Comprehensive epigenetic analysis on 5-methylcytosine modification of circRNAs in the mouse cerebral ischemia model.

A growing body of research suggests that circular RNAs (circRNAs) and 5-methylcytosine (m5C) play crucial roles in the onset and progression of stroke. This includes their potential regulation of neuronal damage and repair under cerebral ischemic conditions. However, the specific features of m5C modification in circRNAs associated with stroke remain unclear. In this study, we established a mouse model of cerebral ischemia (MCAO) and utilized methylated RNA immunoprecipitation sequencing (MeRIP-Seq) to identify m5C peaks. In comparison to the control group, MCAO exhibited significant changes in

METTL3 Promotes Lipid Deposition and Pulmonary Fibrosis by Destabilizing PLIN2 in a m6A-Dependent Manner.

Idiopathic pulmonary fibrosis (IPF) is a lethal lung disorder that is associated with aberrant lipid metabolism. N6-methyladenosine (m6A) methylation is involved in IPF progression. However, whether METTL3, a m6A methyltransferase, modulates lipid metabolism in IPF remains unknown. In this study, we aimed to investigate the effects of METTL3 on fibrosis and lipid accumulation and identify the molecular mechanism. PF mouse model was generated by bleomycin. WI-38 cells were stimulated with TGF-β1 to mimic the IPF-caused damage. Lipids were observed using Nile Red staining and marker detection. F

Cortical organoids reveal human-specific role of METTL5 in neurodevelopment via regulation of CHCHD2.

Methyltransferase-like 5 (METTL5) catalyzes N6-methyladenosine (m6A) modification on 18S rRNA. In humans, loss-of-function mutations in METTL5 cause severe microcephaly and intellectual disability, whereas Mettl5 knockout (KO) animal models display inconsistent and milder phenotypes. To better model human disease, we generated METTL5-KO human-induced pluripotent stem cell (hiPSC)-derived cortical organoids, which exhibit impaired neural progenitor cell (NPC) proliferation and differentiation, leading to reduced ventricle-like structures and significant reductions in cortical organoid diameter.

METTL14 alleviates pyroptosis of placental trophoblasts in gestational diabetes mellitus through the lncRNA MEG8/WNT7A axis via m6A modification.

Gestational diabetes mellitus (GDM) is a pregnancy complication associated with abnormal placental trophoblast function. Pyroptosis has been implicated in GDM pathogenesis, yet the role of m6A modification in this process remains unclear. We hypothesized that METTL14 regulates trophoblast pyroptosis through m6A-dependent modulation of the lncRNA MEG8/WNT7A axis. This study investigated the mechanism of METTL14 in pyroptosis of placental trophoblasts in GDM. HG-treated HTR8/SVneo cells were used as a cell model. METTL14, WNT7A, and lncRNA MEG8 expression was detected by RT-qPCR and western blot

ABI5-activated ALKBH10B demethylates RAP2.6 mRNA to modulate Arabidopsis salt tolerance.

Salt stress induces ABI5 → ALKBH10B transcription; the demethylase removes m6A from RAP2.6 mRNA, accelerating its decay and attenuating salt-responsive genes, thus linking ABA signaling to reversible m6A control of Arabidopsis salt tolerance. N6-methyladenosine (m6A) is the most prevalent internal modification of RNA and plays an important role in regulating RNA metabolism that governs development and environmental adaptation of plants. Here we dissect how the Arabidopsis m6A demethylase ALKBH10B is integrated into abscisic-acid (ABA)-mediated salt-stress signaling. Under salt treatment, loss

m6A RNA modification guides alternative polyadenylation to maintain T cell quiescence.

N6-Methyladenosine (m6A) is primarily enriched in the last exons and 3' untranslated regions (3'UTRs) of messenger RNAs (mRNAs) and is associated with T cell homeostasis. Upon T cell activation, global mRNA 3'UTR shortening is facilitated through alternative polyadenylation (APA). However, it is unclear how T cells coordinate these two important posttranscriptional regulatory events to maintain quiescence. Here, we found that the m6A "writer" METTL3 directly interacts with APA factor NUDT21 and guided poly(A) site selection. Deletion of Nudt21 in T cells resulted in simultaneous overactivation

METTL3 facilitates the malignant progression of prostate cancer by inhibiting GDF15-mediated ferroptosis via a YTHDF1-dependent m6A mechanism.

