A new calcium channel antagonist, lomerizine, alleviates secondary retinal ganglion cell death after optic nerve injury in the rat.

PURPOSE: We investigated whether lomerizine, a new diphenylmethylpiperazine calcium channel blocker, exerted a neuroprotective effect on axonal or retinal damage induced by optic nerve injury in the rat. METHODS: A partial crush lesion was inflicted unilaterally on the optic nerve, 2 mm behind the globe, in adult Wistar albino rats. Animals were treated with the vehicle, 10 or 30 mg/kg lomerizine. Each solution was given orally twice daily for 4 weeks. One week before euthanization, Fluoro-Gold (FG) was injected into both superior colliculi to retrogradely label surviving retinal ganglion cel

Anesthetic Preconditioning as Endogenous Neuroprotection in Glaucoma.

Blindness in glaucoma is the result of death of Retinal Ganglion Cells (RGCs) and their axons. RGC death is generally preceded by a stage of reversible dysfunction and structural remodeling. Current treatments aimed at reducing intraocular pressure (IOP) are ineffective or incompletely effective in management of the disease. IOP-independent neuroprotection or neuroprotection as adjuvant to IOP lowering in glaucoma remains a challenge as effective agents without side effects have not been identified yet. We show in DBA/2J mice with spontaneous IOP elevation and glaucoma that the lifespan of fun

Midbrain atrophy in subcortical ischemic vascular dementia.

Magnetic resonance imaging (MRI) has revealed age-related changes in midbrain volume in normal subjects. Atrophy of the midbrain in patients with progressive supranuclear palsy (PSP) has been demonstrated using conventional brain MRI, and it was recently reported that some patients with vascular dementia also have midbrain atrophy. The aim of the present study was to investigate if the patients with subcortical ischemic vascular dementia (SIVD) have atrophic changes in the midbrain. MRIs of 23 SIVD patients, 18 probable PSP patients, and 96 controls were retrospectively analyzed. Differences i

Brainstem neuropathology in a mouse model of Niemann-Pick disease type C.

Niemann-Pick disease type C (NPC) is a neurovisceral lipid storage disorder characterized by progressive and widespread neurodegeneration. Although some characteristic symptoms of NPC result from brainstem dysfunction, little information is available about which brainstem structures are affected. In this study, the brainstems of mutant BALB/c NPC1-/- mice with a retroposon insertion in the NPC1 gene were examined for neuropathological changes. In the midbrain, the integrated optic density (IOD) and cell count density of tyrosine-hydroxylase (TH) immunostained neurons were decreased in the subs

Endothelin-1 impairs retrograde axonal transport and leads to axonal injury in rat optic nerve.

The purpose of this study was to examine the effects of endothelin-1 (ET-1) on retrograde axonal transport in the rat optic nerve. Vehicle or ET-1 (0.2, 1, or 5 pmol/eye) were injected into the vitreous body in Sprague-Dawley rats. Retinal vessels were observed, using a fundus camera, before, and at 10 min, 3 days and 7 days after a single intravitreous injection. Two days after the injection, a neuronal tracer, fluoro gold, was administered via the superior colliculi to retrogradely label active retinal ganglion cells (RGCs). Five days after the tracer administration, retrogradely labeled RGC

The orientating reflex: the "targeting reaction" and "searchlight of attention".

A concept of the orientating reflex is presented, based on the principle of vector coding of cognitive and executive processes. The orientating reflex is a complex of orientating reactions of motor, autonomic, and subjective types, accentuating new and significant stimuli. Two main systems form the orientating reflex: the "targeting reaction" and the "searchlight of attention:" In the visual system, the targeting reaction ensures that the image of the object falls onto the fovea; this is mediated by involvement of premotor neurons which are excited by saccade command neurons in the superior co

The human brain at stage 16, including the initial evagination of the neurohypophysis.

Thirty-nine sectioned embryos of stage 16 were studied. Up to this stage the amygdaloid body is derived entirely from the medial eminence, which was purely diencephalic in stage 14, but now extends also to the telencephalon. The area of the future olfactory bulb is indicated by the presence of olfactory fibres entering the brain wall; the future olfactory tubercle is characterized by cellular islands. The presence of the hippocampal thickening and various histological features make it possible to outline the main, future cortical areas already at this early stage: archi-, paleo-, and neopalliu

A shared speed encoding model for running and backing away behaviours in segregated neural circuits.

How neuronal firing within a circuit encodes behavioural intensity, like running speed, is largely unknown. Projections from temporal association cortex (TeA) and superior colliculus (SC) to dorsal periaqueductal grey (dPAG) circuit can both trigger running behaviour. Using in vivo loose-patch recordings with circuit manipulations in mice, we quantified a firing - speed relationship and established its encoding model. Here, we report two behavioural patterns induced by circuit activation: backing away and rebound running. Mechanistically, dPAG CaMKIIα neurons receiving inputs from either TeA o

Protocadherin 19 regulates axon guidance in the developing Xenopus retinotectal pathway.

Protocadherin 19 (Pcdh19) is a homophilic cell adhesion molecule and is involved in a variety of neuronal functions. Here, we tested whether Pcdh19 has a regulatory role in axon guidance using the developing Xenopus retinotectal system. We performed targeted microinjections of a translation blocking antisense morpholino oligonucleotide to knock down the expression of Pcdh19 selectively in the central nervous system. Knocking down Pcdh19 expression resulted in navigational errors of retinal ganglion cell (RGC) axons specifically at the optic chiasm. Instead of projecting to the contralateral op

Electrophysiological and pharmacological characterization of spreading depolarization in the adult zebrafish tectum.

