Identification of genes directly regulated by LvBlimp1 in shrimp macrophage-like phagocytes.

Litopenaeus vannamei B lymphocyte-induced maturation protein 1 (LvBlimp1) is a transcription factor expressed in macrophage-like phagocytes that modulates immune responses via direct bactericidal activity, opsonization, and phagocytosis. However, the molecular targets and regulatory architecture by which LvBlimp1 controls shrimp disease resistance remain poorly defined. In this study, ChIP-seq was performed in shrimp hemocytes to identify genome-wide binding targets of LvBlimp1. Five core downstream genes (MAP1B, SLC46, TACC1, COL3A1, and LGMN) are directly regulated by LvBlimp1 were identifie

First report of Bonamia ostreae in wild flat oysters (Ostrea denselamellosa) from the west coast of Korea: Molecular and histopathological confirmation.

Bonamia ostreae is a protozoan parasite of significant concern in oyster aquaculture due to its impact on flat oyster species. In this study, we screened 30 wild flat oysters (Ostrea denselamellosa) collected from the western coastal region of South Korea for B. ostreae infection using two molecular assays. Species-specific BOSTRE-F/R primers targeting the ITS region detected 14/30 (46.67 %) positive samples, while Bonamia genus-specific Bo/Boas primers targeting the SSU rRNA region yielded 19/30 (63.33 %) positives. Although the higher detection rate with Bo/Boas primers raised the possibilit

Histopathological characterization and fluorescence in situ hybridization of Cyprinid herpesvirus 2 in cultured Prussian carp, Carassius auratus gibelio in China.

Cyprinid herpesvirus 2 (CyHV-2) is an emerging pathogen in the commercially exploited fish, Prussian carp (Carassius auratus gibelio), which has caused huge economic loss in China and appears to be spreading worldwide. In this article, CyHV-2 infection of Prussian carp was confirmed for the first time by polymerase chain reaction (PCR), which gave positive results from the tissue samples dissected from moribund fish including kidney, spleen, liver, and gill. Histological examination showed systemic inflammatory reactions in the infected tissues, with infiltration of hemocytes, hypertrophied nu

Proventriculus (cardia) plays a crucial role in immunity in tsetse fly (Diptera: Glossinidiae).

Fat body and hemocytes play a central role in cellular and humoral responses for systemic infections in invertebrates, similar to the mammalian liver and blood cells. Epithelial surfaces, in particular the midgut, participate in the initial local immune responses in order to aid in the generation of the terminal cytotoxic molecules that mediate non-self recognition. Here, we describe for the first time the immune responses of a cluster of cells at the foregut/midgut junction--known as proventriculus (cardia) in the medically and agriculturally important insect, tsetse fly (Diptera: Glossinidae

Activation of Toll and IMD pathways in the Drosophila brain following local and systemic bacterial infection.

Brain infections are often life-threatening and have been linked to the development of neurodegenerative diseases. The fruit fly Drosophila melanogaster is a valuable experimental model to study immunity and the pathophysiology of brain infections. The exact cellular pathways through which brain-specific immune responses are mounted in Drosophila, however, remain poorly characterized. Here, we investigated how brain-specific or systemic infection with Micrococcus luteus and Escherichia coli bacteria activates the Drosophila NF-κB innate immune pathways Toll and immune deficiency (IMD) in the c

Long-term exposure of marine mussels to paracetamol: is time a healer or a killer?

Pharmaceuticals pose a major threat to the marine environment, and several studies have recently described their negative effects on marine organisms. Pharmaceutical compounds are constantly being released into aquatic ecosystems, and chronic exposure, even at low concentrations, may have a major impact on marine organisms. The purpose of the present study is to evaluate the biological changes induced by one of the most widely used pharmaceuticals-paracetamol-in the blue mussel Mytilus edulis, after a long-term exposure at environmentally relevant concentrations. We present our data alongside

The structure of a prophenoloxidase (PPO) from Anopheles gambiae provides new insights into the mechanism of PPO activation.

BACKGROUND: Phenoloxidase (PO)-catalyzed melanization is a universal defense mechanism of insects against pathogenic and parasitic infections. In mosquitos such as Anopheles gambiae, melanotic encapsulation is a resistance mechanism against certain parasites that cause malaria and filariasis. PO is initially synthesized by hemocytes and released into hemolymph as inactive prophenoloxidase (PPO), which is activated by a serine protease cascade upon recognition of foreign invaders. The mechanisms of PPO activation and PO catalysis have been elusive. RESULTS: Herein, we report the crystal struct

Coelomocytes and post-traumatic response in the common sea star Asterias rubens.

