A partial deletion of the muscular dystrophy gene transmitted twice by an unaffected male.

A gene of unknown function located in band Xp21 on the short arm of the human X chromosome gives rise to X-linked recessive muscular dystrophy, of either Duchenne or Becker type, when mutated. The gene encodes a large muscle-specific transcript of about 14 kilobases (kb) and its genomic size extends over more than 1,800 kb. The high mutation rate (about 10(-4) per generation) is likely to result from the large target size. Submicroscopic deletions, detectable with one or more of the dozen cloned DNA probes available for regions within the gene, constitute a significant proportion of the mutati

Genetics of Hunter syndrome: carrier detection, new mutations, segregation and linkage analysis.

We have investigated 31 families segregation for Hunter Syndrome in order to advance our understanding of the genetics of this disease. The hair root test for the diagnosis of carriers of Hunter Syndrome was improved by the adoption of a new diagnostic index that distinguishes between carrier and normal females better than previous methods of analysis. One hundred and eleven female relatives of the affected children were tested by such procedures. This showed that seven out of 31 mothers were not carriers (22.6%), thus suggesting a small deficit of new mutation relative to the expectation that

Extracellular vesicles in stem cell-based gonadal regeneration: mechanisms, therapeutic potential, and translational challenges.

Gonadal dysfunction resulting from conditions such as premature ovarian insufficiency, chemotherapy-induced damage, or genetic disorders often leads to infertility and hormone imbalance. Although assisted reproductive technologies and hormone replacement therapies mitigate clinical symptoms, they remain incapable of reinstating native gonadal architecture and physiological function. In recent years, stem cell-based therapies, particularly those employing mesenchymal stem cells (MSCs), have demonstrated regenerative potential. However, limitations including poor engraftment, potential tumorigen

Double jeopardy: how BRCA2, TP53, and DNMT3A redefine the link between infertility and cancer risk.

BackgroundGermline mutations in genes governing DNA repair, cell cycle regulation, and epigenetic modification are now recognized as common etiological factors for both cancer predisposition and reproductive dysfunction. This reveals a profound intersection between reproductive biology and oncogenesis.MethodA systematic narrative review was conducted. The literature search spanned PubMed/MEDLINE, Scopus, and Web of Science using keywords and MeSH terms related to infertility phenotypes, cancer predisposition syndromes, and shared molecular mechanisms (e.g., DNA repair, epigenetics).OutcomesThe

Meiotic Origins of Non-Mosaic Klinefelter Syndrome (47, XXY): Mechanisms, Dimorphism, and Emerging Genetic Susceptibility.

BACKGROUND: Non-mosaic Klinefelter syndrome (47,XXY) arises from sex-specific meiotic mechanisms leading to nondisjunction during gametogenesis. In maternal cases, errors occur predominantly during meiosis I, frequently involving X chromosomes that lack crossovers or exhibit crossovers outside optimal chromosomal locations; this nondisjunction is further exacerbated by advancing maternal age and the subsequent deterioration of cohesins. Conversely, paternal 47,XXY stems primarily from the failure of obligate recombination within the pseudoautosomal region, which is critical for accurate XY seg

Deoxynivalenol exposure-related male reproductive toxicity in mammals: Molecular mechanisms, detoxification and future directions.

An increasing body of evidence indicates that exposure to widespread, environmental and food contaminants such as mycotoxins may cause endocrine disorders and infertility. Deoxynivalenol (DON), which is a toxic secondary metabolite produced by Fusarium fungi, can lead to multiple harmful effects in humans and animals, such as hepatotoxicity, nephrotoxicity, immunotoxicity, gastrointestinal toxicity, neurotoxicity, genetic toxicity and carcinogenicity. Recently, there has been growing concern about DON-induced male infertility. Exposure to DON and its metabolites can damage the structure and fu

Generation of juvenile rainbow trout derived from cryopreserved whole ovaries by intraperitoneal transplantation of ovarian germ cells.

