The carotenoid beta-cryptoxanthin stimulates the repair of DNA oxidation damage in addition to acting as an antioxidant in human cells.

The role of dietary antioxidants in human health remains controversial. Fruits and vegetables in the diet are associated with lower rates of chronic disease, and this is often attributed to their content of antioxidants, and a resulting protection against oxidative stress. However, large-scale human trials with antioxidant supplements have shown, if anything, an increase in mortality. We have investigated the biological properties of beta-cryptoxanthin, a common carotenoid, in cell culture model systems, using the comet assay to measure DNA damage. At low concentrations, close to those found i

Astaxanthin suppresses hepatocellular carcinoma via targeting Wnt/Β-catenin pathway: Experimental study on chemically induced HCC in rats.

Astaxanthin (ATX) is a potent natural antioxidant that shows promise against hepatocellular carcinoma (HCC). We investigated the therapeutic potential of ATX and its impact on Wnt/β-catenin pathway in a rat model, inducing HCC by nitrosodiethylamine (DEN) and carbon tetrachloride (CCl4). Rats were divided into five groups; I: control, II: HCC, III: HCC rats received ATX (5 days/week), IV: HCC rats received doxorubicin (DOX) weekly, V: HCC rats received combination therapy, for 4 weeks. Our results indicate that combination therapy significantly improved serum biomarkers, reducing alanine amino

Multiomics Dissection of the Mechanisms Underlying Eicosapentaenoic Acid Astaxanthin Ester-Mediated Protection Against High-Fat Diet-Induced Gut Barrier Injury.

The intestinal barrier proves exquisitely sensitive to dietary cues and is intimately linked to the progression of intestinal, metabolic, and systemic disorders. This study explored the effects of eicosapentaenoic acid astaxanthin ester (EA) on high-fat-diet (HFD)-induced intestinal barrier damage based on the modification of gut microecology. The results showed that EA inhibited obesity development, dyslipidemia, and colon inflammation, suppressed colon pathological damage, and restored the intestinal barrier by upregulating ZO-1 and Occludin, but these improvement effects were lost in pseudo

Astaxanthin attenuates AFB1-induced hepatotoxicity by activating Nrf2 and inhibiting the NF-κB signaling pathway.

Aflatoxin B1 (AFB1)-induced hepatotoxicity is a common toxic disease in poultry farming. However, there is currently a lack of effective pharmaceutical interventions for treating AFB1. Astaxanthin (AST), a natural carotenoid, exhibits potent antioxidant and immune-enhancing properties. In this study, the effect of AST on AFB1-induced hepatotoxicity was determined using chick embryo liver cells (CELs). The CCK-8 assay results showed that AST significantly attenuated the decrease in viability of CEL cells induced by AFB1, and reduced the activity of the enzymes GOT, GPT, and ALP (P < 0.01). More

Inhibition of IL-21/IL-21R Signaling by Fucoxanthin: Structure-Based and Experimental Analysis.

Dysregulated activation of the interleukin-21 (IL-21)/IL-21 receptor (IL-21R) signaling pathway is strongly associated with inflammatory and autoimmune disorders, which positions the pathway as a promising therapeutic target. Given the current lack of approved inhibitors or monoclonal antibodies targeting IL-21/IL-21R, we employed a structure-based virtual screening strategy coupled with experimental validation to identify potential IL-21 antagonists from a library of marine natural products provided by TargetMol. Our investigation identified fucoxanthin, a marine-derived carotenoid, as a pote

Astaxanthin activates the Nrf2/Keap1/HO-1 pathway to inhibit oxidative stress and ferroptosis, reducing triphenyl phosphate (TPhP)-induced neurodevelopmental toxicity.

Triphenyl phosphate (TPhP) serves as a major organophosphorus flame retardant, and its induced neurodevelopmental toxicity has attracted widespread attention, but the mechanism remains unclear. In this study, we involved zebrafish to explore the new mechanism of TPhP inducing oxidative stress and ferroptosis to promote neurodevelopmental toxicity. The results suggested that TPhP affected the embryonic development, reduced the number of new neurons, and led to abnormal neural behavior in zebrafish larvae. TPhP also induced ROS accumulation, activated the antioxidant defense signal Nrf2 and Keap

Astaxanthin Prevented Acute Alcoholic Cardiomyopathy by Maintenance of Mitophagy-Mediated Mitochondrial Homeostasis.

Acute alcoholism commonly targets the myocardium, triggering acute alcoholic cardiomyopathy (ACM). Strong evidence suggested that mitochondrial dysfunction-induced myocardial oxidative stress is involved in the subcellular pathogenesis of acute ACM. We investigated whether astaxanthin (AST), an antioxidant lutein carotenoid, prevents acute ACM and explored the underlying mechanisms. C57BL/6J mice were used to model ethanol-induced ACM and were treated with AST (100 mg/kg/day) alongside the autophagy inhibitor, 3-methyladenine (10 mg/kg/day).
