iTRAQ-based quantitative proteomics analysis of Sprague-Dawley rats liver reveals perfluorooctanoic acid-induced lipid metabolism and urea cycle dysfunction.

Perfluorooctanoic acid (PFOA) is a typical C8 representative compound of perfluoroalkyl and polyfluoroalkyl substances (PFAS) widely used in industrial and domestic products. It is a persistent organic pollutant found in the environment as well as in the tissues of humans and wildlife. Despite emerging scientific and public interest, the precise mechanisms of PFOA toxicity remain unclear. In this study, male rats were exposed to 1.25, 5, and 20 mg PFOA/kg body weight/day for 14 days by gavage; food intake and bodyweight changes were recorded every day. After 14 days, blood was collected for se

Prenatal exposure to 11 perfluoroalkyl substances and fetal growth: A large-scale, prospective birth cohort study.

BACKGROUND: Prenatal maternal exposure to perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) has been reportedly associated with decreased birth weight. Although a majority of epidemiological studies concerning perfluoroalkyl substances (PFAS) have focused on PFOS and PFOA, epidemiological studies of PFAS with longer carbon chains are scarce. In this study, we investigated whether prenatal maternal exposure to 11 PFAS, in particular those with longer carbon chains, is associated with fetal growth. METHODS: The study included 1985 mother-infant pairs (inclusive of preterm and p

Exposure to Perfluoroalkyl Substances and Metabolic Outcomes in Pregnant Women: Evidence from the Spanish INMA Birth Cohorts.

BACKGROUND: Exposure to perfluoroalkyl substances (PFASs) may increase risk for metabolic diseases; however, epidemiologic evidence is lacking at the present time. Pregnancy is a period of enhanced tissue plasticity for the fetus and the mother and may be a critical window of PFAS exposure susceptibility. OBJECTIVE: We evaluated the associations between PFAS exposures and metabolic outcomes in pregnant women. METHODS: We analyzed 1,240 pregnant women from the Spanish INMA [Environment and Childhood Project (INfancia y Medio Ambiente)] birth cohort study (recruitment period: 2003-2008) with m

Involvement of oxidative stress and inflammation in liver injury caused by perfluorooctanoic acid exposure in mice.

Perfluorooctanoic acid (PFOA) is widely present in the environment and has been reported to induce hepatic toxicity in animals and humans. In this study, mice were orally administered different concentrations of PFOA (2.5, 5, or 10 mg/kg/day). Histological examination showed that the exposure to PFOA for 14 consecutive days led to serious hepatocellular injury and obvious inflammatory cell infiltration. In addition, malondialdehyde formation and hydrogen peroxide generation, indicators of oxidative stress, were significantly induced by PFOA treatment in the liver of mice. Furthermore, hepatic

Evaluating the additivity of perfluoroalkyl acids in binary combinations on peroxisome proliferator-activated receptor-α activation.

Perfluoroalkyl acids (PFAAs) are found globally in the environment, detected in humans and wildlife, and are typically present as mixtures of PFAA congeners. Mechanistic studies have found that responses to PFAAs are mediated in part by PPARα. Our previous studies showed that individual PFAAs activate PPARα transfected into COS-1 cells. The goal of the current study was to determine if binary combinations of perfluorooctanoic acid (PFOA) and another PFAA act in an additive fashion to activate PPARα in the mouse one-hybrid in vitro model. COS-1 cells were transiently transfected with mouse PPAR

Physiological evaluation of residual stomach motility after local resection in conscious dogs.

PURPOSE: To investigate the phenomenon of remnant gastric motility and emptying after local resection. METHODS: Fifteen dogs were divided into three groups: a control (CONT) group, a group that underwent local resection of the greater (GREAT) curvature, and a group that underwent resection of the lesser (LESS) curvature. We conducted a strain gauge force transducer study, a [(13)C]octanoic acid breath test ((13)C-OBT), and a mosapride citrate effect test. Based on these results, we worked out the receptive relaxation (RR), motility index (MI), and postprandial period (PP) in the postprandial

Safety and tolerability of a novel chromatography-based intravenous immunoglobulin when administered at a high infusion rate in patients with immune thrombocytopenic purpura.

