Mechanisms regulating expression of the HPV 31 L1 and L2 capsid proteins and pseudovirion entry.

Human papillomaviruses (HPV) infect stratified epithelia and restrict expression of late capsid genes to highly differentiated cells. In order to begin to understand the processes regulating HPV 31 infection we examined the synthesis of the HPV 31 capsid proteins, L1 and L2, using heterologous expression systems. Similar to studies in HPV 16, expression of wild type HPV 31 L1 and L2 from heterologous promoters resulted in very low levels of synthesis. In contrast, modification of the codons in the capsid genes to ones more commonly used in cellular genes resulted in high-level synthesis. Throu

Potent cytotoxicity of an auristatin-containing antibody-drug conjugate targeting melanoma cells expressing melanotransferrin/p97.

Identifying factors that determine the sensitivity or resistance of cancer cells to cytotoxicity by antibody-drug conjugates is essential in the development of such conjugates for therapy. Here the monoclonal antibody L49 is used to target melanotransferrin, a glycosylphosphatidylinositol-anchored glycoprotein first identified as p97, a cell-surface marker in melanomas. L49 was conjugated via a proteolytically cleavable valine-citrulline linker to the antimitotic drug, monomethylauristatin F (vcMMAF). Effective drug release from L49-vcMMAF likely requires cellular proteases most commonly locat

Regulation of lipid metabolism by soy protein and its implication in diseases mediated by lipid disorders.

Soybeans have a high-quality protein that has been consumed for approximately 5000 years in Oriental countries. The awareness that soy products are healthy has increased their consumption in Western countries. Substantial data from epidemiological surveys and nutritional interventions in humans and animals indicate that soy protein reduces serum total and low-density lipoprotein (LDL) cholesterol and triglycerides as well as hepatic cholesterol and triglycerides. This review examines the evidence on the possible mechanisms for which soy protein has beneficial effects in diabetes, obesity and s

A CAV3 microdeletion differentially affects skeletal muscle and myocardium.

BACKGROUND: Caveolin-3 is the muscle-specific protein product of the caveolin gene family and an integral membrane component of caveolae. Mutations in the gene encoding caveolin-3 (CAV3) underlie four distinct disorders of skeletal muscle: the autosomal dominant form of limb-girdle muscular dystrophy type 1C (LGMD-1C), rippling muscle disease (RMD), sporadic and familial forms of hyperCKemia, and distal myopathy. OBJECTIVE: To characterize a multigenerational Italian family affected by an autosomal dominant myopathic disorder and to assess the expression of caveolin-3, dystrophin, dystrophin-

Subcellular targeting and agonist-induced site-specific phosphorylation of endothelial nitric-oxide synthase.

The endothelial isoform of nitric-oxide synthase (eNOS) undergoes a complex pattern of covalent modifications, including acylation with the fatty acids myristate and palmitate as well as phosphorylation on multiple sites. eNOS acylation is a key determinant for the reversible subcellular targeting of the enzyme to plasmalemmal caveolae. We transfected a series of hemagglutinin epitope-tagged eNOS mutant cDNAs deficient in palmitoylation (palm(-)) and/or myristoylation (myr(-)) into bovine aortic endothelial cells; after treatment with the eNOS agonists sphingosine 1-phosphate or vascular endot

Flotillin 2 is distinct from epidermal surface antigen (ESA) and is associated with filopodia formation.

ECS-1, a monoclonal antibody (MoAb) raised to cultured human keratinocytes, stains the intercellular glycocalyx with a pemphigus-like pattern and recognizes a 35-kDa epidermal surface antigen (ESA) on Western blotting of keratinocyte extracts. When ECS-1 MoAb was used to screen a keratinocyte expression library, a unique cDNA was identified that predicted a 42-kDa globular protein of unknown function. This putative ESA was conserved between mice and humans and was encoded by a gene on chromosome 17q11-12 in linkage with neurofibromin. Homology between the cDNA sequence has been reported with f

Dual-transporter-targeted oral semaglutide nanomicelles enable enhanced intestinal absorption and metabolic reprogramming in obesity and diabetes.