Cryopreservation of fish sperm offers the practical applications in the selective breeding and biodiversity conservation. However, because of the lack of cryopreservation methods for fish eggs and embryos, maternally inherited cytoplasmic compartments cannot be successfully preserved. We previously developed an alternative method to derive functional eggs and sperm from cryopreserved whole testis by transplanting testicular cells into female and male recipients. However, if target fish had ovaries, the previous method employing male-derived germ cells would be ineffective. Here, we aimed to ge

Gestational exposure to PNMC reduces offspring gamete numbers by disrupting oocyte meiosis and spermatogenesis.

3-Methyl-4-nitrophenol (PNMC) is a prevalent nitrophenolic endocrine disruptor found in pregnant women, with known effects on offspring growth and development. However, its impact on offspring fertility remains unexplored. This study investigates the effects of PNMC exposure during pregnancy on offspring fertility and the underlying mechanisms. Our fertility assessments revealed that PNMC exposure during pregnancy reduced the number of follicles and spermatozoa in offspring, though it did not affect their quality. In male offspring, PNMC exposure impaired spermatogenesis by reducing the number

The E3 Ligase UBR5/Hyd Ensures Meiotic Fidelity Through Catalysis-Independent Regulation of β2-Tubulin in Drosophila.

Background: Spermatogenesis depends on precise cytoskeletal regulation, particularly the microtubule system; however, the mechanisms governing tubulin homeostasis during meiosis are poorly defined. While the E3 ubiquitin ligase Hyd (Hyperplastic discs), the Drosophila homolog of UBR5 (Ubiquitin Protein Ligase E3 Component N-Recognin 5), plays roles in diverse cellular processes, its precise role in male meiosis is unknown. This study aims to define the function and expression dynamics of Hyd during Drosophila spermatogenesis and elucidate whether it acts independently of its canonical ligase a

Integrating microarray data and single-cell RNA-seq reveals correlation between kit and nmyc in mouse spermatogonia stem cell population.

Spermatogonial stem cells (SSCs) are essential for the continuous production of sperm and the maintenance of male fertility. Their selection, culture, and molecular characterization provide critical insights into spermatogenesis and potential therapeutic applications for male infertility. This study utilized CD49f-MACS and matrix selection techniques to isolate SSCs from mouse testicular samples. The molecular profile of the selected SSCs was analyzed through immunocytochemistry, gene ontology enrichment, weighted gene co-expression network analysis (WGCNA), and single-cell RNA sequencing (scR

Leucinostatins from fungal extracts block malaria transmission to mosquitoes.

BACKGROUND: Malaria is a mosquito-transmitted disease that kills more than half a million people annually. The lack of effective malaria vaccines and recently increasing malaria cases urge innovative approaches to prevent malaria. Previously, we reported that the extract from the soil-dwelling fungus Purpureocillium lilacinum, a common fungus from the soil, reduced Plasmodium falciparum oocysts in Anopheles gambiae midguts after mosquitoes contacted the treated surface before feeding. METHODS: We used liquid chromatography to fraction fungal crude extract and tract the active fraction using a

Offspring production of ovarian organoids derived from spermatogonial stem cells by defined factors with chromatin reorganization.

INTRODUCTION: Fate determination of germline stem cells remains poorly understood at the chromatin structure level. OBJECTIVES: Our research hopes to develop successful offspring production of ovarian organoids derived from spermatogonial stem cells (SSCs) by defined factors. METHODS: The offspring production from oocytes transdifferentiated from mouse SSCs with tracking of transplanted SSCs in vivo, single cell whole exome sequencing, and in 3D cell culture reconstitution of the process of oogenesis derived from SSCs. The defined factors were screened with ovarian organoids. We uncovered ex

An Extensive Review of Recent Progress In Kinase-targeting Antimalarial Drug Discovery.

Malaria remains a major global health challenge, with the rise of drug-resistant Plasmodium falciparum strains necessitating the development of novel therapeutic strategies. Kinases, which regulate essential processes throughout the parasite life cycle, including invasion, egress, development, proliferation, gametogenesis, and transmission, have emerged as promising drug targets, leading to significant advances in kinase inhibitor development over the past five years. This review examines recent advances in kinase-targeted antimalarial drug discovery, with particular emphasis on inhibitor scaf

Exposure to Estrone Impairs Oogenesis and Spawning in Zebrafish (Danio rerio).

