Effects of Water and Nitrogen Regulation on Alfalfa (Medicago sativa L.) Production Performance Through Optimization of Nitrogen Metabolism.

Water and nitrogen application rates directly affect crop yield formation and protein accumulation. Nitrogen metabolism, as a key physiological process linking water and nitrogen supply with crop growth, plays a critical role in regulating nitrogen uptake, transformation, and accumulation. However, the functional relationships among different nitrogen metabolism indicators in mediating the formation of high-quality and high-yield alfalfa under water-nitrogen regulation remain unclear. In this study, alfalfa (Medicago sativa L.) was subjected to four nitrogen application levels [N0 (0 kg·hm-2),

Exploring the role of Arthrospira platensis and earthworms (Lumbricus castaneus) biosystem in mitigating antimony toxicity and enhancing the biochemical properties of cucurbit plants.

The rising industrial use of antimony (Sb) has led to its widespread pollution, negatively impacting plant growth as an emerging pollutant. Limited studies have explored the integrated biosystems involving Arthrospira platensis (AP) and/or earthworms (EW) on cucurbit plants under Sb-stress. This study uncovered the toxicity of three levels of 'Sb' on soil characters and biochemical changes of cucurbit leaves with/without EW and/or AP. The GC-mass analysis of AP-extract documented the presence of 33 bioactive compounds; including 11,13-dimethyl-12-tetradecen-1-ol acetate, lanostane-7,11-dione,

Highly selective ergosterol binding and impaired redox balance leads to improved antileishmanial efficacy for amphotericin b synthesized silver nanoparticles with reduced toxicity- In vitro and in vivo studies.

We aim to develop a low-cost silver nanoparticle (AgNP)-based delivery of AmB (AmB-AgNP) which can replace the costly AmBisome and toxic Fungizone formulation for applications against visceral leishmaniasis (VL) caused by the parasite Leishmania donovani (LD). Using different molar ratios of AmB and silver nitrate, we have identified a specific NP of ∼110 nm (zeta potential of -36.7 mV and PDI of 0.15) as the ideal antileishmanial agent with increased efficacy than AmB against LD promastigotes and amastigotes. These NPs were characterized by UV-visible, DLS, Zeta potential, FT-IR, DSC, and FE-

Nitric oxide application alleviates fungicide and ampicillin co-exposure induced phytotoxicity by regulating antioxidant defense, detoxification system, and secondary metabolism in wheat seedlings.

Pesticides and antibiotics usually sink into soil, posing serious phytotoxic effects on plants. However, studies are elusive regarding the phytotoxic effects of fungicide Consento (CON) and antibiotic ampicillin (AMP) co-exposure. Nitric oxide (NO) is an important plant signaling molecule known for abiotic stress tolerance in plants. This study investigated the phytotoxic effects of CON and/or AMP on the growth and antioxidant activities of wheat (Triticum aestivum L.) seedlings and unveiled the underlying mechanisms induced by the application of NO as sodium nitroprusside (SNP; 100 μM) in whe

Aerobic exercise protects against pressure overload-induced cardiac dysfunction and hypertrophy via β3-AR-nNOS-NO activation.

Aerobic exercise confers sustainable protection against cardiac hypertrophy and heart failure (HF). Nitric oxide synthase (NOS) and nitric oxide (NO) are known to play an important role in exercise-mediated cardioprotection, but the mechanism of NOS/NO stimulation during exercise remains unclear. The aim of this study is to determine the role of β3-adrenergic receptors (β3-ARs), NOS activation, and NO metabolites (nitrite and nitrosothiols) in the sustained cardioprotective effects of aerobic exercise. An HF model was constructed by transverse aortic constriction (TAC). Animals were treated wi

Probing the catalytic mechanism of sulfite reductase by X-ray crystallography: structures of the Escherichia coli hemoprotein in complex with substrates, inhibitors, intermediates, and products.

To further understand the six-electron reductions of sulfite and nitrite catalyzed by the Escherichia coli sulfite reductase hemoprotein (SiRHP), we have determined crystallographic structures of the enzyme in complex with the inhibitors phosphate, carbon monoxide, and cyanide, the substrates sulfite and nitrite, the intermediate nitric oxide, the product sulfide (or, most likely, an oxidized derivative thereof), and an oxidized nitrogen species (probably nitrate). A hydrogen-bonded cage of ligand-binding arginine and lysine side chains, ordered water molecules, and siroheme carboxylates provi

Experimental Evidence that In Vivo Intracerebral Administration of L-2-Hydroxyglutaric Acid to Neonatal Rats Provokes Disruption of Redox Status and Histopathological Abnormalities in the Brain.