Spreading depolarization (SD) is a slowly propagating wave of neuronal and glial depolarization. A growing number of studies show that SD and SD-like phenomena play a role in neurological disorders such as migraine, stroke, and traumatic brain injury. Despite the clinical importance of SD, its underlying molecular and cellular mechanisms remain elusive, possibly because of insufficient animal model allowing genetic manipulation. Such a model would also allow high-throughput screening for SD-suppressing drug development. To address this, we developed a novel experimental system to study SD usin

Influence of sympathetic fibers on noise-induced hearing loss in the chinchilla.

The influence of the sympathetic efferent fibers on cochlear susceptibility to noise-induced hearing loss is still an open question. In the current study, we explore the effects of unilateral and bilateral Superior Cervical Ganglion (SCG) ablation in the chinchilla on hearing loss from noise exposure, as measured with inferior colliculus (IC) evoked potentials, distortion product otoacoustic emissions (DPOAE), and outer hair cell (OHC) loss. The SCG was isolated at the level of the bifurcation of the carotid artery and removed unilaterally in 15 chinchillas. Another eight chinchillas underwent

Reduced influence of the ipsilateral ear on spatial tuning of auditory neurons in the albino superior colliculus: a knock-on effect of anomalies of the acoustic chiasm?

Auditory brainstem abnormalities affecting decussation patterns and nuclei involved in the acoustic chiasm exist in a variety of albino mammals, suggesting that binaural processes underlying spatial hearing may be disrupted in these mutants. To evaluate this we have compared the contribution of the two ears in albino and normally pigmented guinea pigs to the spatial tuning of auditory neurons in the deep layers of the superior colliculus (SC). Broadband noise stimuli at threshold and at suprathreshold intensities were presented from different azimuthal loudspeaker locations under free-field an

Protective effects of selegiline and desmethylselegiline against N-methyl-D-aspartate-induced rat retinal damage.

Selegiline, a therapeutic agent of Parkinson's disease, and its metabolite, desmethylselegiline, were explored for their neuroprotective effects against N-methyl-D-aspartate (NMDA)-induced cell death in rat retina. Morphometric analysis of the retina revealed that an intravitreal injection of NMDA induced a significant decrease in cell density in the ganglion cell layer and in thickness of the inner plexiform layer, but not of other retinal layers such as the outer nuclear layer. Concurrent intravitreal injection of selegiline with NMDA did not show a significant protective effect, whereas co-

[A model of calibrated optic nerve crush injury in rats].

OBJECTIVE: To establish a calibrated optic nerve crush injury model in rats. METHODS: Twenty-eight healthy Sprague-Dawley (SD) rats were randomly divided into two groups. Seven rats were in normal control group, i.e. group A. In group B, model of partial optic nerve crush injury group, partial optic nerve crush injury model was induced in the right eyes of another 21 rats by a special designed optic nerve clip with 40-gram holding force at optic nerve 2 mm behind the eyeball clipped for four seconds to partially block the optic nerve axoplasmic transport. The left eyes served as a control. Ac

Central nervous system trans-synaptic effects of acute axonal injury: a 1H magnetic resonance spectroscopy study.

N-acetylaspartate (NAA) has previously been proposed as a neuronal marker. 1H magnetic resonance spectroscopy (MRS) is able to detect NAA in brain, and decreases of NAA have been documented after brain injury. The reason for this decrease is not fully understood and neuron loss damage and "dysfunction" have all been proposed. It is hypothesized that acute central nervous system (CNS) deafferentation causes a trans-synaptic NAA decrease and that high resolution 1H MRS is able to detect such a decrease. To test this hypothesis, an experimental model was used in which axonal lesions were obtained

Spatiotemporal response properties of direction-selective neurons in the nucleus of the optic tract and dorsal terminal nucleus of the wallaby, Macropus eugenii.

1. The spatial and temporal response characteristics of direction-selective neurons in the nucleus of the optic tract and dorsal terminal nucleus of the accessory optic system (NOT-DTN) of the wallaby were established using moving sinusoidal gratings. This is the first comprehensive investigation of the spatiotemporal response characteristics of NOT-DTN neurons in any species. 2. The analysis revealed two main classes of cells. The first class, referred to as slow neurons, are maximally sensitive to motion at low temporal frequencies (< 1 Hz) and high spatial frequencies (0.5-1.0 cpd). The sec

FG7142 causes opposite changes in [3H]GABA release from nigrocollicular regions.

The activity of GABAergic neurons projecting from the striatum to the substantia nigra (SN) and from the SN to the superior colliculus (SC) may be involved in regulating seizure sensitivity such that striatonigral transmission is decreased and nigrocollicular transmission is increased in proconvulsant states. To test whether these changes occur in FG7142-treated rats, GABA transmission was assessed by measuring [3H]GABA release from superfused slices of the SN and SC and measuring [35S]TBPS binding to GABAA receptors throughout the brain. Nine daily injections of FG7142 (30 mg/kg IP) greatly i

Initial stages of retinofugal axon development in the hamster: evidence for two distinct modes of growth.

In order to characterize differences in growth patterns of axons as they elongate toward their targets and during the initial stages of terminal arbor formation within the targets, we examined the primary visual system of fetal and newborn hamsters using three morphological methods: the Cajal-deCastro reduced silver method, the rapid Golgi technique, and anterograde transport of HRP. Axons emerge from the retina between the 10th and 11th embryonic days (E10-E11). The front of retinal axons crosses the chiasm, extends over the primitive dorsal nucleus of the lateral geniculate body (LGBd) by E1