Obesity is a chronic, relapsing metabolic disorder associated with insulin resistance, dyslipidemia, and related complications. Although glucagon-like peptide-1 receptor agonists (GLP-1 RAs), particularly semaglutide (SEMA), exhibit robust therapeutic efficacy, their clinical application is limited by low oral bioavailability. Here, we report a dual-transporter-targeted oral nanomicellar platform (SGT-M) designed to overcome these limitations. SEMA was electrostatically complexed with glucosamine and taurolithocholate to enable transporter-mediated recognition, followed by incorporation into a

Engineering a Virus-like Nanovaccine for Durable and Potent Antigen-Specific Adoptive Immunity.

The topological characteristics of microbial surfaces play pivotal roles in immune responses. However, integrating adjuvants and antigens to construct nanovaccines that mimic pathogenic surface nanostructures, especially for antigen delivery and immune activation, remains unexplored. Herein, we systematically investigated the antigen delivery efficiency and immunostimulatory capacity of nanovaccines with distinct morphologies: MO@SP (spherical), MO@ST (fibrous), MO@LA (lamellar), and MO@VLP (rough-surfaced spiky-like), synthesized through the self-assembly of manganese ions (Mn2+) with antigen

Relaxin-2: Shaping the Proteomic Landscape of Skeletal Muscle Physiology, Glucose Trafficking, and Mitochondrial Function in Rat.

Relaxin-2 is a hormone with robust beneficial effects on the heart and blood vessels and potential as a therapy for cardiovascular (CV) disease. Considering the interorgan communication between skeletal muscle and heart, and the relation between muscle quality/composition and CV events, we hypothesize that relaxin-2 may regulate skeletal muscle physiology and metabolism. We aim to evaluate the impact of relaxin-2 on the proteome of skeletal muscle from healthy Sprague-Dawley rats. Animals were treated with 0.4 mg/kg/day of serelaxin (recombinant form of human relaxin-2) or vehicle (PBS) for 2

Designing influenza virus-derived cell-penetrating peptides for antigen delivery: Integrating uptake efficiency, safety, and receptor targeting.

Successful antigen delivery is of paramount importance for successful vaccination. Cell-penetrating peptides (CPPs) offer a highly effective means of delivering antigens, nucleic acids, and other drug compounds to cells. However, their mechanisms of action remain poorly understood and require further investigation. This study sought to identify novel CPPs within the influenza virus proteome using computational screening methods for vaccine and antigen delivery. CPP candidates were screened from major influenza proteins using CellPPD, C2Pred, and PreTP-EL. Their efficiencies in uptake, physicoc

Association of Sleep Duration and Variability With Body Mass Index: Sleep Measurements in a Large US Population of Wearable Sensor Users.

PURPOSE: Ocular infection by human adenovirus species D type 37 (HAdV-D37) causes epidemic keratoconjunctivitis, a severe, hyperacute condition. The corneal component of epidemic keratoconjunctivitis begins upon infection of corneal epithelium, and the mechanism of viral entry dictates subsequent proinflammatory gene expression. Therefore, it is important to understand the specific pathways of adenoviral entry in these cells. METHODS: Transmission electron microscopy of primary and tert-immortalized human corneal epithelial cells infected with HAdV-D37 was performed to identify the means of v

The effects of collagen-rich extracellular matrix on the intracellular delivery of glycol chitosan nanoparticles in human lung fibroblasts.

Recent progress in nanomedicine has shown a strong possibility of targeted therapy for obstinate chronic lung diseases including idiopathic pulmonary fibrosis (IPF). IPF is a fatal lung disease characterized by persistent fibrotic fibroblasts in response to type I collagen-rich extracellular matrix. As a pathological microenvironment is important in understanding the biological behavior of nanoparticles, in vitro cellular uptake of glycol chitosan nanoparticles (CNPs) in human lung fibroblasts was comparatively studied in the presence or absence of type I collagen matrix. Primary human lung fi

Signal transducing molecules and glycosyl-phosphatidylinositol-linked proteins form a caveolin-rich insoluble complex in MDCK cells.

