ATP-citrate lyase is an epigenetic regulator to promote obesity-related kidney injury.

Obesity is a leading cause of chronic kidney disease (CKD), but how obesity promotes renal injury remains poorly understood. Here we showed that ATP-citrate lyase (ACL), an enzyme converting citrate to acetyl-CoA, is highly induced in the kidney of overweight or obese patients with CKD and ob/ob BTBR mice. ACL induction is associated with increased ectopic lipid accumulation (ELA), glomerulosclerosis, and albuminuria. Acetyl-CoA is the substrate for de novo lipogenesis as well as for histone acetylation. By raising acetyl-CoA concentration ACL promotes H3K9/14 and H3K27 hyperacetylation leadin

Apparent inhibition of ATP citrate lyase by L-glutamate in vitro is due to the presence of glutamine synthetase.

Preincubation in assay mixture for 30 min at 37 degrees C of ATP citrate lyase from rat brain and liver results in 65-70% inhibition in the presence of 10 mM L-glutamate. This inhibition is specific since none of the known brain metabolites of glutamate shows this effect. ATP and ammonium sulphate-suspended, commercially purified malate dehydrogenase are both important in the generation of inhibition; citrate and NADH are not. The ATP citrate lyase activity in desalted crude extracts and 11% polyethylene glycol-precipitated fractions is inhibited but the enzyme purified by dye affinity chromat

The role of bempedoic acid in the management of dyslipidaemia in people with diabetes: an expert opinion of Italian diabetologists.

Diabetes mellitus is a rapidly growing global health challenge and a major driver of atherosclerotic cardiovascular disease (ASCVD). Dyslipidaemia, highly prevalent in both type 1 and type 2 diabetes, plays a central role in this excess cardiovascular risk and often persists despite statin therapy. Although statins remain the cornerstone of lipid management, many patients with diabetes do not achieve recommended low-density lipoprotein cholesterol (LDL-C) goals. Therefore, there is a need for effective, safe, and practical adjunctive therapies. Bempedoic acid is a first-in-class oral ATP-citra

Dietary exposure to environmentally relevant pesticide mixtures impairs swimming performance and lipid homeostatic gene expression in Juvenile Chinook salmon at elevated water temperatures.

Aquatic organisms are exposed to complex mixtures of pesticides in the environment, but traditional risk assessment approaches typically only consider individual compounds. In conjunction with exposure to pesticide mixtures, global climate change is anticipated to alter thermal regimes of waterways, leading to potential co-exposure of biota to elevated temperatures and contaminants. Furthermore, most studies utilize aqueous exposures, whereas the dietary route of exposure may be more important for fish owing to the hydrophobicity of many pesticides. Consequently, the current study aimed to det

Metabolic potential of the adipose tissue of rats during hyper- and hypovitaminosis A.

Effect of excess feeding and depletion of vitamin A on the ability of adipose tissue to maintain plasma free fatty acid levels has been studied in rats. Both in hypervitaminosis A (fed 9 mg of retinol for 2 consecutive days) and in vitamin A deficiency (kept on a vitamin A-deficient diet for 6 weeks) the rats showed elevated levels of plasma free fatty acids. Hypervitaminosis A caused a decrease in the fatty acid release from adipose tissue on in vitro incubation, probably due to lowered levels of cyclic AMP. On the other hand, adipose tissue from vitamin A-deficient animals showed an increase

ATP-Citrate Lyase: A Prognostic Marker of Non-Small Cell Lung Cancer Dictates the Progression of Adenocarcinoma in Indian Population.

Tissue biopsies are routinely used for identifying driver mutations in tumors but limitations such as tumor heterogeneity have encouraged the use of liquid biopsies for the identification of new biomarkers. ATP-citrate lyase (ACLY) has been recognized as a potential target for tumor inhibition due to its overexpression in several cancers. However, the clinical significance of this enzyme remains unknown in liquid biopsies. In this study, 31 pleural effusion fluid samples were collected from patients suspected of malignancy for analyzing the gene expression of ACLY and associated metabolic enzy

Pharmacological induction of mesenchymal-epithelial transition via inhibition of H2S biosynthesis and consequent suppression of ACLY activity in colon cancer cells.

Hydrogen sulfide (H2S) is an important endogenous gaseous transmitter mediator, which regulates a variety of cellular functions in autocrine and paracrine manner. The enzymes responsible for the biological generation of H2S include cystathionine-β-synthase (CBS), cystathionine-γ-lyase (CSE) and 3-mercaptopyruvate sulfurtransferase (3-MST). Increased expression of these enzymes and overproduction of H2S has been implicated in essential processes of various cancer cells, including the stimulation of metabolism, maintenance of cell proliferation and cytoprotection. Cancer cell identity is charact

PSMD14-Mediated LDHA Deubiquitination Upregulates ACLY Expression via H3K18 Lactylation to Promote Lipid Synthesis and Pancreatic Cancer Progression.

Aberrant lipid metabolism is intimately linked to tumor progression. As a pivotal post-translational modification, ubiquitination regulates diverse oncogenic processes. However, the interplay between ubiquitination and lipid metabolic dysregulation in pancreatic cancer (PC), along with its underlying molecular mechanisms, remains poorly understood. Here, it is demonstrated that glycolytic enzyme lactate dehydrogenase A (LDHA) potentiates lipid biosynthesis under the regulation of deubiquitinases. Specifically, PSMD14 directly binds and stabilizes LDHA through its deubiquitinase activity, resul

Inhibition of ATP citrate lyase does not affect muscle mitochondrial activity in apoE-/- mice.

