C-type lectins (CTLs) serve as pivotal pattern recognition receptors (PRRs) within the innate immune system of invertebrates. In this study, we identified and characterized a novel CTL from the echiuran worm (Urechis unicinctus), termed UuCTL6, to elucidate its function in antimicrobial defense. Sequence analysis revealed that the open reading frame (ORF) of UuCTL6 comprises 1074 bp, encoding a polypeptide of 358-amino acid, which includes a conserved carbohydrate recognition domain (CRD) stabilized by two disulfide bonds. Sequence identity analysis showed that UuCTL6 shares 26.7%-31.6% amino

Antifouling paints have been used to control biofouling on submerged surfaces for decades. They contain biocidal compounds that may be released into the environment and harm aquatic ecosystems; however, their effects on tropical organisms are little known. Dichlofluanid, or N-dichlorofluoromethythio-N',N'-dimethyl-N-phenylsulfamide (C9H11Cl2FN2O2S2), is a biocide present in antifouling paints and its toxicity to tropical marine organisms is poorly understood. This study aimed to evaluate the effects of dichlofluanid at the cellular and biochemical levels in bivalves exposed to seawater and sed

Understanding more about the risks posed by hazardous pesticides is essential, particularly for ecologically and economically important species such as the Pacific oyster (Magallana gigas). Although dimethoate toxicity has been widely studied and has led to regulatory measures in certain regions, it is still used in others, such as Tunisia. Investigations on sensitive species in such contexts can provide valuable insights for better assessing and managing potential environmental risks. This study offers a novel evaluation of the toxic effects of experimental concentrations of dimethoate after

Peptidoglycan recognition proteins (PGRPs) are evolutionarily conserved innate immune receptors with pivotal roles in regulating host defense responses, yet the functional characterization of PGRPs in marine benthic invertebrates, especially in Echiurioidea and Annelida, remains limited. In this study, we identified and characterized a short-type PGRP from the U. unicinctus, designated UuPGRP, to elucidate its role in antimicrobial immunity. Sequence analysis revealed that UuPGRP consists of 232 amino acid residues, including a conserved C-terminal PGRP domain. Phylogenetic and sequence identi

Toll-like receptors (TLRs), as core pattern recognition receptors (PRRs), play pivotal roles in pathogen detection during innate immune defense and are critical for initiating immune responses. In this study, we investigated a novel member of the TLR1 subfamily, TLR18, in the economically significant species Scophthalmus maximus, aiming to elucidate its molecular mechanisms in antibacterial immunity and provide insights into innate immune regulation in fish. In our results, SmTLR18 (2652 bp) encoded an 883-amino-acid protein sharing high identity with Paralichthys olivaceus TLR14 (84.15%). S

Insect stress-responsive peptides (SRPs) orchestrate development and immunity, yet their roles in the globally invasive pest Spodoptera frugiperda remain poorly understood. Here, we identify three SRP genes, SfSRP1, SfSRP2, and SfSRP3, that are strongly induced after Beauveria bassiana infection. Each encodes a 115-119 aa peptide with a conserved five-β-strand scaffold and divergent loop regions. RT-qPCR analysis reveals that all three are most abundant in hemocytes and head tissues, with stage-specific peaks at 2nd to 4th instars. Upon challenge with fungal or bacterial pathogens, SfSRP1 and

Lectins are a large group of carbohydrate-binding proteins that widely participate in innate immune response, however, our understanding of its subfamily mannose-binding lectins (MBLs) is still in paucity for commercially important fish species such as the large yellow croaker (Larimichthys crocea). In this study, a novel MBL homolog (designated as galactose-binding lectin, LcGalBL-1, due to its galactose preference) was identified in L. crocea through transcriptomic screening. In addition to bioinformatic characterization, expression profiling of LcGalBL-1 in different tissues and upon pathog

Bulinus truncatus snail is one of the most medically important snails. The goal of this study was to evaluate the molluscicidal effect of saponin on these snails and study how it affects their biological functions. The present results showed that saponin had a molluscicidal activity against adult B. truncatus snails after 24h and 72h with LC50 (57.5 and 27.1 ppm, respectively) and had ovicidal acivity on the snails' embryos. By studying the effect of the sublethal concentrations (LC10 48.63 ppm or LC25 52.83 ppm) exposure on B. truncatus snails, they resulted in significant decreases in the su

