Simvastatin prevents vascular complications in the chronic reactive oxygen species murine model of systemic sclerosis.

Aims Systemic sclerosis (SSc) is characterized by vasculopathy and organ fibrosis. Although microvascular alterations are very well characterized, structural and functional abnormalities of large vessels are not well defined. Therefore, we evaluated the effect of simvastatin administration on aortic and small renal arteries thickening, and on myofibroblasts differentiation in a murine model of SSc. Methods and results SSc was induced in BALB/c mice by daily subcutaneous injections of hypochlorous acid (HOCl, 100 μl) for 6 weeks. Mice (n = 23) were randomized to receive: HOCl (n = 10); HOCl plu

Pathogenic effects of antimyeloperoxidase antibodies in patients with microscopic polyangiitis.

OBJECTIVE: Microscopic polyangiitis (MPA) is a small-vessel vasculitis associated with antimyeloperoxidase (MPO) antibodies in 70% of patients. Anti-MPO antibodies can trigger the release of MPO by neutrophils and monocytes, but their involvement in the pathogenesis of MPA is still questioned. The aim of this study was to investigate whether anti-MPO antibodies can activate MPO to generate an oxidative stress that is potentially deleterious to the endothelium. METHODS: MPA sera, purified IgG from MPA sera, normal control sera, and purified IgG from normal sera were incubated with MPO coated o

Generation of neoantigenic epitopes after posttranslational modification of type II collagen by factors present within the inflamed joint.

OBJECTIVE: Collagen-induced arthritis is a commonly accepted model of rheumatoid arthritis (RA). However, it has been difficult to substantiate the involvement of autoimmunity to type II collagen (CII) in the pathogenesis of RA. The aim of this investigation was to determine if CII, modified by reactive oxidant species present within the inflamed joint, could generate neoantigenic epitopes. METHODS: Oxidants that play a role in acute and chronic inflammation and are present in the rheumatoid joint (hydroxyl radical, hypochlorous acid, and peroxynitrite) were used for modification of native CI

Comparable effects of HOCl and of FMLP-stimulated PMN on the circulation in an isolated lung model.

Polymorphonuclear leukocytes (PMN) are involved in acute lung injury during adult respiratory distress syndrome (ARDS) via several mechanisms. This study focuses on neutrophil-derived oxidative stress. The influence of (A), continuous hypochlorous acid (HOCl) infusion over 105 min and (B) stimulation of PMN having been delayed in the pulmonary microvasculature were studied. Therefore pulmonary artery pressure (PAP), capillary filtration coefficient (Kf,c), and fluid retention (delta W) were monitored using isolated rabbit lungs. These models (A/B) were compared with each other to assess the re

Zinc-doped carbon dots for hypochlorite ultrasensitive determination: from aqueous monitoring to live-cells imaging.

To overcome the limitations of conventional detection methods for Hypochlorite (ClO⁻), bright blue-emitting zinc-doped carbon dots (Zn-CDs) were constructed via a streamlined and rapid hydrothermal method (within 2 h). The Zn-CDs possessed excellent water dispersibility and physicochemical stability with a quantum yield (QY) of 10.05%. Based on the Zn-O coordination structure could provide specific binding sites for ClO⁻, the Zn-CDs as fluorescent probes enabled ultrasensitive and selective detection of ClO⁻ in range of 0.1-50 µM with an exceptionally low limit of detection (LOD) of 8.14 nM. Investigation of ion binding in chlorite dismutases by means of molecular dynamics simulations.

Chlorite dismutases are prokaryotic heme b oxidoreductases that convert chlorite to chloride and dioxygen. It has been postulated that during turnover hypochlorite is formed transiently, which might be responsible for the observed irreversible inactivation of these iron proteins. The only charged distal residue in the heme cavity is a conserved and mobile arginine, but its role in catalysis and inactivation is not fully understood. In the present study, the pentameric chlorite dismutase (Cld) from the bacterium Candidatus Nitrospira defluvii was probed for binding of the low spin ligand cyanid

The antioxidant role of thiocyanate in the pathogenesis of cystic fibrosis and other inflammation-related diseases.

