Hemoglobin I from Lucina pectinata: a model for distal heme-ligand control.

Lucina pectinata hemoglobin I (HbI), which is a ferric sulfide-reactive hemeprotein, contains a distal pocket characterized by the presence of GlnE7 and PheB10. To elucidate the structural-functional properties of HbI, oxygen binding kinetics and FTIR studies with recombinant HbI (rHbI) and a set of mutants were conducted using CO and CN- as sensors of the hemeprotein environment. Three nuCO modes were observed for rHbI at 1936 cm(-1) (A3, closed conformer) 1950 cm(-1) (A1,2, closed conformer) and 1960 cm(-1) (A0, open conformer). These nuCO were affected by substitution of GlnE7 and PheB10 in

POPs in edible clams from different Italian and European markets and possible human health risk.

Human intake of persistent organic pollutants (POPs) occurs primarily through diet and fish and other seafood represent the principal means of contamination. Despite this, few legal limits have been established to protect human health from the various chemicals that are alleged to be carcinogenic or endocrine disruptor compounds. Several pools of edible clams (Tapes philippinarum and Venus gallina) bought in different Italian and European markets were examined in order to perform a risk assessment of some of these contaminants and also to identify possible health issues. POP concentrations in

Interference of heavy metal cations with fluorescent Ca2+ probes does not affect Ca2+ measurements in living cells.

In studies about the effects of heavy metals on intracellular Ca2+, the use of fluorescent probes is debated, as metal cations are known to affect the probe signal. In this study, spectrofluorimetric experiments in free solution, using Fluo-3 and Fura-2, showed that Zn2+ and Cd2+ enhanced the probe signal, Cu2+ quenched it, and Hg2+ had no effect. Addition of GSH prevented most of these effects, suggesting the occurrence of a similar protective role in living cells. Digital imaging of living mussel haemocytes loaded with Fura-2/AM or Fluo-3/AM showed that Hg2+, Cu2+ and Cd2+ induced a rise in

Cyanobacterial toxins: occurrence, modes of action, health effects and exposure routes.

Cyanobacterial toxins are produced by terrestrial- fresh-, brackish- and sea-water cyanobacteria of cosmopolitan occurrence. These toxins present acute and chronic hazards to human and animal health and are responsible for isolated, sporadic animal fatalities (mammals, fish, birds) each year. Human health problems are associated with the ingestion of, and contact with cyanobacterial blooms and their toxins. Modes of action of cyanobacterial neurotoxins, hepatotoxins and skin irritants are considered. Recent indications of the accumulation of cyanobacterial toxins in fish, their effect on crop

First bivalve sperm vitrification technique developed with the model species dwarf surfclam Mulinia lateralis.

Vitrification is a fast, simple and cost-effective cryopreservation technique which has emerged as a promising approach for sperm cryopreservation in a variety of animals, including aquaculture species. However, sperm vitrification in bivalves remains undeveloped, although molluscan sperm cryopreservation has been studied for more than 50 years. This investigation aimed to develop a sperm vitrification technique in dwarf surfclam Mulinia lateralia (a model species in bivalve research) and to assess the cryodamage at different levels. The results showed that the synergistic cryoprotective effec

Evidence of differences in the biotransformation of organic contaminants in three species of freshwater invertebrates.

Acute static bioassays were performed using three freshwater invertebrate species (the oligochaete Lumbriculus variegatus, the fingernail clam Sphaerium corneum and the larvae Chironomus riparius) exposed separately to a variety of 14C radiolabelled contaminants. The aim of this work was to investigate if the chemicals remained as parent compounds after the treatments. Chemicals used were 2,4-dichlorophenol; 2,4,5-trichlorophenol; pentachlorophenol; pyrene; Fenpropidin, and Trifluralin. Homogenates of the whole body tissue of each organism were prepared and total radioactivity was measured. Co

Effects of toxic dinoflagellates and toxin biotransformation in bivalves.

