Functional characterization of two delta-class glutathione S-transferases genes in detoxifying acaricides in Sarcoptes scabiei.

Scabies (or Sarcoptic mange) is a highly contagious skin disease caused by the ectoparasite Sarcoptes scabiei. The resistance of S. scabiei to common acaricides like permethrin has been reported, yet their precise mechanisms remain elusive. Glutathione S-Transferase (GSTs) constitute an important class of detoxification enzymes. In this study, we found that the expression levels of SsGSTd3 and SsGSTd5 in S. scabiei were significantly up-regulated after exposure to LC30 of permethrin, amitraz, and selamectin, suggesting that SsGSTd3 and SsGSTd5 are involved in the detoxification of these acaric

Application of ethnobotanical repellents and acaricides in prevention, control and management of livestock ticks: A review.

Ticks transmit at least the same number or even more pathogens than any other group of blood-feeding arthropods worldwide affecting humans and animals. The eco-friendly control and management of tick vectors in a constantly changing environment is a crucial challenge. Besides the development of vaccines against ticks, IPM practices aimed at reducing tick interactions with livestock, emerging pheromone-based control tools, and few biological control agents, the extensive employment of acaricides and tick repellents still remain the most effective and ready-to-use strategies. However, the former

Design, synthesis and acaricidal activity of tetrahydrothiophene derivatives against Psoroptes cuniculi.

A series of sulfur-containing five-membered heterocycles were devised and prepared, with substituted chalcones and elemental sulfur acting as precursors. They were subsequently oxidized into the relevant tetrahydrothiophene sulfone derivatives. In vitro assessments were carried out to gauge the acaricidal activity of all 44 synthesized compounds against Psoroptes cuniculi. Based on mass concentration and molar concentration, 16 and 14 compounds, respectively, exhibited significantly superior acaricidal activity compared to the commercial drug ivermectin. Acaricidal and repellent activity of terpenoids from seaweeds Collected in Pernambuco, Brazil.

n-Hexane and dichloromethane extracts obtained from Laurencia dentroidea (Rhodophyta) and Canistrocarpus cervicornis (Phaeophyta) were investigated for their acaricidal and repellent properties against Tetranychus urticae under laboratory conditions. The two extracts displayed moderate toxicity and good repellent proprieties, and were significantly more toxic (36-fold) than the positive control (eugenol), whereas eugenol was tenfold more repellent than either seaweed extract. The sesquiterpenoid elatol (1) was isolated from L. dentroidea and the diterpenoid seco-dolastane (4R,9S,14S)-4alpha-ac

Resistance monitoring, sublethal and transgenerational effects of different acaricides on the European red mite, Panonychus ulmi (Koch).

BACKGROUND: Panonychus ulmi (Koch) is an important phytophagous mite pest in apple orchards, and acaricide resistance increasingly compromises its control. Sublethal acaricide exposure may further affect reproduction and population dynamics. This study surveyed resistance to six acaricides in 15 field populations from Xinjiang, China, during 2024-2025 and assessed the LC20-induced sublethal and transgenerational effects of pyridaben, bifenazate and bifenthrin in susceptible and resistant populations. RESULTS: Resistance monitoring showed clear regional differences in susceptibility to the six

Acaricidal effects of diatomaceous earth on Haemaphysalis longicornis under laboratory conditions.

Ticks transmit various diseases to humans and animals, posing a significant global threat to public health. The primary method currently employed to control tick infestations is the use of chemical acaricides. However, improper use leads to environmental contamination, residue in meat and milk, and the development of acaricide-resistant ticks. Therefore, alternative tick control methods that are safe for humans, animals, and the environment and that do not induce resistance are urgently needed. Diatomaceous earth (DE), a naturally occurring physical insecticide, has been proven to effectively

Discovery of Novel 2,4,5-Trisubstituted Oxazoline Derivatives as Potent Acaricidal Agents.

To overcome the high toxicity of etoxazole to aquatic invertebrates and its limited structural diversity while preserving its potent acaricidal activity, we successfully designed and synthesized 27 novel 2,4,5-trisubstituted oxazoline derivatives and systematically assessed their acaricidal activity. Among these, compounds 12q (LC50 = 0.84 mg/L) and 12u (LC50 = 0.74 mg/L) exhibited moderate ovicidal activity. Notably, compound 12aa exhibited good ovicidal (LC50 = 0.14 mg/L) and larvicidal activities (LC50 = 0.09 mg/L), highlighting its potential as a lead compound. Potential surface calculatio

Effects of residential acaricide treatments on patterns of pathogen coinfection in blacklegged ticks.