Coelomocytes are recognized as the main cellular component of the echinoderm immune system. They are the first line of defense and their number and type can vary dramatically during infections or following injury. Sea stars have been used as a model system to study the regeneration process after autotomy or predation. In the present study we examined the cellular and biochemical responses of coelomocytes from the European sea star Asterias rubens to traumatic stress using immunochemical and biochemical approaches. In terms of trauma and post-traumatic stress period, here we consider the experi

Remote Control of Intestinal Stem Cell Activity by Haemocytes in Drosophila.

The JAK/STAT pathway is a key signaling pathway in the regulation of development and immunity in metazoans. In contrast to the multiple combinatorial JAK/STAT pathways in mammals, only one canonical JAK/STAT pathway exists in Drosophila. It is activated by three secreted proteins of the Unpaired family (Upd): Upd1, Upd2 and Upd3. Although many studies have established a link between JAK/STAT activation and tissue damage, the mode of activation and the precise function of this pathway in the Drosophila systemic immune response remain unclear. In this study, we used mutations in upd2 and upd3 to

Pathological evidence links white feces syndrome in farmed Penaeus vannamei to severe hepatopancreatic Ecytonucleospora hepatopenaei infection in China.

White feces syndrome (WFS) remains a major health problem in global Penaeus vannamei aquaculture, yet its etiology and pathogenesis remain debated. In this study, WFS outbreaks were investigated in 24 shrimp ponds across four provinces in China between 2013 and 2024, using light microscopy, histopathology, in situ hybridization, transmission electron microscopy, and molecular diagnostics. Ecytonucleospora hepatopenaei (EHP) was detected in all WFS-affected ponds examined. Microscopic and ultrastructural analyses showed that both fresh white feces from intestinal tract and pond-floating white f

Histological and transcriptional responses of the Chilean mussel Mytilus chilensis to the toxic dinoflagellates Alexandrium catenella and Protoceratium reticulatum.

Harmful algal blooms (HABs) are natural phenomena caused by toxin-producing microalgae whose occurrence appears to have increased worldwide. Although microalgae constitute a natural food source for bivalve molluscs, certain species can exert negative effects. The present study assessed the molecular and histological effects of exposure to the toxic dinoflagellates Alexandrium catenella and Protoceratium reticulatum on adult Chilean mussels (Mytilus chilensis). Mussels were exposed for 15 days to 500 cells mL-1 of A. catenella and for 10 days to 100 cells mL-1 of P. reticulatum, with 50,000 cel

Field evidence of nylon fishing-net macroplastic embedded in the tissue of a wild pearl oyster (Pinctada fucata): A histological case observation.

The objective of this study was to document and characterise an opportunistic case of macroplastic embedding within the soft tissues of a wild pearl oyster (Pinctada fucata) and to describe the associated tissue-level responses. Most existing research on plastic pollution in bivalves has focused on microplastic (<5 mm) ingestion, whereas the physical incorporation of macroplastic (>5 mm) debris into bivalve tissues remains poorly documented under field conditions. Four adult P. fucata specimens, obtained as bycatch from artisanal gill-net fishing off the southeastern coast of India (Nagapattin

Chemosensory protein 3 is a brain host factor for the induction of enhanced-locomotory activity in the BmNPV-silkworm infection model.

Although it is reported that the protein tyrosine phosphatase gene of baculovirus (group I nucleopolyhedrovirus) can induce enhanced locomotory activity (ELA) in caterpillars, our understanding of the host factors that are involved in the regulation of the behavioral change is still limited. Previously, single-nucleus RNA sequencing (snRNA-seq) was used to identify 19 distinct clusters representing Kenyon cell, glial cell, olfactory projection neuron, optic lobes neuron, hemocyte, muscle cell types and other unannotated cells in the silkworm larvae brains. Differential bioaccumulation and histopathological changes induced by iron oxide nanoparticles and ferric chloride in the neotropical snail Biomphalaria glabrata.

Iron oxide nanoparticles (IONPs) are prominent in nanomedicine, cosmetics, and environmental applications. However, their increasing production, use, and release into the environment raises concerns about their potential risks to aquatic life and human health. This study aimed to evaluate the bioaccumulation, as concentration of small, medium and large iron aggregates in the digestive tubules, histopathological changes, and inflammatory responses in the freshwater snail Biomphalaria glabrata following chronic exposure to gluconic-acid functionalized IONPs (GLA-IONPs) compared to their dissolve

Natural genetic variation in Drosophila melanogaster reveals genes associated with Coxiella burnetii infection.

The gram-negative bacterium Coxiella burnetii is the causative agent of Query (Q) fever in humans and coxiellosis in livestock. Host genetics are associated with C. burnetii pathogenesis both in humans and animals; however, it remains unknown if specific genes are associated with severity of infection. We employed the Drosophila Genetics Reference Panel to perform a genome-wide association study to identify host genetic variants that affect host survival to C. burnetii infection. The genome-wide association study identified 64 unique variants (P < 10-5) associated with 25 candidate genes. We e