BACKGROUND: Prostate cancer (PCa) represents one of the most common tumors in males. Growth differentiation factor 15 (GDF15), a divergent member of the transforming growth factor β (TGF-β) family, exhibits elevated expression that correlates with cancer progression (including PCa). The RNA N6-methyladenosine (m6A) modification, an epigenetic regulatory mechanism, has been implicated in oncogenesis and tumor advancement. Therefore, this study aims to delineate the functional role of GDF15 in PCa progression. METHODS: Differentially expressed genes (DEGs) and GDF15 levels in tumor tissues were

N6-methyladenosine-modified miR-873 promotes receptor-interacting protein kinase 3-mediated necroptosis after intracerebral hemorrhage in mice and HT22 cells.

BACKGROUND: Intracerebral hemorrhage (ICH) causes a severe form of stroke characterized by high morbidity, mortality, and long-term disability. Neuronal cell death is influenced at the posttranscriptional level. Certain microRNAs influence neuronal cell death at the posttranscriptional level by regulating receptor-interacting protein kinase 3 ( RIPK3 ), a key mediator of necroptosis. The specific mechanism by which miR-873 mediates neuronal necroptosis following ICH remains unclear. Epigenetic abnormalities, particularly N6-methyladenosine (m6A) modification, are increasingly recognized as cri

METTL3 drives malignant progression in TP53-mutant prostate cancer.

BACKGROUND: Prostate cancer patients harboring TP53 mutations exhibit a more aggressive and chemo-resistant phenotype. Unfortunately, attempts to identify the vulnerabilities that could be exploited to overcome these aggressive malignancies have made only minimal progress in recent years. Consequently, there is an immediate requirement to investigate novel therapeutic strategies for this subclass. METTL3 complex, knowing to govern m6A dynamic alteration, has been suggested to be a critical therapeutic target across human cancer. However, the role of METTL3 in prostate cancer harboring TP53 mut

Macrophage Polarization and Metabolic Reprogramming in Endometriosis: Mechanisms and Therapeutic Implications.

This review synthesizes evidence on macrophage roles, polarization (M1/M2), and immunometabolic/signaling pathways in endometriosis, outlining therapeutic implications. We searched PubMed, Cochrane Library, ClinicalTrials.gov, Scopus, Embase, and Google Scholar. Available evidence demonstrates increased macrophage accumulation and substantial heterogeneity across the eutopic endometrium, peritoneal fluid, and established endometriotic lesions. Rather than undergoing a uniform shift from an M1 to an M2 phenotype, endometriosis-associated macrophages acquire overlapping inflammatory, immunoregul

Sinoatrial node dysfunction: molecular mechanisms, arrhythmogenesis, and translational pathways toward precision therapy.

Sinoatrial node dysfunction (SND) is a major cause of bradyarrhythmia, chronotropic incompetence, tachy-brady syndrome, and a common indication for permanent pacemaker implantation, particularly in older adults. Although SND has diverse etiologies, this review focuses on aging-associated intrinsic SND, while considering other common causes where they inform differential diagnosis, arrhythmogenic mechanisms, or therapeutic development. Although current medical interventions provide effective rhythm support, they does not correct the molecular and tissue-level processes that drive sinoatrial nod

Immunogenic cell death: synergizing ferroptosis and cuproptosis to overcome immunotherapy resistance.

The advent of immune checkpoint inhibitors has fundamentally changed the face of oncology, but the prevalence of primary and acquired resistance has led to the need for the development of novel approaches to remodel the immunosuppressive tumor microenvironment. Central to this challenge is the dichotomy between hot inflamed and cold (immune-excluded) tumors, which is increasingly associated with the immunogenicity of cell death modalities used by therapeutic agents. While apoptosis is largely immunologically silent, regulated cell death pathways driven by the catastrophic failure of mitochondr
