We previously demonstrated that botulinum neurotoxin A (BoNT-A) exerts bilateral antinociceptive effects, involving trans-synaptic transport at the level of the lumbar spinal cord. However, the potential distribution of the toxin to supraspinal sites has not yet been investigated. In the present study, we examined the distribution of cleaved SNAP-25 (cl-SNAP-25), a marker of BoNT-A activity, in the rat brain following peripheral unilateral BoNT-A administration. Brain tissues from rats treated with BoNT-A (7 U/kg, into the hind paw) were analyzed using immunofluorescent tyramide signal amplifi

BACKGROUND: Chronic migraine is a highly disabling neurological disorder. A subset of patients develop chronic daily headaches refractory to all standard therapies. MR-guided focused ultrasound (MRgFUS) ventral intermediate nucleus (VIM) thalamotomy is an incisionless treatment for medication-refractory essential tremor (ET).
OBSERVATIONS: A 76-year-old woman with a 40-year history of chronic migraine with near-daily attacks, characterized predominantly by right-sided headaches and unresponsive to multiple preventive and acute treatments, underwent MRgFUS VIM thalamotomy for severe ET. After

BACKGROUND/AIM: Chronic migraine is a common debilitating disease with limited treatment options. We aimed to develop a novel model for chronic migraine by ligating the nasociliary nerve (NCL) and administering nitroglycerin (NTG) to exacerbate acute headache attacks.
MATERIALS AND METHODS: Exacerbation of the headache was induced by NTG (10 mg/kg, subcutaneously) administered to male Wistar rats (n = 36) 14 days following unilateral NCL. Cutaneous and cold allodynia was tested using Von Frey (VF) filaments and acetone, respectively. Elevated plus maze (EPM) results and c-fos immunoreactivity

The coronavirus SARS-CoV-2 (SCV2) causes acute respiratory distress, termed COVID-19 disease, with substantial morbidity and mortality. As SCV2 is related to previously-studied coronaviruses that have been shown to have the capability for brain invasion, it seems likely that SCV2 may be able to do so as well. To date, although there have been many clinical and autopsy-based reports that describe a broad range of SCV2-associated neurological conditions, it is unclear what fraction of these have been due to direct CNS invasion versus indirect effects caused by systemic reactions to critical illn

Earlier studies have shown that intranasal instillation of vesicular stomatitis virus (VSV), a negative-sense RNA virus, in mice and rats can result in infection of the brain, hind-limb paralysis and death. Using an antiserum directed against VSV proteins, we sought to determine the potential neuronal and non-neuronal pathways VSV utilize, for central nervous system dissemination in BALB/c mice. Within 12 h following intranasal inoculation of VSV, VSV antigen could be detected in the olfactory nerve layer of the ipsilateral olfactory bulb. Within 3-4 days post-inoculation (p.i.), VSV had disse

OBJECTIVE: The objective of this research was to clarify the effect of High Mobility Group Box 1 (HMGB1) in chronic migraine (CM).
METHODS: We established a CM model by repeated intraperitoneal injection of nitroglycerin (NTG). Subsequently, Western blot was used to detect changes in HMGB1 protein expression at the trigeminal nucleus caudalis (TNC), and immunofluorescence was performed to determine HMGB1 intracellular localization. After model establishment, HMGB1 inhibitors Glycyrrhizin were administered intraperitoneally for 9 consecutive days. The paw withdrawal threshold (PWT) were measur

Vocalizations are often elaborate, rhythmically structured behaviors. Vocal motor patterns require close coordination of neural circuits governing the muscles of the larynx, jaw, and respiratory system. In the elaborate vocalization of Alston's singing mouse (Scotinomys teguina) each note of its rapid, frequency-modulated trill is accompanied by equally rapid modulation of breath and gape. To elucidate the neural circuitry underlying this behavior, we introduced the polysynaptic retrograde neuronal tracer pseudorabies virus (PRV) into the cricothyroid and digastricus muscles, which control fre

The neuromodulation of the greater occipital nerve (GON) has proved effective to treat chronic refractory neurovascular headaches, in particular migraine and cluster headache. Moreover, animal studies have shown convergence of cervical and trigeminal afferents on the same territories of the upper cervical and lower medullary dorsal horn (DH), the so-called trigeminocervical complex (TCC), and recent studies in rat models of migraine and craniofacial neuropathy have shown that GON block or stimulation alter nociceptive processing in TCC. The present study examines in detail the anatomy of GON a

Rodents shift their nose from side to side when they actively explore and lateralize odors in the space. This motor action is driven by a pair of muscles, the deflector nasi. We studied the premotor control of this motion. We used replication-competent rabies virus to transsynaptically label inputs to the deflector nasi muscle and find putative premotor labeling throughout the parvocellular, intermediate, and gigantocellular reticular formations, as well as the trigeminal nuclei, pontine reticular formation, midbrain reticular formation, red nucleus, and superior colliculus. OBJECTIVE: To explore the surgical indications and efficacy of lateral approaches in the removal of brainstem cavernous malformations retrospectively.
METHODS: From July 1999 to June 2008, 10 cases of brainstem cavernous malformations were removed via lateral approaches. Four of them had reached the lateral pial surface and 6 were covered by parenchyma or glial tissue. Three lesions were located dorsal to the cranial nerve roots and 4 lesions rostral to the facial nerve root. All 7 lesions were removed via the retrosigmoidal approach. Far lateral approach was used for 2 cases with the lesions

