Effects of combined carotid chemoreceptor and atrial receptor stimulation on renal blood flow in anaesthetized dogs.

In dogs anaesthetized with chloralose and artificially ventilated, carotid chemoreceptors were stimulated by changing the perfusate of the vascularly isolated carotid bifurcations from arterial to venous blood. Left atrial receptors were stimulated by distending balloons in two pulmonary vein-left atrial junctions and in this left atrial appendage. The left renal blood flow was measured by an electromagnetic flow meter at a constant systemic (renal) arterial pressure in preparations in which heart rate changes were prevented by administration of propranolol hydrochloride (0.5 mg kg-1) and atro

Arterial baroreceptor reflex control of renal sympathetic nerve activity following chronic myocardial infarction in male, female, and ovariectomized female rats.

There is controversy regarding whether the arterial baroreflex control of renal sympathetic nerve activity (SNA) in heart failure is altered. We investigated the impact of sex and ovarian hormones on changes in the arterial baroreflex control of renal SNA following a chronic myocardial infarction (MI). Renal SNA and arterial pressure were recorded in chloralose-urethane anesthetized male, female, and ovariectomized female (OVX) Wistar rats 6-7 wk postsham or MI surgery. Animals were grouped according to MI size (sham, small and large MI). Ovary-intact females had a lower mortality rate post-MI

Apnea produces neuronal degeneration in the pons and medulla of guinea pigs.

Obstructive sleep apnea and other sleep-related breathing disorders result in recurrent periods of oxygen deprivation (hypoxia), hypercapnia and an increase in the cellular production of reactive oxygen species (oxidative stress-related injury). Individuals with these disorders suffer from a variety of cellular abnormalities that result in cardiopulmonary dysfunctions, disturbances in sleep and other pathologies. In the present experiment, using an animal model of sleep apnea, we determined that the degeneration of neurons and glia, due to apoptosis, occurs in specific regions of the pons and

Cocaine increases the intracellular calcium concentration in brain independently of its cerebrovascular effects.

Cocaine abuse increases the risk of life-threatening neurological complications such as strokes and seizures. Although the vasoconstricting properties of cocaine underlie its cerebrovascular effects, the mechanisms underlying its neurotoxicity remain incompletely understood. Here, we use optical techniques to measure cerebral blood volume, hemoglobin oxygenation (S(t)O(2)), and intracellular calcium ([Ca(2+)](i)) to test the hypothesis that cocaine increases [Ca(2+)](i) in the brain. The effects of cocaine were compared with those of methylphenidate, which has similar catecholaminergic effects

A fMRI study of brain activations during non-noxious and noxious electrical stimulation of the sciatic nerve of rats.

An acute pain animal model for fMRI study would provide useful spatial and temporal information for studying the supraspinal nociceptive neuronal responses. The aim of the present study was to investigate whether the nociceptive responses in different brain areas can be differentiated by using functional magnetic resonance imaging (fMRI) in anesthetized rats. Functional changes in brain regions activated by noxious or non-noxious stimuli of the sciatic nerve were investigated using fMRI in a 4.7 T MR system in alpha-chloralose anaesthetized rats. To determine the electrical intensity for noxio

Naloxone reverses inhibitory effect of electroacupuncture on sympathetic cardiovascular reflex responses.

Acupuncture and electroacupuncture (EA) have been used in traditional Chinese medicine to treat a wide range of diseases and conditions, including angina pectoris and myocardial infarction. In a feline model of reflex-induced reversible myocardial ischemia, electrical stimulation of the median nerves to mimic EA (Neiguan acupoint) significantly improved ischemic dysfunction, secondary to an inhibitory effect of EA on reflex pressor effects evoked by bradykinin (BK). The central mechanism of EA's inhibitory effect in this model is unknown. Accordingly, in alpha-chloralose-anesthetized cats, BK

The M cell: its contribution to the ECG and to normal and abnormal electrical function of the heart.

The discovery and characterization of the M cell, a unique cell type residing in the deep layers of the ventricular myocardium, has opened a new door in our understanding of the electrophysiology and pharmacology of the heart in both health and disease. The hallmark of the M cell is the ability of its action potential to prolong much more than that of other ventricular myocardial cells in response to a slowing of rate and/or in response to agents that act to prolong action potential duration. Our goal in this review is to provide a comprehensive characterization of the M cell, its contribution

Prolonged increase in micturition threshold volume by anogenital afferent stimulation in the rat.

OBJECTIVE: To investigate whether anogenital afferent stimulation induces a prolonged increase in the micturition threshold volume of anaesthetized rats. MATERIALS AND METHODS: Thirteen female rats, anaesthetized by alpha-chloralose and paralysed by pancuronium bromide were used for the experiments. The micturition threshold volume was determined by repeated cystometry. In two experiments, afferent activity was recorded from the exposed pudendal nerve; vaginal and anal afferents were stimulated electrically by ring electrodes. In one experiment, the dorsal clitoris nerves were exposed bilater

Ethanol inhibits sphincter of Oddi motility.

Patients with alcohol-induced liver disease are at increased risk for pigment gallstones, which are known to be particularly associated with biliary stasis. Although the effects of ethanol on the sphincter of Oddi are thought to contribute to alcoholic pancreatitis, the precise effects of ethanol on the biliary component of the sphincter of Oddi are unclear. In the prairie dog the common bile and pancreatic ducts enter the duodenum separately, facilitating pressure measurement in the sphincter choledochus in isolation. We therefore used this model to test the hypothesis that ethanol administra

Contribution of bed nucleus of the stria terminalis to the cardiovascular responses elicited by stimulation of the amygdala.

