Development and evaluation of chitosan-xanthan gum-based controlled release nanoparticles of mebeverine hydrochloride for targeted IBS therapy: in silico, in vitro, and in vivo investigations.

BACKGROUND: Irritable bowel syndrome (IBS) requires long-term, site-specific therapy. Mebeverine hydrochloride is effective but limited by rapid metabolism and frequent dosing. This study aimed to develop colon-targeted, controlled-release nanoparticles to improve therapeutic efficacy and compliance. METHODS: Nanoparticles were prepared via chitosan-xanthan gum polyelectrolyte complexation and optimized using a Box-Behnken design. Molecular docking predicted strong binding of mebeverine to the muscarinic acetylcholine receptor (-7.4 kcal/mol). Optimized nanoparticles were enteric-coated with

Groove binding phenomenon of ajmalicine with HT DNA: An exothermic-entropy driven interaction involving hydrogen bonds and hydrophobic forces.

Many bioactive compounds target DNA, making ligand-DNA binding studies essential for developing new therapeutics. These interactions provide insight into how small molecules can bind to DNA and inhibit processes like replication and transcription, influencing gene-expression. Ajmalicine (AJM), while extensively studied for its pharmacological properties, has not been fully elucidated in its nucleic acid(s) binding mode. DNA binding of AJM can impact the structure, stability and function of the DNA leading to significant implications for drug development. In this study, we explored the interact

A facile microfluidics based polysaccharide-functionalized virus-mimicking nanovaccine for circumventing pulmonary innate physiological barriers and augmenting immunity.

The escalating prevalence of COVID-19 variants and influenza has garnered substantial attention to lung vaccines lately. Nevertheless, the intrinsic immune barriers present within the lung significantly tend to compromise the efficacy of nanovaccines. Moreover, the deficiency of safe and efficacious inhalable adjuvants also impedes the advancement of nanovaccines. Herein, we devised polysaccharide-functionalized virus-mimicking nanovaccines via a facile microfluidics approach, wherein chitosan-based polyelectrolyte complexes serve as the inner core (CS-OVA), and heparan sulfate (SH) modificati

Chitin nanocrystals/alginate complex for tuning stability, rheology and bioavailability of cholecalciferol in Pickering emulsions.

In this paper we investigate polyelectrolyte complexes of sodium alginate (Alg) and chitin nanocrystals (ChNC). Formation, stability and transport properties of sunflower oil-in-water emulsions stabilized by ChNC-Alg complex were studied using dynamic light scattering (DLS), laser Doppler electrophoresis, optical microscopy, potentiometric titration, rheology and simulated digestion. It has been established that during emulsions formation, the ChNC-Alg complex is rearranged at the interface and the formation of a two-layer coating of the droplet occurs. Stabilized O/W emulsions are stable duri

Biopolymer Drug Delivery Systems for Oromucosal Application: Recent Trends in Pharmaceutical R&D.

Oromucosal drug delivery, both local and transmucosal (buccal), is an effective alternative to traditional oral and parenteral dosage forms because it increases drug bioavailability and reduces systemic drug toxicity. The oral mucosa has a good blood supply, which ensures that drug molecules enter the systemic circulation directly, avoiding drug metabolism during the first passage through the liver. At the same time, the mucosa has a number of barriers, including mucus, epithelium, enzymes, and immunocompetent cells, that are designed to prevent the entry of foreign substances into the body, w

Optimization, physicochemical stability and in vivo study of alginate-chitosan composites as nanocarriers for low molecular weight angiotensin I-converting enzyme (ACE)-inhibitory peptide.

Chitosan and alginate, are non-toxic and biodegradable polymers used to enhance the stability of biotherapeutics by loading them into nanocarriers. In this study, the stone fish-derived low molecular weight peptide (Ala-Leu-Gly-Pro-Gln-Phe-Tyr), exhibited an in vitro ACE-inhibitory activity of 94.43 ± 2.05% and an IC50 of 0.012 ± 0.001 mM. The peptide was encapsulated via ionic gelation with alginate followed by polyelectrolyte complexation with chitosan. The resulting ACE-inhibitory peptide-loaded alginate-chitosan nanoparticles (ACE-I-ALG-CS NPs) were optimized to achieve small particle size

Chitosan-coated alginate (CCA) nanoparticles for augmentation of topical antihistaminic activity of diphenhydramine: in-vitro optimization, skin histopathology and pharmacodynamic studies with in vitro/in vivo correlation.

OBJECTIVE: The aim of the present study was to formulate chitosan-coated alginate nanoparticles containing the drug diphenhydramine hydrochloride (DHH). SIGNIFICANCE: Diphenhydramine hydrochloride (DHH) is the prototype of H1-antihistaminic drugs. It is a lipophilic drug, that easily crosses the blood-brain barrier when taken orally causing decrements in alertness and performance. Multiple applications of topical drug products are required. Thus, drug incorporation in nanocarriers would increase the skin penetration powers increasing the drug efficacy. METHODS: Chitosan-coated alginate (CCA)

Fabrication techniques for the preparation of orally administered insulin nanoparticles.

The development of orally administered protein drugs is challenging due to their intrinsic unfavourable features, including large molecular size and poor chemical stability, both of which limit gastrointestinal (GI) absorption efficiency. Nanoparticles can overcome the GI barriers effectively and improve the oral bioavailability of proteins in the GI tract. They possess large surface area to volume ratio, and can facilitate the GI absorption of nanoparticles via the paracellular and transcellular routes. Nanoparticles can be prepared by various fabrication techniques that can encapsulate the f

Two-Step Optimization to Develop a Transdermal Film Loaded With Dapoxetine Nanoparticles: A Promising Technique to Improve Drug Skin Permeation.

