Magnetic-Optical Dual-Modality Imaging Monitoring Oxaliplatin through the Crystalline Mn-Metal-Organic Framework Sensor.

To support image-assisted evaluation of anticancer drugs in biological settings, functional sensing materials are needed to integrate sensitive molecular detection with imaging-compatible readouts. Oxaliplatin (OHP) is widely used in chemotherapy, and accurate OHP assessment in biological matrices remains important for dosage optimization and toxicity management. Herein, a nanosized Mn-MOF was prepared as a fluorescence sensor and MRI-responsive platform for OHP-associated detection. The structure, morphology, elemental distribution, and Mn surface electronic state of the Mn-MOF were character

Regulatory-compliant detection of tetracycline food contaminants: A polyindole-based chemosensoric approach for food safety monitoring and toxicological risk mitigation.

The widespread contamination of tetracycline (TC) in aquatic environments and food chains poses significant risks to human health and ecosystem sustainability, necessitating the development of rapid, sensitive detection methods. Herein, we report polyindole (PIN) fluorescent sensor for highly selective TC detection. PIN exhibited a quantum yield of 0.27 in ethanol with characteristic blue emission at 428 nm upon excitation at 350 nm. Under optimized conditions (H2O/EtOH = 1:1, pH 7), PIN demonstrated exceptional selectivity and sensitivity toward TC over various interfering antibiotics and met

A novel 2,6-diformyl-4-methylphenol based chemosensor for Zn(II) ions by ratiometric displacement of Cd(II) ions and its application for cell imaging on human melanoma cancer cells.

A new chelating ligand [4-methyl-2,6-bis-(pyridin-2-yl-hydrazonomethyl)-phenol] (1) was prepared by the condensation of 2-hydrazinylpyridine with 2,6-diformyl-p-cresol. Compound 1 exhibits weak fluorescence due to intramolecular photoinduced electron transfer (PET). The sensor (1) demonstrates Zn(2+)-specific emission enhancement due to the “PET off” process through a 1:1 binding mode with the metal ion. The fluorescence quantum yield of chemosensor 1 is only 0.020, and it increases more than 14-fold (0.280) in the presence of one equivalent of the zinc ion. Interestingly, the introduction of

A DNA-based nanosensor for in situ precise imaging of lysosomal potassium ions via pH activation.

For subcellular imaging, activating probes by specific species in targeted organelles is an attractive strategy to eliminate nonspecific signals during transport. The characteristic pH (∼4.5-5.0) of lysosomes enables selective probe activation within lysosomes. In this work, a pH-activated K+ nanosensor for lysosomal K+ imaging was designed by capturing Y-typed DNA probe on linker modified gold nanoparticle. The DNA probe could be conveniently prepared by assembling dual fluorophores-labelled G-quadruplex (G4) strand, Sgc8c modified i-motif structure and capturing DNA sequence. The folding cap

Visualizing hypochlorous acid as a key oxidative stress biomarker in drug-induced liver injury with a red-emitting probe.

Drug-induced liver injury (DILI) is a common form of hepatotoxicity resulting from adverse drug reactions. Extensive studies indicate that reactive oxygen species (ROS) levels undergo significant alterations during the progression of DILI. Among various ROS, hypochlorous acid (HClO)-produced in substantial amounts during oxidative stress in liver injury-has emerged as a potential biomarker for DILI. In this work, we developed a fluorescent probe based on a benzopyran-derived fluorophore with diaminomaleonitrile as the HClO-specific recognition unit. Upon reaction with HClO, the probe emits int

Spectrophotometric Studies of a Fluorescent Naphthalenediimide PET and ICT pH Indicator and DNA Intercalator.

Three naphthalenedimides (NDIs) were studied by UV-visible absorbance, fluorescence and circular dichroism spectroscopy in aqueous methanolic solution as DNA intercalators. Distinct solution colours are observed for the compounds under room lighting and upon irradiation with 365 nm UV light. The tetra-substituted dimethylaminoethylene NDI is designed as a fluorescent pH indicator with a 'fluorophore-spacer-receptor' construct based on a photoinduced electron transfer (PET) mechanism. This NDI derivative exhibits a significant change in the ellipticity with calf-thymus DNA. Fluorescence DNA tit

A coumarin-tethered 1,2,3-triazole-based fluorescent "turn-ON" probe via metal-free click chemistry for selective sensing of serum albumin: quantitative detection of HSA in urine.

A series of novel highly substituted coumarin-based 1,2,3-triazoles (CTZs) were synthesised by a metal-free organo-click reaction. Under environmentally benign, atom-economical conditions, the desired products were obtained in moderate to excellent yields. All derivatives exhibited good binding affinity toward bovine serum albumin (BSA), with binding constants on the order of Kb ∼ 104 L mol-1. Among them, compound 3f, a nitro analogue of CTZ, demonstrated a selective fluorescence "turn-on" response upon binding to both BSA and human serum albumin (HSA). Simultaneous Visualization of Glutathione and Hydrogen Peroxide with a Dual-Responsive Fluorescent Probe to Elucidate Redox Homeostasis in Alzheimer's Disease Models.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder in which oxidative stress plays a critical role in pathogenesis. As key reactive species governing oxidative stress, the dynamic interplay between glutathione (GSH) and hydrogen peroxide (H2O2) is tightly coupled to redox homeostasis. To enable simultaneous tracking of these two crucial species, we developed a dual-responsive fluorescent probe, TCS, for the discriminative visualization of GSH and H2O2. TCS features a methylsulfonyl group that is selectively reduced by GSH, triggering green fluorescence at 500 nm, and a borona

Activatable NIR-IIb Nanosensor for Visualizing Brain HClO to Monitor Parkinson's Disease.