Tissue accumulation of L-2-hydroxyglutaric acid (L-2-HG) is the biochemical hallmark of L-2-hydroxyglutaric aciduria (L-2-HGA), a rare neurometabolic inherited disease characterized by neurological symptoms and brain white matter abnormalities whose pathogenesis is not yet well established. L-2-HG was intracerebrally administered to rat pups at postnatal day 1 (P1) to induce a rise of L-2-HG levels in the central nervous system (CNS). Thereafter, we investigated whether L-2-HG in vivo administration could disturb redox homeostasis and induce brain histopathological alterations in the cerebral

Nitrite Generating and Depleting Capacity of the Oral Microbiome and Cardiometabolic Risk: Results from ORIGINS.

Background The enterosalivary nitrate-nitrite-nitric oxide (NO3-NO2-NO) pathway generates NO following oral microbiota-mediated production of salivary nitrite, potentially linking the oral microbiota to reduced cardiometabolic risk. Nitrite depletion by oral bacteria may also be important for determining the net nitrite available systemically. We examine if higher abundance of oral microbial genes favoring increased oral nitrite generation and decreased nitrite depletion is associated with a better cardiometabolic profile cross-sectionally. Methods and Results This study includes 764 adults (m

Enzymes involved in the anaerobic degradation of phenol by the sulfate-reducing bacterium Desulfatiglans anilini.

BACKGROUND: The sulfate-reducing bacterium Desulfatiglans anilini can grow with phenol as sole source of carbon and energy under strictly anaerobic, sulfate-reducing conditions. In the nitrate-reducing bacterium Thauera aromatica, the enzymes involved in phenol degradation have been well elucidated, whereas the anaerobic phenol degradation pathway by D. anilini was not studied in detail yet. RESULTS: The pathway of anaerobic phenol degradation by the sulfate-reducing bacterium Desulfatiglans anilini was studied by identification of genes coding for phenylphosphate synthase (encoded by pps gen

Transcription activation at Escherichia coli FNR-dependent promoters by the gonococcal FNR protein: effects of a novel S18F substitution and comparisons with the corresponding substitution in E. coli FNR.

The Neisseria gonorrhoeae genome encodes a homologue of the Escherichia coli FNR protein (the fumarate and nitrate reductase regulator). Despite its similarity to E. coli FNR, the gonococcal FNR only partially complemented an E. coli fnr mutation. After error-prone PCR mutagenesis of the gonococcal fnr gene, we identified four mutant fnr derivatives carrying the same S18F substitution, and we showed that the mutant FNR could activate transcription from a range of class I and class II FNR-dependent promoters in E. coli. Prompted by the similarities between gonococcal and E. coli FNR, we made ch

Abnormal levels of antioxidant defenses in a large sample of patients with hypertensive disorders of pregnancy.

Increased levels of oxidative stress have been demonstrated in Preeclampsia in previous studies, but this finding has not been established in other hypertensive disorders in pregnancy (HDP). We measured different markers of lipid peroxidation and antioxidant defenses by spectrophotometry or enzymoimmunoanalysis in 339 pregnant women: 85 with gestational hypertension (GH), 88 chronic hypertension (CH), 104 Preeclampsia and 62 healthy pregnant control women (PCW). Lower activity of superoxide dismutase and higher levels of catalase were found in GH, CH and preeclampsia compared with PCW (964.4±1

Conventional and novel drug therapeutics to relief myocardial ischemia.

Effective management of stable angina usually includes drug therapy. There are several agents that are considered vasculoprotective such as aspirin, angiotensin converting enzyme inhibitors and statins. Conventional anti ischemic therapy includes nitrates, beta-blockers and calcium-channel blockers. In recent years, several other drugs with novel anti ischemic mechanisms have become available including ranolazine, ivabradine, nicorandil and many others. This article reviews drugs that alleviate the symptoms of chronic angina with emphasis on several novel pharmacological agents.

Detection of the His-heme Fe2+-NO species in the reduction of NO to N2O by ba3-oxidase from thermus thermophilus.

Reaction pathways in the enzymatic formation and cleavage of the N-N and N-O bonds, respectively, are difficult to verify without the structure of the intermediates, but we now have such information on the heme a(3)(2+)-NO species formed in the reaction of ba(3)-oxidase with NO from resonance Raman spectroscopy. We have identified the His-heme a(3)(2+)-NO/Cu(B)(1+) species by its characteristic Fe-NO and N-O stretching frequencies at 539 and 1620 cm(-)(1), respectively. The Fe-NO and N-O frequencies in ba(3)-oxidase are 21 and 7 cm(-)(1) lower and higher, respectively, than those observed in M