Rel/nuclear factor (NF)-κB family of transcription factors paly vital roles in innate immunity response to bacterial and viral infection. Here, we cloned and identified a dorsal homologue (named as MnDorsal) from Macrobrachium nipponense. The full-length cDNA of MnDorsal is 2573 bp with a 1986 bp open reading frame that encodes 661 amino acids. Predicted MnDorsal protein contained a RHD (Rel homology domain), an IPT (Iglike, plexins, and transcriptions factors) domain, and two low complexity regions. Phylogenetic analysis showed that MnDorsal has a closer genetic distance with dorsal homologue

Aplastic anemia (AA) is characterized as hypoplasia of bone marrow hematopoietic cells and hematopenia of peripheral blood cells. Though the supplement of exogenous erythropoietin (EPO) has been clinically approved for AA treatment, the side-effects hinder its further application. Here a robust treatment for AA induced by chemotherapy drugs is explored using gadofullerene nanoparticles (GFNPs). Methods: The gadofullerene were modified with hydrogen peroxide under alkaline conditions to become the water-soluble nanoparticles (GFNPs). Priming in invertebrates is the acquired capacity to better combat a pathogen due to a previous exposure to sub-lethal doses of the same organism. It is proposed to be functionally analogous to immune memory in vertebrates. Previous studies with Anopheles gambiae mosquitoes provide evidence that the inhibitory response to a second challenge by the malaria parasite Plasmodium berghei resulted from a sustained activation of hemocytes by midgut bacteria. These bacteria probably accessed the hemolymph during a first aborted infection through lesions produced by parasites invading the midgut. Since

The anti-lipopolysaccharide factor ALF-Pm3 is a 98-residue protein identified in hemocytes from the black tiger shrimp Penaeus monodon. It was expressed in Pichia pastoris from the constitutive glyceraldehyde-3-phosphate dehydrogenase promoter as a folded and (15)N uniformly labeled rALF-Pm3 protein. Its 3D structure was established by NMR and consists of three alpha-helices packed against a four-stranded beta-sheet. The C(34)-C(55) disulfide bond was shown to be essential for the structure stability. By using surface plasmon resonance, we demonstrated that rALF-Pm3 binds to LPS, lipid A and t

BACKGROUND: Armigeres subalbatus is a natural vector of the filarial worm Brugia pahangi, but it rapidly and proficiently kills Brugia malayi microfilariae by melanotic encapsulation. Because B. malayi and B. pahangi are morphologically and biologically similar, the Armigeres-Brugia system serves as a valuable model for studying the resistance mechanisms in mosquito vectors. We have initiated transcriptome profiling studies in Ar. subalbatus to identify molecular components involved in B. malayi refractoriness.
RESULTS: These initial studies assessed the transcriptional response of Ar. subalb

Transplantation surgery induces severe immune stress in pearl oysters Pinctada fucata martensii, yet effective immunomodulatory strategies remain limited. This study establishes a proteomics-guided strategy for immunomodulation by identifying cyclophilin as a candidate mediator through iTRAQ-based quantitative analysis and validating cyclosporin A (CsA) through serum immune indicator and transcriptome analysis. Serum iTRAQ analysis identified 1069 differentially expressed proteins following transplantation, among which cyclophilin exhibited significant up-regulation and was selected as a candi

Schistosomiasis is a serious public health concern, with transmission facilitated by a small number of freshwater snail intermediate host species. Infection outcomes vary greatly across the primary vector genera, Biomphalaria (for Schistosoma mansoni), Bulinus (for S. haematobium), and Oncomelania (for S. japonicum), even within species, ranging from full resistance to high compatibility. Omics methods have altered this field by correlating host genotype, baseline immunological status, and time-resolved responses to whether invading miracidia are eliminated or develop sporocysts. Evidence from

Extracellular vesicles (EVs), as antimicrobial factors, load a variety of immune-related molecules, play a complex and critical role in host-pathogen interactions. The regulatory mechanism of how EVs respond to external stimuli by regulating immune regulatory elements in the innate immune response of marine invertebrates remains unresolved. To address this, we conducted a global transcriptomic analysis of hemocytes in Chlamys farreri stimulated by EVs derived from different sources. Transcriptomic analysis identified 892 specifically upregulated and 246 specifically downregulated genes in the

Ozone nanobubbles (NB-O3) have recently emerged as a promising technology in aquaculture for improving water quality and disease control. However, their impacts on shrimp remain poorly understood, particularly regarding their effects on biochemical processes. This study evaluated the effects of NB-O3 on growth performance, immune responses, oxidative stress, and HSP70 gene expression in whiteleg shrimp (Penaeus vannamei). Shrimp were cultured for 21 days in control and NB-O3-treated tanks. Growth indices, water quality variables, total hemocyte counts (THC), antioxidant enzyme activities (cata