Cystic fibrosis (CF) is a pleiotropic disease, originating from mutations in the CF transmembrane conductance regulator (CFTR). Lung injuries inflicted by recurring infection and excessive inflammation cause approximately 90% of the morbidity and mortality of CF patients. It remains unclear how CFTR mutations lead to lung illness. Although commonly known as a Cl(-) channel, CFTR also conducts thiocyanate (SCN(-)) ions, important because, in several ways, they can limit potentially harmful accumulations of hydrogen peroxide (H(2)O(2)) and hypochlorite (OCl(-)). First, lactoperoxidase (LPO) in t

Analysis of 3-nitrotyrosine in biological fluids and protein hydrolyzates by high-performance liquid chromatography using a postseparation, on-line reduction column and electrochemical detection: results with various nitrating agents.

Nitric oxide reacts rapidly with superoxide to form the strong nitrating agent peroxynitrite, which is responsible for much of the tissue damage associated with diverse pathophysiological conditions such as inflammation. The occurrence of free or protein-bound nitrotyrosine (NTYR) has been considered as evidence for in vivo formation of peroxynitrite. However, various agents can nitrate tyrosine, and their relative significance in vivo has not been determined due to lack of a sensitive method to analyze NTYR in tissue proteins and biological fluids. We have developed a new HPLC-electrochemical

Chemical disinfection of duck hepatitis B virus: a model for inactivation of infectivity of hepatitis B virus.

The susceptibility of duck hepatitis B virus (DHBV) to the virucidal effects of sodium hypochlorite (NaOCl) and sodium dichloroisocyanurate (NaDCC) was compared to hepatitis B virus (HBV) with the aim of using the duck as a model for studying HBV disinfection. Using viral DNA polymerase (DNAP) as a target, inhibition of DNAP activity by chlorine disinfectants was found to be concentration-dependent but independent of contact time. Two minute exposure of minimal effective concentrations of sodium hypochlorite (domestic bleach: 3600 ppm and industrial bleach: 3180 ppm) and sodium dichloroisocyan

CD22 and CD72 contribute to the development of scleroderma in a murine model.

BACKGROUND: Systemic sclerosis (SSc) is a systemic autoimmune disease that is characterized by excessive fibrosis. CD22 and CD72 are B cell-specific cell surface molecules that negatively regulate B cell function. OBJECTIVE: The aim of the present study was to investigate the roles of CD22 and CD72 in a murine scleroderma model. METHODS: The experimental fibrosis model was generated by subcutaneous injection of bleomycin or hypochlorous acid (HOCL) into wild-type (WT), CD22-deficient (CD22-/-), CD72-deficient (CD72-/-) and CD22 and CD72 double-deficient (CD22-/-/CD72-/-) mice. PF-1355, a mechanism-based myeloperoxidase inhibitor, prevents immune complex vasculitis and anti-glomerular basement membrane glomerulonephritis.

Small vessel vasculitis is a life-threatening condition and patients typically present with renal and pulmonary injury. Disease pathogenesis is associated with neutrophil accumulation, activation, and oxidative damage, the latter being driven in large part by myeloperoxidase (MPO), which generates hypochlorous acid among other oxidants. MPO has been associated with vasculitis, disseminated vascular inflammation typically involving pulmonary and renal microvasculature and often resulting in critical consequences. MPO contributes to vascular injury by 1) catabolizing nitric oxide, impairing vaso

Myeloperoxidase interaction with peroxynitrite: chloride deficiency and heme depletion.

