The small bovine amelogenin LRAP fails to rescue the amelogenin null phenotype.

Amelogenins are the most abundant secreted proteins in developing dental enamel. These evolutionarily-conserved proteins have important roles in enamel mineral formation, as mutations within the amelogenin gene coding region lead to defects in enamel thickness or mineral structure. Because of extensive alternative splicing of the primary RNA transcript and proteolytic processing of the secreted proteins, it has been difficult to assign functions to individual amelogenins. To address the function of one of the amelogenins, we have created a transgenic mouse that expresses bovine leucine-rich am

Heterochronic quantitative microevolution: dental divergence in aboriginal Americans.

Studies of tooth crown morphology alone have proven valuable in defining human population differentiation. We test the hypothesis that quantitative comparisons of more complex whole tooth structure may prove informative in understanding human diversity. Three disparate populations in Native American genetic history were compared: Kodiak Island Western Eskimos, Peruvian Inca Amerindians, and Southeast Asians. Enamel depth (an increasing gradient extended from Southeast Asians to the Inca) and root parameters were the most discriminating variables. The observed microevolution appears to result f

Development, growth and eruption of permanent incisor teeth in Romney sheep.

The development of permanent incisor teeth of sheep was followed from the age of six weeks to 116 weeks. The teeth were observed through the bud, cap and bell stages, and the development of enamel and dentine. At the same time that amelogenesis started, the papillary layer developed from the intermediate layer and the outer dental epithelium began to form the cell rests of Malassez. The ameloblasts went through secretory, transitional and maturational phases, and in due course disappeared, after which cementum was laid down over the enamel surface. Initially the connective tissue of the follic

TNAP-mediated hydrolysis of phosphorylated enamel matrix proteins drives local phosphate generation and enamel mineralization.

Dental enamel is the most highly mineralized tissue in the body, and its formation requires a precisely regulated supply of Pi. Although phosphate transport in ameloblasts has been studied, how phosphate is generated within the enamel remains unclear. Tissue-nonspecific alkaline phosphatase (TNAP) is a key enzyme in skeletal mineralization, but its functional contribution to enamel formation has not been fully established. Herein, we examined the presence, activity, and catalytic potential of TNAP in porcine enamel matrix. To determine the localization and functional contribution of TNAP withi

Effects of C-terminal amelogenin peptides on the metabolism of osteoblasts.

OBJECTIVES: Amelogenins, enamel matrix proteins secreted by ameloblasts, comprise 90% of the developing extracellular enamel matrix. Recent evidence suggests that amelogenins might induce the proliferation of various cells. However, the residues comprising the active site of amelogenin remain unclear. Therefore, this study aimed to examine the effects of a human amelogenin C-terminal peptide (amgCP) on the metabolism of osteoblasts. MATERIALS AND METHODS: Mouse calvarial osteoblastic cells (MC3T3-E1) were cultured and treated with amgCP. Cell proliferation was measured using MTS and BrdU assa

Five-minute Apgar score ≤ 5 and Molar Incisor Hypomineralisation (MIH) - a case control study.

BACKGROUND: The aetiology of molar incisor hypomineralisation (MIH) is unclear. The asymmetric distribution of MIH in the dentition may indicate that an insult of short duration that affects ameloblasts at a vulnerable stage could be a causative factor. Apgar ≤ 5 at 5 min may indicate asphyxia (hypoxic-ischemic insult) during birth. It was hypnotised that low Apgar score during birth may cause MIH. The present study aimed to examine a possible association between Apgar ≤ 5 at 5 min and the occurrence of MIH. METHOD: Two study groups were selected for examination. The cases comprised 67 childr

The influence of infantile thiamine deficiency on primary dentition.

OBJECTIVES: The present study explored the histological and chemical effects of infantile thiamine deficiency (ITD) on enamel development through the examination of exfoliated deciduous teeth from a patient who had been fed during his first year of life with a thiamine-deficient milk substitute. MATERIALS AND METHODS: Ground sections derived from six exfoliated primary teeth were examined. Slices from a light microscope were photographed for histological analysis. We calculated the time when the amelogenesis insults occurred, and the data were cross-examined with the patient's medical history

Odontoblast cells immortalized by telomerase produce mineralized dentin-like tissue both in vitro and in vivo.

The formation of dentin provides one well accepted paradigm for studying mineralized tissue formation. For the assembly of dentin, several cellular signaling pathways cooperate to provide neural crest-derived mesenchymal cells with positional information. Further, "cross-talk" between signaling pathways from the mesenchymal derived odontoblast cells and the epithelially derived ameloblasts during development is responsible for the formation of functional odontoblasts. These intercellular signals are tightly regulated, both temporally and spatially. When isolated from the developing tooth germ,

Structural changes in fluorosed dental enamel of red deer (Cervus elaphus L.) from a region with severe environmental pollution by fluorides.

