Sinapoylglucose: malate sinapoyltransferase SNG1 negatively regulates drought resistance by crosstalk with abscisic acid signaling in Arabidopsis.

Drought stress has a profound impact on yield, with massive repercussions for agricultural production. In addition to ABA, the central hormone controlling the plant response to drought, secondary metabolites also play important functions in drought stress; however, the precise nature of their roles is still obscure. Metabolomics analysis of ABA- and PEG-treated Arabidopsis wild-type (WT) seedlings as well as ABA signal transduction mutants snrk-triple (snrk2.2 snrk2.3 snrk2.6) and pyl-quadruple (pyr1 pyl1 pyl2 pyl4) identified a number of drought-related metabolites. Specifically, we observed

The H2S-responsive transcription factor ERF.D3 regulates tomato abscisic acid metabolism, leaf senescence, and fruit ripening.

Hydrogen sulfide (H2S) is a signaling molecule that regulates plant senescence. In this study, we found that H2S delays dark-induced senescence in tomato (Solanum lycopersicum) leaves. Transcriptome and reverse transcription quantitative PCR (RT-qPCR) analyses revealed an ethylene response factor ERF.D3 is quickly induced by H2S. H2S also persulfidated ERF.D3 at amino acid residues C115 and C118. CRISPR/Cas9-mediated gene editing, and gene overexpression analyses showed that ERF.D3 negatively regulates leaf senescence and fruit ripening. Abscisic acid (ABA) levels were reduced by ERF.D3 overex

A promising product: Abscisic acid-producing bacterial agents for restricting cadmium enrichment in field vegetable crops.

Soil heavy metal contamination and its enrichment in the edible parts of crops have gained global concern. In this study, a compound bacterial agent possessing the ability to produce the plant hormone, abscisic acid (ABA), was applied to contaminated farmland in Hunan province. Its application reduced the concentration of Cd in radish, cabbage, mustard, and lettuce by 15-144%. Accordingly, the Cd contents in these vegetables were found to be below the maximum limits set by GB 2762-2017. Meanwhile, bacteria agents also led to a significant increase in crops yield by 45-82%. Furthermore, the nut

Arabidopsis ABCG27 plays an essential role in flower and leaf development by modulating abscisic acid content.

Abscisic acid (ABA) is a phytohormone that mediates stress responses and regulates plant development. Several ATP-binding cassette (ABC) transporters in the G subfamily of ABC (ABCG) proteins have been reported to transport ABA. We investigated whether there are any other ABCG proteins that mediate plant developmental processes regulated by ABA in Arabidopsis (Arabidopsis thaliana). The ABCG27 gene was upregulated in response to exogenous ABA treatment. The abcg27 knockout mutant exhibited two developmental defects: epinastic leaves and abnormally long pistils, which reduced fertility and sili

Hydrogen Sulfide-Linked Persulfidation Maintains Protein Stability of ABSCISIC ACID-INSENSITIVE 4 and Delays Seed Germination.

Hydrogen sulfide (H2S) is an endogenous gaseous molecule that plays an important role in the plant life cycle. The multiple transcription factor ABSCISIC ACID INSENSITIVE 4 (ABI4) was precisely regulated to participate in the abscisic acid (ABA) mediated signaling cascade. However, the molecular mechanisms of how H2S regulates ABI4 protein level to control seed germination and seedling growth have remained elusive. In this study, we demonstrated that ABI4 controls the expression of L-CYSTEINE DESULFHYDRASE1 (DES1), a critical endogenous H2S-producing enzyme, and both ABI4 and DES1-produced H2S

Abscisic acid suppresses the activation of NLRP3 inflammasome and oxidative stress in murine allergic airway inflammation.

Abscisic acid (ABA), a well-known natural phytohormone reportedly exerts anti-inflammatory and anti-oxidative properties in diabetes and colitis. However, the efficacy of ABA against allergic airway inflammation and the underlying mechanism remain unknown. Herein, an OVA-induced murine allergic airway inflammation model was established and treated with ABA in the presence or absence of PPAR-γ antagonist GW9662. The results showed that ABA effectively stunted the development of airway inflammation, and concordantly downregulated OVA-induced activation of NLRP3 inflammasome, suppressed oxidative

Antagonistic interaction between systemic acquired resistance and the abscisic acid-mediated abiotic stress response in Arabidopsis.