GPI-linked protein molecules become Triton-insoluble during polarized sorting to the apical cell surface of epithelial cells. These insoluble complexes, enriched in cholesterol, glycolipids, and GPI-linked proteins, have been isolated by flotation on sucrose density gradients and are thought to contain the putative GPI-sorting machinery. As the cellular origin and molecular protein components of this complex remain unknown, we have begun to characterize these low-density insoluble complexes isolated from MDCK cells. We find that these complexes, which represent 0.4-0.8% of the plasma membrane,

Chemokine Transfer by Liver Sinusoidal Endothelial Cells Contributes to the Recruitment of CD4+ T Cells into the Murine Liver.

Leukocyte adhesion and transmigration are central features governing immune surveillance and inflammatory reactions in body tissues. Within the liver sinusoids, chemokines initiate the first crucial step of T-cell migration into the hepatic tissue. We studied molecular mechanisms involved in endothelial chemokine supply during hepatic immune surveillance and liver inflammation and their impact on the recruitment of CD4(+) T cells into the liver. In the murine model of Concanavalin A-induced T cell-mediated hepatitis, we showed that hepatic expression of the inflammatory CXC chemokine ligands (

Intracellular Fate of a Dual-Fluorescent Hydrophobic Ion Pair: Comparison of Lipid-Based Nanocarriers.

Effective intracellular trafficking and delivery of hydrophilic drugs remain challenging due to poor membrane permeability and limited encapsulation in lipid-based nanocarriers. To address this, we developed a dual-fluorescent hydrophobic ion pair (HIP) by pairing a model fluorescent hydrophilic drug, Cascade Blue hydrazide, with the lipophilic probe DiA. The HIP was subsequently incorporated into three lipid-based nanocarriers─self-emulsifying drug delivery systems (SEDDS), nanoemulsions, and liposomes─to enable visualization and comparison of how formulation composition influences intracellu

A ternary zein/soy lecithin/dextran sulfate nanocarrier for improved resveratrol bioactivity delivery: Enhanced solubility, stability, and bioactivity.

Resveratrol (RES), a natural compound widely used as a food additive and preservative, exhibits anti-inflammatory and antioxidant activities. However, its poor solubility and stability severely limit its translational potential for clinical applications. To address these challenges, we developed a core-shell nanoparticle drug delivery system (DDS) composed of zein, soy lecithin, and dextran sulfate, fabricated via a built-in ultrasonic dialysis process (BUDP). The optimized RES/ZLDS NPs exhibited uniform spherical morphology, small particle size (252.00 ± 14.64 nm), high negative zeta potentia

Nanoplastic Translocation Across Biological Barriers (Blood-Brain, Placental, Intestinal): Transport Mechanisms, Tissue-Specific Vulnerabilities, and a Corona-Driven Barrier Selectivity Framework.

Nanoplastics (NPs; ≤1 µm) have been detected in human placenta, blood, lung, atherosclerotic plaque, testis, semen, olfactory bulb, and brain, shifting the field from environmental description toward mechanistic interrogation of barrier crossing. This review synthesises current evidence on NP translocation across the intestinal epithelium, the blood-brain barrier (BBB) and the placental syncytiotrophoblast. We distinguish four evidence categories throughout the review: detection, association, mechanism, and causality. We also apply model-system labels (in silico, in vitro, ex vivo, animal, and

Role of the Insulin Receptor in Mediating Cytosolic Delivery of Proteins by a Modified Cell-Penetrating Peptide.

Background: Intracellular delivery of high-molecular-weight proteins is limited by the cell membrane. Cell-penetrating peptides (CPPs) offer a potential solution, but effective cytosolic delivery remains hindered by endosomal sequestration. Pas2r12, a CPP-derived peptide, facilitates cytosolic delivery of proteins including immunoglobulin G. Because Pas2r12 internalization occurs via caveolae-dependent endocytosis, we hypothesized that cell-surface receptors contribute to uptake. Methods: HEK293 cells were treated with Pas2r12 alone or complexed with enhanced green fluorescent protein (EGFP).