Intravenous immunoglobulin (IGIV) therapy is generally considered to be a safe and effective treatment for idiopathic thrombocytopenic purpura (ITP). The usual initial treatment dose is 1-2 g/kg body weight, which results in an extended infusion time, significantly impacting patients' day-to-day activities. Two crossover studies assessed the safety and tolerability of a novel IGIV preparation (IGIV-C; Gamunex, 10%) when infused at rates ranging from 0.08 mL/kg/min (the standard maximum licensed rate) to 0.14 mL/kg/min in patients with ITP. The first study included 28 patients and 3 infusion ra

Enhanced potential for oxidative stress in livers of senescent rats by the peroxisome proliferator-activated receptor alpha agonist perfluorooctanoic acid.

Aging sensitizes the liver to the hepatocarcinogenic effect of PPARalpha agonists via unknown mechanisms. This study was designed to investigate age-dependent, hepatic effects of the non-metabolizable PPARalpha agonist perfluorooctanoic acid (PFOA) on the delicate balance between activities of pathways involved in H(2)O(2) production and elimination. Male Fischer-344 rats, ranging in age from juvenile (4 weeks old), post puberty (10 weeks old), mature adulthood (20 weeks old), middle age (50 weeks old), to senescence (100 weeks old), were treated intragastrically with either 150mg PFOA/kg in 0

Effects of therapeutic agents in a rabbit model of Reye syndrome.

Six potential therapeutic agents were evaluated in an experimental model simulating Reye syndrome produced by infusion of the short-chain fatty acid, sodium octanoate, into rabbits. Administration of carnitine, dexamethasone, or fatty acid-free albumin resulted in prolongation of survival, less rise in intracranial pressure, and amelioration of some of the metabolic abnormalities found typically during octanoate infusion. Treatment with pentobarbital or dimethyl sulfoxide prevented intracranial pressure elevations but had no protective effect on survival. Hypertonic glucose administration prod

Culture of the terminally differentiated ventricular cardiac muscle cell. Characterization of exogenous substrate oxidation and the adenylate cyclase system.

Oxidation of several exogenous substrates by cultured adult rat ventricular cardiac muscle cells has been assessed. Unlike freshly isolated cardiac muscle cells which oxidize glucose preferentially, the cultured cells more closely resemble metabolically the in situ heart and the isolated perfused heart, in that their preference for exogenous substrates is in the order of fatty acid greater than glucose. This switch in metabolic preference from glucose to fatty acid is complete within 12 h after placing freshly isolated cells in culture. Glucose oxidation is stimulated by insulin and isoprotere

Metabolic targeting of pantothenate by medium chain fats in glioblastoma cells.

Ketogenic diets are commonly used for epilepsy therapy and are increasingly being considered in cancer treatment, where metabolic reprogramming triggered by glucose restriction and ketosis alters energy provision as the therapeutic mechanism. However, a recently developed medium chain triglyceride product, with a high decanoic acid (DA) to octanoic acid (OA) ratio (DA: OA) has enabled a much less restrictive diet and is clinically effective as an epilepsy treatment, yet no studies have explored the product in cancer models. Here we investigate metabolic mechanisms of DA, OA and DA: OA using a

Prenatal PFAS exposure and early childhood sleep quality in the ECHO Cohort.

Per- and polyfluoroalkyl substances (PFAS) are persistent endocrine-disrupting chemicals that readily cross the placenta and may interfere with hormonal systems essential for neurodevelopment and circadian regulation. We examined associations between prenatal exposure to individual PFAS and early childhood sleep quality. The analytic sample included 2836 mother-child pairs from 14 study sites in the Environmental influences on Child Health Outcomes (ECHO) Cohort. We assessed Child Behavior Checklist (CBCL) Sleep Problems T-scores from ages 1-5 years. Maternal serum concentrations of perfluoroo

Effect of the Spacer Length of Acid-base Complexes Forming Gemini-like Surfactant on the Phase Behavior.

The surface chemical properties and phase behaviors of the three acid-base 2:1 complexes were systematically studied. It was composed of tertiary amine as the main chain and three dicarboxylic acids as the spacer and functioned as a gemini-type surfactant. N-methyl-N-(2,3-dioxypropyl)hexadecylamine (C16amine) was used as the main chain, and octanoic acid (C8), decanoic acid (C10), and dodecanoic acid (C12) were combined to form complexes. In dilute solutions, the surface tension and luminescence behaviors of solubilized pyrene were consistent across all three complexes, indicating similar mice

Multi-dimensional insights into the association between PFOA exposure and NAFLD: An integrative analysis of epidemiology, transcriptomics, and molecular docking.