Medically important ixodid ticks often carry multiple pathogens, with individual ticks frequently coinfected and capable of transmitting multiple infections to hosts, including humans. Acquisition of multiple zoonotic pathogens by immature blacklegged ticks (Ixodes scapularis) is facilitated when they feed on small mammals, which are the most competent reservoir hosts for Anaplasma phagocytophilum (which causes anaplasmosis in humans), Babesia microti (babesiosis) and Borrelia burgdorferi (Lyme disease). Here, we used data from a large-scale, long-term experiment to ask whether patterns of sin

Increased transcription of Glutathione S-transferases in acaricide exposed scabies mites.

BACKGROUND: Recent evidence suggests that Sarcoptes scabiei var. hominis mites collected from scabies endemic communities in northern Australia show increasing tolerance to 5% permethrin and oral ivermectin. Previous findings have implicated detoxification pathways in developing resistance to these acaricides. We investigated the contribution of Glutathione S-transferase (GST) enzymes to permethrin and ivermectin tolerance in scabies mites using biochemical and molecular approaches. RESULTS: Increased in vitro survival following permethrin exposure was observed in S. scabiei var. hominis comp

[Acaricidal activity of "Medifox" against the human itch mites Sarcoptes scabiei].

The acaricidal activity of Medifox was evaluated in terms of the itch mites to lend support to the validity of its regimen to treat human scabies. Experiments using mainly females, eggs, and larvae withdrawn from the burrows on the skin of patients with scabies were performed. Permethrin contained in 0.4% aqueous Medifox emulsion was found to affect an itch mite by contact and intestinal routes, without a marked knock-down effect; however, the signs of intoxication are irreversible and lead to the death of Medifox-treated mites. In an in vitro experiment, 100% death of specimens occurred 24-48

Trials for control of ixodid ticks using pheromone acaricide tick decoys.

Experimental application of Hyalomma dromedarii pheromone-impregnated in cyfluthrin (pyrethroid) decoys on experimentally infested camels revealed efficacy rate of 85.33%, while the control group showed normal pattern of tick engorgement. This study proved the rapid movement of males toward decoys and some males took the mounting position with such decoys. On the other side, the same decoys (H. dromedarii) when used on naturally infested cattle with Boophilus annulatus did not even disturb the Boophilus ticks indicating that B. annulatus contain specific pheromones which should be furtherly id

[The acaricidal activity of pyrethroids against Ixodes persulcatus P. Sch. nymphs].

The efficiency of 9 drugs based on cipermethrin, deltamethrin, permethrin, lambdacigalothrin, ciphenothrin, phenothrin against hungry I. persulcatus nymphs was studied in the laboratory setting. The values LD50 and LD99 (in g of an active ingredient per m2) were found when the nymphs got into contacts with the treated filter paper for 10 sec and 1 min. The paper discusses whether the pyrethroids as part of attractant-acaricidal granules can be used to suppress the size of ticks in the tick-borne encephalitis foci.

Characterization of the Dermanyssus gallinae octopamine receptor DgOctβ2R as the molecular target for synthetic and natural acaricides.

BACKGROUND: The poultry red mite (Dermanyssus gallinae) seriously threatens the global poultry industry. Reliance on chemical acaricides has led to resistance, and plant essential oils represent an eco-friendly alternative, but their mode of action remains unclear. Amitraz targets the β2-adrenergic-like octopamine receptor (Octβ2R). This study aimed to clone and characterize Octβ2R from D. gallinae, elucidate the mechanism of amitraz and essential oils, and support resistance monitoring. RESULTS: We cloned DgOctβ2R. Evolutionary analysis revealed that DgOctβ2R clusters with other mite Octβ2Rs

Elucidating the molecular basis of ATP synthase inhibition: A virtual screening framework for next-gen selective acaricides.

The rational design of pesticides that selectively target harmful species while sparing beneficial organisms remains a major challenge in sustainable agriculture. Progress in this field has been limited by an incomplete understanding of the molecular basis of selectivity and the lack of effective strategies to exploit specific targets. Here, we provide direct molecular evidence that naturally occurring polymorphisms in the CV-a subunit of mitochondrial adenosine 5'-triphosphate (ATP) synthase are the primary determinants of differential sensitivity between pest mites and predatory mites.