Anatomical and physiological studies were done in the rat to investigate the possibility that the cardiovascular responses elicited by stimulation of central nucleus of the amygdala (ACe) were mediated via projections to bed nucleus of the stria terminalis (BST). In the first series, to determine the distribution of neurons in ACe that projected to the cardiovascular region of BST, the retrograde tracer Fluorogold (FG) or rhodamine latex micro-beads (Rd) were injected into BST. FG and Rd injections that overlapped the cardiovascular region of BST resulted in retrogradely labelled neurons throu

Electrophysiological effects of high cocaine concentrations on intact canine heart. Evidence for modulation by both heart rate and autonomic nervous system.

BACKGROUND: Previous clinical reports have suggested that cocaine intoxication may produce severe ventricular arrhythmias due to a direct effect on the heart. However, the effects of high plasma levels of cocaine on the electrophysiology of the heart have not been well characterized and remain poorly understood. METHODS AND RESULTS: The purpose of this study was to characterize the electrophysiological effects of high doses of cocaine on the in situ dog heart. In dogs anesthetized with morphine and alpha-chloralose, cocaine (2-11 micrograms/mL) increased both atrial and ventricular refractory

Dorsal column input to thalamic VL neurons: an intracellular study in the cat.

This study investigated the role of the ventral lateral (VL) nucleus of the thalamus as a lemniscal relay to motor cortex. Intracellular recordings were obtained from thalamic VL relay neurons in cats anesthetized with chloralose, following stimulation of the dorsal column nuclei. VL neurons were identified by their short-latency input from the cerebellar nuclei, their antidromic activation from motor cortex and their anatomical location. A total of 105 neurons was studied. The occurrence of temporal facilitation to double volleys was also examined. It was found that 80/105 (75%) neurons respo

Centrally mediated cardiovascular actions of dynorphin A(1-8) on rat hippocampal formation.

The effects of dynorphin A(1-8) (DA1-8) microinjected into various areas of the hippocampal formation (HF) on the mean blood pressure (MBP) and heart rate (HR) were investigated in the alpha-chloralose-anesthetized rat. Intra-HF injection of DA1-8 dose-dependently (0.5-50 nmol) reduced MBP and HR. Depressor and bradycardic responses also occurred following microinjection of the excitatory amino acid l-glutamate (1 M, 0.2-0.4 microliter) into the DA1-8-sensitive sites. By contrast, administration of 4% lidocaine (0.2-0.4 microliter) into the same HF sites failed to affect MBP and HR. Pretreatme

Neural reflex of the canine pylorus to intraduodenal acid infusion.

In 29 chloralose-urethane anesthetized dogs, a manometric assembly was inserted through a gastrostomy to monitor pressure of the pyloric region with a sleeve sensor. Antral and duodenal contractions were monitored with both manometric sideholes and serosal strain gauges. An additional tube channel allowed intraduodenal infusions 1-2 cm aborad from the pylorus. Intraluminal infusion of hydrochloric acid (0.1 N, 0.92 ml/min, for 2 min) reproducibly caused activation of motor activity in the pyloric region and peristaltic duodenal activity. Proximal duodenal activity probably contributed to the t

The role of the glycine sensitive area of the ventral medulla in cardiovascular responses to carotid chemoreceptor and peripheral nerve stimulation.

The present study on cats anaesthetised with Althesin, which unlike more commonly used anaesthetics does not prevent reflex activation of the brain-stem defence areas, reaffirmed that carotid chemoreceptor stimulation and radial nerve stimulation can evoke the visceral components of the alerting stage of the defence response (visceral alerting response). This includes tachycardia, mesenteric vasoconstriction but vasodilatation in skeletal muscle which is not secondary to the hyperventilation. However, mild chemoreceptor stimulation which evoked but a weak hyperventilation elicited bradycardia

The responses of duodenal tension receptors in sheep to pentagastrin, cholecystokinin and some other drugs.

The proposal that some post-prandially released alimentary hormones modify ingestive behaviour and gastric emptying by altering impulse activity in alimentary enteroceptors has been tested using a number of gastrointestinal peptide hormone analogues. These and other drugs were applied to single-unit afferent preparations of duodenal tension receptors in chloralose-anaesthetized sheep. In separate experiments the effect of pentagastrin and cholecystokinin on duodenal motor activity was recorded without unitary afferent activity measurements. Local intra-arterial bolus injections of pentagastrin

Sympathetic nervous system response to mechanical stress of the spinal column in rats.

The effects of mechanical stimulation of the spine on blood pressure, heart rate and the activity of selected sympathetic nerves (renal and adrenal) were examined in alpha-chloralose/urethane anesthetized rats. Spinal segments from T10 to T13 or from L4 to L7 were isolated from surrounding muscle and the upper and lower segments of the four segment units were fixed by means of spinal clamps. Forces from 0.5 to 3.0 kg were applied to the lateral aspect of the two mobile segments. Stimulations of the thoracic or the lumbar region produced large decreases in blood pressure (-29.8 +/- 3.1 mmHg) al

The boundary of proximal hindlimb representation in the dorsal horn following peripheral nerve lesions in cats: a reevaluation of plasticity in the somatotopic map.

Some investigators have reported that deafferentation markedly alters the somatotopic organization of the dorsal horn in adult mammals, whereas others, including the present investigator, have not. Failure to observe changes in somatotopy might be attributable to insufficient deafferentation, inadequate sampling, limitation of the observations to a population of neurons that does not exhibit the phenomenon, or a variety of technical factors. The most striking changes in somatotopy have been reported following total deafferentation of the medial dorsal horn by sciatic and saphenous nerve lesion
