Decreased cerebrospinal fluid kynurenic acid in epileptic spasms: A biomarker of response to corticosteroids.

BACKGROUND: Epileptic (previously infantile) spasms is the most common epileptic encephalopathy occurring during infancy and is frequently associated with abnormal neurodevelopmental outcomes. Epileptic spasms have a diverse range of known (genetic, structural) and unknown aetiologies. High dose corticosteroid treatment for 4 weeks often induces remission of spasms, although the mechanism of action of corticosteroid is unclear. Animal models of epileptic spasms have shown decreased brain kynurenic acid, which is increased after treatment with the ketogenic diet. We quantified kynurenine pathwa

Discovery of urinary biosignatures for tuberculosis and nontuberculous mycobacteria classification using metabolomics and machine learning.

Nontuberculous mycobacteria (NTM) infection diagnosis remains a challenge due to its overlapping clinical symptoms with tuberculosis (TB), leading to inappropriate treatment. Herein, we employed noninvasive metabolic phenotyping coupled with comprehensive statistical modeling to discover potential biomarkers for the differential diagnosis of NTM infection versus TB. Urine samples from 19 NTM and 35 TB patients were collected, and untargeted metabolomics was performed using rapid liquid chromatography-mass spectrometry. The urine metabolome was analyzed using a combination of univariate and mul

Research on tryptophan metabolites "via kynurenine" in epidermis of man and mouse.

The pattern of tryptophan metabolites "via kynurenine" was determined in the epidermis of man and mouse using semiquantitative method of one- and two-dimensional thin-layer-chromatography. In the epidermis of healthy man the following metabolites were present: tryptophan, kynurenine, anthranilic acid, 3-hydroxyanthranilic acid, 3-hydroxykynurenine, kynurenic acid, nicotinic acid; the same metabolites, with the exception of kynurenine, were present in the epidermis of healthy mouse. In the epidermis of patients with porphyria cutanea tarda and of those with pellagra, the above pattern was modif

Ommochrome genesis in an albino strain of a terrestrial isopod.

The contents of tryptophan (Trp) metabolites and the activities of the enzymes involved in ommochrome biosynthesis were measured in an albino strain of a terrestrial isopod Armadillidium vulgare. There was little difference between the Trp content in the albino mutant and that in the wild type, although the contents of 3-hydroxykynurenine (3-OH-Kyn), 3-hydroxyanthranilic acid (3-OH-AA) and xanthommatin in the albino were significantly lower than those in the wild type. Tryptophan 2,3-dioxygenase (TDO) activity in the albino was extremely low, while the activities of Kyn-3-hydroxylase and kynur

Comprehensive assessment of multiple tryptophan metabolites as potential biomarkers for immune checkpoint inhibitors in patients with non-small cell lung cancer.

PURPOSE: Tryptophan metabolites have immunomodulatory functions, suggesting possible roles in cancer immunity. METHODS: Plasma tryptophan metabolites were measured using liquid chromatography/mass spectrometry before immune checkpoint inhibitors (ICIs) in patients with non-small cell lung cancer (NSCLC). RESULTS: The 19 patients with NSCLC had significantly lower levels of tryptophan (p = 0.002) and xanthurenic acid (p = 0.032), and a significantly higher level of 3-hydroxyanthranilic acid (3-HAA) (p = 0.028) compared with the 10 healthy volunteers. The patients achieving objective responses

A comprehensive examination of circadian rhythm and tryptophan pathway parameters: Assessing their role in predicting bipolar disorder in patients, siblings, and controls.

The complex interplay between sleep, circadian rhythms, and tryptophan pathway metabolites in bipolar disorder (BD) remains poorly understood. This study examined these mechanisms in euthymic individuals with BD (n = 20), their unaffected siblings (n = 20), and healthy controls (n = 24). Subjective and objective sleep and circadian rhythm parameters were assessed using questionnaires and actigraphy, alongside plasma measurements of tryptophan (TRP), kynurenine (KYN), kynurenic acid (KYNA), 3-hydroxykynurenine (3-HK), 3-hydroxyanthranilic acid (3-HAA), and quinolinic acid (QUIN). Elevation of Kynurenine Metabolites in Rat Liver and Serum: A Potential Additional Mechanism of the Alcohol Aversive and Anti-cancer Effects of Disulfiram?

AIMS: The tryptophan metabolites 3-hydroxykynurenine (3-HK) and 3-hydroxyanthranilic acid (3-HAA) inhibit the liver mitochondrial low Km aldehyde dehydrogenase and possess alcohol-aversive and immunosuppressant properties. As the disulfiram (DS) metabolite carbon disulphide activates enzymes forming 3-HK and 3-HAA, we investigated if repeated disulfiram treatment increases the hepatic and serum levels of these 2 metabolites. METHODS: Livers and sera of male Wistar rats were analysed for tryptophan and kynurenine metabolites after repeated DS treatment for 7 days. RESULTS: DS increased liver

Pneumococcal meningitis causes accumulation of neurotoxic kynurenine metabolites in brain regions prone to injury.

