkaloid of lotus seed germ (Lian Zi Xin), a traditional Chinese medicine long used for 'clearing heart fire' and ' eliminating internal heat, and regulating body fluid metabolism (e.g., promoting diuresis and reducing edema)'. From the perspective of modern medical concepts, 'internal heat' and 'fluid imbalance' often correspond to conditions such as impaired kidney function. AIM OF THE STUDY: To investigate the protective effects of Nef against renal injury in mice and identify the underlying mechanisms. MATERIALS AND

Nuciferine Ameliorates Ovariectomy-Induced Bone Loss via TFEB-Mediated Autophagy-Lysosomal Pathway.

Background: Postmenopausal osteoporosis is a common metabolic bone disease that leads to reduced bone formation and increased fracture risk. Nuciferine, an aporphine alkaloid from the lotus leaf, can regulate the TFEB-mediated autophagy-lysosomal pathway. This pathway is critical for osteogenic differentiation. However, it remains unclear whether nuciferine promotes bone formation and alleviates bone loss through this pathway. We therefore hypothesized that nuciferine activates the TFEB-mediated autophagy-lysosomal pathway to promote osteogenic differentiation, thereby counteracting estrogen d

Activation of branched chain amino acid catabolism protects against nephrotoxic acute kidney injury.

Acute kidney injury (AKI) is a major risk factor for chronic kidney disease (CKD), and there are currently no therapies for AKI. Proximal tubules (PTs) are particularly susceptible to AKI, due to nephrotoxins such as aristolochic acid I (AAI). Normal PTs use fatty acid oxidation and branched chain amino acid (BCAA; valine, leucine, and isoleucine) catabolism to generate ATP; however, in AKI, these pathways are downregulated. Our aim was to investigate the utility of a pharmacological activator of BCAA catabolism, BT2, in preventing nephrotoxic AKI. Mice were administered two injections of AAI

Vulvo-vaginal reconstruction after radical excision for treatment of vulvar cancer: Evaluation of feasibility and morbidity of different surgical techniques.

BACKGROUND: Vulvar cancer is a rare disease accounting for approximately 5% of female genital tract tumors worldwide. Currently surgery represents the mainstay alone or sometimes, in combination with chemo-radiotherapy, for locally advanced tumors and recurrent disease. However, significant physical and sexual impairment mostly due to anatomical distortion of external genitalia are a consequence of radical surgical treatment. Postoperative reconstruction after demolitive surgeries improves aesthetic and functional results, guarantees an adequate coverage of large tumors and assures safe surgic

Single centre experience with transapical transcatheter mitral valve implantation.

OBJECTIVES: A transcatheter heart valve technique can be used in failed mitral valve repairs with annuloplasty rings, deterioriated bioprostheses and in mitral annular calcification, all serving as 'docking stations' for balloon-expandable valves. Specially designed transcatheter mitral valve platforms are used in ongoing studies for native mitral valve regurgitation. We present our single centre experience with transcatheter mitral valve implantation-transapical approach procedures in eleven patients. METHODS: Eleven patients were treated between 2011 and 2016. They had severe mitral regurgi

Difference in the ability of blood group-specific lectins and monoclonal antibodies to recognize the ABH antigens in human tissues.

Twelve different kinds of blood group-specific lectins have been used along with monoclonal anti-A, -B and -H antibodies for detecting the corresponding antigens in selected human tissues. Although most of the lectins recognized the antigens in the tissue sections examined, they displayed marked differences in their recognition patterns in certain tissues. Helix asparsa agglutinin (HAA), Helix pomatia agglutinin (HPA) and monoclonal anti-A antibody recognized A antigens in the mucous cells of salivary glands from blood group A or AB nonsecretor as well as secretor individuals, whereas Dolichos

Breaking the Barriers: Machine-Learning-Based c-RASAR Approach for Accurate Blood-Brain Barrier Permeability Prediction.

The intricate nature of the blood-brain barrier (BBB) poses a significant challenge in predicting drug permeability, which is crucial for assessing central nervous system (CNS) drug efficacy and safety. This research utilizes an innovative approach, the classification read-across structure-activity relationship (c-RASAR) framework, that leverages machine learning (ML) to enhance the accuracy of BBB permeability predictions. The c-RASAR framework seamlessly integrates principles from both read-across and QSAR methodologies, underscoring the need to consider similarity-related aspects during the

Self-Cleaning of Hydrophobic Rough Surfaces by Coalescence-Induced Wetting Transition.

The superhydrophobic leaves of a lotus plant and other natural surfaces with self-cleaning function have been studied intensively for the development of artificial biomimetic surfaces. The surface roughness generated by hierarchical structures is a crucial property required for superhydrophobicity and self-cleaning. Here, we demonstrate a novel self-cleaning mechanism of textured surfaces attributed to a spontaneous coalescence-induced wetting transition. We focus on the wetting transition as it represents a new mechanism, which can explain why droplets on rough surfaces are able to change fro

Immortalized epithelial cells from human autosomal dominant polycystic kidney cysts.

Autosomal dominant polycystic kidney disease (ADPKD) is the result of mutations in one allele of the PKD1 or PKD2 genes, followed by "second hit" somatic mutations of the other allele in renal tubule cells. Continued proliferation of clonal cells originating from different nephron segments leads to cyst formation. In vitro studies of the mechanisms of cyst formation have been hampered by the scarcity of nephrectomy specimens and the limited life span of cyst-derived cells in primary culture. We describe the development of a series of immortalized epithelial cell lines from over 30 individual r

A Comprehensive Study Utilizing QSAR, Virtual Screening, Molecular Docking, Molecular Dynamics, and MM/GBSA Analyses Reveals Natural Diterpenoids as Promising Caspase-1 Inhibitors.

Caspase-1 is a crucial inflammatory cysteine protease that facilitates the maturation of pro-inflammatory cytokines such as interleukin-1β and interleukin-18, making it a significant therapeutic target for inflammatory diseases. However, existing caspase-1 inhibitors often face challenges like toxicity and suboptimal drug-like properties, underscoring the need for new inhibitors. This study employed an integrated computational strategy, combining quantitative structure-activity relationship (QSAR) modeling and application of this validated model to a large natural product database followed by

In-silico evaluation of natural alkaloids against the main protease and spike glycoprotein as potential therapeutic agents for SARS-CoV-2.

Severe Acute Respiratory Syndrome Corona Virus (SARS-CoV-2) is the causative agent of COVID-19 pandemic, which has resulted in global fatalities since late December 2019. Alkaloids play a significant role in drug design for various antiviral diseases, which makes them viable candidates for treating COVID-19. To identify potential antiviral agents, 102 known alkaloids were subjected to docking studies against the two key targets of SARS-CoV-2, namely the spike glycoprotein and main protease. The spike glycoprotein is vital for mediating viral entry into host cells, and main protease plays a cru

Woven-like change following intracoronary thrombosis recanalization: a case report.

BACKGROUND: A woven coronary artery is a rare congenital coronary anomaly and incidentally found in coronary angiogram. Coronary angiography is the major diagnostic modality, which shows the main trunk of coronary divides into several channels which later reconnect with normal blood flow (J Int Cardiol 113:121-123 2006). However, some cases and reviews inferred that this characteristic might be mimicked by recanalized coronary thrombus, which occurs following thrombotic occlusion. In some case, the multiple intraluminal channels have a 'Swiss cheese', a 'Spider web-like', a 'Honeycomb' or a 'L
