Sleep patterns, inflammation, and genetic susceptibility with incidence of cardiac conduction disease and its subtypes: A prospective cohort study.

BACKGROUND: Currently, the association between sleep pattern and cardiac conduction disease (CCD) remain largely unexplored. In our study, we aimed to investigate the association between sleep patterns and CCD, the mediating role of inflammation, and the modifying effect of genetic susceptibility. METHOD: Our study included 316,667 participants from UK Biobank. The healthy sleep score (0-5) was calculated through chronotype, sleep duration, insomnia, snoring, and daytime sleepiness. Subsequently, participants were categorized into healthy (≥4), intermediate (2-3), or poor (≤1) sleep patterns.

Childhood maltreatment, circadian preference, and hs-CRP in first-episode, drug-naïve major depressive disorder: total- and symptom-level associations.

BACKGROUND: Major depressive disorder (MDD) is clinically heterogeneous, and psychosocial, circadian, and inflammatory-metabolic factors may relate differently to depressive symptom profiles. We examined childhood maltreatment, circadian preference, and high-sensitivity C-reactive protein (hs-CRP) in relation to overall and symptom-level depressive severity in first episode drug-naïve patients with MDD. METHODS: This cross-sectional study included 133 first episode drug-naïve patients with MDD after excluding participants with hs-CRP ≥10 mg/L. Depression severity was assessed using the Patien

Plasma proteomics of sleep traits reveals systemic immune-metabolic pathways and genetically prioritized proteins.

The plasma proteomic signatures of sleep disturbance remain poorly characterized. Using data from 43,709 predominantly European-ancestry, middle-aged and older UK Biobank participants, we depict a large-scale atlas of plasma proteomic signatures of seven self-reported sleep traits (sleep duration, chronotype, insomnia symptoms, daytime napping, daytime sleepiness, snoring, and ease of getting up in the morning) and a derived sleep health score. We identify 935 proteins associated with at least one sleep trait, converging on lipid metabolism, immune function and inflammation, cell adhesion, and

Beyond One-Size-Fits-All: Individually Tailored Dietary Interventions in Modern Obesity Care.

Obesity is a complex, multifactorial chronic disease characterized by excessive and/or aberrant adiposity that requires interventions beyond conventional calorie-restriction models. Current clinical guidelines converge on behavioral modification, encompassing nutritional therapy, physical activity, stress reduction, and sleep improvement, as the cornerstone of obesity management, with psychological therapy, pharmacotherapy, and bariatric procedures as adjunctive or escalating options. While traditional "one-size-fits-all" dietary approaches frequently yield suboptimal long-term outcomes, preci

Sleep-Related Factors in Shift Workers: A Cross-Sectional Cohort Pilot Study to Inform Online Group Therapy for Insomnia.

Shift workers face a heightened risk of insomnia. Recent research has yielded promising insights, but further progress is necessary to better treat insomnia in this group. The present pilot study evaluates how different characteristics impact sleep in shift workers to develop an innovative therapeutic approach. An online survey was administered to an ad hoc sample of N = 225 (112 shift workers), and correlations were calculated between sleep variables and specific characteristics (e.g., psychological impairment, personality traits, sleep-related behavior, attitudes towards sleep and shift work

Sleep Patterns and Risk of New-Onset Stress Urinary Incontinence: The UK Biobank Prospective Cohort Study.

OBJECTIVE: The association between sleep behaviors and stress urinary incontinence (SUI) risk remains uncertain. We aimed to evaluate the relationship between sleep patterns, based on a combination of five major sleep behaviors, and the risk of new-onset SUI. METHODS: The study included 406 921 participants from the UK Biobank who were free of SUI at baseline. A healthy sleep score was created by combining five major sleep behaviors: chronotype, sleep duration, insomnia, snoring, and daytime sleepiness. The primary outcome was new-onset SUI. RESULTS: During a median follow-up of 12.0 years,

Classifying weekday-weekend sleep pattern subtypes via latent class analysis: Associations with daytime functioning.

Individuals commonly exhibit different sleep patterns between weekdays and weekends, notably weekend catch-up sleep (wCUS: increased duration) and social jet lag (SJL: delayed timing). This study simultaneously examined both sleep timing and duration to identify distinct patterns of weekday-weekend sleep differences using latent class analysis and to explore their associations with chronotype, sleep disturbance, and well-being. A total of 850 adults (aged 20-55) without diagnosed sleep or psychiatric disorders and not engaged in shift work participated in an online survey assessing their sleep

Within-subject and between-subject variability in responses to a mind-body intervention in electrophysiological brain activity.