INTRODUCTION: Lipid-lowering therapy is a cornerstone in preventing coronary disease. Bempedoic acid is recommended in patients who are unable to take statin therapy to achieve the low-density lipoprotein cholesterol goal. Based on the hypothesis that statins impact skeletal mitochondrial activity, this study aimed to evaluate the effect of bempedoic acid on this aspect in apoE-/- mice fed a high-fat high-cholesterol (HFHC) diet and compare it to the results obtained using atorvastatin. METHODS: Female apoE-/- mice. The following techniques were used: En face analysis of aorta, mitostress ana

Gene expression of fatty acid synthesizing enzymes in fetal rat lung in prolonged pregnancy.

In fetal lung the amounts of mRNAs encoding fatty acid synthase (FAS), acetyl-CoA carboxylase (ACC) and ATP citrate lyase (ACL) increase in late gestation and drop around birth. To study the mechanism of the perinatal decrease, pregnancy was prolonged from 22 (term) to 25 days in rats with daily injections of progesterone. Progesterone did not affect the levels of lipogenic enzyme mRNAs in fetal lung prior to term, but significantly delayed the perinatal decrease in the levels of lung FAS and ACC mRNA. Although for ACL mRNA abundance the differences were not statistically significant, its patt

Aerobic exercise reduces lipid accumulation via ACLY-mediated metabolic remodeling to alleviate cardiac remodeling.

BACKGROUND: Pathological cardiac remodeling, a key contributor to heart failure, is characterized by significant reprogramming of cardiac energy metabolism. ATP-citrate lyase (ACLY), which connects glucose metabolism with lipid synthesis, plays an important regulatory role in this process. However, the role of ACLY in exercise-induced metabolic remodeling and cardiac remodeling attenuation remains unclear. METHODS: Cardiac remodeling was induced in H9c2 cells with isoproterenol or urea, or in rats by left anterior descending artery (LAD) ligation or mice by isoproterenol. Interventions includ

ACLY regulates autolysosome acidification through tubulin acetylation-mediated assembly of V-ATPase subunits in Alzheimer's disease model mice.

INTRODUCTION: Faulty autolysosome acidification leads to dystrophic neurites-an early event propelling Alzheimer's disease (AD) progression-yet the underlying mechanism remains elusive. METHODS: To elucidate the physiological functions of neuronal ATP citrate lyase (ACLY) expression, its impact on amyloid beta (Aβ) pathology, and molecular mechanisms, we used intracerebroventricular ACLY inhibitor administration, adeno-associated virus-mediated ACLY modulation in the dorsal hippocampus, and N2a-swAPP695 cell line. RESULTS: Inhibition or knockdown ACLY reduced microtubule stability and impair

The pathological significance and potential mechanism of ACLY in cholangiocarcinoma.

BACKGROUND AND AIM: Cholangiocarcinoma (CCA) is a rare cancer, yet its incidence and mortality rates have been steadily increasing globally over the past few decades. Currently, there are no effective targeted treatment strategies available for patients. ACLY (ATP Citrate Lyase), a key enzyme in de novo lipogenesis, is aberrantly expressed in several tumors and is associated with malignant progression. However, its role and mechanisms in CCA have not yet been elucidated. METHODS: The expression of ACLY in CCA was assessed using transcriptomic profiles and tissue microarrays. Kaplan-Meier curv

Neutrophil extracellular traps in rheumatoid arthritis: Activating fibroblast-like synoviocytes via ATP citrate lyase.

Synovial hyperplasia and inflammation are the main pathological features of rheumatoid arthritis (RA). Fibroblast‑like synoviocytes (FLSs) are the major effector cells contributing to chronic inflation and joint injury in the synovial microenvironment. Neutrophil extracellular traps (NETs) play an important role in the pathogenesis of RA, but their downstream regulatory mechanisms remain unclear. In order to improve the therapeutic prospects for RA, it is crucial to identify the targets of NETs and the molecular mechanisms of synovial dysfunction. ATP-citrate lyase (ACLY) directs glucose metab

Acetate Production from Glucose and Coupling to Mitochondrial Metabolism in Mammals.

Acetate is a major nutrient that supports acetyl-coenzyme A (Ac-CoA) metabolism and thus lipogenesis and protein acetylation. However, its source is unclear. Here, we report that pyruvate, the end product of glycolysis and key node in central carbon metabolism, quantitatively generates acetate in mammals. This phenomenon becomes more pronounced in the context of nutritional excess, such as during hyperactive glucose metabolism. Conversion of pyruvate to acetate occurs through two mechanisms: (1) coupling to reactive oxygen species (ROS) and (2) neomorphic enzyme activity from keto acid dehydro

ACLY facilitates alanine flux in the livers of db/db mice: a hyperpolarized [1-13C]pyruvate MRS study.

INTRODUCTION: Non-alcoholic fatty liver disease (NAFLD) and type 2 diabetes mellitus (T2DM) feature paradoxical increases in both gluconeogenesis and lipogenesis. ATP citrate lyase (ACLY) supports both processes by generating cytosolic acetyl-CoA and oxaloacetate from citrate. While ACLY's role in lipogenesis is well established, its involvement in amino acid-driven gluconeogenesis remains unclear. METHODS: Using hyperpolarized [1-13C]pyruvate magnetic resonance spectroscopy (MRS), we observed [1-13C]alanine labeling in the livers of db/db mice. To test the effect of ACLY inhibition, mice wer