The central pattern generator (CPG) for masticatory movements has been located in the medial bulbar reticular formation, by using cortically induced rhythmical jaw and tongue movements as a model. To analyze how stationary input from the cortex is transformed into rhythmical output in the neuronal population comprising the CPG, rhythmical neural activities representing rhythmical food ingestive movements were experimentally induced in vitro. Bath-application of N-methyl-D-aspartate (NMDA) induced rhythmical activities in the trigeminal (V), facial (VII) and hypoglossal (XII) nerves of in vitro

1. The embryonic hindbrain of chick is segmented until stage 24, when it starts to generate rhythmic activities in cranial nerves. In order to recognize a possible influence of segmentation on the organization of neuronal systems generating motor rhythms, the activity of trigeminal, facial, glossopharyngeal, vagal and hypoglossal nerves was studied during embryonic stages 24-36, by simultaneous recording of different cranial nerves in an isolated, superfused chick hindbrain preparation. 2. Highly correlated recurrent episodes of cyclical burst discharges occurred in all nerves studied (correla

The distribution of the growth associated protein GAP-43 has been described in the brain and spinal cord of rats and other placental mammals but not marsupials. In order to provide such information, we employed a monoclonal antibody to immunostain for GAP-43 in the central nervous system of adult and developing opossums, Didelphis virginiana. The GAP-43 immunoreactivity was widely distributed in the brain of adult opossums, but it was particularly dense within specific layers of the olfactory bulb and hippocampal formation, layer I of the cerebral cortex, the bed nucleus of the stria terminali

The distribution of muscarinic receptor binding was examined in the ferret brainstem vagal nuclei using the non-selective ligand [3H]quinuclidinyl benzilate and the relatively M1 receptor-selective ligand [3H]pirenzepine. The highest density of receptor sites are found in the subnucleus gelatinosus and lower levels in the other subnuclei of the nucleus of the tractus solitarius and in the area postrema and dorsal motor nucleus of the vagus nerve. Dense binding was also seen in the adjacent hypoglossal nucleus. Following unilateral cervical nodose ganglion excision binding in the subnucleus gel

This study examined whether glial cells in the trigeminal nucleus caudalis (Sp5C) were necessary for orofacial nociception and nociceptive processing induced by subcutaneously (s.c.) injection of 5% formalin into left mystacial vibrissae. The immunohistochemical, immunoelectron microscopical methods and behavior assessment were used in this study. Two hours after administration of carbenoxolone (CBX, a gap junction blocker) or fluorocistrate (FCA, a glail metabolic inhibitor) into the cerebellomedullary cistern, the nociceptive behavior and scratching-cumulative time reduced significantly (P<0

The present study was conducted to determine the effect of acute (1 h) and chronic (daily dose for 30 days) paracetamol administration on the development of cortical spreading depression (CSD), CSD-evoked cortical hyperaemia and CSD-induced Fos expression in cerebral cortex and trigeminal nucleus caudalis (TNC). Paracetamol (200 mg/kg body weight, intraperitonealy) was administered to Wistar rats. CSD was elicited by topical application of solid KCl. Electrocorticogram and cortical blood flow were recorded. Results revealed that acute paracetamol administration substantially decreased the numb

Pursuit of endogenous functions for various members of the alcohol dehydrogenase (ADH) enzyme family has led to exploration of gene expression patterns. Herein, we have used transgenic mice to examine the mouse gene encoding class IV ADH (ADH4), an enzyme that is weakly effective as an ethanol dehydrogenase, but highly effective as a retinol dehydrogenase in vitro. ADH4 promoter and upstream regulatory sequences were fused to lacZ and stably introduced into mice so that embryonic expression of ADH4 could be easily monitored by examination of beta-galactosidase activity in situ. Several indepen

This paper describes audiologic, electrophysiologic, and medical test results for a now 10-year-old girl who has had 45 episodes of reversible, sudden sensorineural hearing loss over the last 8 years. Episodes have lasted from 6 to 72 hours and often have been accompanied by a mild illness. Acoustic immittance measures have been consistent with normal middle-ear function with the exception of absent ipsilateral and contralateral acoustic reflexes. Mechanically evoked perioral reflex activity was markedly asymmetric following lower lip stimulation. The asymmetry of R1 activation between right a

Auricular neuromodulation (ANM) is a non-invasive method of stimulating the trigeminal, spinal, and vagal nerves that innervate the auricle, the outer part of the ear. ANM, and the vagal nerve stimulation in particular, has been shown to be effective in treating a number of symptoms, including pain and anxiety. However, the mechanisms by which ANM exerts its beneficial effects are still under investigation. We hypothesized that ANM produces its effects by modulating interoception, a physiological function involved in homeostatic regulation and awareness of internal physical and emotional sensa

The blink reflex (BR) is integrated at the brainstem; however, it is modulated by inputs from various structures such as the striatum, globus pallidus, substantia nigra, and nucleus raphe magnus but also from afferent input from the peripheral nervous system. Therefore, it provides information about the pathophysiology of numerous peripheral and central nervous system disorders. The BR is a valuable tool for studying the integrity of the trigemino-facial system, the relevant brainstem nuclei, and circuits. At the same time, some neurophysiological techniques applying the BR may indicate abnorm