INTRODUCTION: Mind-body interventions (MBIs) increasingly used in health care include alterations in the ordinary state of consciousness that might be essential for their effects on psychological and physical patterns encoded in the unconscious. Little is known about the effects of repeated practice or the time of the repetitions. METHODS: EEG-derived Bispectral-Index (BIS) is clinically used to monitor anesthetic depth, a deviation from the normal waking consciousness. We used BIS-monitoring in six participants who repeated a MBI ten times within 3-4 days at different times of day. RESULTS:

Additive framework of hormonal waves accounts for species and age differences in circadian intraocular pressure rhythm.

Elevated intraocular pressure (IOP) is the primary risk factor for glaucoma, yet IOP demonstrates significant circadian rhythms whose underlying mechanisms remain incompletely understood, and disruption of this rhythm heightens disease susceptibility. A paradox exists in that both diurnal and nocturnal animals experience nocturnal IOP elevation despite their contrasting behavioral chronotypes. Here, we developed a minimal mathematical framework in which IOP rhythms arise from the linear superposition of two sinusoidal signals: adrenal glucocorticoids (GC) and norepinephrine (NE) from the super

Healthy Lifestyles Modify the Association of Melatonin Receptor 1B Gene and Ischemic Stroke: A Family-Based Cohort Study in Northern China.

Limited research has reported the association between MTNR1B gene polymorphisms and ischemic stroke (IS), and there is insufficient evidence on whether adopting a healthy lifestyle can mitigate genetic risks in this context. This study aimed to investigate the associations between MTNR1B gene variants (rs10830963 and rs1387153) and IS, examining the potential effect of gene-lifestyle interactions on IS risk. Conducted in northern China, this family-based cohort study involved 5116 initially IS-free subjects. Genotype data for rs10830963 and rs1387153 in MTNR1B were collected. Eight modifiable

Multi-omics correlates of insulin resistance and circadian parameters mapped directly from human serum.

While it is generally known that metabolic disorders and circadian dysfunction are intertwined, how the two systems affect each other is not well understood, nor are the genetic factors that might exacerbate this pathological interaction. Blood chemistry is profoundly changed in metabolic disorders, and we have previously shown that serum factors change cellular clock properties. To investigate if circulating factors altered in metabolic disorders have circadian modifying effects, and whether these effects are of genetic origin, we measured circadian rhythms in U2OS cell in the presence of ser

Circadian Disruption Alters the Effects of Lipopolysaccharide Treatment on Circadian and Ultradian Locomotor Activity and Body Temperature Rhythms of Female Siberian Hamsters.

The effect of circadian rhythm (CR) disruption on immune function depends on the method by which CRs are disrupted. Behavioral and thermoregulatory responses induced by lipopolysaccharide (LPS) treatment were assessed in female Siberian hamsters in which circadian locomotor activity (LMA) rhythms were eliminated by exposure to a disruptive phase-shifting protocol (DPS) that sustains arrhythmicity even when hamsters are housed in a light-dark cycle. This noninvasive treatment avoids genome manipulations and neurological damage associated with other models of CR disruption. Circadian rhythmic (R

Demographic factors, fatigue, and driving accidents: An examination of the published literature.

This article reviews the literature pertaining to the association between demographic variables (e.g., age, sex, race, socio-economic status) with fatigue, and when feasible, accident risk. It also explores their potential influence and interaction with some working arrangements, commute time, personality characteristics, and circadian chronotype. Fatigue has been implicated in a range of impairments that can have detrimental effects on individuals, and it is differentially associated with conventional demographic variables. However, several major methodological limitations prevent clear concl

Melatonin treatment of winter depression following total sleep deprivation: waking EEG and mood correlates.

Patients with winter depression (seasonal affective disorder (SAD)) commonly complain of sleepiness. Sleepiness can be objectively measured by spectral analysis of the waking electroencephalogram (EEG) in the 1-10 Hz band. The waking EEG was measured every 3 h in 16 female SAD patients and 13 age-matched control women throughout a total sleep deprivation of 30 h. Melatonin (or placebo) under double-blind conditions was administered subsequently (0.5 mg at 1700 h for 6 days), appropriately timed to phase advance circadian rhythms, followed by reassessment in the laboratory for 12 h. The increas

Rewinding the gut clock: from chronodisruption to targeted therapies.

Circadian rhythms orchestrate daily oscillations in physiology and behaviour, enabling optimal alignment between internal biological processes and environmental cycles. A hierarchical system centred in the suprachiasmatic nucleus and supported by peripheral clocks across tissues ensures temporal alignment of metabolic, immune and GI functions. Within the GI tract, local clocks drive rhythmic programmes that regulate digestion, absorption, motility, hormone secretion, immune responses and interactions with the gut microbiota. Feeding-fasting cycles and microbial metabolites act as powerful zeit