Myeloperoxidase (MPO) is a hemoprotein involved in the leukocyte-mediated defense mechanism and uses hydrogen peroxide (H2O2) and chloride (Cl(-)) to produce hypochlorous acid. In human saliva and in hypochloremic alkalosis syndrome occurring in breast-fed infants, the MPO-H2O2 system functions in a lower Cl(-) concentration (10-70 mM) compared to plasma levels (100 mM) as part of the antibacterial defense system. The impact of low Cl(-) concentration and exposure to high peroxynitrite (ONOO(-)) synthesized from cigarette smoke or oxidative stress on MPO function is still unexplored. Rapid mix

3-Chlorotyrosine as a marker of protein damage by myeloperoxidase in tracheal aspirates from preterm infants: association with adverse respiratory outcome.

Oxidative injury is implicated in the development of chronic lung disease in preterm infants with respiratory distress. However, direct evidence of a causal role is limited and the source of reactive oxidants has not been identified. We have previously shown that protein carbonyl levels in tracheal aspirates correlate positively with myeloperoxidase, suggesting that neutrophil oxidants could be the source of this protein injury. We have extended these observations by measuring 3-chlorotyrosine, a specific biomarker of the neutrophil oxidant, hypochlorous acid, in tracheal aspirate proteins (14

Tracking ClO- Dynamics during the Progression of Parkinson's Disease in MPTP Mouse Model Using In Vivo Brain Microdialysis.

Oxidative stress induced by hypochlorite (ClO-) is significant in Parkinson's disease (PD) pathogenesis, yet the dynamic variation in the brain's ClO- level during PD progression remains unclear. Herein, we developed a new reliable method combining in vivo microdialysis brain sampling with fluorescence detection for accurate and real-time tracking of local ClO- levels in the striatum. In the strategy, N,P-co-doped fluorescent carbon dots (N,P-CDs+) with a high positive charge (22.33 mV) and an average particle size (4.4 nm) were synthesized, exhibiting membrane-impermeability, fast response, h

A dual-channel lipid droplet-targeting probe for the detection of polarity and HOCl in models of NAFLD, inflammation, and cancer.

Abnormal lipid droplet accumulation and metabolism can precipitate various diseases. Understanding the dynamics of intracellular lipid droplets, particularly hypochlorite acid (HOCl) and polarity, is crucial for elucidating the onset and progression of these diseases. In this study, we synthesized a near-infrared fluorescent probe, NSSP, featuring a D-π-A structure, which facilitates the independent dual-response detection of HOCl and polarity. NSSP not only reveals the polarity and HOCl fluctuations in inflammatory cells and inflammation in live mice but also differentiates between normal and

Novel NIR fluorescent probe based on BODIPY for diagnosis and treatment evaluation of alcoholic liver disease via visualizing HClO fluctuation.

Alcoholic liver disease (ALD) is gradually becoming common due to the increasing number of drinkers worldwide, which is a serious threat to human physical and mental health. In the process of ALD, it is often accompanied by the occurrence of inflammation, which induce high expression of reactive oxygen species including HClO. In this work, we successfully fabricated a NIR fluorescent probe BDP-ENE-Fur-HClO for real-time imaging alcoholic liver disease via tracing HClO. The probe displayed good sensitivity and specificity, rapid recognition speed and NIR emitting (700 nm) for detection of HClO

Efficacy of selected hand hygiene agents used to remove Bacillus atrophaeus (a surrogate of Bacillus anthracis) from contaminated hands.

CONTEXT: The intentional use of Bacillus anthracis transmitted via the US mail in October-November 2001 resulted in 22 people developing inhalation or cutaneous anthrax. Glove use with handwashing prior to and after contact with potential contaminated environmental surfaces and cutaneous lesions has been recommended. However, only limited data are available on the susceptibility of B anthracis to antiseptics. OBJECTIVE: To evaluate the efficacy of several hand antiseptics (interventions) and soap and water (control) against Bacillus atrophaeus, a surrogate of B anthracis. DESIGN, SETTING, AN