Pneumococcal meningitis (PM) is characterized by an intense inflammatory host reaction that contributes to the development of cortical necrosis and hippocampal apoptosis. Inflammatory conditions in the brain are known to induce tryptophan degradation along the kynurenine pathway, resulting in accumulation of neurotoxic metabolites. In the present study, we investigated the contribution of the kynurenine pathway to brain injury in experimental PM by measuring the concentration of its metabolites and the enzymatic activities and mRNA levels of its major enzymes in the vulnerable brain regions. The Association of Kynurenine Pathway Metabolites with Neurocognitive Function in Unaffected Offspring of Parents with Bipolar Disorder.

INTRODUCTION: The study aimed to investigate whether alterations in tryptophan (TRP) metabolites reflect dysregulation of the kynurenine (KYN) pathway, which has been implicated in bipolar disorder (BD), and to examine their relationship with cognitive functioning by assessing TRP metabolite levels alongside executive performance in non-affected offspring of individuals diagnosed with BD. METHODS: The study included 32 healthy offspring of parents with BD type I as the case group and 31 healthy offspring of parents without any psychiatric disorders as controls. Metabolomics Distinguishes DOCK8 Deficiency from Atopic Dermatitis: Towards a Biomarker Discovery.

Bi-allelic mutations in the dedicator of cytokinesis 8 (DOCK8) are responsible for a rare autosomal recessive primary combined immunodeficiency syndrome, characterized by atopic dermatitis, elevated serum Immunoglobulin E (IgE) levels, recurrent severe cutaneous viral infections, autoimmunity, and predisposition to malignancy. The molecular link between DOCK8 deficiency and atopic skin inflammation remains unknown. Severe atopic dermatitis (AD) and DOCK8 deficiency share some clinical symptoms, including eczema, eosinophilia, and increased serum IgE levels. Increased serum IgE levels are chara

Increased conversion of tryptophan to nicotinamide in rats by dietary valproate.

Valproic acid (VPA) is a short-chained, branched fatty acid that is widely used in humans as an anticonvulsant and mood stabilizer, and has been reported to increase the liver NAD concentration. We investigated the effects of VPA on the conversion of tryptophan to nicotinamide. Rats were fed diets containing various amounts of VPA (0, 0.5, and 1.0% in the diets) for 14 d, 24-h urine samples were collected, and tryptophan and its catabolites were measured. We found that the conversion of tryptophan to nicotinamide was increased by feeding a diet containing VPA (p<0.01; 0% vs. 1.0% VPA). Of the

Decolouration of the lens pigment in senile nuclear cataract.

Human cataract lens proteins can be bleached by exposure to sodium borohydride (NaBH4), sodium cyanoborohydride (NaCNBH3), or hydrogen peroxide (H2O2). The decolouration resulting from these treatments could be monitored by a change in absorbance at 350 nm. At pH 12 the magnitude of the absorbance change increased in proportion with the severity of the nuclear cataract in the case of NaBH4 and H2O2 treatments, but not in the case of NaCNBH3 treatment. The rate of change in absorbance at 350 nm following exposure to the different reagents was used to evaluate three model systems for senile nucl

The correlation between certain tryptophan metabolites and the N-nitrosamine content in the urine of bilharzial bladder cancer patients.

The present work is an up-to-date approach to study the correlation between the excretion pattern of tryptophan metabolites along the kynurenine pathway (after loading with 2 gm. L-tryptophan), and the N-nitrosamine content in urine of bilharzial bladder cancer patients. The control group was composed of healthy subjects who had no reported history of S. haematobium infection and no current bacterial cystitis. The N-nitrosamine content was determined by the colorimetric method of Eisebrand and Preussmann (1970). It was demonstrated that 64 per cent of the patients metabolized the tryptophan lo

Sirtuin 2 regulates neutrophil functions through NAD+ synthesis pathway in virus infection.

Neutrophils play an important role in antiviral immunity, but the underlying mechanisms remain unclear. Here, we found that SIRT2 deficiency inhibited the infiltration of neutrophils, as well as the secretion of inflammatory cytokines and the formation of neutrophil extracellular traps (NETs), ameliorating disease symptoms during acute respiratory virus infection. Mechanistically, SIRT2 deficiency upregulates quinolinic acid (QA)-producing enzyme 3-hydroxyanthranilate oxygenase (3-HAO) and leads to expression of quinolinate phosphoribosyltransferase (QPRT), which promotes the synthesis of QA f

The Role of Downstream Kynurenine Pathway Metabolites in the Modulation of Cardiovascular Disease Development in Chronic Kidney Disease.

Cardiovascular diseases (CVDs) represent a significant and escalating health challenge in patients with chronic kidney disease (CKD). In this population, the cardiovascular incidents are markedly elevated, and CVD represents the leading cause of mortality. The pathogenesis of CVD in the course of CKD is multifactorial, and some evidence indicates that disturbances in the kynurenine pathway (KP), the major route of tryptophan metabolism, can also play a significant role in this process. The enhanced activation of the KP and reduced clearance of its metabolites contribute to their accumulation d

The multiple faces of tryptophan in bone biology.

Osteoporosis is highly prevalent in older persons. While many advances have been made in the field of osteoporosis, current treatments have been affected by unexpected side effects and limited efficacy; therefore, new approaches to identify disease mechanisms and pathways are required. This review focuses on the influence of tryptophan metabolites, particularly kynurenines and serotonin on bone. The kynurenine (KYN) pathway is associated with osteoblastogenesis and can be linked to the pathophysiology of osteoporosis. The activity of osteoblasts is reduced by 3-hydroxykynurenine (3-HKYN), a pr