Dapoxetine (DPX) is an orally administered drug for the treatment of premature ejaculation (PE). One of the challenges of administering DPX orally as a tablet is its poor bioavailability (ie, 42%) due to extensive first-pass metabolism. Thus, it is vital to develop a new formulation and mode of delivery to achieve the unmet needs of PE treatment. In this study, an optimized DPX polymeric nanoparticle (PNP) was developed and subsequently loaded into a transdermal film. The Box-Behnken design was utilized to optimize 3 formulation factors affecting the particle size and entrapment efficiency (EE

Complexation of novel thiomers and insulin to protect against in vitro enzymatic degradation - towards oral insulin delivery.

A significant barrier to oral insulin delivery is its enzymatic degradation in the gut. Nano-sized polymer-insulin polyelectrolyte complexes (PECS) have been developed to protect insulin against enzymatic degradation. Poly(allylamine) (Paa) was trimethylated to yield QPaa. Thiolation of Paa and QPaa was achieved by attaching either N-acetylcysteine (NAC) or thiobutylamidine (TBA) ligands (Paa-NAC/QPaa-NAC and Paa-TBA/QPaa-TBA thiomers). PEC formulations were prepared in Tris buffer (pH 7.4) at various polymer: insulin mass ratios (0.2:1-2:1). PECS were characterized by %transmittance of light

Intra-articular delivery of a nanocomplex comprising salmon calcitonin, hyaluronic acid, and chitosan using an equine model of joint inflammation.

Polyelectrolyte nanoparticle constructs (NPs) comprising salmon calcitonin (sCT), chitosan (CS), and hyaluronic acid (HA) were previously established as having anti-inflammatory potential when injected via the intra-articular (i.a.) route to a mouse model. We attempted to translate the formulation to a large animal model, the lipopolysaccharide (LPS)-stimulated equine model of joint inflammation. The aim was to manufacture under aseptic conditions to produce sterile pyrogen-free NPs, to confirm physicochemical characteristics, and to test toxicity and efficacy in a pilot study. Structure and properties of polycaprolactone/chitosan nonwovens tailored by solvent systems.

Electrospinning of chitosan blends is a reasonable idea to prepare fibre mats for biomedical applications. Synthetic and natural components provide, for example, appropriate mechanical strength and biocompatibility, respectively. However, solvent characteristics and the polyelectrolyte nature of chitosan influence the spinnability of these blends. In order to compare the effect of solvent on polycaprolactone/chitosan fibres, two types of the most commonly used solvent systems were chosen, namely 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) and acetic acid (AA)/formic acid (FA). Results obtained by

Preparation, property of the complex of carboxymethyl chitosan grafted copolymer with iodine and application of it in cervical antibacterial biomembrane.

Cervical erosion is one of the common diseases of women. The loop electrosurgical excisional procedure (LEEP) has been used widely in the treatment of the cervical diseases. However, there are no effective wound dressings for the postoperative care to protect the wound area from further infection, leading to increased secretion and longer healing time. Iodine is a widely used inorganic antibacterial agent with many advantages. However, the carrier for stable iodine complex antibacterial agents is lack. In the present study, a novel iodine carrier, Carboxymethyl chitosan-g-(poly(sodium acrylate

Formation and mechanical characterization of ionically crosslinked membranes at oil-water interfaces.

Here we report on the preparation and mechanical characterization of a 2-D self-assembled membrane formed by ionically crosslinking the polyelectrolyte parts of a gradient amphiphilic copolymer at oil and water interfaces. To fabricate these membranes, chloroform solutions of styrene-acrylic acid copolymers were suspended as pendant drops in an aqueous embedding phase. Due to the amphiphilic nature of these molecules, the copolymer chains migrate to the oil-water interface creating an interfacial layer. Upon the addition of zinc acetate to the embedding phase, crosslinks between copolymer mole

Role, metabolism, chemical modifications and applications of hyaluronan.

Hyaluronan (hyaluronic acid, HA) is a linear naturally occurring polysaccharide formed from repeating disaccharide units of N-acetyl-D-glucosamine and D-glucuronate. Despite its relatively simple structure, HA is an extraordinarily versatile glycosaminoglycan currently receiving attention across a wide front of research areas. It has a very high molar mass, usually in the order of millions of Daltons, and possesses interesting visco-elastic properties based on its polymeric and polyelectrolyte characteristics. HA is omnipresent in the human body and in other vertebrates, occurring in almost al

Safety and biodistribution of intrathecal administration of mesenchymal stem cells (MSCs) and neurotrophin-releasing nanoparticles in a porcine CSF-guided delivery model for amyotrophic lateral sclerosis (ALS) drug discovery.

Amyotrophic lateral sclerosis (ALS) is a multifactorial neurodegenerative disorder that complicates the identification of effective therapeutic targets. The potential of stem cells and neurotrophins as promising candidates has become increasingly evident, owing to their neuroprotective and anti-inflammatory properties. In this study, a preclinical evaluation of the safety and biodistribution of mesenchymal stromal/stem cells (MSCs) combined with neurotrophin-releasing polyelectrolyte nanoparticles (NTs) was conducted in a porcine intrathecal delivery model relevant to ALS therapy development.

Full-length brain-derived α-synuclein fibril models reveal fuzzy-coat control of peptide recognition.

α-Synuclein (aSyn) fibrils in Parkinson's disease are generally represented by their ordered cross-β cores, although nearly half of the protein remains disordered in the fibrillar state. The unresolved N- and C-terminal segments form a fuzzy coat that is expected to shape the surface encountered by molecular partners. Here, we reconstructed full-length, brain-derived aSyn Lewy-fold fibrils by extending a patient-derived cryo-EM core spanning residues 31-100, with disordered termini, and sampling the resulting assemblies with the CALVADOS coarse-grained force field. The acidic C-terminal tails