A macroscopic, microradiographic and scanning electron microscope study was performed on the structure of fluorosed dental enamel in red deer from a fluoride polluted region (North Bohemia, Czech Republic). As was revealed by analysis of mandibular bone fluoride content, the rate of skeletal fluoride accumulation in the fluorotic deer was about 6 times that in controls taken from a region not exposed to excessive fluoride deposition. In all fluorosed mandibles, the 1st molar was consistently less fluorotic than the other permanent teeth. This was related to the fact that crown formation in the

Tooth induction and temporal patterning in palatal epithelium of fetal mice.

The present study examined the effect of aging on epithelium and on its ability to respond to an inductive stimulus provided by murine dental papillae. In fetal CD-1 mice, 15- to 17-day molar mesenchyme was combined with 15- to 19-day epithelium from the secondary palates. Enamel organs were separated from the dental papillae, and palatal epithelium was peeled away from its underlying mesenchyme after treatment with 1% trypsin. Recombinants of epithelium and papillae were initially cultured on a solidified complex medium for 24 hr followed by an additional 10-14 days of intraocular explanation

Influence of colchicine and vinblastine on the intracellular migration of secretory and membrane glycoproteins: I. Inhibition of glycoprotein migration in various rat cell types as shown by light microscope radioautography after injection of 3H-fucose.

Previous studies have shown that colchicine and vinblastine inhibit secretion in many cell types by interrupting the normal intracellular migration of secretory products. In the present work, radioautography has been used to study the effects of these drugs on migration of membrane and secretory glycoproteins in a variety of cell types. Young (40 gm) rats were given a single intravenous injection of colchicine (4.0 mg) or vinblastine (2.0 mg). At 10 min after colchicine and 30 min after vinblastine administration, the rats were injected with 3H-fucose. Control rats received 3H-fucose only. All

Diagnostic dilemma of unicystic ameloblastoma: novel parameters to differentiate unicystic ameloblastoma from common odontogenic cysts.

AIM: Diagnostic criteria that have been specified for unicystic ameloblastomas (UAs) are not always helpful to differentiate these cystic tumors from common odontogenic cysts. The aim of this study therefore was to identify additional histopathological features (other than the features considered for the diagnosis of UA at present) that would be helpful to differentiate UA from odontogenic cysts. METHODS: One hundred histopathologically confirmed unicystic ameloblastomas and 20 cases each of radicular, inflamed dentigerous and non-inflamed dentigerous cysts were selected. Immunolocalization and activity of the MMP-9 and MMP-2 in odontogenic region of the rat incisor tooth after post shortening procedure.

MMP-9 and MMP-2 are metalloproteinases which degrade the denatured collagen fibers. However, there is no report about roles of these MMPs in the odontogenic region of the adult rat incisor tooth under different eruption conditions. Male Wistar rats were divided in a normofunctional group (NF) in which their lower teeth remained in a normal eruption. In a hypofunctional group (HP) rats underwent shortening of their lower left incisor tooth every 2 days during 12 days. The eruption rate as well as the expression and activities of MMP-9 and MMP-2 were evaluated using imunohistochemistry and zymog

Early Life Microplastic Exposure Impairs Mouse Incisor Enamel Formation.

Microplastics (MPs) are pervasive environmental contaminants with growing evidence of systemic toxicity; however, their effects on tooth development during early life remain poorly understood. In the present study, we investigated whether early-life exposure to polystyrene microplastics (PS-MPs) affects tooth eruption, root formation, and enamel development in mice. Pregnant mice were administered 1, 4, and 10 μm PS-MPs (10 μg/kg/day) from gestation through lactation, and offspring continued exposure post-weaning. Mandibles were analyzed at postnatal days 14, 25, and 42 using micro-computed to

Hemimaxillectomy defect reconstruction with a maxillary obturator prosthesis using a 3D-model stereolithography: A case report.

INTRODUCTION: Complete or partial resection of the maxilla during tumor surgery causes oronasal defects, leading to oral-maxillofacial dysfunction, for which the obturator prosthesis is an important treatment option. Traditional manufacturing of obturators involves great complexity, often results in impaired function and fit, and is time-consuming. This paper presents a case of using 3D stereolithography as a guide design for defect reconstruction with a maxillary obturator prosthesis. CASE PRESENTATION: We report a 14-year-old girl who presented with facial deformities followed by swelling a

Intrauterine exposure to omeprazole increases the risk of teeth morphological anomalies in the offspring of a murine model.

Conditions experienced in early life have long-lasting effects on offspring health. Despite this, little is known about how maternal exposure to drugs during pregnancy affects offspring teeth morphogenesis. In humans, omeprazole is a common drug used to mitigate Gastroesophageal Reflux Disease. Importantly, omeprazole is a non-specific proton-pump inhibitor, which may inhibit the proton pumps expressed in the developing tooth germ. To date, however, the effects of intrauterine life exposure to omeprazole on offspring tooth development remain unknown. In this study, we addressed this gap in a m
