Photodynamic therapy of tumour cells mediated by the natural anthraquinone parietin and blue light.

Photodynamic therapy (PDT) is a treatment for superficial tumours involving the administration of a photosensitiser followed by irradiation. The potential of the natural anthraquinone parietin (PTN) in PDT is still relatively unexploited. In the present work, PTN isolated from the lichen Teoloschistes nodulifer (Nyl.) Hillman (Telochistaceae) was evaluated as a potential photosensitiser on tumour cells employing UVA-Vis and blue light. Blue light of 2 J/cm2 induced 50% death of K562 leukaemic cells treated 1 h with 30 μM PTN (Protocol a). Higher light doses (8 J/cm2) were needed to achieve the

Plant Secondary Metabolites on Efflux-Mediated Antibiotic Resistant Stenotrophomonas Maltophilia: Potential of Herbal-Derived Efflux Pump Inhibitors.

During the process of adapting to metal contamination, plants produce secondary metabolites that have the potential to modulate multidrug-resistant (MDR) phenotypes; this is achieved by inhibiting the activity of efflux pumps to reduce the minimum inhibitory concentrations (MICs) of antimicrobial substrates. Our study evaluated the effect of secondary metabolites of belowground parts of Pteris vittata L. and Fallopia japonica, two metal-tolerant plants from northern Vietnam, on six antibiotic-resistant Stenotrophomonas maltophilia strains possessing efflux pump resistance mechanisms that were

Limited Nrf2 activation and heterogeneous thyroidal effects in a 424-compound multi-assay screen call for rigorous testing of purported antioxidant and health-promoting supplements.

We screened 424 natural compounds, including many marketed as dietary supplements, to assess their antioxidant and thyroidal effects. In rat PCCL3 thyroid follicular cells, we performed parallel primary screens for antioxidant response element (ARE) activation, thyroglobulin (Tg) promoter activity, sodium/iodide symporter (NIS) promoter activity and iodide uptake. Hits were tested in downstream analyses, including ARE activation in Nrf2-knockout cells, an Nrf1 stabilization reporter, endogenous Nqo1, Tg and NIS mRNA and protein levels, and relationships between these readouts. Of 424 compounds

Therapeutic potential of traditional herbal plants and their polyphenols in alleviation of mercury toxicity.

Mercury (Hg) is a major environmental contaminant significantly impacting human health. As a naturally occurring element, mercury has been extensively mobilized into aquatic and terrestrial ecosystems over thousands of years, largely due to anthropogenic activities such as mining and metal extraction. Acute mercury toxicity causes extensive physiological damage, affecting vital organs including the kidneys, heart, liver, brain, and skin. Phytochemicals, known for their diverse pharmacological properties, have shown promise in mitigating metal-induced toxicities, including mercury. These compou

Mild synthesis of biomass-derived fluorescent Fe,N-carbon dots for fluorometric and smartphone-assisted visual detection of aloin.

Herein, a mild one-pot synthesis of fluorescent Fe,N-doped carbon dots (Fe-NCDs) was developed from biomass waste in a choline chloride/urea deep eutectic solvent (DES) with FeCl3 as a promoter. The DES disrupts the lignocellulosic network while FeCl3 catalyzes carbonization and provides in situ Fe doping. The resulting Fe-NCDs exhibited bright blue emission with a high quantum yield of 38.5% owing to the introduction of Fe and N elements. The probe showed outstanding environmental stability but could be selectively fluorescence quenched toward aloin because of static coordination and the inne

Aloin attenuates high-glucose-induced oxidative and inflammatory damage in renal cells via AMPK/Nrf2 signaling.

BACKGROUND: Diabetic nephropathy (DN) is a leading cause of end-stage renal disease, with proximal tubular injury and oxidative stress playing pivotal roles in its progression. The AMPK-Nrf2 signaling axis is a key regulator of antioxidant defense, but its modulation by Aloin in DN-relevant renal models remains insufficiently characterized. Aloin, a natural anthraquinone glycoside from Aloe species, exhibits antioxidant, anti-inflammatory, and anti-fibrotic properties in non-renal systems. This study investigated whether Aloin protects renal cells from high-glucose-induced injury and whether A

Natural-derived compounds and their mechanisms in potential autosomal dominant polycystic kidney disease (ADPKD) treatment.

BACKGROUND: Autosomal dominant polycystic kidney disease (ADPKD) is a monogenic kidney disorder that impairs renal functions progressively leading to kidney failure. The disease affects between 1:400 and 1:1000 ratio of the people worldwide. It is caused by the mutated PKD1 and PKD2 genes which encode for the defective polycystins. Polycystins mimic the receptor protein or protein channel and mediate aberrant cell signaling that causes cystic development in the renal parenchyma. The cystic development is driven by the increased cyclic AMP stimulating fluid secretion and infinite cell growth. I

Fungal quinones: diversity, producers, and applications of quinones from Aspergillus, Penicillium, Talaromyces, Fusarium, and Arthrinium.

Quinones represent an important group of highly structurally diverse, mainly polyketide-derived secondary metabolites widely distributed among filamentous fungi. Many quinones have been reported to have important biological functions such as inhibition of bacteria or repression of the immune response in insects. Other quinones, such as ubiquinones are known to be essential molecules in cellular respiration, and many quinones are known to protect their producing organisms from exposure to sunlight. Most recently, quinones have also attracted a lot of industrial interest since their electron-don