Systemic acquired resistance (SAR) is a potent innate immunity system in plants that is effective against a broad range of pathogens. SAR development in dicotyledonous plants, such as tobacco (Nicotiana tabacum) and Arabidopsis thaliana, is mediated by salicylic acid (SA). Here, using two types of SAR-inducing chemicals, 1,2-benzisothiazol-3(2H)-one1,1-dioxide and benzo(1,2,3)thiadiazole-7-carbothioic acid S-methyl ester, which act upstream and downstream of SA in the SAR signaling pathway, respectively, we show that treatment with abscisic acid (ABA) suppresses the induction of SAR in Arabido

Abscisic acid (ABA) inhibits light-induced stomatal opening through calcium- and nitric oxide-mediated signaling pathways.

Nitric oxide (NO) is an important signaling component of ABA-induced stomatal closure. However, only fragmentary data are available about NO effect on the inhibition of stomatal opening. Here, we present results supporting that, in Vicia faba guard cells, there is a critical Ca2+-dependent NO increase required for the ABA-mediated inhibition of stomatal opening. Light-induced stomatal opening was inhibited by exogenous NO in V. faba epidermal strips. Furthermore, ABA-mediated inhibition of stomatal opening was blocked by the specific NO scavenger cPTIO, supporting the involvement of endogenous

Salicylic acid-mediated establishment of the compatibility between Alternaria brassicicola and Brassica juncea is mitigated by abscisic acid in Sinapis alba.

This work addresses the changes in the phytohormonal signature in the recognition of the necrotrophic fungal pathogen Alternaria brassicicola by susceptible Brassica juncea and resistant Sinapis alba. Although B. juncea, S. alba and Arabidopsis all belong to the same family, Brassicaceae, the phytohormonal response of susceptible B. juncea towards this pathogen is unique because the latter two species express non-host resistance. The differential expression of the PR1 gene and the increased level of salicylic acid (SA) indicated that an SA-mediated biotrophic mode of defence response was trigg

[Level of cytokines, abscisic and salicylic acids in the leafs of Phlox under the effect of invasion by conidia of phytopathogens].

We studied the effects of the invasion of Phlox paniculata L. and Ph. setacea L. by causative agents of the phlox powdery mildew (compatible combination) and lupine powdery mildew (incompatible combination) on the level of endogenous cytokinins and abscisic and salicylic acids. In all experimental variants, the level of zeatin-riboside and abscisic and salicylic acids in the leaves of invaded plants increased within 48 h. The highest level of phytohormones and salicylic acid was recorded in the absolutely resistant species Ph. setacea.

Guard cell Signal Transduction Mechanisms Controlling Stomatal Movements by Light, Abscisic Acid, CO2, VPD and Heat.

Guard cell pairs form dynamic stomatal gas exchange pores in aerial tissues of plants. Guard cells regulate CO2 influx and water loss through stomata by integrating multiple environmental and internal signals. Guard cells serve as highly specialized sensory and signal processing cells, thereby orchestrating stomatal movements in response to changing conditions. This review describes recent advances and early discoveries in elucidation of guard cell signaling networks. Present day understanding of the molecular and cellular mechanisms mediating stomatal opening by blue light, red light, heat st

Lanthanum supplementation with abscisic acid modulates rhizosphere dynamics through changes in nitric oxide synthesis in wheat.

Lanthanum (La), a rare earth element (REE) found its wide applications in the agricultural land practices. Utilization of La as a fertilizer is based on its concentration for staple crops, including wheat (Triticum aestivum). We have investigated the role of a beneficial dose of La (60 μM) along with ABA (10 μM) in improving wheat root dynamics and defense systems (ascorbate-glutathione pathway, secondary metabolites, and nitric oxide (NO) biosynthesis). Co-application of La60 and ABA significantly enhanced all the root dynamic attributes along with defense systems and reduced production of re

Abscisic Acid Enhances Motor and Cognitive Function in the 3-Acetylpyridine Mouse Model of Cerebellar Ataxia.

Cerebellar ataxia is a debilitating neurodegenerative disorder characterized by impaired motor coordination and balance with limited treatment options. Abscisic acid (ABA), a phytohormone detected in mammalian brains, has shown neuroprotective properties. This study investigated the effects of ABA on motor, cognitive, and affective deficits in a mouse model of cerebellar ataxia in which male Swiss mice received a single intraperitoneal injection of 3-acetylpyridine (3-AP; 60 mg/kg), which leads to the loss of climbing fiber input to Purkinje neurons leading to cerebellar degeneration. In ABA-t

Abscisic acid ameliorates cognitive dysfunctions, but not facial allodynia, in a male rat nitroglycerin migraine model.