Parkinson's disease (PD) is a complex neurodegenerative disorder. Currently, the early diagnosis and treatment of PD are often hindered by significant subjectivity in clinical assessments from both patients and physicians. Hypochlorous acid (HClO), a representative reactive oxygen species, has been widely recognized as closely linked to the pathological mechanisms underlying PD. Thus, we developed a HClO-activated NIR-IIb fluorescent probe, SQ6-RENPs, which is functionalized with the VHP peptide to enable blood-brain barrier (BBB) crossing for monitoring cerebral HClO levels associated with PD

Lysosome-targeted near-infrared fluorescent probes for monitoring aβ plaques in vivo and aβ monomers dynamic degradation in vitro.

The aggregation of β-amyloid (Aβ) is a central pathological feature of Alzheimer's disease (AD), with its dynamic changes closely linked to disease progression. Lysosomes play a critical role in the clearance and degradation of Aβ, making them an important focus in AD research. In this study, a series of multifunctional lysosome-targeting near-infrared fluorescent probes based on a dicyanoisophorone scaffold were developed to enable simultaneous targeting of lysosomes and monitoring of Aβ aggregates. All probes effectively localized within cellular lysosomes, with NCM-4 demonstrating the most

Red-emissive carbon dot (RCDs@Ag+) nanohybrid as a dual-functional platform for glutathione sensing and antibacterial applications.

BACKGROUND: Biothiols, especially glutathione (GSH), are crucial for cellular redox balance, detoxification, and disease progression, and require highly sensitive and selective detection methods in biological, clinical, and food-safety contexts. Red-emitting carbon dots are ideal due to their photostability, biocompatibility, and adjustable optical features. Meanwhile, silver-based nanocomposites demonstrate potent antimicrobial properties and improve analytical signals by facilitating efficient electron transfer and fluorescence control. Combining these components into a single nanoplatform o

Drug Screening of Nitroreductase Modulators Using an Atomic-Engineered Activatable Fluorescent Probe for Tumor Imaging.

Nitroreductase (NTR), a hypoxia-upregulated enzyme, is a promising biomarker for solid tumors, driving demand for effective biosensors. Cyanine dyes are prominent fluorophores widely used in fluorescence imaging, tumor phototheranostics, and image-guided surgery. Guided by atomic engineering through strategic heteroatom substitution (C, O, and S), we designed four NTR-activated probes. Optical characterization identified HCy-NS-4 as the lead candidate, exhibiting near-infrared emission (λem = 712 nm) and over 18-fold fluorescence enhancement upon activation. The probe enabled precise imaging o

A hydrogen sulfide activated fluorescence probe for screening tumor cells and diagnosis of non-small cell lung carcinoma.

Fluorescence probes are applied as chemical tools for the diagnosis and treatment of cancer. Despite the successful development of certain fluorescence probes, significant demand for enhancement remain in sensitive diagnosis and effective treatment. In this work, we developed a novel H2S-activable fluorescence probe, CerYT, utilizing nitrobenzoxadiazole (NBD) as the imaging fluorophore and ceritinib as a potent ALK kinase inhibitor for tumor cells. The probe exhibits good sensitivity (LOD = 121 nM) and specificity, and it can also detect H2S with precision in cells and tumor. Moreover, probe C

A benzopyran-based near-infrared fluorescent probe with a large stokes shift for visualizing cysteine dynamics in hepatic ischemia-reperfusion injury and tumor tissues.

Cysteine (Cys) is a key biomarker of oxidative stress and is closely associated with pathologies such as hepatic ischemia-reperfusion injury (HIRI) and cancer. However, existing fluorescent probes for Cys detection often lack practicality in complex disease models. Herein, we developed a near-infrared fluorescent probe, Cys398, with a large Stokes shift, for high-fidelity in vivo imaging of Cys. The probe demonstrated high sensitivity, selectivity, and anti-interference capability, enabling real-time visualization of both exogenous and endogenous Cys in live cells and mice. In HIRI models, it

A lipid droplet-targeted viscosity-sensitive fluorescent probe for visualization of tumor and NAFLD imaging.

Non-alcoholic fatty liver disease (NAFLD) and tumors remain major clinical challenges, characterized by aberrant lipid droplets (LDs) accumulation and elevated microenvironmental viscosity as key pathological features. In this study, we developed a series of carbazole-based fluorescent probes, LJL1-LJL6. Through systematic screening of their photophysical properties and bioimaging performance, LJL1 was identified as the optimal candidate due to its superior viscosity sensitivity and LDs-targeting specificity. Specifically, LJL1, featuring a carbazole-thiophene electron-donating unit and a pyri

A turn-on fluorescent biosensor based on CdTe quantum dots for sensitive and selective detection of APE1 activity in cervical cancer cell lysates.

Apurinic/apyrimidinic endonuclease 1 (APE1) plays a critical role in the base excision repair (BER) pathway, and its aberrant activity is closely associated with various cancers, including cervical cancer, as well as neurodegenerative diseases. Herein, we report a novel "turn-on" fluorescent biosensor for the sensitive and selective detection of APE1 activity based on CdTe quantum dots (QDs) and silver ions (Ag+). The sensing strategy relies on the inner filter effect (IFE)-induced fluorescence quenching of CdTe QDs by Ag+, followed by fluorescence recovery upon specific cleavage of an AP site