Estrone (E1) is one of the major natural estrogens influencing the reproductive system of vertebrates. However, it contaminates aquatic environments due to runoff from livestock waste and the discharge of domestic sewage. The aim of this study was to investigate the impact of exposure to environmentally relevant concentrations of E1 on oogenesis and spawning in zebrafish (Danio rerio). Fish were exposed, in duplicate, to 20, 200, and 2000 ng/L of E1 for 49 days for oogenesis analysis evaluating histology, sex steroids, morphometry, cell proliferation, apoptosis, immunohistochemistry for insuli

De-extinction of the Northern white rhinoceros (Ceratotherium simum cottoni).

The last survivors of the northern white rhinoceros (NWR, Ceratotherium simum cottoni), once widespread across Central and East Africa, are two non-reproductive females under continuous human protection. In order to prevent the extinction of this white rhinoceros subspecies, the BioRescue consortium founded in 2019 has developed an innovative reproductive biotechnology program. This holistic rescue strategy implements (i) advanced assisted reproductive technologies (aART) including in vitro fertilization (IVF) and embryo transfer, (ii) stem cell associated techniques (SCAT) for establishing in

Comparative analysis of immunocontraceptive vaccines in wild animals: Modes of action, molecular targets, and immunological responses.

Immunocontraceptive vaccines have emerged as a promising, non-invasive strategy for regulating fertility in wildlife, domestic animals, and humans, particularly in species such as elephants where conventional population control methods are often impractical or ethically challenging. These vaccines act by eliciting immune responses against critical reproductive targets, thereby disrupting gametogenesis, fertilization, or endocrine regulation. The major categories include gonadotropin-releasing hormone (GnRH)-based vaccines, zona pellucida (ZP)-based vaccines, and gamete-specific antigen vaccine

Current research on adhesion regulating molecule 1: A Review.

Adhesion regulating molecule 1 (ADRM1), also known as regulatory particle non-ATPase subunit 13 (Rpn13), is a multifunctional proteasome-associated ubiquitin receptor that bridges protein degradation and cellular signaling networks. Structurally, ADRM1 comprises an N-terminal pleckstrin-like receptor for ubiquitin (Pru) domain responsible for ubiquitin and proteasome recognition, and a C-terminal DEUBAD domain that activates the deubiquitinating enzyme UCH37, jointly coordinating substrate recognition and deubiquitination within the 26S proteasome. Functionally, ADRM1 regulates multiple biolog

Non-neuronal acetylcholine: from cellular signaling to clinical implications.

For over a century, acetylcholine (ACh) was viewed primarily as a wired neurotransmitter of cholinergic nerves. However, accumulating evidence supports the view of the non-neuronal cholinergic system (NNCS) as an evolutionarily ancient autocrine and paracrine network operating across peripheral tissues. Rather than an extension of neuronal signaling, non-neuronal ACh can be viewed as a tissue-embedded chemical communication system positioned at the interface of cellular homeostasis and systemic neural reflexes. This review frames the NNCS as a network of tissue-resident ACh-producing cells tha

Comprehensive transcriptome analysis of somatic and gonadal tissues for identification of sex-biased genes in spotted scat (Scatophagus argus).

The spotted scat (Scatophagus argus), an economically valuable aquaculture species in southern China, exhibits a pronounced sexual dimorphism in growth performance, with females growing significantly faster than males. Nevertheless, progress in sex-controlled breeding remains limited due to limited understanding of its sex determination and differentiation mechanisms. To investigate sex-biased gene expression in this species, a comprehensive transcriptomic analysis was performed. A total of 62 transcriptomic libraries were analyzed, comprising 18 newly sequenced libraries derived from female g

Seasonal immune dynamics and host-microbiota interactions in the mussel Mytilus galloprovincialis under a warming Mediterranean Sea.

Marine bivalves are increasingly threatened by disease outbreaks and mass mortality events driven by climate change and polymicrobial infections. Mytilus galloprovincialis is the main farmed bivalve in the Mediterranean, one of the world's fastest-warming marine regions, with the Ligurian Sea experiencing particularly rapid increases in temperature and heatwave frequencies and intensities. Bivalve hemocytes integrate environmental and microbial cues, making their seasonal dynamics a key determinant of host resilience under changing climate conditions. Previous studies identified surface seawat