OBJECTIVE: Abscisic acid (ABA), found in fruits and vegetables and produced in the human body, has anti-inflammatory, anti-oxidative, and pro-cognitive effects. The impact of ABA on trigeminal nerve hyperalgesia and cognitive behaviors in a rat migraine model was evaluated. DESIGN: Male rats (n = 8) were randomly distributed into 5 groups (Control, Sham, Abscisic acid (ABA), Nitroglycerin (NTG), and Nitroglycerin + Abscisic acid (NTG+ABA). Migraine was induced through 4 injections of NTG (5 mg/kg intraperitoneal (i.p.)). ABA (10 µg/rat intracerebroventricular (i.c.v)) was administered 4 times

N6-methyladenosine-mediated feedback regulation of abscisic acid perception via phase-separated ECT8 condensates in Arabidopsis.

N6-methyladenosine (m6A) is the most abundant internal modification in eukaryotic mRNAs, yet how plants recognize this chemical modification to swiftly adjust developmental plasticity under environmental stresses remains unclear. Here we show that m6A mRNA modification and its reader protein EVOLUTIONARILY CONSERVED C-TERMINAL REGION 8 (ECT8) act together as a key checkpoint for negative feedback regulation of abscisic acid (ABA) signalling by sequestering the m6A-modified ABA receptor gene PYRABACTIN RESISTANCE 1-LIKE 7 (PYL7) via phase-separated ECT8 condensates in stress granules in respons

Stabilization of dimeric PYR/PYL/RCAR family members relieves abscisic acid-induced inhibition of seed germination.

Abscisic acid (ABA) is the primary preventing factor of seed germination, which is crucial to plant survival and propagation. ABA-induced seed germination inhibition is mainly mediated by the dimeric PYR/PYL/RCAR (PYLs) family members. However, little is known about the relevance between dimeric stability of PYLs and seed germination. Here, we reveal that stabilization of PYL dimer can relieve ABA-induced inhibition of seed germination using chemical genetic approaches. Di-nitrobensulfamide (DBSA), a computationally designed chemical probe, yields around ten-fold improvement in receptor affini

Insulin-independent stimulation of skeletal muscle glucose uptake by low-dose abscisic acid via AMPK activation.

Abscisic acid (ABA) is a plant hormone active also in mammals where it regulates, at nanomolar concentrations, blood glucose homeostasis. Here we investigated the mechanism through which low-dose ABA controls glycemia and glucose fate. ABA stimulated uptake of the fluorescent glucose analog 2-NBDG by L6, and of [18F]-deoxy-glucose (FDG) by mouse skeletal muscle, in the absence of insulin, and both effects were abrogated by the specific AMPK inhibitor dorsomorphin. In L6, incubation with ABA increased phosphorylation of AMPK and upregulated PGC-1α expression. LANCL2 silencing reduced all these

Transcript analyses reveal a comprehensive role of abscisic acid in modulating fruit ripening in Chinese jujube.

BACKGROUND: Chinese jujube (Ziziphus jujuba Mill.) is a non-climacteric fruit; however, the underlying mechanism of ripening and the role of abscisic acid involved in this process are not yet understood for this species. RESULTS: In the present study, a positive correlation between dynamic changes in endogenous ABA and the onset of jujube ripening was determined. Transcript analyses suggested that the expression balance among genes encoding nine-cis-epoxycarotenoid dioxygenase (ZjNCED3), ABA-8'-hydroxylase (ZjCYP707A2), and beta-glucosidase (ZjBG4, ZjBG5, ZjBG8, and ZjBG9) has an important ro

Up-regulation of abscisic acid signaling pathway facilitates aphid xylem absorption and osmoregulation under drought stress.

The activation of the abscisic acid (ABA) signaling pathway reduces water loss from plants challenged by drought stress. The effect of drought-induced ABA signaling on the defense and nutrition allocation of plants is largely unknown. We postulated that these changes can affect herbivorous insects. We studied the effects of drought on different feeding stages of pea aphids in the wild-type A17 of Medicago truncatula and ABA signaling pathway mutant sta-1. We examined the impact of drought on plant water status, induced plant defense signaling via the abscisic acid (ABA), jasmonic acid (JA), an

Abscisic acid activates the murine microglial cell line N9 through the second messenger cyclic ADP-ribose.

Abscisic acid (ABA) is a phytohormone regulating important functions in higher plants, notably responses to abiotic stress. Recently, chemical or physical stimulation of human granulocytes was shown to induce production and release of endogenous ABA, which activates specific cell functions. Here we provide evidence that ABA stimulates several functional activities of the murine microglial cell line N9 (NO and tumor necrosis factor-alpha production, cell migration) through the second messenger cyclic ADP-ribose and an increase of intracellular calcium. ABA production and release occur in N9 cel