Attempts were made to elucidate the different responses of shellfish to paralytic shellfish poison (PSP) and the PSP donor Alexandrium minutum T1. Five species of edible bivalves (Crassostrea gigas, Meretrix lusoria, Mytilus edulis, Ruditapes philippinarum, and Soletellina diphos) were collected and examined for susceptibility to PSP and PSP donor. It was determined that all five bivalves had low susceptibility to PSP following an intramuscular injection (> 300 MU/20 g). The abnormal effects on bivalves were species-specific and varied with the concentration of A. minutum T1. Judging from the

Assessment of Escherichia coli as an indicator of antimicrobial resistance and monitoring of ESBL/AmpC-Producing Enterobacteriaceae in retail seafood commonly consumed in Italy.

This study assessed Escherichia coli as an indicator of antimicrobial resistance (AMR) in retail seafood, integrating microbiological and genomic analyses within a One Health perspective. Overall, E. coli was cultured from 55 (13%) of 422 raw samples from seven seafood categories commonly consumed in Italy. E. coli was detected across multiple seafood types, but particularly in bivalves (23%) and crustaceans (20%). Because these species can accumulate or acquire faecal contaminants from the aquatic environment or along the post-harvest chain, they represent relevant matrices for AMR monitoring

Antimicrobial resistance and genetic diversity of Escherichia coli isolated from marine bivalves.

Escherichia coli can contaminate the marine environment through sewage and accumulate in bivalve molluscs. We assessed antimicrobial resistance (AMR) mechanisms and genetic diversity of 69 E. coli isolates recovered from in natura, boiled, and purchased Perna perna mussels in Niterói city, Brazil. All isolates were sensitive to cefoxitin, imipenem, meropenem, and fosfomycin. In contrast, 61 (88.4%) isolates were not susceptible to streptomycin. We found ten (14.5%) multidrug-resistant (MDR) isolates. Extended-spectrum beta-lactamase (ESBL) production was detected in 30 (43.5%) isolates, with 1

Clam-associated vibriosis, USA, 1988-2010.

Infections with Vibrio spp. have frequently been associated with consumption of bivalve molluscs, especially oysters, but illness associated with clams has also been well documented. We describe the 2312 domestically acquired foodborne Vibrio infections reported to the Cholera and Other Vibrio Illness Surveillance system from 1988 to 2010. Clams were associated with at least 4% (93 persons, 'only clams') and possibly as many as 24% (556 persons, 'any clams') of foodborne cases. Of those who consumed 'only clams', 77% of infections were caused by V. parahaemolyticus. Clam-associated illnesses w

Mechanistic target of rapamycin (mTOR) regulates extracellular trap production in clam Ruditapes philippinarum.

In the present study, a mechanistic target of rapamycin (RpmTOR) gene was identified and characterized in clam Ruditapes philippinarum and its role in extracellular traps (ETs) formation was investigated. Sequence and structural analyses showed that RpmTOR encodes a conserved mTOR family protein containing the characteristic DUF3385, FAT, FRB, PI3Kc, and FATC domains, and phylogenetic analysis clustered it with other molluscan mTOR homologs. RpmTOR is highly expressed in immune-related tissues and is significantly upregulated following infection with Vibrio parahaemolyticus. Functional experim

Investigating the effectiveness of economically sustainable carrier material complexes for marine oil remediation.

The application of bioremediation to marine oil spills is limited due to dilution of either nutrients or hydrocarbonoclastic organisms. This study investigated the effectiveness of three unique natural carrier materials (mussel shells, coir peat and mussel shell/agar complex) which allowed nutrients, hydrocarbonoclastic organisms and oil to be in contact, facilitating remediation. TPH analysis after 30 d showed that mussel shells exhibited the greatest capacity to degrade oil with a 55% reduction (123.3 mg l(-1) from 276 mg l(-1)) followed by mussel shell/agar complex (49%) and coir peat (36%)

Temporal lobe epilepsy caused by domoic acid intoxication: evidence for glutamate receptor-mediated excitotoxicity in humans.

We describe the development of temporal lobe epilepsy in an 84-year-old man who had suffered domoic acid intoxication. Following intoxication he had nausea, vomiting, confusion, and coma. Generalized convulsions and complex partial status epilepticus progressively developed. After 3 weeks he improved and was seizure free with severe residual memory deficit. Electroencephalograms initially showed periodic epileptiform discharges, later evolving to epileptic abnormalities over frontotemporal regions with diffuse slow waves. Eight months after the intoxication the electroencephalogram was normal.