Perfluorooctanoic acid (PFOA), a persistent organic pollutant from the PFAS family, is widely detected in the environment and human blood, raising significant concerns about its long-term health effects. This study explores the potential association between serum PFOA exposure and non-alcoholic fatty liver disease (NAFLD) by integrating data from the NHANES 2003-2012 cohort, global disease burden data (GBD 2021), and toxicogenomic analyses. Weighted logistic regression revealed that individuals in the highest tertile of PFOA exposure had a significantly increased risk of NAFLD (adjusted OR = 1

Insight into perfluorooctanoic acid-induced impairment of mouse embryo implantation via single-cell RNA-seq.

Perfluorooctanoic acid (PFOA) is an environmentally persistent chemical that poses significant risks to human health. Studies have shown that PFOA affects female reproduction, but the specific impact on endometrial receptivity and the underlying mechanisms remain poorly understood. In this study, we investigated the effects of low-dose PFOA exposure through drinking water on endometrial receptivity in a murine model. Our results demonstrate that PFOA exposure significantly impaired endometrial receptivity, which led to a marked decrease in embryo implantation rates. Utilizing single-cell RNA s

Transcriptomic dose response assessment of PFAS chemicals 3:3 fluorotelomer carboxylic acid, 7:3 fluorotelomer alcohol, and perfluorohexanesulfonamide.

Per- and polyfluoroalkyl substances (PFAS) are a diverse class of anthropogenic chemicals, and their widespread use in manufacturing and commerce has led to introduction of these chemicals into the environment. Owing to the lack of traditional toxicology data on the majority of PFAS, novel testing methods that provide supporting information to inform human health impacts in a relatively short time frame will be increasingly important. The US Environmental Protection Agency's (EPA) Transcriptomic Assessment Process (ETAP) was recently implemented by the Agency as an efficient and cost-effective

Per- and polyfluoroalkyl substances serum concentrations during pregnancy and gestational weight gain in the Healthy Start Study.

BACKGROUND: Per- and polyfluoroalkyl substances (PFAS) are widespread and persistent contaminants detected in most humans. PFAS exposure has been linked to adverse health conditions, including excessive gestational weight gain (GWG), though results have been inconsistent. METHODS: Participants (n = 539) were enrolled in the Healthy Start Study, a Colorado-based cohort (2009-2014). Eleven PFAS were measured in serum collected during pregnancy (median 27 weeks of gestation). GWG was quantified in four ways: absolute change in weight during pregnancy; GWG adjusted for the duration of pregnancy (

Perfluorooctanoic acid triggers premature ovarian insufficiency by impairing NAD+ synthesis and mitochondrial function in adult zebrafish.

High-dose perfluorooctanoic acid (PFOA) impairs oocyte maturation and offspring quality. However, the physiological concentrations of PFOA in follicular fluids of patients with premature ovarian insufficiency (POI) were detected at lower levels, thus the relationship between physiological PFOA and reproductive disorders remains elusive. Here, we investigated whether physiological PFOA exposure affects gonad function in adult zebrafish. Physiological PFOA exposure resulted in POI-like phenotypes in adult females, which exhibited decreased spawning frequency, reduced number of ovulated eggs, abn

Immunotoxicity and allergenic potential induced by topical application of perfluorooctanoic acid (PFOA) in a murine model.

Perfluorooctanoic acid (PFOA) is a per- and polyfluoroalkyl substance (PFAS) once used as a surfactant in the polymerization of chemicals. Because of its ubiquitous nature and long half-life, PFOA is commonly detected in the environment, wildlife, and humans. While skin exposure to PFOA is of concern, studies evaluating the immunotoxicity of dermal exposure are lacking. These studies evaluated the immunotoxicity of PFOA (0.5-2% w/v, or 12.5-50 mg/kg/dose) following dermal exposure using a murine model. PFOA (0.5-2%) was not identified to be an irritant or sensitizer using the local lymph node
